IL-18 Induces Early Activation of NK Cells and Tumor-Specific CTLs in Vitro

Abstract:Objective Applying rhIL-18 in an in vitro cell co-culture(CC) system of human peripheral mononuclear cell(hPBMC) preparations and tumor cells to observe the bioactivities of IL-18 for inducing cell-mediated cytotoxicities. Methods hPBMCs were separated from human peripheral blood by density centrifugation. The cell subsets of DCs, NK, and T cells were further fractionated by negative screening with the StemSep TM immunomagnetic beads. Phenotypes of the sorted cells were analyzed by FCM using FITC-labelled CD moAbs. The effluent cells were reconsoluted and co-cultured with mitomycin C-inactivated tumor cells for 24-96 h. Isotope release assay was used to examine cell-mediated cytotoxicities of the cultured effector cells. The expression and amount of IFN-γ produced by the activated cells was measured with RT-PCR and ELISA. Results It was found that rhIL-18 rapidly activated the hPBMC preparations to kill various tumor target cells within 24 h induction in the cell co-culture system. The early phase activation of cytolytic effector cells was not inhibited by addition of moAbs against IFN-γ or MHC molecules into the cell culture. Depletion of NK cells but neither T cells nor DCs by negative screening abrogated subsequent target cell cytolysis. When the effector cell preparations were continually stimulated with rhIL-18 for 96 h in the same culture condition, the cytotoxicity mediated by CTLs was induced, which was characterized by tumor cell specificity for the cytotoxicity and blockage by moAbs against both MHCs and T cell surface markers. Conclusion IL-18 can induce activation of NK cells and CTLs consecutively under the stimulation of tumor antigen. This may provide a pathway to induce cytotoxic cells for cancer immunotherapy by adoptive transfer.
