Inhibitory effects of JAK inhibitor AG490 on invasion and migration in MCF-7 cells

Abstract:Objective: To explore the cross-talking between JAK/STAT3 and MAPK/ERK1/2 signaling pathways and effects of janus kinase (JAK) inhibitor AG490 on invasion and metastasis in MCF-7. Methods: MCF-7 was treated with AG490. Invasion and migration of MCF-7 were investigated with transwell chamber test. The secretion of MMP-2 and MMP-9 was tested with gelatin zymography. Meanwhile the expression and transcription of STAT3 and ERK1/2 in MCF-7 were detected by Western blot and RT-PCR. Results: In MCF-7, it was detected that expressions of p-STAT3, STAT3, p-ERK1/2 and ERK1/2, as well as transcription levels of STAT3 and ERK1/2 mRNA were remarkably decreased after treatment with AG490. In addition, it was observed that the secretion of MMP-2 and MMP-9 in MCF-7 treated with AG490 diminished sharply. MCF-7 suppressed the invasion and migration of MCF-7 by down-regulating MMP-2 and MMP-9, which was supported by the results of transwell chamber test in this experiment. Conclusion: Our study demonstrates that AG490 can block STAT3 and ERK1/2 signaling pathways. The inhibition of JAK could block the invasion and metastasis of MCF-7 through regulating MMP-2 and MMP-9.
