Abstract:A Pt prodrug polyphenol and gadolinium ion loaded cancer theranostics nanoplatform based on a mild acidic pH and thermal sensitive polymer was designed for photoacoustic (PA)/magnetic resonance (MR)/positron emission tomography (PET) multimodal imaging-guided chemo-photothermal combination therapy. The Pt drug release can be controlled by tumour-specific acidic pH and heat generated by external NIR irradiation. The nanoparticles were stable under normal physiological environments and released the drug under the acidic pH of tumours and NIR laser irradiation, which can reduce the side effects of the drug to normal organs. Moreover, the MR signal can be significantly enhanced (approximate to 3-fold increase in T-1 relaxivity) under the acidic tumour microenvironment, which is favorable for cancer diagnosis. The nanoparticles exhibited excellent tumour accumulation and led to complete tumour eradication with low power NIR laser irradiation. This promising approach provides a new avenue for imaging-guided combination therapy.

Abstract:Stimuli-responsive polymers are defined as polymers whose structure and properties can be changed in response to minor changes in the surrounding physical or chemical environment and have become increasingly attractive as biomaterials. Reactive oxygen species (ROS) are a series of chemically reactive oxygen-containing molecules, which play an indispensable physiological role in cell signal transduction, acting as secondary messengers. It has been suggested that ROS levels in the cancer cells are much higher than those in the normal cells, and thus, ROS have been selected as a starting point or target for cancer therapy. Tellurium belongs to the chalcogen along with sulfur and selenium. Compared to sulfur and selenium, the relatively large atomic size and low electronegativity of tellurium give it many unique properties, such as redox sensitivity and coordination ability. In recent years, our research group pioneered in the research of ROS-responsive tellurium-containing polymers and their application in drug delivery. The work presented in this feature article, starting from the difference of oxidation responsiveness of tellurium and selenium, explored the ultra-sensitive ROS-response of tellurides. Then we studied the application of tellurium-containing polymers in drug delivery, focusing on the delivery of platinum drugs. For example, polymers with tellurium in the main chain can simultaneously deliver Cisplatin and indocyanine green to achieve a combination of cancer chemotherapy and photodynamic therapy, while side-chain tellurium/platinum-containing polymer can realize a combination of cancer chemotherapy and radiotherapy. Besides, the tellurium/platinum assembly can achieve controlled drug release through a dual response mechanism of ROS and coordination. Finally, we put forward the prospect of the development of tellurium-containing polymers, such as the development of controllable synthesis of tellurium-containing polymers based on chain polymerization, the development of tellurium-containing polymers and tellurium/platinum-containing assembly with different topological structures, and the exploration of the long-term toxicity of tellurium-containing molecules in biological applications.

Abstract:Tellurium-containing photoresponsive polyelectrolyte multilayer films were fabricated by layer-by-layer assembly of a tellurium-containing polymer, photosensitizer, and poly(styrenesulfonate). The resulting films were investigated by UV/vis spectroscopy, XPS, EPR, and fluorescence spectroscopy. Under visible light, the photosensitizer in the film is excited and transforms triplet oxygen into singlet oxygen in aqueous solution. Singlet oxygen oxidizes -Te- to high valence state (Te═O) on the polymer backbone. The generated (Te═O) group makes the micelles more hydrophilic and looser, thereby facilitating the controlled release of the loaded cargo of micelles. These results show that the film has the potential to be used for cargo loading and controlled release, thus may provide a new way to combine photodynamic therapy and chemotherapy.
Assembly Regulates Gamma Radiation Polymerization of Polytelluoxane

Abstract:Regulating chemical drug's responsiveness to gamma radiation is crucial for achieving better therapeutic effects in cancer treatment. Most research focused on thermodynamic chemical structure design, while little attention was paid to kinetic regulate strategy, which possesses greater universality and security. In this study, we achieved a kinetic-based regulate strategy of gamma radiation reaction, through the construction of microphase environment during polymerization of polytelluoxane (PTeO). We designed hydrophobic segments forming large compound micelles (LCMs) assembly to create kinetically favorable higher concentration for radiation-induced reaction. It exhibited a > ten times higher responsiveness and, as far as we know, merely required a minimum dosage of 5 Gy for polymerization to occur. What's more, by taking advantages of the assembly change with Te-O hydrophilic segments and gamma radiation, polymerization became milder with lower polydispersity than previous methods. Such kinetic-based regulate strategy could offer a novel perspective on the design of radiation-responsive chemoradiotherapy and other radiation-induced chemical process.

