Abstract:Lactic acid bacteria (LAB) are a group of bacteria that are naturally present in the digestive tract of vertebrates, including fish such as Channa sp. Lactic acid bacteria are considered the most suitable microbes for use as probiotics. Probiotics can produce antimicrobial metabolites so that can improve the microbial balance in the digestive tract. It is believed that Channa sp. harbors LAB in its digestive tract, which may have probiotic potential and produce antimicrobial metabolites that can inhibit pathogenic bacteria. The aim of this study was to isolate and identify LAB from the intestines of Channa sp. The research involved several steps, including sample preparation, LAB isolation, characterization, and purification of bacterial isolates, biochemical tests, temperature resistance tests, antimicrobial tests, haemolysis tests, lactic acid production tests, and cholesterol-lowering activity tests. Eight LAB isolates with potential probiotic characteristics were isolated and identified as the genus Lactobacillus, which are gram-positive bacteria that produce lactic acid during culture

Abstract:To study the taxonomic,morphologic,physiological and biochemical characteristics of lactic acid bacteria(LAB) in silage,five strains of LAB were isolated from alfalfa(Medicago sativa L.)and maize(Zea mays L.)silage.The 16S rRNA sequence analysis indicate that two strains are Pediococcus pentosaceus(GI7,GI24),and the other three are Pediococcus acidilactici,Lactobacillus curvatus and Lactobacillus plantarum(GI11,GI44,GI62).Each is homofermentative lactic acid bacteria and can grow well or weakly at 5,10,40,45℃,and with pH3.0~8.0,3.0% and 6.5% NaCl except that GI44 does not grow at pH3.0,4.0,8.0.Through measuring the rate of lactic acid production and the growth curve,GI62 was selected as eximious strain used for preparing lactic acid bacteria additive due to its rapid acid-production and growth speed.

Abstract:Seven strains of lactic acid bacteria were isolated and purified from the fermented horse meat. Their main fermentation characteristics such as salt and nitrite tolerance,slime and gas production were studied,the growth and acid production on different temperature and pH change in process of fermentation were compared. Three strains of them had good fermentation characteristics in the speed of growth and acid production,resisted 6% NaCl and 150 mg/kg NaNO2,no gas and ammonia producing,and H2O2,H2S,V-P reactian was negative,ect. The results showed that they meet the requirements of fermented meat products bacteria. After preliminary identification on morphologic observation and carbon source utilization test,two of them are C.callins,one is L.delbrueckii.

Abstract:The LCA strain was identified as Lactobacillus pentosus according to the morphology,physiological characteristic and 16S rDNA.The fermentation products of LCA strain were analyzed by means of the chiral column HPLC.The optical purity of D-lactic acid was more than 80%.It was critical important for widening the range of application of D-lactic acid to screen suitable strain for D-lactic acid production and investigated technology of fermentation.
Probiotic potential characterization and clustering using unsupervised algorithms of lactic acid bacteria from saltwater fish samples

Abstract:This research aimed to isolate lactic acid bacteria from the bowel of saltwater fish to assess their potential probiotic properties. Nineteen isolates of LAB including Lactiplantibacillus plantarum, Lactiplantibacillus pentosus, Lactobacillus acidophilus, Levilactobacillus brevis, Pediococcus pentosaceus, and Pediococcus acidilactici were recognized using molecular tools. All the isolates survived in the simulated conditions of the GI tract. Auto-aggregation ranged from 01.3 ± 0.5 to 82.6 ± 1.4% and hydrophobicity with toluene ranged from 3.7 ± 1.6 to 69.4 ± 1.3%, while the range of hydrophobicity with xylene was from 02.2 ± 1.6 to 56.4 ± 2.1%. All the isolates of lactobacilli, pediococci, enterococci, and lactococci indicated variable sensitivity and resistance towards clinical antibiotics. Non-neutralized cell free supernatant of isolates F12 and F15 showed antimicrobial activity against all the 8 evaluated enteric pathogens. Cluster analysis of identified potential probiotic bacteria based on heat-map and PCA methods also highlighted the priority of isolates F3, F7, F12, and F15 as bio-control agents in fishery industry. The findings of this study may essentially contribute to the understanding of the probiotic potential of LAB in saltwater fish, in order to access their probiotic characterization for use as biocontrol in fishery.

