Abstract:Objective:To establish an HPLC method for the simultaneous determination of albiflorin,paeoniflorin,naringin and glycyrrhizic acid in Qizhiweitong granules.Method:The chromatography was performed on a Zorbax SB-C18 column(250 mm×4.6 mm,5 μm)with a mobile phase of water(containing 0.2% acetic acid)-acetonitrile(containing 0.2% acetic acid)and linear gradient elution.The flow rate was 1.0 mL·min-1.The column temperature was 30 ℃.The detection wavelength was 230 nm from 0 to 15 min,284 nm from 15 to 20 min and 254 nm from 20 to 35 min.Results:The calibration curves of albiflorin,paeoniflorin,naringin and glycyrrhizic acid were linear in the ranges of 15.1-226 μg·mL-1(r=0.9998),25.6-383 μg·mL-1(r=0.9998),27.7-277 μg·mL-1(r=0.9998)and 9.43-47.2 μg·mL-1(r=0.9998)respectively.The mean recoveries of low,middle and high concentrations of the four components were 97.4%-101%(RSD=0.8%-1.7%),96.8%-99.7%(RSD=1.4%-2.6%),97.3%-100%(RSD=1.9%-2.4%).Conclusion:The method is simple and accurate,which can be adopted in the quality control of Qizhiweitong granules.
Simultaneous Determination of Five Compounds in Qingreling Granules by HPLC-UV

Abstract:OBJECTIVE To establish a HPLC-UV method for the simultaneous determination of five compounds including baica-lin, phillyrin, wogonoside, baicalein and wogonin in Qingreling granules. METHODS The separation was performed on an Agilent Zorbax SB-C_(18) column (4.6mm×250 mm, 5 μm) with gradient elution. The mobile phases consisted of 0.05% formic acid-aeetoni-till and 0.05% formic acid-water at a flow rate of 1.0 mL·min~(-1) The detective wavelength was 274 nm. RESULTS 5 compounds showed good linearity (r~2≥0.999 7) in the ranges of tested concentration. The recoveries(RSD≤3.77%), reproducibility(RSD≤0.65% ), precision(RSD≤2.78% ) and stability (RSD≤3.19% ) for 5 compounds were satisfactory. CONCLUSION The method is simpple, accurate and reliable, It may be used for the quality control of Qingreling granules.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.

Abstract:OBJECTIVE:To establish a RP-HPLC method for simultaneously determination of swertiamarin, gentiopicroside, sweroside, isoorientin in Gentiana lawrencei from Qinghai province.METHOD:The RP-HPLC method was used. Chromatographic column was the Kromasil C18 column (4.6 mm x 250 mm, 5 microm). The gradient elution solvent system was composed of acetonitrile (A) and (0.1% H3PO4) water (B). The ratio of acetonitrile was as follows: 0.00-27.00 min, 10% -17% (A); 27.00-45.00 min, 17% -33% (A); 45.01-55.00 min, 100%-100% (A). The detective wavelength was 240 nm; the flow rate was 1 mL x min(-1); column temperature was set at 25 degrees C.RESULT:Swertiamarin, gentiopicroside, sweroside and isoorientin were base-isolated. The method had good linearity within the ranges of 2.12-10.6 mg x mL(-1) for swertiamarin (r = 0.999 4), 2.46-12.3 g x L(-1) for gentiopicroside (r = 0.999 7), 2.47-12.4 g x L(-1) for sweroside (r = 0.999 2) and 0.309-1.54 mg x mL(-1) for isoorientin (r = 0.999 5).CONCLUSION:The method is rapid, precise and repeatable, can be applied to control the quality of G. lawrencei.
Determination of Active Constitutents in Folium Eucommiae by RP-HPLC

