Automated Submicroliter Fluid Dispensing Technology for Protein Crystallization

Abstract:Recently,high-throughput protein crystallization requires that the volumes of sample solution in crystallization experiments should be reduced to increase the screening chance with a given amount of sample.This led to the developments of automated liquid-handling instruments that can transfer submicroliter volumes of fluid.In this paper,many kinds of submicroliter fluid-dispensing methodologies and the characteristics and applicability have been expounded.And the rules for selecting appropriate submicroliter fluid-dispensing methodologies in protein crystallization was brought forward.In order to ensure dispensing accurately,reproducibility and reliability for liquid handling in the submicroliter range on protein crystallization,a non-contact solenoid based dispensing system was developed and the dispensing process was investigated with computational fluid dynamics(CFD) models.The dispensing process is a function of the fluid properties and the system parameters such as the pressure,nozzle geometry and the details of the actuation motion.Physical arguments show that this functional dependence can be reduced to two dimensionless numbers:the Weber number which represents the ratio of inertia to surface tension forces,and the Reynolds number which represents the ratio of inertia to viscous forces.A parametric study indicates that there is an optimal regime for the dispensing process in which a drop forms and finally the breakup happens.Finally,the CFD model was verified experimentally by comparing the non-contact fluid dispense behaviour with photographs of a dispense process.
