Abstract:This review highlights the use of bromelain in various applications with up-to-date literature on the purification of bromelain from pineapple fruit and waste such as peel, core, crown, and leaves. Bromelain, a cysteine protease, has been exploited commercially in many applications in the food, beverage, tenderization, cosmetic, pharmaceutical, and textile industries. Researchers worldwide have been directing their interest to purification strategies by applying conventional and modern approaches, such as manipulating the pH, affinity, hydrophobicity, and temperature conditions in accord with the unique properties of bromelain. The amount of downstream processing will depend on its intended application in industries. The breakthrough of recombinant DNA technology has facilitated the large-scale production and purification of recombinant bromelain for novel applications in the future.

Abstract:Food processing industries release a lot of fruit wastes mainly of seed, skin, rind, pomace, crown etc. containing potential bioactive compounds. Bromelain is a crude protein extracted from certain fruits. Bromelain is a sub-type of cysteine protease enzyme extensively investigated for its therapeutic properties such as anti-microbial and anti-oxidant activity with good ferric reducing power. The present study deals with isolating and purifying bromelain from kiwi and avocado fruits and assessing their therapeutic application. Fruit samples were taken from a juice shop in VIT, Vellore, for performing all the experimental analysis. Different parts such as stem, pulp, peel and leaves of the avocado and kiwi fruits were collected for the bromelain extraction process. The extraction of protein was done by the ammonium sulfate precipitation method. Estimation of protein content was performed and the concentration of crude enzyme was found to be 970μg/ml and 900μg/ml for avocado peel and kiwi pulp respectively. Avocado peel showed good activity in ferric reducing power and also a potent radical scavenging activity of 40% at 200μg concentration. Crude bromelain extracted from both fruits showed a broad spectrum of antimicrobial activity against Escherichia coli and Staphylococcus aureus. Hence it can be concluded that bromelain can be used as an effective health supplement.

Abstract:Bromelain is widely used in food industry and pharmaceutical products due to its strong antioxidant properties. Therefore, the extraction of bromelain from pineapple peel may improve the profitability and sustainability of pineapple industry. The aim of this work is to show the purification, stability, and kinetics of bromelain from pineapple peel. By studying the stability of purified bromelain (PB), we found that the activity of PB was inhibited by Fe3+, Al3+, methanol, ethanol, and n-butyl alcohol, while it was increased in the presence of Ca2+, ethylenediamine tetra acetic acid, glucose, D-xylose, maltose, potassium sodium tartrate, sodium citrate, citric acid, and sodium nitrite. These stability tests will expand the application and space acquisition of bromelain. The kinetics study indicated that the thermal inactivation of PB was conforming to the first-order reaction and the half-life (t(1/2)) of PB under different temperature conditions (45, 55, 65, and 75 degrees C) was 81.54, 31.12, 10.28, and 5.23 min, respectively. Therefore, the inactivation time of PB can be predicted at different temperatures for food heating processing.Practical Application The potential of utilizing pineapple peel for bromelain extraction might improve the profitability and sustainability of the pineapple industry.

Abstract:Papain, bromelain, and ficin are commonly used plant proteases used for meat tenderization. Other plant proteases explored for meat tenderization are actinidin, zingibain, and cucumin. The application of plant crude extracts or powders containing higher levels of compounds exerting tenderizing effects is also gaining popularity due to lower cost, improved sensory attributes of meat, and the presence of bioactive compounds exerting additional benefits in addition to tenderization, such as antioxidants and antimicrobial effects. The uncontrolled plant protease action could cause excessive tenderization (mushy texture) and poor quality due to an indiscriminate breakdown of proteins. The higher cost of separation and the purification of enzymes, unstable structure, and poor stability of these enzymes due to autolysis are some major challenges faced by the food industry. The meat industry is targeting the recycling of enzymes and improving their stability and shelf-life by immobilization, encapsulation, protein engineering, medium engineering, and stabilization during tenderization. The present review critically analyzed recent trends and the prospects of the application of plant proteases in meat tenderization.

