Mechanism of Intrahepatic Cholestasis after Obstructive Jaundice Operation

Abstract:Objective To investigate the mechanism of intrahepatic cholestasis after obstructive jaundice operation in rats.Methods The models of bile duct obstruction and bile flow restoration were constructed.The levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),γ-glutamyl transferase(GGT),indirect bilirubin(IBIL),and total bilirubin(TBIL) were measured.Malondialdehyde(MDA) content in hepatic tissues was measured.Pathological changes in hepatic tissues were observed under the light microscope.Expression levels of UDP-glucuronosyltransferase1A1(UGT1Al) mRNA and UGT1Al in hepatic tissues were detected by RT-PCR and Western blot,respectively.Results After biliary obstruction,the levels of serum ALT,AST,GGT,IBIL,TBIL,and MDA content in hepatic tissues were increased significantly.Pathological morphological changes included swelling of hepatocytes,infiltration of inflammatory cells,proliferation of bile duct epithelial cells and connective tissues,and gradual formation of false lobules.Expression levels of UGT1Al mRNA and UGT1Al in hepatic tissues were decreased significantly.When the obstruction time reached 14 days,the above changes were attenuated gradually after restoration of bile flow.The recovery of the above changes was significantly slow after restoration of bile flow when the obstruction time reached 28 days.Conclusion With the biliary obstruction prolonged,the expressions of UGT1Al mRNA and UGT1Al in hepatic tissues are decreased significantly.The expressions of UGT1Al mRNA and UGT1Al slowly recover after restoration of bile flow,which may be one of the major mechanisms of intrahepatic cholestasis after obstructive jaundice operation.
