Effects of olfactory ensheathing cells on neurite outgrowth of embryonic spinal cord dorsal horn neurons

Abstract:Objective:To investigate effects of olfactory ensheathing cells(OECs)on neurite outgrouth of embryonic spinal cord dorsal horn neurons in vivo.Method:The fetal spinal cord was obtained from E15d rats and divided into two halves-dorsal and ventral.The dorsal part was made into cell suspension,then(1)co-cultured with OECs,(2)cultured in supernatant fluid of OECs,(3)co-cultured with fibroblasts or(4)cultured without OECs or fibroblasts as a control.After24hours,the cultures were prepared for anti-neurofilement-200immunostaining as well as image analysis to get the average lengths of all neurites and the longest neurite of one neuron in the4culture groups.Result:In OEC co-culture group,the average length of all neurites was408.62±126.91μm and the length of the longest neurite was180.13±83.54μm,which was longer than that of fibroblast co-culture group(199.37±76.14μm and107.29±43.35μm respectively)and control group(64.61±24.49μm and23.04±5.30μm respectively)significantly(P0.01),but there was no difference between OEC co-culture group and OEC supernatant group(336.24±106.34μm and141.22±53.68μm respectively).Conclusion:When co-cultured,OECs could accelerate neurite outgrouth of embryonic spinal cord dorsal horn neurons obviously,and it's likely that the neurotrophins produced by OECs take the role.
