Abstract:AIM: To investigate whether NF-kappa B is activated in human gastric carcinoma tissues and, if so, to study whether there is any correlation between NF-kappa B activity and heparanase expression in gastric carcinoma.METHODS: NF-kappa B activation was assayed by immunohistochemical staining in formalin-fixed, paraffin-embedded specimens from 45 gastric carcinoma patients. Electrophoretic mobility shift assay (EMSA) method was used for nuclear protein from these fresh tissue specimens. Heparanase gene expression was quantified using quantitative RT-PCR.RESULTS: The nuclear translocation of RelA (marker of NF-kappa B activation) was significantly higher in tumor cells compared to adjacent and normal epithelial cells [(41.3 +/- 3.52)% vs (0.38 +/- 0.22) %, t = 10.993, P = 0.000<0.05; (41.3 +/- 3.52)% vs (0 +/- 0.31)%, t = 11.484, P = 0.000<0.05]. NF-kappa B activation was correlated with tumor invasion-related clinicopathological features such as lymphatic invasion, pathological stage, and depth of invasion (Z = 2.148, P = 0.032<0.05; chi(2) = 8.758, P = 0.033<0.05; chi(2) = 18.531, P = 0.006<0.05). NF-kappa B activation was significantly correlated with expression of heparanase gene (r = 0.194, P = 0.046<0.05).CONCLUSION: NF-kappa B RelA (p65) activation was related with increased heparanase gene expression and correlated with poor clinicopathological characteristics in gastric cancers. This suggests NF-kappa B as a major controller of the metastatic phenotype through its reciprocal regulation of some metastasis-related genes. (C) 2005 The WJG Press and Elsevier Inc. Heparanase Mrna Expression and Point Mutation in Hepatocellular Carcinoma

Abstract:AIM:To explore the expression of heparanase mRNA and point mutation in hepatocellular carcinoma (HCC).METHODS:Reverse transcription polymerase chain reaction was used to measure the expression of heparanase mRNA in the primary tumor tissues and surrounding liver tissues of 33 HCC patients. T-A cloning and sequencing were used to detect whether there was any mutation in the amplified PCR products.RESULTS:The expression of heparanase mRNA was positive in 16 primary tumor tissues of HCC, and the positive rate was 48.5%, which was significantly higher than that in the surrounding liver parenchyma (P<0.01). The positive rate for heparanase gene in high-tendency to metastatic recurrence group (71.4%, 10/14) was obviously higher than that in low-tendency to metastatic recurrence group (31.6%, 6/19) (P = 0.023). The positive rate for heparanase gene in patients with metastatic recurrence during postoperative follow-up (78.6%, 11/14) was also significantly higher than that in those without metastatic recurrence (21.4%, 3/14) (P = 0.003). Sequence analysis of the HPA PCR products was made in 7 patients, and 2-point mutations were found in 4 patients, one of which was sense mutation, neither base insertion nor deletion was detected. The mutation rate was 57.1% (4/7).CONCLUSION:The expression rate of heparanase mRNA increases in HCC, and HPA mRNA may be one of the reliable markers for the metastatic activity gained by the liver tumor cells and could be used clinically in predicting metastatic recurrence of HCC. Point mutation may be one of the causes for enhanced heparanase mRNA expression.

Abstract:Heparanase,a mammalian cell endoglucuronidase,degrades heparan sulfate(HS) at specific intrachain sites.HS is a major constituent of the extracellular matrix(ECM) and basement membranes(BMs),the cleavage of which can affect the integrity and functional state of ECM and BMs.Apart from its direct contribution to ECM degradation,degradation of HS can release and activate HS-binding bioactive molecules such as growth factors,chemotactic factors,and enzymes which can accelerate cell growth and angiogenesis.Thus, heparanase may play important roles in invasion,metastasis and angiogenesis of tumors.

Abstract:Heparanase (HPSE)-directed tumor progression plays a crucial role in mediating tumor-host crosstalk and priming the tumor microenvironment, leading to tumor growth, metastasis and chemo-resistance. HPSE-mediated breakdown of structural heparan sulfate (HS) networks in the extracellular matrix (ECM) and basement membranes (BM) directly facilitates tumor growth and metastasis. Lysosome HPSE also induces multi-drug resistance via enhanced autophagy. Therefore, HPSE inhibitors development has become an attractive topic to block tumor growth and metastasis or eliminate drug resistance. In this review, we summarize HPSE inhibitors applied experimentally and clinically according to interaction with the binding sites of HPSE and participation of growth factors. The antitumor activity and structure-activity relationship (SAR) are also emphasized.