Abstract:Presently in order to excavate suitability of resources of lactic acid bacteria for fermentation of mandarin fish, a strain of lactic acid bacteria (LAB), which was cultivated from stinky mandarin fish via using traditional MRS medium (CMC 1-8), was firstly isolated and purified. Subsequently, it was discovered that the as-harvested LAB was recognized as Lactobacillus sakei subsp. sakei with assistance of technology of phenotypic identifying and 16S rRNA sequencing. Concurrently, relevant characteristics in aspects of growth curve, acid production capacity, temperature and salt tolerance of Lactobacillus sakei subsp. sakei were plainly investigated. Furthermore, the preliminary research on protease properties and biological properties was revealed. The results unfolded that the strain could grow and produce acid normally at 37 °C, and deliver a strong tolerance even under 3% NaCl at 12 °C, which could be significant sufficiently to satisfy the low-temperature and salty fermentation of mandarin fish. The best compatible temperature and pH value for protease production were at 70 °C and 9.0, respectively. Resultantly, the main metabolites, containing polyphenols, flavonoids and soluble polypeptides, performed a remarkable effect on scavenging abilities of 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 1,1-diphenyl-2-picrylhydrazine (DPPH) free radical and ferricyanide reducing ability, of which the IC50 were 1.05, 6.87, and 4.62 mg/mL, respectively. And, the metabolites displayed partial inhibitory effects on Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis. Therefore, CMC 1-8 can potentially serve as a functional microorganism to directionally ferment mandarin fish.

Abstract:In order to obtain lactic acid bacteria using molasses wastewater to produce lactic acid,an activated sludge was sampled from bioreactor with molasses wastewater,and strain MD-9 was isolated by an anaerobic Hungate technique.The morphology,physic-biochemical characteristics and 16S rRNA sequence analysis of strain MD-9 were investigated.The results indicated that 16S rRNA gene homology of strain MD-9 with Lactobacillus fermentum was 99.6%.Strain MD-9 is facultative anaerobe bacillus.Glucose fermentation is heterologous.Strain MD-9 can use molasses wastewater to produce lactic acid.

Abstract:Background: There are still a large variety of microorganisms among aquatic animals, especially probiotic lactic acid bacteria in cold water fishes at high latitudes have not been fully developed. Hence, the present study aims to evaluate the probiotic potential of cold-adapted Lactobacillus strains isolated from the intestinal tract of cold water fishes (Xinjiang) and select candidates to be used as new food preservative agents and/or probiotic additives in feeding of aquaculture. Results: A total of 43 Lactobacillus spp. were isolated from 16 kinds of intestinal tract of cold-water fishes. They were characterized by phenotypic methods, identified using Rep-PCR and 16S rRNA gene sequencing as four species: Lactobacillus sakei (22 isolates), Lactobacillus plantarum (16 isolates), Lactobacillus casei (4 isolates) and Lactobacillus paracasei (1 isolate). The in vitro tests included survival in low pH and bile, antimicrobial activity (against Escherichia coli, Salmonella enterica subsp. enterica serovar Typhimurium, Salmonella enterica subsp. enterica, Listeria monocytogenes, and Listeria innocua), resistance to 15 antibiotics and hemolytic tests. Among all 43 lactobacilli isolates, the 22 isolates showed a wide range of antimicrobial activity against 6 different pathogenic strains. There were twenty isolates growing at optimal temperature ranging 16~20 °C, which were initially considered to be cold-adapted strains. Two (2) Lb. sakei strains and 2 Lb. plantarum strains demonstrated the highest survivability after 4 h of exposure at pH 2.0. Most of the tested strains cannot be cultured after exposed into 0.5% bile solution for 4 h, while 2 Lb. plantarum strains (E-HLM-3, CQ-CGC-2) and 1 Lb. sakei strain M-DGM-2 survived even at 2% bile concentration. In addition, the safety assessment showed that 22 strains without any detectable hemolytic activity and resistant to glycopeptides (vancomycin, teicoplanin), levofloxacin, aztreonam, amikacin and oxacillin, while all the studied lactobacilli showed sensitivity to or semi-tolerant to other antibiotics. Conclusions: Based on all the experiments, 3 strains, including E-HLM-3, CQ-CGC-2, and M-DGM-2 might be a candidate of choice for using in the food preservative agents and/or probiotic additives in feeding of aquaculture.
Isolation and Identification of Lactic Acid Bacteria from Previously Fermented Juice of Italian Ryegrass