Abstract:Selenium/tellurium containing polymers have been attracting growing interest due to their unique properties and potential applications as bio-nanomaterials. The introduction of selenium or tellurium chemistry into block copolymers endows self-assembled nanomaterials with novel stimuli responsiveness and enhanced sensitivity. This review article provides an overview on recent advances of selenium/tellurium containing polymers. We summarize the redox response and reversible self-assembly behaviors, gamma-ray responsive systems for combined chemo- and radio-therapies, and coordination-responsive systems for controlled delivery are highlighted. The review then reveals how selenium-containing polymers can modulate reactive oxygen species and work as gene delivery vehicles or self-delivery therapeutics. (C) 2015 Elsevier Ltd. Abstract:Cisplatin (CDDP) has received worldwide approval for clinical use in the past decades. However, its development in cancer chemotherapy was overshadowed by severe side effects and drug resistance. Herein, we developed a CDDP drug delivery system with high encapsulation efficiency and near-infrared light stimuli-responsive drug release properties based on the coordination of novel tellurium-containing block polymer (PEG-PUTe-PEG) and CDDP. The nanocarriers made from PEG-PUTe-PEG were loaded with CDDP and indocyanine green (ICG) simultaneously. The coordination chemistry between CDDP and tellurium guaranteed the nanocarrier a high stability in plasma and prolonged circulation time in vivo by reducing possible penetration of water molecule into the nanoparticles. Under the stimuli of a near-infrared laser, an amount of ROS can be generated by irradiation of ICG. The tellurium is easily oxidized by ROS because of the low electronegativity of tellurium. The CDDP could be rapidly released from the nanocarriers along with the oxidation of the tellurium at the tumor sites as the oxidized tellurium will weaken the coordination interaction with CDDP. In addition, the encapsulated ICG played a synergistic antitumor effect through photothermal effect with mild laser irradiation. The integrated strategy achieved higher antitumor efficacy and showed minimal side effects compared with the CDDP alone.

Abstract:We have developed a potential radiation-sensitive drug-delivery system using active diselenide-containing block co-polymer aggregates in aqueous solution that can load and release anticancer drugs. These aggregates were sensitive to even a low dose of γ-radiation, such as 5 Gy, which is close to the radiation dose received by patients during a single radiotherapy treatment. This line of research may open an avenue for the combination of radiotherapy and chemotherapy.

Abstract:Although mortality continues to decline over the past two decades, cancer is still a pervasive healthcare problem worldwide due to the increase in the number of cases, multidrug resistance (MDR) and metastasis. As a consequence of multidrug resistance, cancer treatment must rely on a host of chemotherapeutic agents and chemosensitizers to achieve remission. To overcome these problems, a series of biodegradable triblock copolymers of PEG, guanidinium-functionalized polycarbonate and polylactide (PEG-PGCx(-)PDLA(y)) is designed as chemotherapeutic agents. These copolymers self-assemble into micellar nanoparticles, and are highly effective against various cancer cell lines including human breast cancer (BCap37), liver cancer (HepG2), lung cancer (A549) and epidermoid carcinoma (A431) cell lines as well as MDR Bats-72 and Bads-200 cancer cells that were developed from BCap37. Multiple treatments with the polymers at sub-lethal doses do not induce resistance. The polymers kill cancer cells by a non-apoptotic mechanism with significant vacuolization and subsequent membrane disruption. In vivo antitumor efficacy is evaluated in a metastatic 4T1 subcutaneous tumor model. Treatment with stereocomplexes of PEG-PGC(43)-PLLA(19) and PEG-PGC(43)-PDLA(20) at a dose of 20 mg/kg of mouse body weight suppresses tumor growth and inhibits tumor metastasis in vivo. These polymers show promise in the treatment of cancer without the onset of resistance.