Abstract:Exogenous enzymes are extraneous enzymes that are not intrinsic to the subject. The exogenous enzyme industry has been rapidly developing recently. Successful application of recombinant DNA amplification, high-efficiency expression, and immobilization technology to genetically engineered bacteria provides a rich source of enzymes. Amylase, cellulase, protease, pectinase, glycosidase, tannase, and polyphenol oxidase are among the most widely used such enzymes. Currently, the application of exogenous enzyme technology in the development of natural plant resources mainly focuses on improving the taste and flavor of the product, enriching the active ingredient contents, deriving and transforming the structure of a chosen compound, and enhancing the biological activity and utilization of the functional ingredient. In this review, we discuss the application status of exogenous enzyme technology for the development of natural plant resources using typical natural active ingredients from plant, such as resveratrol, steviosides, catechins, mogrosides, and ginsenosides, as examples, to provide basis for further exploitation and utilization of exogenous enzyme technology.

Abstract:Bromelain is a mixture of proteolytic enzymes found in pineapple (Ananas comosus) plants. It can be found in several parts of the pineapple plant, including the stem, fruit, leaves and peel. High demand for bromelain has resulted in gradual increases in bromelain production. These increases have led to the need for a bromelain production strategy that yields more purified bromelain at a lower cost and with fewer production steps. Previously, bromelain was purified by conventional centrifugation, ultrafiltration and lyophilisation. Recently, the development of more modern purification techniques such as gel filtration, ion exchange chromatography, affinity chromatography, aqueous two-phase extraction and reverse micelle chromatography has resulted in increased industrial bromelain production worldwide. In addition, recombinant DNA technology has emerged as an alternative strategy for producing large amounts of ultrapure bromelain. An up-to-date compilation of data regarding the commercialisation of bromelain in the clinical, pharmaceutical and industrial fields is provided in this review. © 2016 Society of Chemical Industry.

Abstract:In order to quest new potential plant protease and make it industrially applicable and cost-effective, three novel proteases were detected in tomato (Lycopersicum esculentum Mill.) fruit by sodium dodecyl sulfate (SDS)-gelatin-polyacrylamide gel electrophoresis. All three proteases were isolated and partially purified by heat treatment (70C, 5 min) and ethanol precipitation (0-33%, v/v), with ninefold increase in specific activity and 31.6% recovery. These proteases exhibited the highest specificity for gelatin substrate and were indentified as serine protease. The optimum pH and temperature of proteolytic activity were pH 8-10 and 37-60C, respectively. They were stable at high pH (up to 12) and temperature (up to 70C) and in the presence of 20 mM metal ions (Na+, Ba2+, Ca2+, Mg2+, Mn2+), surfactants (SDS and 80 mM Triton X-100) and denaturants (urea and dithiothreitol). These results suggested that proteases from tomato fruit had potential application in food, detergent, tannery and medical industries.PRACTICAL APPLICATIONSPlant cysteine proteases such as papain, bromelain and ficin are extensively applied in many industries. Recently, several plant serine proteases, which were more stable under demanding conditions than cysteine proteases, were exploited for industrial applications. Tomato is one of the highest produced and consumed vegetables in the worldwide. Alkaline serine proteases from tomato fruit in this paper were stable at high pH and temperature and in the presence of metal ions and oxidation. Enzyme activity against a wide variety of nature protein substrates indicated that they would be investigated for a range of potential commercial applications especially for gelatin and casein hydrolysis in the food processing industry as well as for the dissolution of casein and bovine hemoglobin in detergent industry.

Abstract:Pectin encompass a group of acidic heteropolysaccharides with three structural domains: homogalacturonan(HGA),rhamnogalacturonanI(RG-I) and rhamnogalacturonan II(RG-II),covalently linked to one another.The pectolytic enzymes are of great variety,the definition of which had been extended.The pectolytic enzymes can be used in textile,paper,and cosmetic industries in addition to its uses in food processing.Nowadays,their application in extraction of natural products and bioactive pectic oligosaccharides have jumped to new stages.The immobilization of pectolytic enzyme have been practiced in many ways.This short review highlights the progress on pectin and pectolytic enzymes and their application,and suggests their perspectives in application.