Abstract:OBJECTIVE To establish a new reversed phase high-performance liquid chromatography(RP-HPLC) method for the determination of chlorogenic acid and genipoide,the active constitutents in folium eucommiae simultaneously.METHODS YMC-pack ODS-A column(250 mm×4.6 mm,5 μm) was used and the mobile phase was methanol-water-acetic acid(20:80:1.5) at the flow rate of 1.0 mL·min-1.The detection wavelength was 237 nm and the temperature of column was 30 ℃.RESULTS The linear range of chlorogenic acid and genipoide were 0.041 6-0.624 0 μg(r=0.999 2) and 0.081 6-1.224 0 μg(r=0.998 9).The average recoveries were 102.92% and 102.35%,respectively.The RSD were 1.49% and 1.20%.The content of chlorogenic acid and genipoide in folium eucommiae were 2.801 mg·g-1 and 0.273 mg·g-1.CONCLUSION The method is simple,accurate and reliability,with good repeatability,and can be used for the quality control of folium eucommiae.

Abstract:Rutin,quercertin and 1-DNJ(1-deoxynojirimycin) are active medicinal constituents in mulberry leaves.In order to determine these constituents by RP-HPLC(reversed-phase high performance liquid chromatography),rutin and quercertin were extracted with 85% ethanol by ultrasonic-assisted extraction technology and 1-DNJ was extracted with 0.05 mol/L HCl,and derivatized by FMOC-Cl in pH 8.5 borate buffer solution to yield DNJ-FMOC,which can be detected with ultraviolet.Rutin,quercertin and 1-DNJ were separated by Alltima C18 column(4.6 mm×250 mm,5 μm) with THF-0.02% phosphoric acid(25∶75),acetonitile-0.5 mol/L pH 4.0 aqueous acetic acid buffer solution(27∶73) and acetonitile-0.1% aqueous acetic acid(36∶64) as mobile phases.The detecting wavelengths were 360 nm,360 nm and 254 nm respectively.The result indicated that rutin,quercertin and DNJ-FMOC had been separated efficiently.There existed significant positive correlation between the peak area and the concentration.The recovery rates(n=3) were 99.14%,98.89% and 100.85% respectively,and the relative standard deviations were 1.01%,0.33% and 1.24% respectively.Therefore,this determination method is accurate and reliable.

Abstract:Reversed-phase high-performance liquid chromatography (RP-HPLC) with ultraviolet diode array detection (UV-DAD) was used for the simultaneous determination of four flavone C-glucosides, i.e. orientin, homoorientin, vitexin and isovitexin in several food systems fortified by the antioxidant of bamboo leaves (AOB), such as high temperature sterilized milk, sunflower seed oil and extruded rice cake for the first time. The method included extraction of flavone C-glucosides from AOB-fortified foods by methanol aqueous solution, deproteinating with saturated lead acetate solution and potassium oxalate, defatting with n-hexane and clean-up by solid-phase extraction (SPE) with Phenomenex C18 cartridges. Analytes were separated with Luna C18 5μm 250mm×4.6mm column using acetonitrile and 1% (v/v) acetic acid (pH 3.0) as mobile phase. Good results were obtained with respect to repeatability (relative standard deviation (RSD)<2.2%) and recovery (81.4–91.8%) which fulfilled the requirements defined by European Union (EU) legislation. The total amounts of four flavone C-glucosides were 12.56μg/100mL, 881.08μg/100mL and 1420.83μg/100g dry weight in AOB-fortified sterilized milk, sunflower seed oil and extruded rice cake, respectively. The method was successfully applied to the analysis of flavone C-glucosides in AOB-fortified samples. The optimized procedure could also be referenced for the separation of flavone C-glucosides in other fortified foodstuffs.
Simultaneous Determination of Three Flavonoids in Rat Plasma by RP-LC after Oral Administration of the Total Flavonoids of Scutellaria Barbata