Abstract:Materials responsive to reactive oxygen species (ROS) at a physiological level are of great significance in diagnostic and therapeutic applications. We have synthesized a water-soluble tellurium-containing hyperbranched polymer and demonstrated that it forms a kind of ROS-responsive aggregate. In aqueous environment, the polymer aggregates can swell under biologically relevant concentration of H2O2, which could be expected to find applications in fields such as elimination of excess ROS.

Abstract:X-ray-responsive nanocarriers for anticancer drug delivery have shown great promise for enhancing the efficacy of chemoradiotherapy. A critical challenge remains for development of such radiation-controlled drug delivery systems (DDSs), which is to minimize the required X-ray dose for triggering the cargo release. Herein, we design and fabricate an effective DDS based on diselenide block copolymers (as nanocarrier), which can be triggered to release their cargo with a reduced radiation dose of 2 Gy due to their sensitivity to both X-ray and the high level of reactive oxygen species (ROS) in the microenvironment of cancer cells. The underlying molecular mechanism is further illustrated by proton nuclear magnetic resonance (1H NMR) experiments and density functional theory (DFT) calculations. In vivo experiments on tumor-bearing mice validated that the loaded drugs are effectively delivered to the tumor site and exert remarkable antitumor effects (minimum tumor volume/weight) along with X-ray. Furthermore, the diselenide nanocarriers exhibit no noticeable cytotoxicity. These findings provide new insights for the de novo design of radiation-controlled DDSs for cancer chemoradiotherapy.
Antibacterial tellurium-containing polycarbonate drug carriers to eliminate intratumor bacteria for synergetic chemotherapy against colorectal cancer

Abstract:Intratumor microbes have attracted great attention in cancer research due to its influence on the tumorigenesis, progression and metastasis of cancer. However, the therapeutic strategies targeting intratumoral microbes are still in their infancy. Specific microorganisms, such as Fusobacterium nucleatum (F. nucleatum), are abundant in various cancer and always result in the CRC progression and chemotherapy resistance. Here, a combined anticancer and antibacterial therapeutic strategy is proposed to deliver antitumor drug to the tumors containing intratumor microbiota by the antibacerial polymeric drug carriers. We construct oral tellurium-containing drug carriers using a complex of tellurium-containing polycarbonate with cisplatin (PTE@CDDP). The results show that the particle size of the prepared nanoparticles could be maintained at about 105 nm in the digestive system environment, which is in line with the optimal particle size of oral nanomedicine. In vitro mechanism study indicates that the tellurium-containing polymers are highly effective in killing F.nucleatum through a membrane disruption mechanism. The pharmacokinetic experiments confirmed that PTE@CDDP has the potential function of enhancing the oral bioavailability of cisplatin. Both in vitro and in vivo studies show that PTE@CDDP could inhibit intratumor F.nucleatum and lead to a reduction in cell proliferation and inflammation in the tumor site. Together, the study identifies that the CDDP-loaded tellurium-containing nanoparticles have great potential for treating the F.nucleatum-promoted colorectal cancer (CRC) by combining intratumor microbiota modulation and chemotherapy. The synergistic therapeutic strategy provide new insight into treating various cancers combined with bacterial infection. STATEMENT OF SIGNIFICANCE: The synthesized antibacterial polymer was first employed to remodel the intratumor microbes in tumor microenvironment (TME). Moreover, it was the first report of tellurium-containing polymers against F.nucleatum and employed for treatment of the CRC. A convenient oral dosage form of cisplatin (CDDP)-loaded tellurium-containing nanoparticles (PTE@CDDP) was adopted here, and the synergistic antibacterial/chemotherapy effect occurred. The PTE@CDDP could quickly and completely eliminate F.nucleatum in a safe dose. In the CRC model, PTE@CDDP effectively reversed the inflammation level and even restored the intestinal barrier damaged by F.nucleatum. The ultrasensitive ROS-responsiveness of PTE@CDDP triggered the fast oxidation and efficient drug release of CDDP and thus a highly efficient apoptosis of the tumors. Therefore, the tellurium-containing polymers are expected to serve as novel antibacterial agents in vivo and have great potential in the F.nucleatum-associated cancers. The achievements provided new insight into treating CRC and other cancers combined with bacterial infection.

