Studies on Synthesis and Antibacterial Performance of Carboxymethylated Larch Tannin

Abstract:Conditions for synthesis of carboxymethyl larch tannin (CLT) such as reaction temperature, dispersion time and concentration of alkaline reagent were investigated. The optimum synthesis conditions were as follows: molar ratio of chloroacetic acid to hydroxyls of tannin 1.8, dispersion time 2 h, alkalization temperature 60 ℃, ratio of alkaline reagent to hydroxyls in tannin 1.4, concentration of alkaline reagent 0.2 g/mL, alkalization time 30 min, temperature 78 ℃, etherification time 4 h. FT-IR and elemental analysis (EA) were applied to analyse structural changes of larch tannin. EA showed that after carboxymethylation contents of C and H decreased, while an obvious increase of O content was observed. Characteristic functional groups observed in FT-IR spectra approved the carboxymethylation of tannin. Antibacterial experiment indicated that carboxy-methylated larch tannin showed obvious inhibition effects against Bacillus cereus, Escherichia coli and Staphylococcus aureus, with MIC of 62.5 mg/L and MBC of 125 mg/L against B. cereus respectively, which are 1/4-1/2 of those against E. coli and S. aureus respectively, showing higher antibacterial rates than those of larch tannin.
