Abstract:AIM: Adeno-associated virus vector has become the focus as it has good safety, low immunogenicity, could infect separated and unseparated cells as well as mediate stable and long-term expression of exogenous gene. It is the most prospective virus vector for clinical application. In this paper, we observed the effects of recombinant adeno-associated virus vector encoding the report gene of galactosidase LacZ (rAAV-LacZ) transfected bone tissue in vivo. METHODS: The experiment was conducted in the Third Hospital of Peking University Medical School and Yijishan Hospital of Wannan Medical College from January 2005 to January 2006. Eight male SD rats of 8-week old and 220 g were selected and randomly divided into experiment group and control group with 4 in each group. A segmental defect was created in the lateral tibia of four rats in the experiment group, 6×1011 v·g rAAV-LacZ and type Ⅰ collagen sponge composites were implanted into the bone defects, while in the control group, only type Ⅰ collagen sponge composites were implanted. ① After X-gal staining analysis at 6 weeks post-implantation, the gross pathological and histological observation was carried out to detect the β-gal expression. ② The implanted sites, liver and heart tissue mRNA were harvested to detect whether there were specific expressions there by RT-PCR. The same bone defects were created in anther four mature rats and only type Ⅰ collagen sponges were implanted into the bone defects, which served as controls. RESULTS: Eight rats were involved in the result analysis. ① Gross pathological observation showed that the β-gal expressions were found in bone defects and surrounding muscle of the experiment group through X-gal staining, but no marker gene expression was found in the distant tissue (liver and heart) of type Ⅰ collagen sponges implanted defects. ② Histological observation suggested that the β-gal expression was found in the muscle fiber of bone defects in the experiment group without obvious local infiltration in leukomonocytes. In the blue-stained bone cortex, the osteocytes in bone lacuna specifically expressed β-gal, which was not observed in the control group. ③ RT-PCR results appealed that there were specific β-gal expression in the experimental site of the experiment group, while no marker gene expression was found in the distant tissue (liver and heart). There were no gene expression in the experiment site and the distant tissue (liver and heart) of the control group. CONCLUSION: After injected in the bone defects and bone fracture sites, adeno-associated virus vector could effectively transfer the bone and adjacent muscle tissues, and express exogenous gene for at least six weeks; moreover, it is very safe.