Abstract:Nanomaterials capable of achieving tunable cargo release kinetics are of significance in a fundamental sense and various biological or medical applications. We report a competitive coordination system based on a novel tellurium-containing polymer and its ligand-regulated release manners. Tellurium was introduced to water-soluble polymers for the first time as drug delivery vehicles. The coordination chemistry between platinum and tellurium was designed to enable the load of platinum-based drugs. Through the competitive coordination of biomolecules, the drugs could be released in a controlled manner. Furthermore, the release kinetics could be modulated by the competitive ligands involved due to their different coordination ability. This tellurium-containing polymer may enrich the family of delivery systems and provide a new platform for future biomedical nanotechnologies.

Abstract:Tellurium-containing polymers have become increasingly attractive owing to their unique properties as biomaterials and optoelectronic materials. The relatively large atomic size and low electronegativity of tellurium give it with many unique properties, such as coordination ability and redox sensitivity. The introduction of tellurium can endow materials with different responsiveness, which makes it possible for them to be employed as biomaterials. The narrow band gap of polytellurophene also allows its use in optoelectronic materials. In this focus review, we highlight a series of recent advances in tellurium-containing polymers. First, tellurium-containing amphiphilic block copolymers are summarized; these species can self-assemble to form aggregates with coordination-responsive and reactive oxygen species-responsive properties. Then, we describe both tellurophene-containing homopolymers and copolymers, including their synthesis, chemical properties, and applications in different optoelectronic devices. It is anticipated that tellurium-containing polymers will have more applications in material science.
In situ synthesis of multifunctional tellurium nanorods stabilized by polypeptide-engineered for photothermal-sonodynamic combination therapy of tumors

Abstract:Nanoparticles containing elemental tellurium (Te) have been aroused extensive attention in biomedical applications. However, the ideal synthesis methods and effective anti-cancer strategies of Te nanoparticles remain great challenges. In this study, genetically engineered polypeptide (PC10ARGD)-capped Te nanorods (P-TeNRs) was in situ synthesized. The excellent dispersion of P-TeNRs with average lengths of 25.2 ± 4.65 nm and diameters of 6.5 ± 0.83 nm were confirmed by TEM. PC10ARGD was integrated onto the surface of Te nanorods to enhance the stability and biocompatibility of P-TeNRs. P-TeNRs exhibited the excellent photothermal conversion performance under irradiation of NIR laser and the reactive oxygen species (ROS) generating capacity under ultrasound (US) irradiation, which can achieve photothermal therapy and sonodynamic therapy of tumors. Compared with single photothermal therapy and sonodynamic therapy, the results of in vitro and in vivo experiments showed that the combination therapy has a synergistic effect on breast 4T1 tumors. In addition, the results of in vivo toxicity assay suggested that synthesized P-TeNRs presented negligible toxicity. These results demonstrate that this P-TeNRs can be engineered as a novel theranostic nanoplatform for synergistic photothermal-sonodynamic therapy of tumors.

Abstract:Checkpoint blocking-based immunotherapy has been proved to be effective for the treatment of cancer, but its dependence on T cell infiltration has limited its effectiveness across patients. Radiotherapy (RT) is becoming the most common method of clinical tumor treatment but can occasionally cause systemic tumor rejection. Here, we demonstrate a simple one-pot hydrothermal method to synthesize tellurium (Te) nanostars (GTe-RGD) and offer a treatment strategy that combines GTe-RGD-potentiated RT with checkpoint blockade immunotherapy for efficient and systemic tumor elimination. In mouse models of breast cancer, GTe-RGD-potentiated RT not only eradicated primary tumors but also elicited antitumor immunity to inhibit growth at distant sites by enhancing cytotoxic T lymphocytes when combined with an immune checkpoint inhibitor. Furthermore, the triggered release of tumor-associated antigens and cytokines can effectively reduce the percentage of M2 macrophages with enhanced antitumor activity. Thus, this simply synthesized Te-based nanomedicine, with radiation-driven immunotherapy, offers an attractive clinical alternative for tumor treatment.

