Abstract:There have been more and more studies on the excited-state intramolecular proton transfer (ESIPT) reaction involving sulfur atom in recent years. Based on 3-mercapto-2-(naphthalen-2-yl)-4H-chromen-4-one (TFN1), its four derivatives, 6-amino-3-mercapto-2-(naphthalen-2-yl)-4H-chromen-4-one (ATFN1), 6-dimethylamine-3-mercapto-2-(naphthalen-2-yl)-4H-chromen-4-one (DTFN1), 6-trifluoromethyl-3-mercapto-2-(naphthalen-2-yl)-4H-chromen-4-one (FTFN1) and 6-cyano-3-mercapto-2-(naphthalen-2-yl)-4H-chromen-4-one (CTFN1) were designed. Using density functional theory (DFT) and time-dependent DFT (TD-DFT) methods, the ESIPT processes and photophysical properties of TFN1 derivatives were explored, and the substituent effects on the ESIPT behavior and spectral properties of TFN1 were investigated in detail. The introduction of electron-donating groups (–NH 2 , –N(CH 3 ) 2 ) can strengthen the intramolecular hydrogen bond (IHB), decrease the ESIPT barrier and blue-shift the absorption and fluorescence wavelengths, while the role of electron-withdrawing groups (–CF 3 , –CN) is completely opposite to that of electron-donating groups.

Theoretical Insights into the Effect of Different Numbers of Thiophene Groups on Hydrogen Bond Interaction and Excited-State Intramolecular Proton-Transfer Process for Flavonoid Derivatives

Abstract:In this study, we systematically explored the impact of varying the number of thiophene groups on the hydrogen bond interaction and excited-state intramolecular proton-transfer (ESIPT) processes in flavonoid derivatives (STF, DTF, and TTF) using the density functional theory and time-dependent density functional theory methods. Initially, a thorough analysis of the optimized geometric structures revealed that the intramolecular hydrogen bond in the S1 state is enhanced and gradually weakened as the number of thiophene groups increases. To gain a deeper understanding of the hydrogen bond interaction, topological analysis, interaction region indicator scatter plots, and isosurface plots were employed. These images provide further insights that align with the structural analysis. Additionally, we observed a red-shift in the electronic spectra (absorption and fluorescence spectra), which is primarily attributed to the narrowing of the energy gap between the highest occupied molecular orbital and the lowest unoccupied molecular orbital, as elucidated by the frontier molecular orbitals. Furthermore, a combined analysis between the hole-electron distribution and the transition density matrix heat map shows that electron excitation involves the unidirectional charge-transfer mechanism. In the end, by conducting relaxed potential energy curve scans, we found that an increase in the number of thiophene groups elevates the energy barrier for ESIPT, making it more challenging for the reaction. In summary, our study underscores the vital effect of thiophene-substituted numbers in modulating the ESIPT process, which is able to provide valuable insights for the design and synthesis of desired organic fluorescent probes in the future.
DFT Study on Molecular Structures and ROS Scavenging Mechanisms of Novel Antioxidants from Lespedeza Virgata

Abstract:The molecular structure and radical scavenging activity of three novel antioxidants from Lespedeza Virgata, lespedezavirgatol, lespedezavirgatal, and lespedezacoumestan, have been studied using density functional theory with the B3LYP and BhandHLYP methods. The optimized geometries of neutral, radical cation, radical and anion forms were obtained at the B3LYP/6–31G(d) level, in which it was found that all the most stable conformations contain intramolecular hydrogen bonds. The same results were obtained from the MP2 method. The homolytic O—H bond dissociation enthalpy and the adiabatic ionization potential of neutral and anion forms for the three new antioxidants and adiabatic electron affinity and H-atom affinity for hydroxyl radical, superoxide anion radical, and hydrogen peroxide radical were determined both in gas phase and in aqueous solution using IEF-PCM and CPCM model with UAHF or Bondi cavity. The antioxidant activities and reactive oxygen species scavenging mechanisms were then discussed, and the results obtained from different methods are consistent. Furthermore, the antioxidant activities are consistent with the experimental findings of the compounds under investigation.
Optimization of Extraction of Compound Flavonoids from Chinese Herbal Medicines Based on Quantification Theory and Evaluation of Their Antioxidant Activity

