Design and optimization of polymers for gene delivery

Abstract:OBJECTIVE: As the concept of gene therapy expanded, the choice for a safer and more efficient genetic transfer delivery becomes the key to successful gene therapy. In this paper, we summarize the design and optimization of polymers for gene delivery. DATA SOURCES: Using the of "gene therapy, non-viral vectors, cationic polymer/DNA complex", we searched the PUBMED database for relevant articles published in English between January 1990 and December 2005. Meanwhile, the online search of Chinese Journal Full-text Database was undertaken for relevant Chinese articles between January 1990 and December 2005 with the same key words in Chinese. STUDY SELECTION: All articles were selected firstly, and the references of each article were checked in detail. Inclusive criteria: Articles relevant to the design and optimization of polymers for gene delivery. Exclusive criteria: Duplicated articles or Meta analysis. DATA EXTRACTION: Among 60 selected articles, 28 one were included, 32 duplicated or out of date ones were excluded. In the 28 inclusive articles, 26 were relevant to the gene delivery systems, 2 were relevant to problems in gene transfection and optimization of polymers for gene delivery. DATA SYNTHESIS: Firstly, two categories of gene-delivery systems: viral and non-viral gene delivery system, and classification of non-viral gene delivery system were briefly introduced. Secondly, the development of polymers for gene delivery was expounded, and the advantages and disadvantages, the correlation between structures and functions, and their optimization strategies of several recently designed cationic polymers were described. Finally, the cell transfection mechanisms were presented with cationic polymers as the gene delivery vectors and the optimization strategies was discussed with consideration of the mechanisms. CONCLUSION: Cationic polymers, within the non-viral gene delivery system, have great potential for human gene therapy because of their flexibility of design, good biocompatibility and other advantages. However, low genetic transfection efficiency limits their clinical application. With the growing understanding of polymer gene-delivery mechanisms, it is likely that polymer-based gene-delivery systems will become an important tool for human gene therapy.