Abstract:Heparanase plays an important role in invasion and metastasis of tumor cells. In this study, we explored the expression and clinicopathological significance of heparanase protein in hepatocellular carcinogenesis to investigate their roles in invasion and the relationship between biological behavior and prognosis of hepatocellular carcinoma (HCC) in tissue microarrays (TMAs). Heparanase expression was examined by immunohistochemistry in TMAs comprising 120 cases of HCC, 48 cases of adjacent tumor liver, 62 cases of cirrhosis, and 23 cases of normal liver tissues. Statistical analyses were determined to access the correlation between heparanase expression and the clinicopathological features of HCC. The results showed a positive level of heparanase in HCC tissues that was significantly higher than that in adjacent tumor liver, cirrhosis, and normal liver tissue. Heparanase was expressed lower in clinical TNM stages I and II than in III and IV. Moreover, the expression of heparanase in cases without metastasis within 20 months was statistically lower than in those with metastasis. Furthermore, heparanase expression in groups of alpha-fetoprotein (AFP) > or = 400 microg/L, portal vein tumor emboli, multiple tumor nodes, and tumor diameter > or = 5 cm were significantly higher than those of corresponding groups, while it was not associated with patients' age, sex, histological classification, cirrhosis, or tumor capsular infiltration. In conclusion, TMA is a powerful tool for the rapid identification of molecular alterations in HCC. The overexpression of heparanase may play an important role in hepatocarcinogenesis, progression, and metastases of HCC. It could serve as a determining factor for clinical prognosis and curative effect.

Abstract:In addition to the cancer cells,there are many other cells in the tumor microenvironment,such as the stromal cells,endothelial cells,CD4+ and CD8+ lymphocytes,NK cells,macrophages and neutrophils,etc.Recent studies have found that tumor-associated neutrophils(TANs) play important roles in the development and progression of tumors.TANs can promote tumor development and metastasis through various mechanisms,such as TANs promoting tumor proliferation and metastasis by secreting elastase,promoting tumor angiogenesis by secreting matrix metalloproteinase,and highly activated TANs exerting anti-tumor activity by killing tumor cells.Further study on TANs casts new lights on immunotherapy of tumors;and using TANs and molecules secreted by TANs as targets may become an important strategy for inhibiting the development and progression of tumors.
Significance of host heparanase in promoting tumor growth and metastasis

Abstract:Heparanase, the sole heparan sulfate degrading endoglycosidase, regulates multiple biological activities that enhance tumor growth, angiogenesis and metastasis. Much of the impact of heparanase on tumor progression is related to its function in mediating tumor-host crosstalk, priming the tumor microenvironment to better support tumor growth and metastasis. We have utilized mice over-expressing (Hpa-tg) heparanase to reveal the role of host heparanase in tumor initiation, growth and metastasis. While in wild type mice tumor development in response to DMBA carcinogenesis was restricted to the mammary gland, Hpa-tg mice developed tumors also in their lungs and liver, associating with reduced survival of the tumor-bearing mice. Consistently, xenograft tumors (lymphoma, melanoma, lung carcinoma, pancreatic carcinoma) transplanted in Hpa-tg mice exhibited accelerated tumor growth and shorter survival of the tumor-bearing mice compared with wild type mice. Hpa-tg mice were also more prone to the development of metastases following intravenous or subcutaneous injection of tumor cells. In some models, the growth advantage was associated with infiltration of heparanase-high host cells into the tumors. However, in other models, heparanase-high host cells were not detected in the primary tumor, implying that the growth advantage in Hpa-tg mice is due to systemic factors. Indeed, we found that plasma from Hpa-tg mice enhanced tumor cell migration and invasion attributed to increased levels of pro-tumorigenic factors (i.e., RANKL, SPARC, MIP-2) in the plasma of Hpa-Tg vs. wild type mice. Furthermore, tumor aggressiveness and short survival time were demonstrated in wild type mice transplanted with bone marrow derived from Hpa-tg but not wild type mice. These results were attributed, among other factors, to upregulation of pro-tumorigenic (i.e., IL35+) and downregulation of anti-tumorigenic (i.e., IFN-γ+) T-cell subpopulations in the spleen, lymph nodes and blood of Hpa-tg vs. wild type mice and their increased infiltration into the primary tumor. Collectively, our results emphasize the significance of host heparanase in mediating the pro-tumorigenic and pro-metastatic interactions between the tumor cells and the host tumor microenvironment, immune cells and systemic factors.

