Abstract:The phototheranostics in the second near-infrared window (NIR-II) have proven to be promising for the precise cancer theranostics. However, the non-responsive and "always on" imaging mode lacks the selectivity, leading to the poor diagnosis specificity. Herein, a tumor microenvironment (TME) activated NIR-II phototheranostic nanoplatform (Ag2 S-Fe(III)-DBZ Pdots, AFD NPs) is designed based on the principle of Förster resonance energy transfer (FRET). The AFD NPs are fabricated through self-assembly of Ag2 S QDs (NIR-II fluorescence probe) and ultra-small semiconductor polymer dots (DBZ Pdots, NIR-II fluorescence quencher) utilizing Fe(III) as coordination nodes. In normal tissues, the AFD NPs maintain in "off" state, due to the FRET between Ag2 S QDs and DBZ Pdots. However, the NIR-II fluorescence signal of AFD NPs can be rapidly "turn on" by the overexpressed GSH in tumor tissues, achieving a superior tumor-to-normal tissue (T/NT) signal ratio. Moreover, the released Pdots and reduced Fe(II) ions provide NIR-II photothermal therapy (PTT) and chemodynamic therapy (CDT), respectively. The GSH depletion and NIR-II PTT effect further aggravate CDT mediated oxidative damage toward tumors, achieving the synergistic anti-tumor therapeutic effect. The work provides a promising strategy for the development of TME activated NIR-II phototheranostic nanoprobes.

Abstract:The multifunctional plasmon‐pyroelectric nanoheterojunction is fabricated based on 2D BTO@Nb2C‐PEG nanosheets, which is featured with excellent photothermal property and pyroelectric catalysis‐mediated robust reactive oxygen species (ROS) generation, possessing a powerful capacity for tumor inhibition. This research offers a photo‐thermoelectric catalyst for hyperthermia‐/ROS‐mediated cancer precision therapy. Photodynamic therapy (PDT) as a non‐invasive strategy shows high promise in cancer treatment. However, owing to the hypoxic tumor microenvironment and light irradiation‐mediated rapid electron–hole pair recombination, the therapeutic efficacy of PDT is dramatically discounted by limited reactive oxygen species (ROS) generation. Herein, a multifunctional theranostic nanoheterojunction is rationally developed, in which 2D niobium carbide (Nb2C) MXene is in situ grown with barium titanate (BTO) to generate a robust photo‐pyroelectric catalyst, termed as BTO@Nb2C nanosheets, for enhanced ROS production, originating from the effective electron–hole pair separation induced by the pyroelectric effect. Under the second near‐infrared (NIR‐II) laser irradiation, Nb2C MXene core‐mediated photonic hyperthermia regulates temperature variation around BTO shells facilitating the electron–hole spatial separation, which reacts with the surrounding O2 and H2O molecules to yield toxic ROS, achieving a synergetic effect by means of combinaterial photothermal therapy with pyrocatalytic therapy. Correspondingly, the engineered BTO@Nb2C composite nanosheets feature benign biocompatibility and high antitumor efficiency with the tumor‐inhibition rate of 94.9% in vivo, which can be applied as an imaging‐guided real‐time non‐invasive synergetic dual‐mode therapeutic nanomedicine for efficient tumor nanotherapy.

Abstract:Photothermal therapy operated in the second near-infrared (NIR-II, 1000–1700 nm) window and fluorescence imaging in the NIR-IIb (1500–1700 nm) region have become the most promising techniques in phototheranostics. Their combination enables simultaneous high-resolution optical imaging and deep-penetrating phototherapy, which is essential for high-performance phototheranostics. Herein, carboxyl-functionalized small organic photothermal molecules (Se-TC) and multi-layered NIR-IIb emissive rare-earth-doped nanoparticles (NaYF4:Yb,Er,Ce@NaYF4:Yb,Nd@NaYF4, RENP) were rationally designed and successfully synthesized. Then, high-performance hybrid phototheranostic nanoagents (Se-TC@RENP@F) were easily constructed through the coordination between Se-TC and RENP and followed by subsequent F127 encapsulation. The carboxyl groups of Se-TC can offer strong binding affinity towards rare-earth-doped nanoparticles, which help improving the stability of Se-TC@RENP@F. The multilayered structure of RENP largely enhance the NIR-IIb emission under 808 nm excitation. The obtained Se-TC@RENP@F exhibited high 1064 nm absorption (extinction coefficient: 24.7 L g−1 cm−1), large photothermal conversion efficiency (PCE, 36.9%), good NIR-IIb emission (peak: 1545 nm), as well as great photostability. Upon 1064 nm laser irradiation, high hyperthermia can be achieved to kill tumor cells efficiently. In addition, based on the excellent NIR-IIb emission of Se-TC@RENP@F, in vivo angiography and tumor detection can be realized. This work provides a distinguished paradigm for NIR-IIb-imaging-guided NIR-II photothermal therapy and establishes an artful strategy for high-performance phototheranostics.