Abstract:The objective of this study was to compare the effect of papain, bromelain, kiwifruit protease, ficin and ginger extract with different concentration (0, 20, 40, 60, 80, 100 U/g) for improving the quality properties of beef rump steaks. The pH, cooking loss, Warner-Bratzler shear force, texture, colour, myofibrillar fragmentation index (MFI), collagen heat solubility, content of soluble protein (CSP), electrophoresis and sensory evaluation are invested. The result showed that all of plant-origin proteases and ginger extract can improve quality characteristics of rump steaks, and ginger extract was found to be the most effective at reducing cooking loss and contributing to the desirable red color and sensory properties, while papain and bromelain were found to be the most effective at decreasing Warner-Bratzler shear force. Ginger extract can be applied in the industrial scale and household level as a convenient and effective method to improve meat qualities. Furthermore, high-quality rump beef steaks at a keen price would be more available for Chinese consumers.

Abstract:In this study, a comparative analysis on the distribution of protease activities among 90 plant resources, including fruits and vegetables, has been performed. Protease activities of plant extracts were assayed at different pH values (pH 3.0, pH 7.5 and pH 10.5) using casein as a substrate. Ten fruits and thirteen vegetables show protease activities above 10U/g. Pineapple, fig and papaya, which are used for commercial protease production, exhibited high protease activities. Additionally, high protease activities were detected in kiwifruit (28.8U/g), broccoli (16.9U/g), ginger (16.6U/g), leek (32.7U/g) and red pepper (15.8U/g) at different pH values. SDS-PAGE and zymograms confirmed that various types of proteases existed in the five plant extracts and might be explored. Furthermore, five plant extracts were treated by different protease inhibitors. These results show that there are still many plant resources unexplored, which may be promising candidates for plant-derived protease production.
Fractionation and activity profiling of fruit bromelain from pineapples of Phuket variety growing in Thailand

Abstract:Bromelain is a type of protease found in both fruits and stems of pineapples. Stem bromelain has been extensively studied and is commercially available for applications in various industries. In contrast, studies of fruit bromelain are quite limited since most of pineapples have been consumed freshly, canned or juiced. Nowadays, the consumption of canned fruits, including canned pineapples has decreased greatly. Fruit bromelain could be a new growth point for pineapple industry. In this study, fruit bromelain was extracted from the pineapple juice of Phuket variety and some of its properties were studied. The enzyme was purified by precipitation using ammonium sulfate fractionation followed by ion-exchange and gel filtration chromatography. Consequently, the protease purification level was increased by 95.2 fold. The final specific activity was getting to 448,590 U/mg on average, dominated by cysteine proteases, with optimal activity at 45°C and pH ranging from 6 to 8. The study facilitates the molecular and application research of fruit bromelain. PRACTICAL APPLICATIONS: The research has been carried out at Funong Food Technology Co., Ltd., Guangdong, China, which produces primarily pineapple chunks and juice. As plenty of by-products, like peels and cores of pineapples, are produced, the techniques are employed to extract bromelain from the by-products. The techniques reported in this work are not new or advanced, however, they are applicable during the manufacturing process and the related equipment is easy to set up and operate. What's more, the practical application of the techniques is cost-effective for the manufactories. Take Funong Food Technology Co., Ltd. as an example, they was using 80% saturation ammonium sulfate to precipitate protein from pineapple juice and obtained a bromelain with activity of approximately 8,000 U/mg and yield of 1.7 kg per ton of juice. With the application of the techniques reported in this work, bromelain was first extracted by ammonium sulfate gradient precipitation, and then purified through ion-exchange and gel filtration chromatography. Each step of precipitation and purification generates a different level of activity and recovery of bromelain, ranging from around 2,506 to 448,590 U/mg, which allows for the production of bromelain according to the requirement of the market and brings more profits.

Abstract:Papain is one of the widely used enzymes in the industry. This study aimed to extract papain enzyme from various parts (i.e. latex, leaves, peels and seeds) of papaya using distilled water and citric phosphate buffer and subsequently purified with a two-step salt precipitation method. The optimum protein concentration of purified papain was obtained with citric phosphate buffer (0.159-0.319 mg/mL) and distilled water (0.119-0.347 mg/ mL). The protein concentration was almost similar for crude extracted with both the solvent. The purification procedure enhanced up to 1.56 fold when extracted with distilled water and 0.83 fold when extracted with citric phosphate buffer. Latex, leaves, peels and seeds showed maximum enzyme activity at 60oC and pH 8.0 as well as better thermal stability at 60oC for 30 mins. Findings highlighted that extracted papain enzyme from waste parts of papaya could be utilized as a potential source of industrial application in terms of waste to value-added products.