Abstract:OBJECTIVE:To study the expression of heparanase(HPSE)in pancreastic cancer and the correlation between HPSE and angiogenesis and prognosis.METHODS:Immunohistochemistry was used to detect heapranase and microvessel density(MVD)in 47 cases of pancreatic cancer,24 adjacent tissues of cancer and 10 normal tissues,and the correlation of the expression of HPSE protein with the clinicopathological charac teristics were investigated.In addition,CD34 was detected and the association of the HPSE expression with microvessel density(MVD)and extent of proliferation of pancreatic carcinomas was analysised.RESULTS:The rate of the expression of HPSE protein in 47 cases of pancreatic cancer tissues(59.6%,28/47)was significantly higher than those in the adjacent tissues of cancer(20.8%,5/24)and normal tissues(20.0%,2/10),χ2=9.59,3.90;P=0.002 0,0.0487 1.It was shown that HPSE protein expression did not significantly correlate with the size and histological grade of pancreatic cancer,χ2=1.09,2.89,P=0.580 4,0.235 8,but it correlated with the invasion,metastasis and survival time of the patients,χ2=6.52,9.25,6.19,P=0.010 7,0.002 4,0.012 8.The expression of HPSE protein was positively associated with MVD of pancreatic carcinomas,t=5.09,P=0.000 0.CONCLUSIONS:The expression of HPSE protein may play an important role in the growth,invasion,metastasis and angiogenesis of pancreatic cancer.Pancreatic cancer with a high proliferating activity tends to express HPSE.HPSE may be a valuable marker to evaluate biological behavior and prognosis of pancreastic cancer.
[Heparanase and Vascular Endothelial Growth Factor Expression in Non-Small-cell Lung Cancer].

Abstract:OBJECTIVE To study the clinicopathological significance of heparanase and VEGF expression in NSCLC. METHODS Eighty-five lung samples were studied. Each sample was divided into two parts, one used for heparanase mRNA detection by reverse transcription PCR and the other for VEGF detection by immunohistochemistry. RESULTS (1) The expressions of heparanase mRNA and VEGF were higher in NSCLC than in benign pulmonary diseases (P < 0.05). (2) The expression of heparanase mRNA was higher in cases with lymph-node invasion, metastasis, stage III and IV diseases, low-differentiation, and adenocarcinoma, as compared to cases without lymph-node invasion and metastasis, with stage I and II diseases, higher and moderate differentiation, and squamous cell cancer (P < 0.05). Its expression was higher in tumors larger than 5 cm in size (P < 0.05). (3) The expression of VEGF was higher in cases with lymph-node invasion, metastasis, and stage III and IV disease, as compared to cases without lymph-node invasion and metastasis, and with stage I and II diseases (P < 0.05). (4) There was no significant correlation between heparanase and VEGF expression (P > 0.05). CONCLUSIONS Heparanase and VEGF are associated with NSCLC invasion and metastasis, and may be used to evaluate NSCLC metastasis status. Heparanase and VEGF promote NSCLC invasion and metastasis by independent mechanisms. Detection of these two markers may improve the sensitivity and specificity of the measurement of NSCLC metastatic potential.

Abstract:Expression of the heparanase gene is associated with invasive, angiogenic and metastatic potential of diverse malignant tumors and cell lines. Here we used RNA interference strategies to evaluate the role of human heparanase in breast malignancy and to explore the therapeutic potential of its specific targeting. The siRNA targeting human heparanase almost completely inhibited the expression of heparanase in human breast carcinoma MDA-MB-435 cells, whereas the mismatched siRNA showed no effect. Cells transfected with heparanase siRNA expressed significantly less heparanase and profoundly reduced invasion and adhesion in vitro. In MDA-MB-435 cell xenograft model, tumors treated with siRNA were less vascularized and less metastatic than those treated with saline and the mismatched controls. The association of reduced levels of heparanase and altered tumorigenic properties in cells with anti-heparanase siRNA indicates that heparanase is important in cancer progress and has potential use as a target for anticancer drug development.
Heparanase Promotes Human Gastric Cancer Cells Migration and Invasion by Increasing Src and P38 Phosphorylation Expression