Abstract:A liquid chromatographic method for the simultaneous determination of three flavonoids, scutellarin (SCU), isoscutellarein-8-O-glucuronide (ISO) and luteolin (LUT) in rat plasma was developed and validated. Following a single-step liquid–liquid extraction with ethyl acetate, the analytes and internal standard (IS) (rutin) were successfully separated on a Diamonsil C18 column using a mobile phase composed of acetonitrile (A)–0.2% phosphoric acid aqueous solution (B) (0–5 min, 20% A–29% A; 5–25 min, 29% A, v/v) at a flow rate of 1.0 mL min−1. The linear range was 0.044–2.20 μg mL−1 for SCU, 0.042–2.08 μg mL−1 for ISO, and 0.056–2.80 μg mL−1 for LUT, with the correlation coefficients of 0.9995, 0.9989 and 0.9963, respectively. The limit of quantification of SCU, ISO and LUT were 44, 41.6 and 56 ng mL−1, respectively. The accuracy of assay was between 88.4 and 103.0%. The inter-day and intra-day precisions (RSD) were less than 10.5%. The developed method was simple, rapid and applied successfully to study the pharmacokinetics of SCU, ISO and LUT after oral administration of the total flavonoids of Scutellaria barbata.
[Determination of neohesperidin and naringin in Qingfei Paidu Granules by RP-HPLC and their transfer rates in preparation process]

Abstract:The present study established an RP-HPLC method for simultaneous determination of two active components in Qingfei Paidu Granules and investigated the transfer rates of neohesperidin and naringin in the preparation process to provide references for improving the quality control standard and production of Qingfei Paidu Granules.RP-HPLC was performed on a YMC Triart C_(18) column(4.6 mm×150 mm, 5 μm)with column temperature of 30 ℃, acetonitrile(A) and 0.2% phosphoric acid solution(B) as mobile phases for gradient elution at a flow rate of 1.0 mL·min~(-1) and detection wavelength of 284 nm.Good linearity was observed for naringin at 0.10-1.0 μg(R~2=0.999 9) and neohesperidin at 0.12-1.2 μg(R~2=0.999 9).The average recovery of naringin was 99.52% with an RSD of 1.2%, and that of neohesperidin was 100.8% with an RSD of 1.2%.The transfer rates of naringin and neohesperidin between medicinal materials, extracts, concentrates, and granules were measured by this method.The average transfer rate of naringin from medicinal materials to granules was 54.89%±4.38%, and that of neohesperidin was 57.63%±5.88%.The process from medicinal materials to extracts was presumedly the key link affecting the whole preparation process.The established method is simple and sensitive and can be adopted for the quality control of Qingfei Paidu Granules.Meanwhile, it can be used to investigate the transfer rate of neohesperidin and naringin in the preparation of Qingfei Paidu Granules, and further improve the quality control standard of Aurantii Fructus Immaturus in Qingfei Paidu Granules.

Abstract:Objective:To establish a method for simultaneous determination of ginsenoside Rgl,ginsenoside Re,ginsenoside Rbl and notoginsenoside R1 in Renshen Sanqi granules.Method:The four components in Renshen Sanqi granules were determined simultaneously by HPLC.A Sunfire C_(18)(150mm×4.6mm,5μm) analytical column was used with acetonitrile-water as the mobile phase with gradient elution.The detection wavelength was at 203nm,the flow rate was 1.0ml·min~(-1) and the column temperature was at30℃.Result:The relationship between the concentration and the peak area of ginsenoside Rg_1,ginsenoside Re,ginsenoside Rb_1and notoginsenoside R_1 was respectively linear.The average recovery was 99.83%,97.84%,98.43%and 97.34%with RSD of 2.08%,1.66%.1.73%and 1.42%(n=5),respectively.Conclusion:The method can simultaneously determine the four components in Renshen Sanqi granules.

