Abstract:Objective To analyze the expression of macrophage inflammatory protein-1αand the relationship with neovascularization in atheresclerotic plaque.Methods Twenty-four Wistar rats were di- vided randomly into two groups:normal group and model group.The atherosclerosis models were estab- lished with high lipid + vitamin D3 in the model group.Serum lipid was determined.Immunohistochemical staining method was used to detect the expression of MIP-1αand micro-vessel density (MVD).Results The expression of MIP-1αand MVD in the model group was significantly higher than in the normal group (P<0.05),and that in unstable group was significantly higher than in the stable group (P<0.05). There was a positive correlation between MIP-1αand MVD (r=0.671,P<0.05).Conclusion The content of MIP-1α,in the unstable athcrosclerotic phque was higher than in the stable one,suggesting the content of MIP-1αin the atherosclerotic plaque may be an important factor influencing the stabilization of plaque.MIP-1αcan induce the neovascularization in atherosclerotic plaque,and promote the instabiliza- tion of plaque.

Abstract:Transient receptor potential ankyrin 1 channel (TRPA1) plays an important role in the pathogenesis of inflammatory diseases, yet its role and the underlying mechanism in atherosclerosis remain unclear. We aimed to investigate the role of TRPA1 in atherosclerosis and foam-cell formation in vivo in mice and in vitro in mouse macrophages. Histopathology was examined by hematoxylin and eosin staining, levels of cytokines and lipid profile were evaluated by assay kits, and protein expression was determined by western blot analysis. TRPA1 expression was increased in macrophage foam cells in atherosclerotic aortas of apolipoprotein E-deficient (apoE(-/-)) mice. Atherosclerotic lesions, hyperlipidemia and systemic inflammation were worsened with chronic administration of the TRPA1 channel antagonist HC030031 or genetic ablation of TRPA1 (TRPA1(-/-)) in apoE(-/-) mice. Treatment with allyl isothiocyanate (AITC, a TRPA1 agonist) retarded the progression of atherosclerosis in apoE(-/-) mice but not apoE(-/-)TRPA1(-/-) mice. Mouse macrophages showed oxidized low-density lipoprotein (oxLDL) activated TRPA1 channels. OxLDL-induced lipid accumulation of macrophages was exacerbated by HC030031 or loss of function of TRPA1. Inhibition of TRPA1 activity did not alter oxLDL internalization but impaired cholesterol efflux by downregulating the ATP-binding cassette transporters. Furthermore, tumor necrosis factor-α-induced inflammatory response was attenuated in AITC-activated macrophages. TRPA1 may be a pivotal regulator in the pathogenesis of atherosclerosis and cholesterol metabolism of macrophage foam cells.

Abstract:Abstract Atherosclerosis is a main pathological basis for cardiovascular diseases. Macrophages, featured in phenotypic and functional plasticity, play critical roles in initiation, progression, and regression of atherosclerosis. Our previously studies have shown that protein tyrosine phosphatase SHP2 regulates macrophage polarization, the immune homeostasis mediated by the interactions between macrophages and epithelial cells in mouse lung and intestine, and a tumor-favorable immune microenvironment. However, a potential role of SHP2 in atherosclerotic macrophages remains unknown. The expression of SHP2 is increased in macrophages within human atherosclerotic plaques. SHP2 allosteric inhibitor and macrophage-specific SHP2 deletion worsened atherosclerosis in ApoE null mice. Total cholesterol and low-density lipoprotein were increased in SHP2 inefficient mice. SHP2 inefficiency promoted inflammatory M1 polarization and inhibited anti-inflammatory M2 polarization, which was ApoE-null-dependent. Less cholesterol accumulation was observed in SHP2 inefficient macrophages and cholesterol transporter CD 36 was downregulated. To further explore the underlying mechanism, RNA sequencing was performed using SHP2 deleted macrophages with or without oxLDL treatment. Taken together, our studies using SHP2 knockout mice and specific inhibitor suggest that SHP2 is a novel anti-atherosclerotic regulator in promoting inflammatory macrophage phenotype.