Abstract:Photothermal therapy (PTT) is a promising approach for treating tumors that offers multiple advantages. Nevertheless, its practical use in clinical settings faces several limitations, such as suboptimal delivery efficiency, uneven heat distribution, and challenges in predicting optimal treatment duration. In addition, the localized hyperthermia generated by the PTT method to induce cell apoptosis can result in the production of excessive reactive oxygen species (ROS) and the release of inflammatory cytokines, which can pose a threat to the healthy tissues surrounding the tumor. To address the above challenges, we designed an integrated H2 delivery nanoplatform for multimodal imaging H2 thermal therapy. The combination of the second near‐infrared window (NIR‐II) fluorescence imaging (FL) agent (CQ4T) and the photothermal and photoacoustic (PA) properties of Ti3C2 (TC) enables real‐time monitoring of the tumor area and guides photothermal treatment. Simultaneously, due to the acid‐responsive H2 release characteristics of the nanoplatform, H2 can be utilized for synergistic photothermal therapy to eradicate tumor cells effectively. Significantly, acting as an antioxidant and anti‐inflammatory agent, Ti3C2‐BSA‐CQ4T‐H2 (TCBCH) protects peritumoral normal cells from damage. The proposed technique utilizing H2 gas for combination therapies and multimodal imaging integration exhibits prospects for effective and secure treatment of tumors in future clinical applications. Abstract:Photothermal therapy (PTT) has gained significant attention as a non-invasive treatment in clinical oncology. However, the translation of PTT into clinical practice remains constrained by three fundamental limitations: acquired thermal tolerance in tumor cells, restricted light penetration depth in biological matrices, and insufficient therapeutic outcomes from single-modality treatment. To address these issues, a strategy for forming in situ complexes between near-infrared-II (NIR-II) photothermal agents and proteins is developed, aimed at damaging protein conformation and enhancing PTT effectiveness. We developed a nanoplatform called PCy-SF, consisting of the NIR-II photothermal polymer (PCy) and sorafenib (SF). PCy-SF responds to the tumor microenvironment (TME), specifically releasing Cy-CHO and sorafenib from the assemblies. The released Cy-CHO covalently binds to proteins, forming Cy-Protein complexes that activate NIR-II fluorescence, facilitating NIR-II imaging-guided photothermal therapy. Concurrently, the released SF intensifies microvascular damage, synergizing with PTT for enhanced therapeutic efficacy. Notably, PCy-SF induces a strong anticancer immune response, effectively suppressing tumor recurrence and metastasis. This study introduces a promising protein deactivation strategy for achieving mild-temperature PTT, offering broader applicability of PTT and insights for sensitizing tumors to photothermal therapy. Together, this innovative approach combining NIR-II photothermal agents with protein complexation and a responsive nanoplatform enhances PTT precision and efficacy, demonstrating significant potential in the field of cancer nanomedicine.

Abstract:Photothermal therapy is a very promising treatment method in the field of cancer therapy. The photothermal nanomaterials in near-infrared region (NIR-I, 750-900 nm) attracts extensive attention in recent years because of the good biological penetration of NIR light. However, the penetration depth is still not enough for solid tumors due to high tissue scattering. The light in the second near-infrared region (NIR-II, 1000-1700 nm) allows deeper tissue penetration, higher upper limit of radiation and greater tissue tolerance than that in the NIR-I, and it shows greater application potential in photothermal conversion. This review summarizes the photothermal properties of Au nanomaterials, two-dimensional materials, metal oxide sulfides and polymers in the NIR-II and their application prospects in photothermal therapy. It will arouse the interest of scientists in the field of cancer treatment as well as nanomedicine.