Abstract:Gastric cancer is one of the most common cancers and it remains difficult to cure, primarily because most cancer stem like cells possess higher capability of invasion and metastasis. Heparanase acts as a master regulator of the aggressive tumor phenotype in part by enhancing expression of proteins and activating signaling molecules. There were less associated with heparanase of molecular biology mechanism in human gastric cancer. We first evaluated the endogenous expression of heparanase in human gastric cancer cell lines and found Heparanase expression higher in SGC-7901 than MGC-803. Using the technology of RNAi in SGC-7901 cells down regulated heparanase gene, and reduced SGC-7901 cells migration and invasion. On the other hand, recombinant heparanase protein added in MGC-803 cells enhanced MGC-803 cell migration and invasion. The elevated cell migration and invasion were impaired by treatment of Src inhibitor pp2 or p38 inhibitor SB 203580. We further found that Stable knockdown of heparanase in SGC-7901 cells decreased phosphorylation of Src and p38. The phosphorylation of p38 was inhibited in response to pp2 treatment while the addition of SB 203580 to SGC-7901 cells did not change phosphorylation of Src. These data suggest that heparanase facilitates invasion and migration of human gastric cancer cells probably through elevating phosphorylation of Src and p38.
Expression Characteristics of Heparanase in Colon Carcinoma and Its Close Relationship with Cyclooxygenase-2 and Angiogenesis.

Abstract:BACKGROUND/AIMS:Heparanase (HPA) influences tumourigenesis and tumour progression by various mechanisms, including angiogenesis. Cyclooxygenase-2 (COX-2) was strongly correlated with microvessel density, and that COX-2 expression is up-regulated by HPA in esophageal cancer. In this study, we examined the relationship between HPA expression and that of COX-2 in colon carcinoma. The aim of this study was to determine whether the expression of HPA is related to the angiogenesis in colorectal cancer and whether it could be involved in clinical behaviour of colon carcinoma.METHODOLOGY:HPA and COX-2 was analyzed with Immunohistochemistry and Western blot. Microvessels in colon carcinoma were examined by using anti-CD34 antibody. Statistical analysis was applied to test for the prognostic and diagnostic associations.RESULTS:Immunohistochemistry revealed that HPA was expressed at low level in normal colonic mucosa (4/78, 5.1%), but at higher level in tumor tissues (63/78, 80.7%) and closely correlated with tumor lymph node metastasis (p < 0.05). This result was further confirmed by Western blot analysis. Furthermore, carcinomas with high HPA expression demonstrated high COX-2 expression and high MVD (microvesseldensity) labelled with CD34. In addition, mortality was higher in patients with HPA+ phenotype and HPA was an independent predictor of overall survival (p < 0.05).CONCLUSIONS:Our findings indicated that HPA might be an important biomarker for malignant transformation and be involved in promoting colon carcinoma metastasis by increasing angiogenesis.

Abstract:α-Hederin is a monosaccharide pentacyclic triterpene saponin compound derived from the Chinese herb, Pulsatilla. It has garnered considerable attention for its anti-tumor, anti-inflammatory, and spasmolytic pharmacological activities. Given the rising incidence of cancer and the pronounced adverse reactions associated with chemotherapy drugs—which profoundly impact the quality of life for cancer patients—there is an immediate need for safe and effective antitumor agents. Traditional drugs and their anticancer effects have become a focal point of research in recent years. Studies indicate that α-Hederin can hinder tumor cell proliferation and impede the advancement of various cancers, including breast, lung, colorectal, and liver cancers. The principal mechanism behind its anti-tumor activity involves inhibiting tumor cell proliferation, facilitating tumor cell apoptosis, and arresting the cell cycle process. Current evidence suggests that α-Hederin can exert its anti-tumor properties through diverse mechanisms, positioning it as a promising agent in anti-tumor therapy. However, a comprehensive literature search revealed a gap in the comprehensive understanding of α-Hederin. This paper aims to review the available literature on the anti-tumor mechanisms of α-Hederin, hoping to provide valuable insights for the clinical treatment of malignant tumors and the innovation of novel anti-tumor medications.

Abstract:The aim of this study is to explore the effects and specific mechanisms of heparanase on angiogenesis and iron deficiency anemia in TP53 mutant cancer. For this purpose, we conducted in vitro cell experiments and in vivo animal experiments respectively. In this study, we first analyzed the differential expression of heparanase in TP53 wild-type and mutant cells, and analyzed its effects on iron removal and angiogenesis in two types of CALU-1 and NCI-H358 cells. Secondly, we validated whether the mechanism of action of heparanase on TP53 mutant cells for iron removal and angiogenesis is related to VEGF. We applied the iron removal agonist erastin and VEGF inhibitor bevacizumab in both in vitro and in vivo experiments to validate the relationship between heparanase and VEGF in the mechanisms of iron removal and angiogenesis. The experimental results show that heparanase is highly expressed in TP53 mutated cancer cells, and has anti-ferroptosis and pro-angiogenic effects. Our experiment also confirmed that the effect of heparanase on TP53 mutant cancer's iron removal and angiogenesis is related to VEGF. In short, heparanase is highly expressed in p53 mutated lung cancer, and the mechanism of ferroptosis tolerance to TP53 mutated cancer is related to VEGF.