Abstract:The concern about the effective use of agro-industrial residues from the food industry has increased. Bromelain is an enzyme mainly obtained from the pineapple plant (A. comosus), including its parts normally considered as waste. It is a high value-added product with a wide variety of applications such as in food, cosmetic and pharmaceutical industries. Many approaches have been used to purify this enzyme, but an efficient method to produce highly active and pure bromelain is still highly desirable. The investigated methods include ion exchange chromatography, precipitation, an aqueous two-phase system as well as affinity and filtration membranes. This paper gives an overview about the advantages and limitations of each one. Chronologic results and advances for these separation processes are also shown. The major tendency is the use of combined processes, such as hybrid or integrated ones.

Abstract:Bromelain is a mixture of proteolytic enzymes that is extracted primarily from pineapples. It is present in almost all the aerial parts of the plant—the peels, leaves, stems and fruit—although only the fruit and stems contain significant amounts. The market-demand for bromelain is quickly increasing, especially in the health sector as it can be used as a drug and/or nutraceutical. Although the complete molecular mechanism has not been fully identified, bromelain possesses several properties, including anti-inflammatory, anti-oedema, antithrombotic and fibrinolytic, mucolytic, anticancer and cicatrizing effects that have been evaluated in several clinical trials. However, one of the main limitations for the clinical use of this supplement is the varying composition of extracts, which leads to heterogeneity in results and, therefore, difficulty in making evidence-based prescriptions. In fact, different geographic locations, land and modes of cultivation, as well as extraction methods, can all give extracts with different activities, depending on the plant part used. Additionally, although novel extraction techniques have been developed to improve bromelain purification and extraction and give higher yields without loss in enzymatic activity, these methods are still expensive and challenging. This review will describe the state of the art in the main conventional and unconventional extraction and purification methods of bromelain and discuss the advantages and limitations of these strategies. Pharmacokinetic and pharmacodynamic profiles and the clinical applications that have arisen from randomized controlled clinical trials are also discussed. Finally, future perspectives for bromelain extracts will be presented.

Abstract:The shortage of oilseeds resource has existed in China,and the development of cost-effective,environmental friendly,and safe processing technology of edible vegetable oils is an effective way to resolve the current situation. The enzyme technology can not only meet the needs of the development of the oil industry,but also can effectively recover the other nutritional components in the oil during the extraction process. The present review was to introduce the application status of the enzymatic technology in vegetable oil extraction at home and abroad. The key factors of enzymatic extraction including the broken particle size,the type of enzyme,the enzyme concentration,the ratio of material to liquid,the hydrolysis time,the extraction temperature,p H value,etc.,the enzymatic degumming technology and the enzymatic degradation of aflatoxin B1 of edible vegetable oils was also discussed in this review.

Abstract:Bromelain is a concoction of sulfhydryl proteolytic enzymes. Depending upon the site of extraction it can be regarded as either stem bromelain (SBM) (EC 3.4.22.32) or fruit bromelain (FBM) (EC 3.4.22.33). Bromelain remain enzymatic active over a broad spectrumand endure a range of pH (5.5 to 8.0) and temperature (35.5 to 71 ºC). It is one of the extensively investigated proteolytic enzyme owing to its astonishing applications in various industries. This necessitated employing a strategy that result in highest purified bromelain in less steps and lowest cost. Use of modernistic approach such as membrane filtration, reverse micellar systems, aqueous two phase extraction and chromatographic techniques have shown promise in this regard. Besides its industrial applications, bromelain has been widely utilized as a potential phytomedical compound. Some of its reported actions include inhibition of platelet aggregation, anti-edematous, anti-thrombotic, anti-inflammatory, modulation of cytokines and immunity, skin debridement and fibrinolytic activity. It also assist digestion, enhance absorption of other drugs and is a potential postoperatively agent that promote wound healing and reduce postsurgical discomfort and swelling.