Abstract:In the second near-infrared (NIR-II) biowindow, multimodal optical imaging-guided precise antitumor therapy is a novel strategy for high-efficiency tumor theranostics, however, the all-in-one dual NIR-II photoacoustic (NIR-II PA) and NIR-II fluorescence (NIR-II FL) nanoprobes have been rarely reported mainly due to the short of a simple and universal design approach. Herein, a NIR-II PA/NIR-II FL imaging-adjustable nanozyme (HSC-2) is designed and developed to guide precise photothermal-catalytic synergistic therapy. Based on the ionic liquids adsorption capacity, the electronic structure of zeolite nano-Beta (three dimensional 12-ring pore system and large surface area) can be turned from the indirect band gap to direct band gap via doping carbon in the framework, resulting in outstanding NIR-II FL emission characteristics. As the silicon etching reaction proceeds, HSC-2 shows superior dual-modal NIR-II PA/NIR-II FL imaging performance facilitated by the optimal silicon-to-carbon ratio, simultaneously ensuring efficient tumor photothermal therapy (PTT) in the NIR-II window. Impressively, the peroxidase-mimic activity of HSC-2 in the tumor microenvironment could be further remarkably enhanced by its photothermal effect, leading to excellent synergistic PTT/catalytic therapy. Moreover, the HSC-2 exhibits dual-enzyme activity, and its catalase-like property could effectively eliminate excessive ROS for protection of the normal cells.

Abstract:Abstract Photothermal therapy has attracted extensive attentions in cancer treatment due to its precise spatial-temporal controllability, minimal invasiveness, and negligible side effects. However, two major deficiencies, unsatisfactory heat conversion efficiency and limited tissue penetration depth, hugely impeded its clinical application. In this work, hollow carbon nanosphere modified with polyethylene glycol-graft-polyethylenimine (HPP) was elaborately synthesized. The synthesized HPP owns outstanding physical properties as a photothermal agent, such as uniform core-shell structure, good biocompatibility and excellent heat conversion efficiency. Upon NIR-II laser irradiation, the intracellular HPP shows excellent photothermal activity towards cancer cell killing. In addition, depending on the large internal cavity of HPP, the extended biomedical application as drug carrier was also demonstrated. In general, the synthesized HPP holds a great potential in NIR-II laser-activated cancer photothermal therapy.

Abstract:Photothermal therapy is a promising phototherapeutic modality that has been widely studied in cancer therapy. However, because of the influence of heat shock protein (HSP), the therapeutic efficacy of photothermal therapy (PTT) is significantly suppressed. To improve the therapeutic efficacy, different tumor-specific therapeutic modalities have been chosen to combine with PTT. However, most of them rely on endogenous stimuli to trigger combination therapy, which may suffer from the issue of incomplete activation. Herein, we develop a PTT/thermodynamic combination therapeutic nanosystem whose therapeutic process is controlled by an external stimulus, near-infrared (NIR) light. The nanosystem (ADPPTN) is composed of a second NIR (NIR-II) fluorescent semiconducting polymer (SP) (DPPT) as the core, and a carboxyl group-decorated amphiphilic copolymer (PSMA-PEG) as the shell with an azo-containing compound (AIPH) loaded via electrostatic interaction. Under 808 nm laser irradiation, DPPT can generate heat to conduct PTT, while the elevated temperature may further trigger the release of AIPH radicals, conducting thermodynamic therapy (TDT). In addition, the NIR-II fluorescence signal emitted from DPPT can light the tumor. Compared with the nanoparticles without AIPH (DPPTN), ADPPTN has better anticancer efficacy under laser irradiation both in vitro and in vivo. Thus, our study provides an NIR-II fluorescence imaging-guided PTT/TDT combination therapeutic nanosystem for efficient cancer theranostics.
Precise Synergistic Photothermal Therapy Guided by Accurate Temperature‐Dependent NIR‐II Fluorescence Imaging

Abstract:A phototheranostic nanoparticle, 17‐RF@Ag2Se, and a NIR‐II fluorescence imaging‐photothermal therapy linkage apparatus have been developed to achieve high‐precision tumor ablation. This approach significantly improves tumor killing efficacy and reduces damage to normal tissues through synergistic photothermal therapy with an HSP90 inhibitor (17‐AAG) under the supervision of accurate temperature‐dependent NIR‐II fluorescence imaging. Photothermal therapy (PTT) is considered a promising treatment strategy for solid tumors. However, local hyperthermia (over 45°C) during PTT can cause severe side effects in neighboring healthy tissues. PTT with accurate temperature feedback is a compelling strategy to ablate tumors and reduce side effects, but it still faces challenges. Here, a new kind of phototheranostic nanoparticle, namely 17‐RF@Ag2Se is developed, enabling in vivo NIR‐II fluorescence tracking, PTT and fluorescence nanothermometry as well as synergistic heat‐shock protein (HSP) inhibition. Precise PTT with high spatiotemporal resolution is achieved with the help of the designed NIR‐II fluorescence imaging‐photothermal therapy linkage apparatus. Upon intravenous injection, 17‐RF@Ag2Se is specifically accumulated in tumors targeted by the overexpressed integrin αvβ3, which is monitored by NIR‐II fluorescence imaging of Ag2Se QDs. Further, the release of HSP inhibitor, tanespimycin (17‐AAG), enhances the thermosensitivity of tumor cells. Subsequently, the internal temperature of the tumor is precisely monitored and adjusted during PTT via the temperature‐dependent NIR‐II fluorescence feedback of Ag2Se QDs and the linkage apparatus calibration, thereby achieving efficient and safe tumor PTT. Also, the results present a new method for accurate temperature monitoring and control in vivo, which can be applied to other biomedical studies.

