Abstract:An anticancer drug, dasatinib (DAS), is used to treat prostate cancer, chronic myelogenous leukaemia, and acute lymphoblastic leukaemia. In the present work, we describe the preparation of zinc oxide (ZnO) nanoparticles (NPs) using Averrhoa bilimbi leaf extract. These ZnO NPs were combined with functionalized multi-walled carbon nanotubes (f-MWCNT) to prepare a ZnO@f-MWCNT nanocomposite for electrochemical sensing of DAS. The prepared electrode materials, ZnO NPs, f-MWCNT, and ZnO@f-MWCNT, were characterised by powder X-ray diffraction (XRD), scanning electron microscopy (SEM), the Fourier-transform infrared (FT-IR) spectrum, atomic force microscopy (AFM), zeta potential, and the dynamic light scattering (DLS) techniques. DAS exhibited a significant enhancement (18-fold) in the oxidation peak current at ZnO@f-MWCNT, 10-fold at f-MWCNT, and 6-fold at ZnO NPs as compared to that at bare GCE. Under optimised conditions, a linear relationship was noticed among the peak current and concentration of DAS in the range of 0.03-82.65 μM and 0.01-122.45 μM for the differential pulse voltammetric (DPV), and square wave voltammetric (SWV) methods, respectively. Further, the applicability of the proposed sensor was demonstrated by analysing pharmaceutical and biological samples containing DAS. Higher percentage recovery values, and lower relative standard deviation (RSD) values highlighted the proposed electrochemical methods, accuracy, and precision.

Abstract:Herein, we proposed a new approach to design a MIP-based electrochemical sensor with carbon nanofiber (CNF), which could improve its conductivities as well as electrode sensitivity and successful detection of dasatinib (DAS). CNFs are capable of forming high porosity with significant interconnected porous networks. The poly(2-hydroxyethyl-methacrylate-N-methacryloyl-L-tyrosine) (PHEMA-MATyr) copolymer was synthesized in the presence of both CNF and DAS by photopolymerization. After optimization of the parameters, the modified MIP-based electrochemical sensor demonstrated the ability to determine the DAS in the linear working range of 1.0 × 10-14-1.0 × 10-13 M for the standard solution and commercial serum samples with a LOD of 1.76 × 10-15 and 2.46 × 10-15, respectively. Good linearity for DAS was observed with correlation coefficients (r) of 0.996 and 0.997 for the standard solution and commercial serum samples, respectively. The recoveries of the DAS ranged from 99.45 % to 99.53 % for the tablet dosage form and commercial serum samples, with average relative standard deviations below 1.96 % in both cases. The proposed modified sensor demonstrated significant sensitivity and selectivity for the rapid determination of DAS in commercial serum samples and tablet form.

Abstract:In this study, a novel tin oxide/nitrogen-doped graphene quantum dot nanocomposite (SnO2-NDGQD) and 1-methyl-3-octylimidazolinium chloride (1M3OICl) ionic liquid amplified carbon paste electrode (CPE) was fabricated as an efficient and fast-response sensor to determine daunorubicin, an anticancer drug. The electro-chemical characteristics of daunorubicin at the surface of the 1M3OICl/SnO2-NDGQD/CPE was explored via various voltammetric methods. The high-resolution transmission electron microscope (HR-TEM) images were recorded to examine the morphological structure of the as-synthesized nanocomposites. The 1M3OICl/SnO2- NDGQD/CPE offered a wide linear concentration of 0.001-280.0 mu M with a low detection limit of 0.40 nM at the optimized experimental conditions using square wave voltammetric (SWV) method. In a nutshell, the developed electrode illustrated outstanding selectivity in the presence of interfering agents and long-term stability. The1-M3OICl/SnO2-NDGQD/CPE was used as new and powerful analytical tool for determination of daunorubicin in real samples with recovery range 98.75%-104.8%.