Abstract:To develop a HPLC method for simultaneous determination of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid in Spica Prunellae.After ultrasonic extraction with 75% ethanol solution containing 1% formic acid, the ethanol-extract of Spica Prunellae was analyzed on an Elite SinoChrom ODS-AP column using gradient elution of 0.01% phosphoric acid (A) and acetonitrile (B) at a flow rate of 0.9-1.0 mL x min(-1). A wavelength switch program was used for detection at 330 nm (0-33 min) and 203 nm (33-40 min). The column temperature was set at 20 degrees C and injection volume was 50 microL.The calibration curves of all analytes were linear. The average recoveries were 93.7%-105.2% with RSDs not more than 4.5%. The contents of caffeic acid, rosmarinic acid, ursolic acid and oleanolic acid in Spica Prunellae were 0.0401-0.0968, 0.99-2.57, 0.243-0.556, 4.06-8.13 mg x g(-1), respectively.The described method is sensitive, convenient and accurate, and is suitable for the simultaneous determination of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid in Spica Prunellae.

Abstract:Objective To establish a method to control the quality of Xiao'er Changweikang Granules.Methods Herba Kummerowiae Striatae,Radix Paeoniae Rubra,Radix et Rhizoma Glycyrrhizae were indentified by TLC.Paeoniflorin and glycyrrhizic acid were determined by HPLC.The chromatographic separation was performed on the Kromasil C18 column(250 mm×4.6 mm,5 μm) by gradient elution with 0.1%phosphoric acid-acetonitrile at the flow rate of 1.0 mL/min,the detection wavelength was set at 230 nm and 250 nm.Results The developed TLC spots were fairly clear and the blank test showed no interference.The linear range of paeoniflorin was 2.9-174 μg/mL(r2=1.000 0),the average recovery rate was 99.25%,RSD=1.55%(n=6).The linear range of glycyrrhizic acid was 1.3-79 μg/mL(r2=0.999 9),the average recovery rate was 98.38%,RSD=1.27%(n=6).Conclusion The method is effective for the quality control of Xiao'er Changweikang Granules.

Abstract:Fructus gardeniae and Radix scutellariae were identified by thin layer chromatography,and the content of chlorogenic acid was determined by HPLC.The column was Symmetry~ C_(18)(4.6×250 mm,5 μm),mobile phase was acetonitrile-0.4% phosphoric acid water solution(8∶92),and the detective wavelength was 324.5 nm.The method of TLC identification was specific and repeatable.The linearity range for chlorogenic acid was from 0.161 6 mg to 1.292 8 mg.The average recovery of chlorogenic acid was 99.19% and RSD was 1.26%(n=6).The method is simple and feasible,highly accurate and precise.It can be used for the quality control of Gandujing granules.

Abstract:Objective:To establish a reversed phase high performance liquid chromatography with ultro violet detector (RP-HPLC-UV) method for the simultaneous determination of three compounds in A.senticocus leaves.Methods:The Diamonsil C18 column (250 mm × 4.6 mm,5μm) was used for separated and analysis using gradient elution with a mobile phase of acetonitrile as solvent A and 0.3％ (v/v) phosphoric acid in water as solvent B,the flow rate was 1.0 mL·min-1,detection wavelength was 360 nm and the column temperature was maintained at 30 ℃.Results:The three compounds (rutin,hyperin,quercetin) were completely separated within 40 min.The regression equation showed good liner relationship between logarithm of the peak area and logarithm of the content of each compound (r =0.9995-0.9999).the average recoveries was 96.7％～99.1％ and RSD was 2.3％～3.3％.Conclusion:The method is simple,fast and reliable,which can be used to determine the contents of three compounds in A.senticocus leaves.

Abstract:OBJECTIVE To establish a method to determine naringin in Dingxiang Jiangqi granule.METHODS An HPLC method was established with Kromasil C18 column.The mobile phase was consisted of acetonitrile-water(21.1:78.9,pH was adjusted to 3 with phosphoric acid)with the flow rate of 1.0 ml·min-1 and the detective wavelength was 283 nm.The column temperature was 30℃.RESULTS Within the range of 2.68-100.50 μg·ml-1,there was a good linear relationship between the concentration and peak area of naringin(r=0.9999).The average recovery was 99.5% with RSD of 2.41%.CONCLUSION The method is accurate,reliable,reproducible and suitable for the determination of naringinin in Dingxiang Jiangqi granules.

