Primary Study on Callus Induction of Pinus Koraiensis

Abstract:Callus induction of Pinus koraiensis was studied by taking immature and mature zygotic embryos as explants.The results showed that:(1)mature zygotic embryos were more suitable for callus induction than immature zygotic embryos;(2)explant browning occurred mainly on radicle,therefore cotyledon,hypocotyls and embryo without radicle should be selected as explants;(3)effective type II callus can massively regenerated on LM medium plus 2.0 mg/L 2,4-D and 0.5 mg/L 6-BA,and early pre-embryo and quasi-embryogenic callus can be seen in histological observation;(4)LM medium plus 0.5 mg/L 2,4-D and 0.2 mg/L 6-BA was suitable for callus subculture.