Abstract:Plant-derived flavonoids have been attracting increasing research interest because of their multiple health promoting effects, where numerous investigations were carried out on the optimization of extraction and bioactivities. This study aims to optimize the extraction process of compound flavonoids (CFs) from Chinese herbal medicines and detect their antioxidant activity in vitro. CFs were extracted from the raw materials named "medicine food homology," composed of hawthorn, lotus leaf, tartary buckwheat, cassia seed, Lycium barbarum, and Poria cocos in a mass ratio of 4 : 2 : 2 : 1.5 : 1 : 1. L9 (34) orthogonal design, level effect and engineering average estimation, and quantification theory were utilized to improve the extraction method of CFs, and the predictive model for CFs yield was constructed. The 2,2ʹ-diphenyl-1-picrylhydrazyl (DPPH), 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), hydroxyl radical scavenging rate, and reducing power of CFs were measured. The highest CFs yield was obtained under the following extraction condition: liquid-solid ratio of 35 : 1 mL/g, extraction temperature of 75°C, extraction duration of 75 min, and extraction mode enzyme-assisted extraction. The forecasted yield was 37.62%. The result was accurate and the established prediction equation was reliable (R = 0.95). The antioxidant activity of CFs was significantly positively correlated with the concentration from 0.05 to 0.4 mg/mL. The DPPH, ABTS, hydroxyl radical scavenging abilities, and the reducing power of CFs were 81.82 ± 1.75%, 49.35 ± 0.09%, 89.78 ± 0.66%, and 0.232 ± 0.001 at the concentration of 0.4 mg/mL, respectively. CFs could be exploited as natural antioxidants in pharmaceuticals and functional foods.

Abstract:The development of diagnostic and therapeutic drugs for superoxide ions in organisms is extraordinarily vital in the field of biomedical. Herein, we tuned the electronegativity of the atoms (O, S, Se) to study their effect on the excited state intramolecular proton transfer (ESIPT) and antioxidant activity of myricetin (MYR) by density functional theory (DFT) and time-dependent DFT methods. The study revealed that the reduction of atomic electronegativity effectively diminishes the barrier of ESIPT. The reduced potential barrier allows MYR derivatives to have stronger K* state fluorescence compared with the original MYR. By calculating the fluorescence spectra of MYR and its derivatives, the derivatives of MYR exhibit a more reddish K* state photoluminescence. Moreover, the antioxidant activity of MYR derivatives tends to increase as the electronegativity of atoms decreases. Interestingly, the K state antioxidant activity of the three studied molecules is stronger than its own E state. Furthermore, this phenomenon is largely pronounced in the derivatives of MYR. The frontier molecular orbitals, non-covalent interactions, infrared spectra, electronic spectra, potential energy curves, antioxidant activities have cross-validated our results. Our work will provide support for the design and development of a more outstanding diagnosis and treatment drugs in organisms.

Abstract:Flavonoids widely found in natural foods are characterized by acting as antioxidants compounds. There are close relationship between the antiradical activities and structural properties of flavonoids. In this work, density functional theory (DFT) methods were applied to investigate the influence of the H5⋯OC4 intramolecular hydrogen-bond (IHB) on the antiradical activity of flavonoid based on three prevalently accepted radical scavenging mechanisms: hydrogen atom transfer (HAT), single electron transfer-proton transfer (SET-PT) and sequential proton-loss electron-transfer (SPLET). The thermodynamic properties: bond dissociation enthalpy (BDE), ionization potential (IP), proton dissociation enthalpy (PDE), proton affinity (PA) and electron transfer enthalpy (ETE) related with these mechanisms were calculated to elucidate the antiradical activity. The results showed that the 5−OH group is most influenced and its antiradical capacity was weakened by the H5⋯OC4 IHB. In the gas, benzene and chloroform phases, H5⋯OC4 IHB would reduce the antiradical activity of flavonoid via increasing the bond dissociation enthalpy. While, in the DMSO and H2O phases, the opposite result occurs by lowering the proton affinity.
A Dft Study on Reaction of Eupatilin with Hydroxyl Radical in Solution