Abstract:The poly (N-isopropylacrylamide)-co-polystyrene (PNIPAM-co-PS) nanofibers have been fabricated by electrospinning, and the blends of PNIPAM-co-PS nanofibers with titanium dioxide (TiO2) nanoparticles have been characterized and utilized as the new nanocomposites to enhance the relevant detection sensitivity of biomolecular recognition of an anticancer drug daunorubicin. Our observations demonstrate that upon application of the nanoTiO2–PNIPAM-co-PS polymer nanocomposites, the drug molecules could be readily deposited on the surface of the relevant blends so that the remarkable enhancement effect of the new nanocomposites on the respective biorecognition of daunorubicin could be observed, suggesting the potential valuable application of the blending of the nanoTiO2 and PNIPAM-co-PS polymer nanocomposites in high sensitive bioanalysis.

Abstract:In this work, a novel highly sensitive electrochemical sensing platform has been developed for simultaneous determination of anticancer drugs doxorubicin (DOX) and dasatinib (DAS) by means of mesoporous Pd@ Pt core-shell nanoparticles supported on MWCNT (Pd@ Pt/MWCNT). The electrochemical behaviour of the drugs was assessed through cyclic voltammetry, while simultaneous determination was carried out using adsorptive stripping square wave voltammetry (AdSSWV). Under optimized conditions, the method exhibits linear responses towards simultaneous determination of DOX and DAS in the concentration ranges of 4.4-8580 nM and 38-9880 nM, with detection limits of 0.86 and 6.72 nM, respectively. Furthermore, the proposed platform was successfully used for reliable determination of DOX and DAS in urine and blood serum samples with good accuracy and precision. The satisfactory recoveries obtained in blood serum and urine samples showed the potential for application of the proposed platform in quality control in the pharmaceutical industry, clinical analysis and optimization of chemotherapeutic treatment. We found that the established sensor has several advantages in terms of high sensitivity and selectivity, ease of preparation, short analysis time, long-term stability and fast response.
Guanine-Based DNA Biosensor Amplified with Pt/SWCNTs Nanocomposite as Analytical Tool for Nanomolar Determination of Daunorubicin as an Anticancer Drug: A Docking/Experimental Investigation

Abstract:Daunorubicin is a famous anthracycline anticancer chemotherapy drug with many side effects that is very important to measure in biological samples. A daunorubicin electrochemical biosensor was fabricated in this study using ds-DNA as the biorecognition element and glassy carbon electrode (GCE) amplified by Pt/SWCNTs as a sensor. The synthetization of Pt/SWCNTs was done by the polyol method, and their characterization was accomplished via XRD, EDS, and TEM methods. The results showed a diameter of about 5.0 nm for the Pt nanoparticle decorated at the surface of SWCNTs. The morphological and conductivity properties of Pt/SWCNTs/GCE were investigated by EIS and AFM methods, and the results confirmed that Pt/SWCNTs/GCE had a high surface area and high conductivity. ds-DNA/Pt/SWCNTs/GCE showed an oxidation signal relative to that of the guanine base at the potential of 847 mV and a positive shift after interaction with the daunorubicin anticancer drug. This point confirms the intercalation reaction between the guanine base in the ds-DNA structure and the drug that could be used as an analytical factor for the determination of daunorubicin. Furthermore, molecular docking study is used to predict the interaction site of daunorubicin with DNA. It is found that daunorubicin interacts with guanine bases of DNA via an intercalative mode. Kinetic investigation showed an association equilibrium constant (K a) of about 5.044 × 103 M–1 between ds-DNA and daunorubicin. The differential pulse voltammetric results showed a linear dynamic range of 4.0 nM to 250.0 μM with a detection limit of 1.0 nM for determination of daunorubicin on the surface of ds-DNA/Pt/SWCNTs/GCE. Finally, ds-DNA/Pt/SWCNTs/GCE was successfully used for the determination of daunorubicin in injection samples with a recovery range of 98.27–10313%.

Abstract:The voltammetric behavior of doxorubicin at a ZnO nanoparticle/1-butyl-3-methylimidazolium tetrafluoroborate modified carbon paste electrode (ZnO-NPs/BMTFB/CPE) has been investigated. For the goal, ZnO-NPs were biosynthesized by celery extract in aqueous solution. The ZnO-NPs/BMTFB/CPE exhibits a good promoting effect on the oxidation response of doxorubicin. After optimization of pH condition, ZnO-NPs/BMTFB/CPE exhibits two separated oxidation signals for simultaneous analysis of doxorubicin and dasatinib with ∆E ~300 mV. The oxidation currents showed linear relation with doxorubicin or dasatinib concentration in the ranges of 0.07–500 μM and 1.0–1200 μM, respectively. We detected limit of detection 9.0 nM and 0.5 μM for doxorubicin and dasatinib, respectively. The ZnO-NPs/BMTFB/CPE has been applied to the analysis of doxorubicin and dasatinib in injection and pharmaceutical serum samples.
Doxorubicin Anticancer Drug Monitoring by ds-DNA-Based Electrochemical Biosensor in Clinical Samples