Abstract:In this study, we aim to determine cellular mechanisms linking nutrient metabolism to the regulation of inflammation and insulin resistance. The nutrient sensors AMP-activated protein kinase (AMPK) and SIRT1 show striking similarities in nutrient sensing and regulation of metabolic pathways. We find that the expression, activity, and signaling of the major isoform alpha 1AMPK in adipose tissue and macrophages are substantially down-regulated by inflammatory stimuli and in nutrient-rich conditions, such as exposure to lipopolysaccharide (LPS), free fatty acids (FFAs), and diet-induced obesity. Activating AMPK signaling in macrophages by 5-aminoimidazole-4-carboxamide-1-beta 4-ribofuranoside or constitutively active alpha 1AMPK (CA alpha 1) significantly inhibits; although inhibiting alpha 1AMPK by short hairpin RNA knock-down or dominant-negative alpha 1AMPK (DN-alpha 1) increases LPS- and FFA-induced tumor necrosis factor alpha expression. Chromatin immunoprecipitation and luciferase reporter assays show that activation of AMPK by CA-alpha 1 in macrophages significantly inhibits LPS- or FFA-induced NF-kappa B signaling. More importantly, in a macrophage-adipocyte co-culture system, we find that inactivation of macrophage AMPK signaling inhibits adipocyte insulin signaling and glucose uptake. Activation of AMPK by CA-alpha 1 increases the SIRT1 activator NAD(+) content and SIRT1 expression in macrophages. Furthermore, alpha 1AMPK activation mimics the effect of SIRT1 on deacetylating NF-kappa B, and the full capacity of AMPK to deacetylate NF-kappa B and inhibit its signaling requires SIRT1. In conclusion, AMPK negatively regulates lipid-induced inflammation, which acts through SIRT1, thereby contributing to the protection against obesity, inflammation, and insulin resistance. Our study defines a novel role for AMPK in bridging the signaling between nutrient metabolism and inflammation.

Abstract:Objective:Atherosclerosis is an immune and inflammatory disease.Abundant inflammatory and immune mediators and macrophages are two key features of atherosclerotic vulnerable plaques.Since oxHDL and oxLDL have been detected in atherosclerotic plaques,exploration and comparison of the effect of these two factors on production of inflammatory and immune mediators are much helpful to further elucidate the mechanism of atherogenesis.Therefore,the present study is aimed to investigate the influence of oxHDL3 and oxLDL in stimulation of expression of four atherosclerosis-related genes coding for interleukin-1β(IL-1β),interleukin-6(IL-6),class Ⅱ molecule HLA-DR,and costimulatory molecules CD86,which are immune and inflammatory-associated mediators,in THP-1monocyte-derived macrophages.Methods:After treated with oxHDL3 and oxLDL for 0-48 h,IL-1β and IL-6 mRNA levels in THP-1monocyte-derived macrophages were detected by RT-PCR,while surface class Ⅱ MHC and co-stimulatory molecules expression were analyzed by flow cytometry.Results:In THP-1monocyte-derived macrophages treated with oxHDL3 and oxLDL,IL-1β and IL-6 mRNA expression were significantly elevated.oxHDL3 and oxLDL also increased surface expression of HLA-DR and CD86 in a dose-and time-dependent manner.The percentage of HLA-DR+cells and CD86+cells were augmented remarkably.Neither native HDL3 nor native LDL caused an increase of IL-1β and IL-6 mRNA as well as HLA-DRand CD86 surface expression.Conclusion:oxHDL3 and oxLDL might be involved in atherogenesis via stimulating the expression of IL-1β,IL-6,CD86,and HLA-DR in THP-1monocyte-derived macrophages.

Abstract:BACKGROUND AND AIMS:Macrophage is a major contributor to the development of atherosclerosis by taking up deposited lipoprotein and eliciting local inflammation. Previously, we and others have shown C1q/TNF-related proteins (CTRPs) play diverse roles in vascular functions. In this study, we sought to investigate the changes in CTRP expression levels during vital biological processes in macrophages and their relation to inflammatory responses.METHODS:Western blot and real-time PCR were performed to analyze CTRPs expression levels in human peripheral blood mononuclear cells, primary macrophages and lipid-laden foam cells. Mechanisms that regulate CTPR1 expression were further investigated by bioinformatic analysis and chromatin immunoprecipitation. Enzyme-linked immunosorbent assay was performed to measure the concentration of inflammatory cytokines.RESULTS:We found that almost all CTRPs were significantly increased in primary human macrophages after differentiation from peripheral blood mononuclear cells. In particular, CTRP1 was further up-regulated upon exposure to oxidized low-density lipoprotein (oxLDL) in a peroxisome proliferator-activated receptor (PPAR)-dependent manner. Chromatin immunoprecipitation also confirmed the presence of PPAR-γ in the CTRP1 promoter after oxLDL treatment. Stimulation of CTRP1 led to markedly enhanced secretion of pro-atherogenic factors, including MCP-1, TNF-α, IL-1β, and IL-6, whereas oxLDL-induced inflammatory cytokine production was significantly attenuated after the treatment with CTRP1 neutralizing antibody.CONCLUSIONS:These data suggest an essential role of CTRP1 in linking dysregulation of lipid metabolism and inflammatory responses in macrophages.