Abstract:Antioxidants scavenge reactive oxygen species and, therefore, are vitally important in the living cells. The antioxidant properties of eupatilin have recently been reported. In this article, the reactions of eupatilin with the hydroxyl radical (OH center dot) in solution are studied using density functional theory calculations and the polarizable continuum model. Three mechanisms are considered including: sequential electron proton transfer (SEPT), sequential proton loss electron transfer (SPLET), and hydrogen abstraction (HA). Three solvents with different polarities, that is, benzene, methanol, and water, are used to investigate the effect of the environment on the mechanisms. The relative Gibbs free energies and enthalpies corresponding to different mechanisms are calculated. Our results show that SEPT is thermodynamically favored in aqueous solution. Once the eupatilin anion is produced, the second step in SPLET mechanism is thermodynamically favored in methanol and water. The HA mechanism is thermodynamically favored in gas, benzene, methanol, and water. This mechanism is more energetically favorable to occur in a more polar solvent. The natural bond orbital charges and spin densities as well as the singly occupied molecular orbital are then analyzed. It is concluded that the HA process is governed by proton coupled electron transfer mechanism. The attack of the radical takes place preferentially at position 7 of eupatilin. Computational Investigation about the Effects of Solvent Polarity and Chalcogen Element Electronegativity on ESIPT Behaviors for the Et2N-Substituted Flavonoid

Abstract:Inspired by the outstanding nature of flavonoid derivatives in the fields of chemistry and medicine, in this work we mainly focus on exploring the photo-induced properties of the novel Et2N-substituted flavonoid (ENF) fluorophore theoretically. Considering the potential photo-induced properties in different solvents and the chalcogen atomic electronegativity-associated photoexcitation, by time-dependent density functional theory (TDDFT) methods we primarily explore the intramolecular hydrogen bonding interactions and photo-induced charge redistribution behaviors. Via comparing geometrical data and the infrared (IR) spectral shifts-associated hydroxy moiety of ENF, we confirm that the intramolecular hydrogen bond O-H···O should be enhanced with facilitating an excited-state intramolecular proton-transfer (ESIPT) reaction. Particularly, the charge reorganization around hydrogen bonding moieties further reveals the tendency of ESIPT behavior. Combined with the construction of the potential energy surface and the search for reaction transition states, we finally confirmed the solvent-polarity-regulated behaviors as well as the chalcogen elements' electronegativity-dependent ESIPT mechanisms for the ENF fluorophore. We sincerely wish our work could accelerate the further development and applications of flavonoid derivatives.

Abstract:Modulating the excited-state intramolecular proton transfer (ESIPT) reaction to achieve anticipant performance is always fascinating for chemists. However, feasible methods and a definite mechanism for tuning the ESIPT reaction remain insufficient. In this work, we reported the feasibility of continuously modulating the ESIPT dynamics in 2-(2'-hydroxyphenyl)oxazole (HPO) derivatives with different substitutions on the positions 5 and 5' of the core HPO through steady-state/transient spectroscopy and theoretical calculations. We found that the main factor affecting the tendency of the ESIPT reaction is the variation of electron population on proton donor and acceptor. An index Δpdif was proposed to evaluate the overall promotion effect on proton transfer caused by the variation of electron population on proton donor and acceptor, which shows high reliability in interpreting the ESIPT tendency. This method, for its capacity to quickly estimate the tendency of ESIPT, shows great potential in ESIPT molecular design with chemical substitution of electron-donating/withdrawing moieties.