Abstract:AIM To develop an HPLC method for simultaneously determining homoprotocatechuic acid,protocatechuic acid,p-hydroxy benzeneacetic acid,chlorogenic acid,caffic acid and hyperoside in Fanhuncao Granule(Senecio cannabifolius Less or Senecio cannabifolius var.integrilifolius).METHODS Agilent Zorbax SB-C18 column(4.6 mm ×250 mm,5 μm) was used with acetonitrile-1% trifluoroacetic acid as the mobile phase in gradient elution.The flow rate was 1 mL/min.The column temperature was maintained at 30 ℃,and the detection wavelength was set at 280 nm.RESULT Six active components were in baseline separation.Homoprotocatechuic acid was linear in the range from 1.14 to 114 μg/mL(r=0.999 9) and the average recovery was 100.4%(RSD=1.3%).Protocatechuic acid was linear in the range from 0.74 to 186 μg/mL(r=0.999 2) and the average recovery was 99.4%(RSD=2.2%).p-Hydroxy benzeneacetic acid was linear in the range from 5.26 to 526 μg/mL(r=0.999 8) and the average recovery was 98.4%(RSD=2.2%).Chlorogenic acid was linear in the range from 2.88 to 288 μg/mL(r=0.999 8) and the average recovery was 99.5%(RSD=3.6%).Caffic acid was linear in the range from 1.50 to 148 μg/mL(r=0.999 9) and the average recovery was 96.2%(RSD=1.8%).Hyperoside was linear in the range from 1.42 to 205 μg/mL(r=0.999 9) and the average recovery was 97.6%(RSD=3.4%).CONCLUSION The method is accurate,simple and feasible.It can be used as a quality evaluation for Fanhuncao Granules.

Abstract:Objective:To establish HPLC method for determination of Gastrodin in Shenan granules .Methods:HPLC was performed on Kromasil C18 column,the mobile phase consisted of acetonitrile -0.05% phosphoric acid solution (0.5∶99.5),the detection wave-length is 220 nm, flow rate 1.0mL /min, column temperature room temperature , sample size 10μL.Results:Gastrodin is linear with the peak area in the range of0.0518~0.5180μg (r=0.9993), the average recovery was 98.05%(RSD=2.92%).Conclusion:The established method is simple , shortcut, well specificity, reproducible, thus can be used for the quality control of Glycoside constituent quality controlling of Shenan granules .

Abstract:OBJECTIVE:To develop a method for the determination of salvianolic acid B and paeoniflorin in Shuangdan granules.METHOD:The chromatographic separation was performed on a Zorbax SB C18 column with a linear gradient elution of phosphoric acid-water (0.05:100) and phosphoric acid-acetonitrile-methanol (0.05:4:96) was used. The flow rate was 1 mL x min(-1) and column temperature was set at 25 degrees C. The UV detection wavelength was set at 230 nm.RESULT:The linear ranger was of 0.710-22.7 microg for salvianolic acid B and 0.103-3.30 microg for paeoniflorin. Three batches of Shuangdan granules were analyzed, and the content of salvianolic acid B ranged from 6.46-11.6 mg x g(-1) and paeoniflorin ranged from 1.44-1.78 mg x g(-1).CONCLUSION:The method was accurate, reproducible, reliable, and could be used for quantitative control of Shuangdan granules.

Abstract:A simple, rapid, and sensitive liquid chromatographic method has been established for the simultaneous analysis of three compounds (narirutin, hesperidin and naringin), in granule decoctions of Fructus Aurantii-type formulae. The compounds were separated in less than 10 min using a C18 column with gradient elution using (A) acetonitrile, (B) water, and (C) acetic acid at a flow rate of 0.3 mL/min, and with a PDA detector. The method was validated for specificity, accuracy, precision, and limits of detection. Good linear regression data (r2 > 0.9980) were obtained for all the calibration plots within the ranges tested. The method is an attractive alternative for quality control and clinical monitor of granule decoctions of Fructus Aurantii-type formulae.