Abstract:Objective To investigate the effects of apolipoproteinA-I (apoA-I) on the expression of monocyte chemoattractant protein-1 (MCP-1), vascular cell adhesion molecule-1 (VCAM-1) and ATP-binding cassette transporter A1 (ABCA1) in macrophage derived foam cells. Methods The cultured human acute mono cytic leakemia (THP-1) cells were induced into foam cells by exposing to phorbol myristate acetate (PMA) for 48 h, and then to oxidized low-density lipoprotein (ox-LDL) for 48 h. The expression of MCP-1, VCAM-1 and ABCA1 in macrophages and foam cells were examined by immunohistochemistry method before and after treatment with apoA-I (10 mg/L) or ABCA1 antibody for 24 hours. Results ① After treatment with ox-LDL(50 mg/L), the expression of MOP-1, VCAM-1 and ABCA1 was obviously increased in the foam cells compared with that in the macrophages. ② Compared with the non-interventional group, the expression of MCP-1 and CVAM-1 in the foam cells was significantly decreased after treatment with apoA-I 10 mg/L, and the expression of ABCA1 was obviously decreased; after intervention with ABCA1 antibody (10 mg/L), the expression of MCP-1 was significantly increased. Conclusion ApoA-I has anti-arthrosclerosis and anti-inflammatory effect, which is through increasing the expression of ABCA1, MOP-1 and VCAM-1 protein expression. ABCA1 channel may play a role in decreasing the expression of MCP-1 in foam cells.
The Role of Monocyte Chemotactic Protein-Induced Protein 1 (MCPIP1) in Angiotensin II-induced Macrophage Apoptosis and Vulnerable Plaque Formation.

Abstract:Atherosclerotic plaque rupture is the main cause of acute coronary syndrome (ACS). Angiotensin II (Ang II) and macrophage apoptosis are involved in the pathogenesis of atherosclerosis. However, the underlying mechanisms remain unclear. We aimed to address the role of monocyte chemotactic protein-induced protein 1 (MCPIP1) in Ang II-induced macrophage apoptosis and vulnerable plaque formation. In mouse peritoneal macrophages, Ang II promoted endoplasmic reticulum (ER) stress-dependent macrophage apoptosis. Ang II markedly upregulated the expression of MCPIP1 via activating p38 mitogen-activated protein kinase (p38 MAPK). Treatment with MCPIP1 shRNA downregulated ER stress-related proteins and decreased macrophage apoptosis induced by Ang II. Ang II also activated the AMP-activated protein kinase (AMPK) signaling in macrophages. Inhibition of AMPK reduced macrophage apoptosis by inhibiting the p38 MAPK/MCPIP1/ER stress pathway. Furthermore, blocking the Ang II type 1 receptor (AT1R) with losartan effectively inhibited Ang II-induced macrophage apoptosis and AMPK/p38 MAPK/MCPIP1/ER pathway activation. In the atherosclerotic vulnerable plaque model in mice, losartan inhibited the progression of atherosclerosis and transformed vulnerable plaque into a more stable phenotype. Moreover, losartan markedly decreased the number of CD68+TUNEL+, CD68+MCPIP1+, CD68+p-eIF2α+ and CD68+CHOP+ cells in the lesion area. Taken together, Ang II promotes macrophage apoptosis via the AMPK/p38 MAPK/MCPIP1/ER stress pathway in macrophages via its receptor AT1R, which may contribute to vulnerable plaque formation. Our study clarifies a novel regulatory role of MCPIP1 in Ang II-induced macrophage apoptosis and plaque instability, providing a potential therapeutic target for prevention of ACS.

