Roles of Primary Cilia and Hedgehog Pathway in the Proliferation of Keratinocytes of Psoriasis

Abstract:Objective: To investigate the roles of primary cilia related genes and the hedgehog(Hh) pathway in the proliferation of the keratinocytes(KC),and to find a therapeutic approach specifically targeting to psoriasis.Methods: The model of the excessively proliferating state of psoriasis KC was established through inducing the excessive proliferation of HaCaT cells in vitro by tumor necrosis factor-α(TNF-α). Specific siRNA expressing vector of Ift88 which is necessary for cilia was constructed and transfected into the TNF-α-induced cultural HaCaT cells in vitro.The growth of the cilia on the surface of KC was detected by the scanning electron microscope after the transfection for 24 h.The protein expression levels of the cilia-related genes and Ptch-1 and Gli-1 in the Hh pathway were detected by Western blotting,and cell proliferation was also measured.Results: There were more primary cilia in the TNF-α-induced cul-tural HaCaT cells than in normal HaCaT cells.Incubation with TNF-α increased the protein levels of cilia-related genes,as well as Ptch-1 and Gli-1 in Hh pathway.Inhibition of Ift88 was shown to be effective in reducing not only the proportion of cells with primary cilia,but also the expression of Ptch-1 and Gli1.Meanwhile,the proliferation of HaCaT cells was distinctly inhibited.Conclusion: Inhibition of the conformation of cilia can inhibit the excessive proliferation of KC by blocking the Hh pathway.Primary cilia might be involved in the pathogenesis of psoriasis and enhancing the occurrence and development of which through the activation of the Hh signaling pathway.