Abstract:In order to study lactic acid bacteria species which plays a major role in the fermentation process of Italian ryegrass(Lolium multiflorum)silage,6lactic acid bacteria strains(L-1,L-3,L-4,L-5,L-8and L-9)were isolated from previously fermented juice of Changjiang No.2Italian ryegrass.The results of traditional biochemical identification,experiment of acid-production rate,acid tolerance test,salt tolerance test,and the 16SrRNA sequence analysis indicated that L-1,L-3,L-4,L-5,L-8and L-8are Lactobacillus plantarum,Lactococcus lactis,Lactococcus garvieae,Leuconostoc lactis,Weissella confuse and Weissella cibaria,respectively,and all of them have good acid productivity,strong acid and salt tolerance.Further studies on using these strains as feed additives can be made.

Abstract:According to Lactobacillus 16 S-23 S rRNA intergenic spacer region sequences in GenBank,primers were designed to further identify Lactobacillus HZ521 isolated from pig intestines.Meanwhile,the present research,considering L.acidophilus ATCC4356 as reference strain,was aimed to examine the probiotic activities of HZ521 in vitro.Results of homology of partial 16 S-23 S rRNA gene sequence indicated that the strain belonged to L.helveticus.L.helveticus HZ521 possessed strong aci-producing ability and was resistant to acidic condition,and strian HZ521 was more adherent to HeLa cells than strain ATCC 4356 with statistical difference(P0.01).These results suggested that L.helveticus HZ521 possessed probiotic properties and could be a potential candidate of strains for probiotic application.

Abstract:A strain of high-yield lactic acid bacterium,designated as TD175,was isolated from food waste.The strain TD175 produced 78.56 g/L of lactic acid from 100 g/L of glucose after 72 h fermentation.Based on its morphology and physio-biochemical properties,the strain TD175 was tentatively identified as Lactobacillus sp..Analysis of 16S rDNA sequence showed that TD175 had high homology(99%) with Lactobacillus sp.MR2,L.plantarum and L.pentosus.An acid-tolerance strain TD175-1 was obtained by breeding of the strain TD175.Lactic acid output for the strain TD175-1 was 10.7% higher than the strain TD175.The strain TD175-1 was capable of enhancing lactic acid fermentation of food waste,with 29.65 g/L of lactic acid produced after 48 h of anaerobic fermentation,which was 34.3% higher than the spontaneous fermentation without inoculum.

Abstract:Six strains of lactic acid bacteria were obtained from the fermentation broth of Chinese rice wine.By using methods of physiological-biochemical and 16S rDNA tests,strains R1,R4,R5,R8 and R20 were identified as Lactobacillus rossiae,and strain R2 was identified as Lactobacillus casei.The ability of producing biogenic amines by these strains was evaluated by MRS plates and showed that only strain R1 could produce biogenic amines.HPLC confirmed that strain R1 could produce 598.61mg/L putrescine,and the others could not produce biogenic amines.The consistency of the results indicated that MRS plates analysis could be used as an effective,simple and economical method to evaluate the lactic acid bacteria's ability of producing biogenic amines.

Abstract:The isolation and screening of lactic acid bacteria (LAB) from natural sources have been one of the powerful means to obtain strains for the food industry. A total of 275 indigenous isolates were obtained from 43 samples of traditional fermented foods in specific ecological niches throughout the northwestern China, and among which 13 strains of LAB were selected for their potential in food preservation and production. Among the 13 isolates, Lactobacillus (10) was dominant over Lactococcus (3). The distribution of the isolates was as follows: Lactobacillus paracasei ssp. paracasei (J23, M10, M20, M22), Lactobacillus rhamnosus (J20, M18), Lactococcus lactis ssp. lactis (X20, Q7), Lactobacillus casei (Q1, Q12), Lactobacillus plantarum (J11), Lactococcus lactis ssp. cremoris (X8), Lactobacillus delbrueckii ssp. bulgaricus (Q5). All 13 isolates produced bacteriocin with a broad inhibitory spectrum against selected Gram-positive as well as Gram-negative pathogenic and spoilage species. Biochemical analysis revealed that they possessed high acidification and coagulation activity. Several strains possessed the high activity of 2 or 3 technological characteristics, related to novel starters and food preservatives.

