Effect of Iaspp on the Anti-Apoptosis and Genomic Instability of MCF-7

Abstract:OBJECTIVE: To investigate the effect of iASPP-full length(iASPP-FL) and iASPP-splice variant(iASPP-SV) on apoptosis and the genomic instability.METHODS: MCF-7 cell line was transfected with iASPP-FL and iASPP-SV plasmid,and cell lines that stably expressing iASPP-FL and iASPP-SV were identified.Flow cytometry was used to detect the cell apoptosis with DNA damage caused by radiation or etoposide(Vp-16).By alkaline single cell gel electrophoresis(comet assay) and Western Blot,we observed the existence of DNA double strand break damage and the difference between these cell lines.RESULTS: After 200 μg/mL Vp-16 treated,SV1 and SV2 cell lines that stably expressing iASPP-SV showed significantly decreasing early apoptosis rate from(42.70±2.38)% to(24.40±1.12)% and(17.52±1.08)%,and late apoptosis rate from(28.94±1.38)% to(18.78±0.95)% and(19.58±1.04)% compared with the cell line transfected with empty vector,while the expression of H2AX increased to 1.48 and 2.10 folds.FL1 and FL2 cell lines that stably expressing iASPP-FL showed significant reduction of early apoptosis rate from(31.22±1.64)% to(14.22±0.68)% and(15.82±0.74)%,and late apoptosis rate from(36.90±2.08)% to(11.16±0.88)% and(9.88±0.56)% compared with the cell line transfected with empty vector,while the expression of H2AX increased to 2.09 and 1.99 folds.After 4 Gy 137Cs iradiation,the early apoptosis rate of SV1 and FL1 decreased to(6.42±0.24)% and(7.05±0.24)% compared with the(16.56±0.81)% of empty vector group.The late apoptosis rate decreased to(15.10±0.50)% and(15.14±0.58)% compared with the(29.08±1.55)% of empty vector.Comet assay also showedmore DNA damages in iASPP-FL-or iASPP-SV-expressing cell lines than that in control cell line.CONCLUSION: iAPSS-FL/iASPP-SV an reduce apoptosis induced by radiation or Vp-16,with DNA damage accumulating which may promote the genomic instability and pathogenesis and/or progression of cancer.