Exploring the excited state proton transfer mechanisms of flavonoid probe in three representative solvents

Abstract:The role of intermolecular hydrogen bond is often ignored in the study of the solvent effect on the excited state proton transfer (ESPT) processes. The ESPT mechanisms of 2-(4-(dimethylamino)phenyl)-3‐hydroxy-6,7-dimethoxy-4H-chromen-4-one (HOF) in three representative solvents, which were toluene, dimethyl sulfoxide (DMSO) and ethanol (EtOH), were investigated through time-dependent density functional theory (TD-DFT) method, and the effect of intermolecular hydrogen bond on the ESPT was fully considered in this paper. The results show that the enhancement of intra- and inter-molecular hydrogen bonds is the key factor of proton transfer process. The change of electron density distribution is also more conducive to the ESPT processes. Due to the existence of intermolecular hydrogen bond in HOF-DMSO complex, the energy barrier of ESPT in DMSO is higher than that in toluene. In EtOH solvent, the excited state double proton transfer (ESDPT) reaction can occur between HOF and EtOH along two different reaction channels with the assistance of dual intermolecular hydrogen bonds. Although the ESPT modes in DMSO and EtOH solvents are different, the energy barriers are basically the same. The energy barriers of ESPT processes in two polar solvents increase obviously due to intermolecular hydrogen bond. Moreover, in polar solvents, a relatively large red-shift was observed in the fluorescence spectra.

Abstract:Understanding the role of substituents is of great importance for the preparation of novel phenolic compounds with enhanced antioxidative properties. In this work, the antioxidative activity of isoflavonoid derivatives with different substituents placed at the C2 position was determined by density functional theory (DFT) calculations. The bond dissociation enthalpy (BDE), ionization potential (IP), and proton affinity (PA) related to hydrogen atom transfer (HAT), single electron transfer-proton transfer (SET-PT), and sequential proton loss electron transfer (SPLET) mechanisms were calculated. The strongest antioxidative group of isoflavonoid is not altered by the substituents. Excellent correlations were found between the BDE/IP/PA and Hammett sigma constants. Equations obtained from linear regression can be useful in the selection of suitable candidates for the synthesis of novel isoflavonoids derivatives with enhanced antioxidative properties. In the gas and benzene phases, the electron-donating substituents would enhance the antioxidative activity of isoflavonoids via weakening the BDE of 4′−OH. In water phase, they will reduce the antioxidative by strengthening the PA of 7−OH. Contrary results occur for the electron-withdrawing groups. In addition, the electronic effects of substituents on the BDE/IP/PA have also been analyzed.
Evaluation of the Antioxidant Effects of Four Main Theaflavin Derivatives Through Chemiluminescence and DNA Damage Analyses.

Abstract:Theaflavins (TFs) are the dimers of a couple of epimerized catechins, which are specially formed during black tea fermentation. To explore the differences among four main TF derivatives (theaflavin (TF(1)), theaflavin-3-gallate (TF(2)A), theaflavin-3'-gallate (TF(2)B), and theaflavin-3,3'-digallate (TF(3))) in scavenging reactive oxygen species (ROS) in vitro, their properties of inhibiting superoxide, singlet oxygen, hydrogen peroxide, and the hydroxyl radical, and their effects on hydroxyl radical-induced DNA oxidative damage were systematically analyzed in the present study. The results show that, compared with (-)-epigallocatechin gallate (EGCG), TF derivatives were good antioxidants for scavenging ROS and preventing the hydroxyl radical-induced DNA damage in vitro. TF(3) was the most positive in scavenging hydrogen peroxide and hydroxyl radical, and TF(1) suppressed superoxide. Positive antioxidant capacities of TF(2)B on singlet oxygen, hydrogen peroxide, hydroxyl radical, and the hydroxyl radical-induced DNA damage in vitro were found. The differences between the antioxidant capacities of four main TF derivatives in relation to their chemical structures were also discussed. We suggest that these activity differences among TF derivatives would be beneficial to scavenge different ROS with therapeutic potential.