Abstract:In this study, three Lactiplantibacillus plantarum, namely 3-14-LJ, M22, and MB1, with high acetate esterase activity, acid, salt, and high-temperature tolerance were selected from 708 strains isolated from fermented food. Then, L. plantarum strains MB1, M22, and 3-14-LJ were inoculated at 107 CFU/mL in the model and 107 CFU/g in actual Suanzhayu systems, and the effects during fermentation on the physicochemical properties, amino acid, and volatile substance were investigated. The results showed that the inoculated group had a faster pH decrease, lower protein content, higher TCA-soluble peptides, and total amino acid contents than the control group in both systems (p < 0.05). Inoculation was also found to increase the production of volatile compounds, particularly esters, improve the sour taste, and decrease the bitterness of the product (p < 0.05). L. plantarum M22 was more effective than the other two strains in stimulating the production of isoamyl acetate, ethyl hexanoate, and ethyl octanoate. However, differences were discovered between the strains as well as between the model and the actual systems. Overall, the isolated strains, particularly L. plantarum M22, have good fermentation characteristics and have the potential to become excellent Suanzhayu fermenters in the future.

Abstract:Lactic acid bacteria are commonly used in food industry,mainly in the fermentation process of meat products,prickles and diary products. They can add special flavor and color to the products,depress the growth of pathogens,and reduce nitrite concentration in the products,which makes the products safer to the customers.12 strains of lactic acid bacteria were isolated from the pickled vegetables.After amplification and sequencing of 16S rDNA gene,the isolated strains were identified by sequence homology as follows: 9 strains of Lactobacillus plantarum,2 strains of Lactobacillus sakei and 1 strain of Pediococcus pentosaceus.Some of their biochemical characteristics were studied and compared,as well as their abilities on the tolerance and degradation of nitrite.A L.plantarum strain,named J-10,was approved to degrade nitrite quickly with good biochemical characteristics. When grown in 0.25 mg/ml nitrite sodium added medium,strain J-10 could degrade 99.2% nitrite sodium at 37℃ and pH 6.2 for 24 h.

Abstract:The study was conducted to isolate, select, and identify the lactic acid bacteria strain with the highest lactic acid fermentation ability from pickled lotus root products in Giang Thanh district, Kien Giang province. From three samples of pickled lotus root collected in Giang Thanh district, 11 lactic acid bacteria strains were isolated and preliminarily identified through colony morphology and some biochemical reactions such as Gram staining and catalase reaction. The results of determining the fermentation type showed that all 11 isolated bacterial strains had a homomorphic fermentation type, the main fermentation product was lactic acid. Of which, strain D15 had the highest lactic acid content, reaching 1.87 mg/mL and had the highest biomass growth ability, with an OD600nm value of 0.947. The 16S rRNA gene sequence of strain D15 was determined with a molecular size of 1469 bp. Comparing the 16S rRNA gene sequence of strain D15 on the gene bank, it has a similarity level of 100% with some lactic acid bacteria strains of the species Lactobacillus plantarum and 99.68% when comparing with the 16S rRNA gene sequence of the standard strain Lactobacillus plantarum. Constructing a phylogenetic tree and evaluating the genetic relationship of strain D15, the standard strain Lactobacillus plantarum, and 9 strains with the highest similarity to strain D15 based on the 16S rRNA gene sequence. The results were classified into 2 large groups with a similarity level of 92%, in which strain D15 is in the same group as the standard strain Lactobacillus plantarum and also has a similarity level of 92%.
Antimicrobial Characteristics of Lactic Acid Bacteria Isolated from Homemade Fermented Foods