Abstract:Effective drug delivery with precise tumour targeting is crucial for cancer treatment. To address the challenges posed by the specificity and complexity of the tumour microenvironment, we developed a poly(tetraethylene glycol)-based disulfide nanoparticle platform and explored its potential in cancer treatment, focusing on drug loading and controlled release performance. Poly(tetraethylene glycol) nanoparticles were characterised using nuclear magnetic resonance spectroscopy, mass spectrometry, and ultraviolet-visible spectroscopy. Additionally, we evaluated physicochemical properties, including dynamic light scattering, zeta potential analysis, drug loading capacity, and drug loading efficiency. The impact of nanoparticles on the mouse colorectal cancer cell line (CT26) and NIH3T3 cells was assessed using a cytotoxicity assay, live/dead staining assay, flow cytometry, and confocal fluorescence microscopy. The experimental results align with the expected chemical structure and physicochemical properties of poly(tetraethylene glycol) nanoparticles. These nanoparticles exhibit high drug loading efficiency (78.7%) and drug loading capacity (12%), with minimal changes in particle size over time in different media. In vitro experiments revealed that the nanoparticles can induce significant cytotoxicity and apoptosis in CT26 cells. Cellular uptake notably increases with increasing concentration and exposure time. The confocal microscopic analysis confirmed the effective distribution and accumulation of nanoparticles within cells. In conclusion, poly(tetraethylene glycol) nanoparticles hold promise for improving drug-delivery efficiency, offering potential advancements in cancer treatment.

Responsive Degradable Theranostic Agents Enable Controlled Selenium Delivery to Enhance Photothermal Radiotherapy and Reduce Side Effects

Abstract:Radiotherapy (RT) is a popular clinical therapy method for extending cancer patient survival, but is hampered by severe side effects and the weak therapy effect. Herein, responsive degradable selenium (Se) theranostic agents (Se@SiO2@Bi nanocomposites (NCs)) are fabricated, which combine computed tomography (CT) imaging and simultaneously enhance the therapeutic effects of photothermal therapy (PTT) and RT, while reducing the side effects of radiation. The Se@SiO2@Bi theranostic agents can accumulate at the tumor site, and responsively decompose to releease Se, avoiding systemic toxicity by the element. Se enhances the effect of PTT/RT, simultaneously reducing the side effects of RT. The Se@SiO2@Bi NCs as CT agents also exhibit significantly enhanced contrast imaging performance due to the high atomic number of Bi. More importantly, the Se@SiO2@Bi NCs can be rapidly excreted without long‐term toxicity, owing to responsive degradation into ultrasmall particles (<5 nm) at the tumor site. In vitro and in vivo results show that the Se@SiO2@Bi NCs can remarkably inhibit tumor cells, without causing appreciable toxicity during the treatment. This study opens a new perspective in rationally designing responsive degradable theranostic agents for future tumor therapy with enhanced therapeutic efficacy and lesser side effects.

Highly Effective Radioisotope Cancer Therapy with a Non-Therapeutic Isotope Delivered and Sensitized by Nanoscale Coordination Polymers

Abstract:Nuclear medicine with radioisotopes is extremely useful for clinical cancer diagnosis, prognosis, and treatment. Herein, polyethylene glycol (PEG)-modified nanoscale coordination polymers (NCPs) composed of hafnium (Hf4+) and tetrakis (4-carboxyphenyl) porphyrin (TCPP) are prepared via a one-pot reaction. By chelation with the porphyrin structure of TCPP, such Hf-TCPP-PEG NCPs could be easily labeled with 99mTc4+, an imaging radioisotope widely used for single-photon emission computed tomography (SPECT) in a clinical environment. Interestingly, Hf, as a high- Z element in such 99mTc-Hf-TCPP-PEG NCPs, could endow nontherapeutic 99mTc with the therapeutic function of killing cancer cells, likely owing to the interaction of Hf with γ rays emitted from 99mTc to produce charged particles for radiosensitization. With efficient tumor retention, as revealed by SPECT imaging, our 99mTc-Hf-TCPP-PEG NCPs offer exceptional therapeutic results in eliminating tumors with moderate doses of 99mTc after either local or systemic administration. Importantly, those biodegradable NCPs could be rapidly excreted without much long-term body retention. Our work, showing the success of applying NCPs for radioisotope therapy (RIT), presents a potential concept for the realization of highly effective cancer treatment with 99mTc, a short-half-life (6.0 h) diagnostic radioisotope, which is promising for cancer RIT with enhanced efficacy and reduced side effects.
