Observation on the Gametes and Sperm Penetration with Seanning Electron Microscope and a Preliminary Study of the Embryonic Development in Pacific Oyster (crassostrea Gigas)

Abstract:The eggs and sperms of the Pacific oyster ( Crassostrea gigas ) were observed during developing stages with a scanning electron microscope. The results indicated that the total length of the sperm was about 21.6 μ m, the diameters of the head and the middle part of the sperm were about 2.6 μ m and 1.1 μ m, respectively. The length of the tail was about 18.2 μ m and the eyelet shaped structure was observed at the end. The middle part of the immature sperm tail was swollen, the diameter of this part was about 0.3 μ m. The egg was pear shaped, its diameter about 41.9 μ m. Vitelline membrane holes and a fold structure formed with microvillies were seen on the egg surface. The evident arborization structure was observed at the egg handle on egg surface when the egg protoplasm drew back. Many cortical alveoli were observed at the part where the yolk membrane was peeled off on the zygote surface after fertilization. At the water temperature 21.2~23.5℃, Polar bodies(PBI) were observed after sperms and eggs were mixed for 17min, and Polar bodies(PBII)were observed after 1h22min. After 21h55min, the embryo developed to early stage D shaped larvae.