Abstract:Objective. Lactic acid bacteria (LAB) were isolated from fermented foods, such as glutinous rice dough, corn noodle, chili sauce, potherb mustard pickles, and stinky tofu, in northeast China. LAB strains with antimicrobial activities were screened, and seven of these Lactobacillus strains were identified as L. plantarum , L. pentosus , and L. paracasei through 16S rRNA gene analysis. After the supernatant of LAB was treated with proteinase K, pepsin, and papain, their antibacterial effect almost disappeared. Most strains with antibacterial activities were highly resistant to heat (65°C–121°C), acidity (pH 2–6), and alcohol. The antimicrobial effect of most strains treated with the Tween-80 surfactant was significantly reduced, and the antibacterial property of T4 was even lost. Ammonium sulfate precipitation, PCR, and nanoLC-ESI-MS/MS results confirmed that T8 produced antibacterial substances belonging to a protein family, and its zone of inhibition against pathogens significantly increased (>13 mm). In bacterial growth inhibition experiments, the colony count of Staphylococcus aureus was up to 10 15 CFU/mL in the 3 ⁎ de Man, Rogosa, and Sharpe (MRS) group, and this value was more than that in the 3 ⁎ S6 supernatant group (10 12 CFU/mL) and the control group (10 10 CFU/mL) at 12 h. This study provided a basis for the selection of antimicrobial peptides and the development and utilization of LAB.

Abstract:Leymus chinensis is an important forage in China. To investigate the characteristics of lactic acid bacteria (LAB) and select strains for enhancing fermentation quality L. chinensis silage, 167 LAB strains were isolated from L. chinensis silage prepared on steppes in China. Based on physiological, biochemical characteristics and 16S rRNA gene sequence, the isolates were divided into 10 groups (A–J), including Lactobacillus plantarum (species ratio: 44.9%), Lactobacillus brevis (40.7%), Lactobacillus casei (6.6%), Weissella hellenica (2.4%), Enterococcus faecium (1.2%), Enterococcus faecalis (1.2%), Enterococcus mundtii (1.2%), Enterococcus durans (0.6%), Enterococcus sulfureus (0.6%) and Lactococcus garvieae (0.6%). Lactobacillus plantarum and Lb. brevis were the dominant species in L. chinensis silage. Lactobacillus plantarum NG 2 was selected and its effect on fermentation quality of L. chinensis silage was evaluated. NG 2 decreased pH, ammonia N, butyric acid contents, count of coliform bacteria, yeasts and moulds, and increased lactic acid content (P < 0.05). Overall, L. chinensis silage contains plenty of LAB species and these LAB inoculants could improve the fermentation quality. Lactobacillus plantarum NG2 can be used to enhance fermentation quality of L. chinensis silage.

Abstract:A total of 83 acid-producing strains were isolated from fresh Lentinus edodes, Pleurotus eryngii, Flammulina velutipes, Agaricus bisporus, Pleurotus ostreatus, Pleurotus djamor, Pleurotus abalones, and Pleurotus citrinopileatus by CaCO3-MRS plate medium. These 83 strains were divided into 9 species, including 52 strains of Lactococcus lactis, 13 strains of Pediococcus pentosaceus, 8 strains of Enterococcus faecium, 3 strains of Lactococcus garvieae, 2 strains of Enterococcus casseliflavus, 2 strains of Lactobacillus plantarum, 1 strain of Enterococcus lactis, 1 strain of Pediococcus acidilactici, and 1 strain of Lactobacillus pentosus based on the catalase test, Gram staining, and 16S rDNA molecular identification. Among these P. acidilactici, P. pentosus, and L. plantarum could be used in food. P. acidilactici had a strong acid-producing capacity and fast growth rate, thus showing preferable fermentation characteristics in MRS broth and edible mushroom medium than P. pentosus and L. plantarum. In the medium of L. edodes, P. eryngii, and F. velutipes, the best sensory state can be reached within 12-24 h, among them P. eryngii had the best fermentation effect, which was characterized by uniform and bright color, moderate acidity, good flavor, and tight tissue state. This study investigated the types and fermentation characteristics of lactic acid bacteria (LAB) derived from edible mushrooms, enriched the resources of LAB suitable for the fermentation, and provided a theoretical reference for the application of LAB fermentation technology of edible mushrooms.
