Antiarrhythmic Effects of Carvedilol on Aconitine-induced Arrhythmia in Rats and Its Mechanisms

Abstract:To investigate the antiarrhythmic effects of carvedilol on aconitine-induced arrhythmia in rats and its mechanisms of the antiarrhythmic effects. So as to provide some evidence of carvedilol in clinical antiarrhythmic use. Rat cardiomyocytes were isolated with collagenase to acquire single ventricular myocytes. Sodium channel currents (I_ Na ) were recorded using whole-cell recording patch clamp technique. The influences of aconitine and carvedilol on I_ Na were studied. Results: The doses of aconitine (μg)to produce arrhythmia in control group were smaller than that in carvedilol group (all P 0.05) .I_ Na current density was 58.63±11.65pA/pF in control group;and was 73.35±12.80pA/pF and 10.19±0.02(pA/pF),respectively,when aconitine and 50μmol carvedilol was added to extracellular bath solution( P 0.05). Current-voltage relationship (I-V) curve was shifted to downward and upward,respectively,in aconitine group and carvedilol group. Conclusions: Carvedilol has antiarrhythmic effects on aconitine-induced arrhythmia in rats. One mechanism of its antiarrhythmic effects is related to inhibition of sodium channel.
