Identification of 25 000 Protein Decreased in Skeletal Muscle of Patients with Myasthenia Gravis

Abstract:Aim:To identify the molecular structure of 25 000 protein decreased in skeletal muscles of patients with myasthenia gravis (MG).Methods:25 000 protein was separated and purified from skeletal muscle by two-dimensional electrophoresis, and its precise molecular mass and N-terminal partial amino acid sequence were measured by the methods of electrospray ionization mass spectrometry (ESIMS) and Edman degradation, respectively. The peptide mass fingerprinting (PMF) of 25 000protein was analyzed by matrix assisted laser desorption ionization time-of flight mass spectrometry (MALDI-TOF-MS). The possibility of 25 000protein for carbonic anhydrase (CAⅢ) was demonstrated by immuno-Western blot.Results:①The ESIMS showed that the precise molecular mass of 25 000protein was 29 632.05; ② Analysis of Edman degradation showed that the N-terminal of 25 000protein may be acylation so that the sequence determination was blocked. ③ All of the results from analysis of PMF and the data of protein bank and 25 000protein characteristics showed there were quite a few similarities in the molecular chemical and physical properties and biological characteristics between 25 000protein and CAⅢ. ④ The immuno-Western blot showed that these bands for immuno-responsible to the antibodies were much similar. Conclusion:The 25 000protein decreased in skeletal muscle of patients with myasthenia gravis was identified as a known protein “carbonic anhydrase Ⅲ”, which has not been reported in the studies of MG. It is necessary to further clarify that the deficiency of CAⅢ in MG skeletal muscle was associated with development of MG muscular weakness and/or pathogeneses MG.