Abstract:In this research, glassy carbon electrode (GCE) amplified with single-wall carbon nanotubes (SWCNTs) and ds-DNA was fabricated and utilized for voltammetric sensing of doxorubicin with a low detection limit. In this technique, the reduction in guanine signal of ds-DNA in the presence of doxorubicin (DOX) was chosen as an analytical factor. The molecular docking study revealed that the doxorubicin drug interacted with DNA through intercalation mode, which was in agreement with obtained experimental results. The DOX detection performance of ds-DNA/SWCNTs/GCE was assessed at a concentration range of 1.0 nM–20.0 μM. The detection limit was found to be 0.6 nM that was comparable and even better (in many cases) than that of previous electrochemical reported sensors. In the final step, the ds-DNA/SWCNTs/GCE showed powerful ability for determination of the DOX in injection samples with acceptable recovery data.

Abstract:The determination of doxorubicin and topotecan as two important breast anticancer drugs with high side effects is significant in pharmaceutical samples. 2D nitrogen doped reduced graphene (NrG) nano-sheet and 1-methyl-3-octylimidazolium chloride (MOICI) were suggested as two high conductive modifiers for fabrication of anticancer drug sensor based carbon paste electrode (CPE). The electrical conductivity of NrG/MOICl/CPE as a new analytical approach was probed by voltatnmetric methods. The NrG/MOICl/CPE showed accelerating the exchange of electrons for oxidation of doxorubicin and this improving helps to simultaneous determination of doxorubicin and topotecan with separation gap of 315 mV. The NrG/MOICl/CPE as an anticancer sensor displayed detection limits of 3.1 nM and 0.27 pM for doxorubicin and topotecan, respectively. The NrG/MOICl/CPE was found to be practically applicable in the determination of doxorubicin and topotecan in drug samples. (C) 2018 Published by Elsevier B.V.
A Label-Free Fluorescent Sensor for Rapid and Sensitive Detection of Ctdna Based on Fluorescent PDA Nanoparticles.

Abstract:Technological advances in the detection of circulating tumor DNA (ctDNA) have made new options available for diagnosis, classification, biological studies, and treatment selection. However, effective and practical methods for analyzing this emerging class of biomarkers are still lacking. In this work, a fluorescent biosensor was designed for the label-free detection of ctDNA (EGFR 19 del for non-small cell lung cancer, NSCLC). The biosensor was based on the fact that MnO2 nanosheets (MnO2 NSs) have stronger affinity towards single-stranded DNA (ssDNA), as compared with double-stranded DNA (dsDNA). As a high-performance nanoenzyme, MnO2 NSs could oxidize dopamine (DA) into fluorescent polydopamine nanoparticles (FL-PDA NPs), which could be used as a fluorescence signal. The probe ssDNA could be adsorbed on the surface of MnO2 NSs through π-π stacking, and the active site would be masked, causing a lower fluorescence. After the targets were recognized by probe ssDNA to form dsDNA, its affinity for MnO2 NSs decreased and the active site recovered, causing a restored fluorescence. It was verified that Mn ions, •OH radicals and electron transfer were the important factors in the catalytic oxidation of DA. Under the optimal experimental conditions, this biosensor exhibited a detection limit of 380 pM and a linear range of 25-125 nM, providing reliable readout in a short time (45 min). This sensor exhibited outstanding specificity, stability and reproducibility. In addition, this sensor was applied to the detection of ctDNA in serum samples and cell lysates. It is demonstrated that FL-PDA NPs can be used as a fluorescence signal for easy, rapid and label-free detection of ctDNA without any other amplification strategies, and the proposed strategy has great potential for biomarker detection in the field of liquid biopsy.
The High Stability and Selectivity of Electrochemical Sensor Using Low-Cost Diamond Nanoparticles for the Detection of Anti-Cancer Drug Flutamide in Environmental Samples