Abstract:The implication of the heterogeneous spectrum of pro- and anti-inflammatory macrophages (Macs) has been an important area of investigation over the last decade. The polarization of Macs alters their functional phenotype in response to their surrounding microenvironment. Macs are the major immune cells implicated in the pathogenesis of atherosclerosis. A hallmark pathology of atherosclerosis is the accumulation of pro-inflammatory M1-like macrophages in coronary arteries induced by pro-atherogenic stimuli; these M1-like pro-inflammatory macrophages are incapable of digesting lipids, thus resulting in foam cell formation in the atherosclerotic plaques. Recent findings suggest that the progression and stability of atherosclerotic plaques are dependent on the quantity of infiltrated Macs, the polarization state of the Macs, and the ratios of different types of Mac populations. The polarization of Macs is defined by signature markers on the cell surface, as well as by factors in intracellular and intranuclear compartments. At the same time, pro- and anti-inflammatory polarized Macs also exhibit different gene expression patterns, with differential cellular characteristics in oxidative phosphorylation and glycolysis. Macs are reflective of different metabolic states and various types of diseases. In this review, we discuss the major differences between M1-like Macs and M2-like Macs, their associated metabolic pathways, and their roles in atherosclerosis.

Abstract:Macrophages accumulated in the arterial intima play an important role in the development of atherosclerosis by producing a large number of proinflammatory cytokines which accelerate the disease. Recent studies show that adipophilin might be involved in inflammatory processes in macrophages. In this study, we observe the effect of adipophilin on proinflammatory cytokine expression and secretion in THP-1 macrophages. SiRNA and adipophilin gene overexpression mediated by an pEGFP-C3 vector were used to observe the effect of adipophilin on proinflammatory cytokines in THP-1 macrophages in vitro. Realtime PCR and enzyme-linked immunosorbent assay (ELISA) were applied to detect the production of tumor necrosis factor alpha (TNF-alpha), monocyte chemoattractant protein 1 (MCP-1), and interleukin-6 (IL-6). It was found that acetylated low-density lipoprotein (AcLDL), pioglitazone [a peroxisome proliferator-activated receptor gamma (PPARgamma) agonist] increased adipophilin expression in macrophages, while glucose had no such affect. It was also shown that adipophilin augments TNF-alpha, MCP-1, and IL-6 expression in AcLDL induced macrophages. Our results suggest that adipophilin augment inflammation in macrophages, which might be one role of adipophilin in atherosclerosis.

Abstract:RATIONALE:The mammalian target of rapamycin complex 1 inhibitor, rapamycin, has been shown to decrease atherosclerosis, even while increasing plasma low-density lipoprotein levels. This suggests an antiatherogenic effect possibly mediated by the modulation of inflammatory responses in atherosclerotic plaques. OBJECTIVE:Our aim was to assess the role of macrophage mammalian target of rapamycin complex 1 in atherogenesis. METHODS AND RESULTS:We transplanted bone marrow from mice in which a key mammalian target of rapamycin complex 1 adaptor, regulatory-associated protein of mTOR, was deleted in macrophages by Cre/loxP recombination (Mac-Rap(KO) mice) into Ldlr(-/-) mice and then fed them the Western-type diet. Atherosclerotic lesions from Mac-Rap(KO) mice showed decreased infiltration of macrophages, lesion size, and chemokine gene expression compared with control mice. Treatment of macrophages with minimally modified low-density lipoprotein resulted in increased levels of chemokine mRNAs and signal transducer and activator of transcription (STAT) 3 phosphorylation; these effects were reduced in Mac-Rap(KO) macrophages. Although wild-type and Mac-Rap(KO) macrophages showed similar STAT3 phosphorylation on Tyr705, Mac-Rap(KO) macrophages showed decreased STAT3Ser727 phosphorylation in response to minimally modified low-density lipoprotein treatment and decreased Ccl2 promoter binding of STAT3. CONCLUSIONS:The results demonstrate cross-talk between nutritionally induced mammalian target of rapamycin complex 1 signaling and minimally modified low-density lipoprotein-mediated inflammatory signaling via combinatorial phosphorylation of STAT3 in macrophages, leading to increased STAT3 activity on the chemokine (C-C motif) ligand 2 (monocyte chemoattractant protein 1) promoter with proatherogenic consequences.