Abstract:Isoflavonoids are one of the most important groups of naturally occurring antioxidants. Their structural features are important for evaluating their antioxidative activity. In this work, density functional theory (DFT) methods were applied to investigate the influence of the C2=C3 double bond on the antioxidative activity of isoflavonoids based on three currently accepted radical scavenging mechanisms from the viewpoint of thermodynamics. The C2=C3 double bond can make the compounds more flat, which would extend the conjugated system in the molecule and make the isoflavonoids higher antioxidant activity. The C2=C3 double bond would not alter the strongest antioxidative hydroxyl group of the isoflavonoids. In the gas, benzene and CHCl3 phases, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids by lowering the bond dissociation enthalpies of the hydroxyl groups in the B ring that are the strongest antioxidative sites for the hydrogen atom transfer (HAT) mechanism. In polar phases, a similar result is obtained by weakening the proton affinity of 7-OH that is the strongest antioxidative hydroxyl group in the sequential proton loss electron transfer (SPLET), mechanism. Thus, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids irrespective of the studied phases.
Substituent effects on the ESIPT process and the potential applications in materials transport field of 2'-aminochalcone derivatives

Abstract:This work investigates the different charge transfer characteristics and excited state intramolecular proton transfer process (ESIPT) of 2'-aminochalcones derivatives carrying different electron-withdrawing groups. Four new molecules are designed in the experiment and named as 2c, 3c, 4c and 5c, respectively. (Dyes and Pigments, 2022, 202.) Based on these four molecules, the effect of substituents on the ESIPT process and the charge transfer process are discussed in detail in our work. According to the study of the related parameters at the hydrogen bond site, infrared vibration spectrum, interaction region indicator isosurface (IRI) and scatter plots, it is concluded that the hydrogen bond interaction is enhanced under photoexcitation, and the descending order of the excited state hydrogen bond strength is 3c > 5c > 4c > 2c. The hydrogen bond energy is calculated by atoms in moleculs (AIM) topological analysis and core-valence bifurcation (CVB) index. The potential energy curve reveals the ESIPT mechanism. Frontier molecular orbital and electron-hole analyses explain the reasons for the changes in the ESIPT process at the electronic level. In addition, the ionization potentials (IPa and IPv), affinity energies (EAa and EAv) and reorganization energies are calculated to evaluate the potential application value of organic molecules in material transport field.
Comparative Study of Fluorescence Emission of Fisetin, Luteolin and Quercetin Powders and Solutions: Further Evidence of the ESIPT Process

Abstract:Fisetin and Luteolin are important flavonoids produced in plants and known for their antioxidant, anti-inflammatory, neuroprotective, and analgesic properties. They are also good candidates for different types of biosensors. The model used to describe the fluorescence (FL) emission of these flavonoids involves an excited-state intermolecular proton transfer (ESIPT) process that causes a change in the molecule configuration and a corresponding decrease in the emission energy. Due to the different molecular structures of Fisetin and Luteolin, only one possible proton transfer within the molecule is allowed for each of them: transfer of the H3 proton for Fisetin and of the H5 for Luteolin. Here, we compare their calculated emission wavelengths, obtained using TDDFT/M06-2X/6-31++G(d,p), with their FL emission spectra measured on the corresponding powders and solutions and show that the experimental data are consistent with the presence of the ESIPT process. We also compare the emission wavelengths found for Fisetin and Luteolin with those calculated and measured for Quercetin, where, under photoexcitation, the transfers of both H3 and H5 protons are possible. We analyze the difference in the processes associated with the H3 and H5 proton transfers and discuss the reason for the predominance of the H5 proton transfer in Quercetin. Additionally, a new system of notation for flavonoid molecules is developed.

