Abstract:Objective To investigate the immune response against Hepatitis B virus (HBV) in- duced by HBsAg recombinant adenovirus (Ad-S)-transduced dendritic cells (DCs) in HBV transgenic mice (HBV-Tg),and evaluate its potential in treating chronic HBV infection.Methods DCs were generated from bone marrow cells from the HBV Tg mice.and transfected with Ad-S (DC/Ad-S) or pulsed with HBsAg protein (DC/HBsAg).HBV-Tg mice were randomly assigned to receive either DC/Ad-S (n=10) or DC/HBsAg(n=10) or pcDNA3.1(+) S plasmid (n=6),and the same im munization schedule was repeated twice at 3 week intervals.Intracellular cytokines of splenic T cells, HBsAg-specific activity of splenic cytotoxic T lymphocyte (CTL),and HBsAg and anti-HBs and HBV DNA titers in sera were detected after immunization by flow eytometry,LDH release assay, ELISA and quantitative PCR,respectively.Hepatic injury was monitored at various time points after immunization by measuring serum alanine aminotransferase (ALT) activity.Tissue samples of the liver were stained with hematoxylin and eosin for histological analysis,and the expression of hepatitis B core Ag(HBcAg) and HBsAg was assessed by immunohistochemical analysis.Results DC/Ad-S stimulated splenic Tc more efficiently to secrete IFNγthan DC/HBsAg or pcDNA3. 1 (+)-S (P＜0.05 or P＜0.01),and induced stronger splenic HBsAg-specific CTL responses than that induced by DC/HBsAg (P＜0.05 or P＜0.01 at 10:1,20:1,40:1 of E:T ratio),which were stronger than that induced by DNA vaccination (all P＜0.01 at 10:1,20:1,40:1 of E:T ratio),at 1 or 2 weeks after immunization, respectively. The CTL response elicited by DC/Ad-S was transient, rising to strongest at 1 to 2 weeks after immunization and waning gradually thereafter,but reduced more slowly than that elicited by DC/HBsAg.The suppression of serum HBsAg and HBV DNA and the reduction of expression of HBcAg and HBsAg in liver from HBV-Tg mice induced by DC/Ad-S were the strongest in all groups of immunization,and the second was that induced by DC/HBsAg from 1 to 4 weeks after immunization.The serum anti-HBs titer and ALT activity were slightly elevated,and histological analysis of the liver displayed only a very slight focal inflammatory infiltration up to 4 weeks after immunization,and the inter-group differences were not significant.Conclusions HBsAg recombinant adenovirus-transduced DCs can induce Tcl and CTL response,reduce the titer of serum HBsAg and HBV DNA,and inhibite the expression of HBcAg and HBsAg in liver in HBV-Tg mice, stronger than that induced by HBsAg-pulsed DCs or plasmid DNA.