C1q/TNF-related protein-levels were higher in sera, endarterectomy specimens, aortic atherosclerotic plaques, and peripheral blood mononuclear cells (PBMCs) from CAD patients compared with controls, and were related to CAD severity. The production of CTRP1 was profusely induced by inflammatory cytokines and itself caused a concentration-dependent expression of adhesion molecules and inflammatory markers in human endothelial cells, human peripheral blood monocytes, and THP-1 cells. C1q/TNF-related protein-1 induced p38-dependent monocyte-endothelium adhesion in vitro and the recruitment of leucocytes to mesenteric venules in C57BL/6 mice. Immunohistochemistry of atherosclerotic femoral arteries exhibited CD68 and VE-cadherin loci-associated increased CTRP1 expression in plaques. Compared with saline, intraperitoneal injection of recombinant CTRP1 protein (200 μg/kg) every other day promoted atherogenesis in apoE(-/-) mice at 24 weeks. However, pro-atherogenic effects were significantly attenuated in CTRP1(-/-)/apoE(-/-) double-knockout mice compared with apoE(-/-) mice, with a consistent decrease in vascular adhesion molecule, phospho-p38 and TNF-α expression and macrophage infiltration in plaque in CTRP1(-/-) and double-knockout mice. Tumour necrosis factor-α-induced expression of adhesion molecules and cytokines were lower in primary endothelial cells and macrophages from CTRP(-/-) mice than in those from C57BL/6 mice.CONCLUSION:C1q/TNF-related protein-1 is a marker of atherosclerosis in humans and promotes atherogenesis in mice.

Abstract:Background. Atherosclerosis is an inflammatory cardiovascular disease. However, whether the association of immune cells in plaques promotes the progression of this disease has not yet been completely elucidated. Materials and Methods. Thus, this study aimed to investigate the relationship between C1q+ macrophages and CD8T cells through scRNA‐seq data reanalysis, quantitative real‐time PCR, and flow cytometry. Chromatin immunoprecipitation‐quantitative polymerase chain reaction, western blot, and antibody‐blocking experiments were performed to investigate the role of macrophage–CD8T interaction in atherosclerosis. An atherosclerotic mouse model was developed to confirm our findings. Results. Mechanistically, Spi1 expression induced by granulocyte–macrophage colony‐stimulating factor promoted C1q expression in the macrophages. Moreover, C1q+ macrophages suppressed CD8T cell apoptosis by upregulating Slc7a7 expression to enhance the L‐arginine uptake of CD8T cells. CD8T‐derived interferon‐γ promoted macrophage activation to induce atherosclerosis. Blockade of the C1q–C1qbp axis attenuated atherosclerosis. Conclusion. In conclusion, macrophages interacting with CD8T promote atherosclerosis development via the C1q–C1qbp axis.

Abstract:Aim To explore the effects and mechanism of ApoAⅠ on TNF-α expression in macrophages and plasma TNF-α levels from atherosclerotic mice.Methods Male ApoE-/-mice were fed with high cholesterol diet for 12 weeks and then randomly divided into control group,ApoAⅠ group and ApoAⅠ+AG490(a specific JAK2 inhibitor)group;each group were separately received treatment with PBS,ApoAⅠ (40 μg/g),ApoAⅠ (40μ,g/g) +AG490(4 μg/g) on the third and the first day before sacrifice,12 h before sacrifice all groups were administered with LPS via intraperitoneal injection,TNF-α levels in plasma were measured by ELISA.THP-1 macrophage-derived foam cells were randomly divided into control group,ApoAⅠ(10 mg/L) group and ApoAI(10 mg/L)+AG490(25 μmol/L) group;all groups incubated with LPS (10 μg/L).TNF-α level in supernate were measured by ELISA and TNF-α mRNA expression were examined by RT-PCR.Results Compared with the control group,ApoAⅠ can significandy decrease TNF-α levels in mice plasma,after administered with AG490 ApoAⅠ-inhibition of TNF-α secretion induced by LPS markedly weakened(P＜0.05).In vitro study demonstrated ApoAⅠ can inhibit TNF-α transcription and expression induced by LPS (P＜0.01),AG490 abrogated the anti-inflammatory effect of ApoAⅠ(P＜0.05).Conclusion ApoA Ⅰ inhibited TNF-α transcription and expression through JAK2/STAT3 signaling pathway.