Abstract:In this research, antioxidant potency (in terms of peroxyl radical scavenging and catalytic iron ions sequestration) of α-tocopherol and its analogues, has been theoretically investigated. A DFT based kinetic estimation of HOO • radical scavenging potency of PMHC (lipophilic α-TOH derivative) and Trolox (hydrophilic α-TOH derivative) was performed in eight solvents of different polarity. Calculations employing M06-2X functional, accompanied by 6–311++G(d,p) basis set and SMD solvation model, were found as more accurate in reproducing experimentally determined rate constants than M05-2X functional. Rate constant of Trolox in pentyl ethanoate also was estimated by using Snelgrove − Ingold equation and Abraham's β 2 H solvent parameter. Scavenging of CH 3 OO • and HOO • radicals, which is influenced by location, orientation, and dynamics of α-TOH and its derivatives in the membrane lipid bilayer, was estimated by using M06-2X functional. Position of α-TOH phenolic OH group is opposite to that of its metabolite α-CEHC: α-TOH's OH group is located at the membrane-water interface, and α-CEHC's OH group is deeply buried into the bilayer. In the aqueous phase both fHAT and SET were investigated, and inside the bilayer only fHAT was considered as feasible. The spontaneous disproportionation of HOO • /O 2 •− in aqueous solutions should be taken into account to predict potency of antioxidant in scavenging of HOO • radicals. Iron chelating ability of α-TOH and α-CEHC was also investigated. Obtained results indicate α-CEHC, α-tocopherol final catabolite, as more potent scavenger of CH 3 OO • and HOO • radicals and iron ions chelator than the parent molecule.

Preparation of Two Sets of 5,6,7-Trioxygenated Dihydroflavonol Derivatives As Free Radical Scavengers and Neuronal Cell Protectors to Oxidative Damage.

Abstract:An unusual class of 5,6,7-trioxygenated dihydroflavonols (3a–e and 4a–j) were designed and prepared. Their antioxidative properties were assessed by examining their capacities in several in vitro models, including superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, rat liver homogenate lipid peroxidation inhibition, PC12 cells protection from oxidative damage, and xanthine oxidase inhibition. These dihydroflavonols displayed positive quenching abilities towards O2- and DPPH free radicals, in which the majority exhibited superior antioxidant properties to Vitamin C. cis-Configurated compound (±)-3e demonstrated remarkable inhibition to LPO with an IC50 value of 1.9±0.3μM, which was apparently stronger than that of quercetin (IC50=6.0±0.4μM). trans-Configurated dihydroflavonol (±)-4h exhibited significant protective effect on PC12 cells against oxidative damage with an EC50 value of 41.5±5.3μM, more effective compared to that of quercetin (EC50=81.8±8.7μM). The 6-OH-5,7-dimethoxy analogue (±)-3d showed significant inhibition of xanthine oxidase with an IC50 value of 16.0±0.8μM, which is superior to that of allopurinol (IC50=23.5±2.0μM). In addition to the hypothesized action mechanism of the bio-active compounds, 3D modeling was used to analyze the relationship between the minimized-energy structures and antioxidant activities.
The antityrosinase and antioxidant activities of flavonoids dominated by the number and location of phenolic hydroxyl groups

Abstract:BackgroundCompounds with the ability to scavenge reactive oxygen species (ROS) and inhibit tyrosinase may be useful for the treatment and prevention from ROS-related diseases. The number and location of phenolic hydroxyl of the flavonoids will significantly influence the inhibition of tyrosinase activity. Phenolic hydroxyl is indispensable to the antioxidant activity of flavonoids. Isoeugenol, shikonin, baicalein, rosmarinic acid, and dihydromyricetin have respectively one, two, three, four, or five phenolic hydroxyls. The different molecular structures with the similar structure to l-3,4-dihydroxyphenylalanine (l-DOPA) were expected to the different antityrosinase and antioxidant activities.MethodsThis investigation tested the antityrosinase activity, the inhibition constant, and inhibition type of isoeugenol, shikonin, baicalein, rosmarinic acid, and dihydromyricetin. Molecular docking was examined by the Discovery Studio 2.5 (CDOCKER Dock, Dassault Systemes BIOVIA, USA). This experiment also examined the antioxidant effects of the five compounds on supercoiled pBR322 plasmid DNA, lipid peroxidation in rat liver mitochondria in vitro, and DPPH, ABTS, hydroxyl, or superoxide free radical scavenging activity in vitro.ResultsThe compounds exhibited good antityrosinase activities. Molecular docking results implied that the compounds could interact with the amino acid residues in the active site center of antityrosinase. These compounds also exhibited antioxidant effects on DPPH, ABTS, hydroxyl, or superoxide free radical scavenging activity in vitro, lipid peroxidation in rat liver mitochondria induced by Fe2+/vitamin C system in vitro, and supercoiled pBR322 plasmid DNA. The activity order is isoeugenol < shikonin < baicalein < rosmarinic acid < dihydromyricetin. The results showed the compounds with more phenolic hydroxyls have more antioxidant and antityrosinase activities.ConclusionThis was the first study of molecular docking for modeling the antityrosinase activity of compounds. This was also the first study of the protective effects of compounds on supercoiled pBR322 plasmid DNA, the lipid peroxidation inhibition activity in liver mitochondria. These results suggest that the compounds exhibited antityrosinase and antioxidant activities may be useful in skin pigmentation and food additives.