Abstract:BACKGROUND & OBJECTIVE:Heparanase, a kind of endo-D-glucuronidase, degrades heparin sulfate proteoglycans, and plays important roles in invasion and metastasis of many kinds of malignant tumors. This research was designed to investigate the expression of heparanase in esophageal squamous cancer cell line TE-13 and the effect of heparanase antisense oligodeoxynucleotide (ASODN) transfection on invasion of TE-13 cells.METHODS:Synthesized heparanase ASODN was transfected into TE-13 cells. Before and after transfection, the expression of heparanase mRNA and protein in TE-13 cells was detected by reverse transcription-polymerase chain reaction (RT-PCR), Western blot, and immunocytochemistry. Matrigel invasion assay was used to determine the invasive capability of TE-13 cells.RESULTS:In TE-13 cells, heparanase gene (585 bp) was detected by RT-PCR, and heparanase protein (50 ku) by Western blot. Heparanase protein mainly located in cytoplasm and on cell membrane. After heparanase ASODN transfection, heparanase gene and protein expression and the invasive TE-13 cells were significantly reduced along with the ascending concentration of heparanase ASODN (P<0.05, P<0.05).CONCLUSIONS:Heparanase gene is expressed in TE-13 cells. Heparanase ASODN can obviously inhibit heparanase gene expression, and restrain invasive activity of TE-13 cells.

Abstract:Objective To investigate the relationship between the expression of heparanase gene in peripheral blood lymphocytes and lymph nodes metastasis in patients with non-small cell lung cancer.Methods The expression of heparanase gene in peripheral blood lymphocytes from 62 patients with non-small cell lung cancer was detected by reverse transcriptase-polymerase chain reaction(RT-PCR),and 15 healthy subjects served as normal controls.Results The expression of heparanase gene in patients with lymph node metastasis was significantly higher than those without lymph node metastasis(P0.01).There was no significant difference in the expression before and after operation(P0.05).The expression of heparanase in normal control group was negative.The expression of heparanase gene in patients with squamous carcinoma was obviously lower than those with adenocarcinoma(P0.05).Conclusion The expression of heparanase gene in peripheral blood lymphocytes may be related with the metastasis of non-small cell lung cancer,which may be used as a prognostic marker for the metastasis of non-small cell lung cancer.
Expression of Heparanase and Nuclear Factor Kappa B in Pancreatic Adenocarcinoma

Abstract:OBJECTIVE:To detect the expressions of heparanase and nuclear factor kappa B p65 (NF-kappaB p65) in pancreatic adenocarcinomas and analyze their relation to patients' prognosis and the regulatory mechanism of NF-kappaB on heparanase expression.METHODS:Heparanase and NF-kappaB p65 proteins in the tumor and adjacent tissues were detected by immunohistochemistry in 48 patients with pancreatic adenocarcinoma and analyzed for their clinicopathological significance.RESULTS:Heparanase and NF-kappaB p65 proteins were found in 30 (62.5%) and 22 (45.9%) tumor specimens, respectively, a rate significantly higher than that in the adjacent tissues. High heparanase expression was closely related to advanced TNM stage (P=0.031), lymph node metastasis (P=0.003) and decreased 3-year postoperative survival (20.0% vs 0%, P=0.001). NF-kappaB p65 expression was associated with lymph node metastasis (P=0.017) and distant metastasis (P=0.031), but had a higher positive rate in heparanase-positive cases than in heparanase-negative cases (P=0.018). Multivariate analysis showed that neither heparanase nor NF-kappaB p65 was the independent prognostic factors.CONCLUSION:Heparanase is overexpressed in pancreatic adenocarcinomas in association with decreased postoperative survival. NF-kappaB may up-regulate heparanase expression and promote heparanase-dependent tumor invasion and metastasis.

Abstract:Hyaluronan (HA) is a kind of mucopolysaccharide existing in extracellular matrix, playing a critial role in maintaining the extracellular matrix structure and regulating a variety of cellular functions. Recent studies found that the balance of HA changes when tumor occurs, not only the concentration of HA fluctuates, but also the molecular weight of HA alters, resulting in a series of changes of cellular functions. Therefore, it is of great significant to explicit the mechanism of native high molecular weight hyaluronan (nHA) and oligosaccharides of hyaluronan (oHA) invovled in tumor initiation, development, examination and treatment. This article is focused on the relationship between the expression of nHA/oHA and the diagnosis of tumor.