Abstract:Cardiovascular diseases, the notorious killer, are mainly caused by atherosclerosis (AS) characterized by lipids, cholesterol, and iron overload in plaques. Macrophages are effector cells and accumulate to the damaged and inflamed sites of arteries to internalize native and chemically modified lipoproteins to transform them into cholesterol-loaded foam cells. Foam cell formation is determined by the capacity of phagocytosis, migration, scavenging, and the features of phenotypes. Macrophages are diverse, and the subsets and functions are controlled by their surrounding microenvironment. Generally, macrophages are divided into classically activated (M1) and alternatively activated (M2). Recently, intraplaque macrophage phenotypes are recognized by the stimulation of CXCL4 (M4), oxidized phospholipids (Mox), hemoglobin/haptoglobin complexes [HA-mac/M(Hb)], and heme (Mhem). The pro-atherogenic or anti-atherosclerotic phenotypes of macrophages decide the progression of AS. Besides, apoptosis, necrosis, ferroptosis, autophagy and pyrotopsis determine plaque formation and cardiovascular vulnerability, which may be associated with macrophage polarization phenotypes. In this review, we first summarize the three most popular hypotheses for AS and find the common key factors for further discussion. Secondly, we discuss the factors affecting macrophage polarization and five types of macrophage death in AS progression, especially ferroptosis. A comprehensive understanding of the cellular and molecular mechanisms of plaque formation is conducive to disentangling the candidate targets of macrophage-targeting therapies for clinical intervention at various stages of AS.

[Modulation of Inflammatory Cytokines by ATP-binding Cassette A1 in THP-1 Macrophages].

Abstract:OBJECTIVE:To observe the effects of ATP-binding cassette A1 (ABCA1) on intercellular cell adhesion molecule type 1 (ICAM-1), monocyte chemoattractant protein-1 (MCP-1) and interleukin-1beta (IL-1beta) in THP-1 macrophages stimulated with 8-Br-cAMP to identify a possible new mechanism that ABCA1 contributes to atherosclerogenesis (AS).METHODS:Monocytic THP-1 cells were cultured in the presence of 100 nmol/L phorbol myristate acetate (PMA) for 72 h to transform the cells into THP-1 macrophages. After the macrophages were stimulated with 8-Br-cAMP (final concentration 0.5 mmol/L) for 3, 6, 12 and 24 h respectively, the amounts of ABCA1, ICAM-1 and MCP-1 mRNA were examined by real-time fluorescent quantitative RT-PCR, and the protein amounts of ABCA1, ICAM-1, MCP-1 and IL-1beta were determined by Western blotting and enzyme-linked immunosorbent assay (ELISA). Phosphorothioate antisense oligonucleotides of ABCA1 were add into the culture media at a final concentration of 100 nmol/L and the experiments were repeated.RESULTS:ABCA1, ICAM-1 and MCP-1 mRNA and protein and IL-1beta protein were increased in the macrophages after stimulation with 8-Br-cAMP for 6 and 12 h. The mRNA expressions of ABCA1, ICAM-1 and MCP-1 were decreased significantly at 3 and 6 h (P<0.01), and the protein expressions of ABCA1, ICAM-1, MCP-1 and IL-1beta declined significantly at 12 and 24 h (P<0.01) after transfection of the macrophages with antisense oligonucleotides of ABCA1.CONCLUSION:ABCA1 can increase the expressions of the inflammatory cytokines in THP-1 macrophages stimulated by 8-Br-cAMP and plays a role in the pathogenesis of AS.

Abstract:Atherosclerosis (AS) is the main pathological basis of cardiovascular diseases, which is mainly characterized by lipid deposition and inflammatory response. Macrophages (MΦ), as the key mediators of the inflammatory response, run through all stages of the occurrence and development of AS, from plaque initiation to the transition to vulnerable plaques, and are regarded as important therapeutic targets. It was previously thought that the atherogenic mechanism of MΦ was mainly due to their phagocytosis of lipids, resulting in excessive foam cells aggregated on the arterial wall. However, increasing evidence has revealed the diversity of AS mechanisms caused by MΦ. For example, MΦ present a continuum phenotypic spectrum, and their polarization has been demonstrated to play a vital role in the regulation of AS-related inflammatory response. MΦ apoptosis and the ability of MΦ to clean up apoptotic cells (also known as efferocytosis) are crucial determinants of AS lesion progression and plaque stability. Hence, this review probes into the contradictory regulation of MΦ on AS based on polarization, apoptosis, and efferocytosis, designed to highlight the complex and interrelated regulated network of MΦ promoting AS.
