Assessment of the Effect of Tetrandrine-Loaded Solid Lipid Nanoparticles on Plasma Metabolite Profile Using Metabolomics

Abstract:The aim of our study was to characterize the metabolism of Tetrandrine (TET) and Tetrandrine-Loaded Solid Lipid Nanoparticles (TET-SLN). TET-SLN was prepared by high temperature emulsified-low temperature curing method. Composition of prescription was glycerol monostearate, soy lecithin, twain-80 and TET, oil phase organic solvent was anhydrous ethanol. The Box-Behnken response surface analysis determined the optimal prescription: quantity of reagent: 5.71 mg, emulsifier concentration: 3.1%, Soya Lecithin-Glyceryl Monostearate (SL-GMS) ratio: 1.31:1.TET-SLN was translucent liquid and had blue-opalescent obviously. TET-SLN content was determined by High Performance Liquid Chromatography (HPLC) method. Nanoparticles were round or class round solid ball under electron microscopy, and dispersion was good. Particle size was (93.6 +/- 2.87) nm, polydispersity index (PDI) was (0.276 +/- 0.01), zeta potential was -(18.71 +/- 0.85) mV, pH was 7.14. The hemolysis of TET-SLN was measure by rabbit blood, and determined the concentration of injected quality was within 11.42 mu g/mL. Ultrafiltration centrifugation method determined encapsulation efficiency and drug loading capacity, encapsulation efficiency was (90.70 +/- 0.52)% and drug loading capacity was (1.99 +/- 0.01)%. The data indicate that TET-SLN had good physical properties. Finally, we determined the metabolite profile in mice receiving TET-SLN or SLN using Nuclear Magnetic Resonance (NMR). Subsequent Principal Component Analysis (PCA) and Partial Least Squares-Discrimination Analysis (PLS-DA) revealed that both experimental groups were well separated.