Abstract:In this study, a novel electrochemical sensor was created by fabricating a screen-printed carbon electrode with diamond nanoparticles (DNPs/SPCE). The successful development of the sensor enabled the specific detection of the anti-cancer drug flutamide (FLT). The DNPs/SPCE demonstrated excellent conductivity, remarkable electrocatalytic activity, and swift electron transfer, all of which contribute to the advantageous monitoring of FLT. These qualities are critical for monitoring FLT levels in environmental samples. Various structural and morphological characterization techniques were employed to validate the formation of the DNPs. Remarkably, the electrochemical sensor demonstrated a wide linear response range (0.025 to 606.65 μM). Additionally, it showed a low limit of detection (0.023 μM) and high sensitivity (0.403 μA μM−1 cm−2). Furthermore, the practicability of DNPs/SPCE can be successfully employed in FLT monitoring in water bodies (pond water and river water samples) with satisfactory recoveries.

Abstract:The establishment of a monitoring technique for imatinib is necessary in clinical and environmental toxicology. Leaf extracts of Lycoris longituba were used as reducing agent for the one-step synthesis of reduced graphene oxide-Ag nanocomposites. This nanocomposite was characterized by TEM, FTIR, XRD, and other instruments. Then, the graphene/Ag nanocomposite was used as a modifier to be cemented on the surface of the glassy carbon electrode. This electrode exhibited excellent electrochemical sensing performance. Under the optimal conditions, the proposed electrode could detect imatinib at 10 nM−0.28 mM with a low limit of detection. This electrochemical sensor also has excellent anti-interference performance and reproducibility.

Abstract:Determination of pharmaceuticals especially anticancer drugs is one of the important issues in environmental and medical investigation and creating good information about human health. The presence sturdy introducing an electroanalytical sensor based on molecularly imprinted polymer (MIP)/Multi-walled carbon nanotubes (MWCNTs)/Au@Fe3O4 nanoparticles modified carbon paste electrode (PE) to determine imatinib (IMA). The MIP/MWCNTs/Au@Fe3O4/PE showed catalytic activity and also a sensitive strategy to sensing IMA in the concentration range 1-1000 μM with a limit of detection of 0.013 μM. The MIP/MWCNTs/Au@Fe3O4/PE has shown interesting results in the analysis of IMA in real samples, and the interference investigations results show the high selectivity of the MIP/MWCNTs/Au@Fe3O4/PE in the monitoring of IMA in complex fluids such as tablet and blood serum and results approved by F-test and t-test as statistical methods.

Abstract:Nanocomposite materials have provided a wide range of conductivity, sensitivity, selectivity and linear response for electrochemical biosensors. However, the detection of rare cells at single cell level requires a new class of nanocomposite-coated electrodes with exceptional sensitivity and specificity. We recently developed a construct of gold nanoparticle-grafted functionalized graphene and nanostructured polyaniline (PANI) for high-performance biosensing within a very wide linear response and selective performance. Further, replacing the expensive gold nanoparticles with low-cost silver nanoparticles as well as optimizing the nanocomposite synthesis and functionalization protocols on the electrode surface in this work enabled us to develop ultrasensitive nanocomposites for label-free detection of breast cancer cells. The sensor presented a fast response time of 30 min within a dynamic range of 10 - 5 × 106 cells mL-1 and with a detection limit of 2 cells mL-1 for the detection of SK-BR3 breast cancer cell. The nano-biosensor, for the first time, demonstrated a high efficiency of > 90% for the label-free detection of cancer cells in whole blood sample without any need for sample preparation and cell staining. The results demonstrated that the optimized nanocomposite developed in this work is a promising nanomaterial for electrochemical biosensing and with the potential applications in electro-catalysis and super-capacitances.
A Green and Sensitive Guanine-Based DNA Biosensor for Idarubicin Anticancer Monitoring in Biological Samples: A Simple and Fast Strategy for Control of Health Quality in Chemotherapy Procedure Confirmed by Docking Investigation.