Abstract:Hydroxyl radical (•OH) scavenging and the regeneration of Fe2+ may inhibit or enhance peroxidative damage induced by a Fenton system, respectively. Plant polyphenols reveal the afore-mentioned activities, and their cumulative net effect may determine anti- or pro-oxidant actions. We investigated the influence of 17 phenolics on ultra-weak photon emission (UPE) from a modified Fenton system (92.6 μmol/L Fe2+, 185.2 μmol/L EGTA (ethylene glycol-bis(β-aminoethyl-ether)-N,N,N′,N,-tetraacetic acid) and 2.6 mmol/L H2O2 pH = 7.4). A total of 8 compounds inhibited (antioxidant effect), and 5 enhanced (pro-oxidant effect) UPE at all studied concentrations (5 to 50 μmol/L). A total of 4 compounds altered their activity from pro- to antioxidant (or vice versa) along with increasing concentrations. A total of 3 the most active of those (ferulic acid, chlorogenic acid and cyanidin 3-O-glucoside; mean UPE enhancement by 63%, 5% and 445% at 5 μmol/L; mean UPE inhibition by 28%, 94% and 24% at 50 μmol/L, respectively) contained catechol or methoxyphenol structures that are associated with effective •OH scavenging and Fe2+ regeneration. Most likely, these structures can determine the bidirectional, concentration-dependent activity of some phenolics under stable in vitro conditions. This is because the concentrations of the studied compounds are close to those occurring in human fluids, and this phenomenon should be considered in the case of dietary supplementation with isolated phenolics.

Abstract:Inspired by the distinguished photochemical characteristics of new organic molecule containing the chalcogenide substitution that could be potentially applied across various disciplines, in this work, the effects of atomic electronegativity of chalcogen (O, S and Se) on hydrogen bond interactions and proton transfer (PT) reaction. We present the characteristic 2,8-diphenyl-3,7-dihydroxy-4H,6H-pyrano[3,2-g]-chromene-4,6-dione (D3HF), which is based on 3-hydroxyflavone (3HF) and contains intramolecular double hydrogen bonds that is the main objective of this study to explore in detail the influence of the change of atomic electronegativity on the dual hydrogen bond interaction and the excited state proton transfer (ESPT) behavior by photoexcitation. By comparing the structural changes and infrared (IR) vibrational spectra of the D3HF derivatives (D3HF-O, D3HF-S and D3HF-Se) fluorophores in S0 and S1 states, combined with the preliminary detection of hydrogen bond interaction by core-valence bifurcation (CVB) index, we can conclude that the hydrogen bond is strengthened in S1 state, which is favorable for the occurrence of ESPT reactions. The charge recombination behavior of hydrogen bond induced by photoexcitation also further illustrates this point. Via constructing potential energy surfaces (PESs) based on restrictive optimization, we finally clarify the excited state single PT mechanism for D3HF derivatives. Specially, we confirm change of atomic electronegativity has a regulatory effect on the ESIPT behavior of D3HF and its derivatives, that is, the lower the atomic electronegativity is more conducive to the ESIPT reaction.