Abstract:(GCE) was modified by Pt- and Pd-incorporated ZnO nanoparticles-decorated single-wall carbon nanotubes (Pt-Pd-ZnO/SWCNTs) nanocomposites, and ds-DNA (Calf Thymus) that was a biological recognition element, and it was aimed to be utilized as an ultrasensitive and effective electroanalytical biosensor for idarubicin (IDR) monitoring. Various physicochemical characterization techniques including transmission electron microscopy (TEM), field-emission scanning electron microscopy (FE-SEM) with energy-dispersive X-ray spectroscopy (EDS) were used to investigate the morphology and structure of the Pt-Pd-ZnO/SWCNTs nanocomposite, which was produced via straightforward chemical precipitation combined with the one-pot method. The layer-by-layer modification technique was implemented to fabricate the ds-DNA/Pt-Pd-ZnO/SWCNTs/GCE to be further utilized as a voltammetric sensor for sensitive monitoring of idarubicin in biological fluids and pharmaceutical substances. The electroanalytical method implemented to detect idarubicin was based to detect the ds-DNA's guanine base signal on the surface of the modified electrode in the absence and presence of the anticancer drug. The results explicated that the developed biosensor performed well in determining idarubicin in concentrations ranging from 1.0 nM to 65 mu M, with a detection limit of 0.8 nM. The idarubicin detection ability of the modified electrode in real samples was evaluated, and the recovery data was acquired in the range of 98.0% and 104.75%. Abstract:A sophisticated electrochemical sensor is presented employing a glassy carbon electrode (GCE) modified with a novel composite of synthesized graphitic carbon nitride (g-C3N4) and CoNiO2 bimetallic oxide nanoparticles (g-C3N4/CoNiO2). The sensor's electrocatalytic capabilities for Sunitinib (SUNI) oxidation were demonstrated exceptional performance with a calculated detection limit (LOD) of 52.0 nM. The successful synthesis and integrity of the composite were confirmed through meticulous characterization using various techniques. FT-IR analysis affirmed the successful synthesis of g-C3N4/CoNiO2 by providing insights into its molecular structure. XRD, FE-SEM, SEM-EDX, and BET analyses collectively validated the material's structural integrity, surface morphology, and electrocatalytic performance. Optimization of key analytical parameters, such as loading volume, concentration, electrolyte solution type, and pH, enhanced the electrocatalytic sensing capabilities of g-C3N4/CoNiO2. The synergistic interaction between g-C3N4 and CoNiO2 bimetallic oxide nanoparticles executed the sensor highly effective in the electrical oxidation of SUNI. Across a concentration range of 0.1-83.8 µM SUNI, the anodic peak current exhibited a linear increase with good precision. Application of the newly developed g-C3N4/CoNiO2 system to detect SUNI in a variety of samples, including urine, human serum, and capsule dosage forms, obtained satisfactory recoveries ranging from 97.1 to 103.0%. This methodology offers a novel approach to underscore the potential of the developed sensor for applications in biological and pharmaceutical monitoring.
A simple voltammetric method for rapid sensing of daunorubicin in the presence of dacarbazine by graphene oxide/metal–organic framework-235 nanocomposite-modified carbon paste electrode

Abstract:In the current work, a novel and simple electrochemical sensing platform was built utilizing a graphene oxide (GO)/metal–organic framework-235 (MOF-235) nanocomposite, which was successfully synthesized and used as an excellent modifier to determine the simultaneous presence of two key anticancer medications, daunorubicin (DNR), and dacarbazine (DTIC). Fourier transform infrared spectroscopy, energy-dispersive X-ray spectroscopy, field-emission scanning electron microscopy, and scanning electron microscopy (SEM) are used to characterize the GO/MOF-235 nanocomposite. The electrocatalytic properties of the modified sensor based on MOF and GO in phosphate buffer solution (PBS) (0.1 M—pH 7.0) toward DNR redox reaction were thoroughly investigated using electrochemical methodologies including cyclic voltammetry, differential pulse voltammetry, and chronoamperometry. This GO/MOF-235-modified CPE demonstrated a wide linear response of 0.005–400.0 μM under optimized conditions. It was determined that 0.002 μM was the detection limit. This sensor displayed two distinct oxidation peaks at 390 mV for DNR and at 750 mV for dacarbazine (DTIC) (peak potential separation = 360 mV), allowing for the simultaneous detection of the two anticancer medications. Determination of these anticancer medicines in samples (DNR injection and DTIC injection) was effectively accomplished in the final step using the GO/MOF-235/CPE sensor.