Abstract:High biocompatibility cell membrane of over‐expression transferrin camouflaged polymer nanoparticles (SPN‐TF) exhibit strong light absorption and excellent photothermal conversion in NIR‐II optical window with superior photostability. Furthermore, SPN‐TF nanoparticles can efficiently be taken up by cancer cells and possess outstanding anticancer ability in vitro and in vivo under 1064 nm laser irradiation. The SPN‐TF nanoparticles exhibit an excellent photothermal conversion ability, predominant targeting efficiency, and superb PTT for anti‐cancer both in vitro and in vivo. Tumor‐selective near infrared‐induced photothermal therapy is a successful approach for ablation of malignant and cancerous cells. In the NIR region, tissue penetration depth in the near‐infrared second region (NIR‐II, 1000–1700 nm) is deeper than that of the traditional near‐infrared first region (NIR‐I, 700–900 nm) for the lower scattering and absorption. However, there are few photothermal conversion agents (PCAs) in the NIR‐II. And these PCAs are quickly identified by the immune system and rapidly cleared by the reticuloendothelial system (RES), leading to low accumulation in tumor, which prevents further advanced clinical practice. In this work, the conjugated polymer, PBTPBFTDPP, is prepared and assembled into nanoparticles (SPN). Then, the genetically engineered cell membranes are coated on the surface of SPN to obtain the positive targeting ability (SPN‐TF). The obtained SPN‐TF nanoparticles exhibit an excellent photothermal conversion ability, predominant targeting efficiency, and superb photothermal therapy for anticancer both in vitro and in vivo. These works will inspire more translational work on SPN for future clinical applications.

Abstract:A variety of approaches have been investigated in the development of photothermal (PTT) and photodynamic (PDT) therapies for cancer treatment. However, when used in isolation, both PTT and PDT suffer from intrinsic drawbacks, resulting in unsatisfactory therapeutic efficacy. We describe here, a multifunctional nanoplatform, enabling the effective combination of PTT and PDT, triggered by a single 808nm-NIR light. This platform is based on MoO2 nanoparticles, which can effectively transform NIR light into heat. By attaching molecules of indocyanine green (ICG), a hydrophilic and biocompatible NIR-mediated photosensitizer, to the surface of the particles, both photothermal and photodynamic effects can be achieved. Using an in vitro study with 4T1 cancer cells, we have demonstrated that irradiating the particles with 808nm NIR can trigger ROS production and create heat within cells. Both mechanisms acting synchronously are likely to result in superior in vivo efficacy in tumor phototherapy. (C) 2019 Elsevier Ltd. All rights reserved.

Abstract:Second near-infrared (NIR-II,1000 ∼ 1700 nm) therapeutic window presents an increased tissue penetration and elevated maximal permissible exposure in the application of photothermal therapy (PTT). However, the lack of NIR-II photothermal conversion agents (PCAs) limit their further development. In this work, we rationally designed and successfully developed three novel indolium-like heptamethine cyanine dyes (NFs) by installing N,N- diethylamino on the terminal ends of a conjugated polyene backbone and replacing the middle chlorine atom with o- mercapto benzoic acid and p- mercapto benzoic acid. Notably, NF2 with stronger rotating group encapsulated in organic nanoparticles (NF2 NPs) exhibited high photothermal conversion efficiency (PCE), which could come up to (61.3 %). Then we conducted serial experiments to further investigate PTT capability of NF2 NPs 4 T1 cell line and nude mice bearing 4 T1 tumor. As expected, the resulting NF2 NPs presented the excellent photothermal conversion ability and superb PTT effect both in vivo and in vitro. This study will inspire more work for future design and clinical applications of NIR-II therapeutic agents.

Reported here is an "all-in-one" phototheranostic platform based on near-infrared II (NIR-II) dual-plasmonic Au@Cu2- x Se core-shell nanocrystals (dpGCS NCs) for combined photoacoustic (PA)/photothermal (PT) imaging-guided chemodynamic therapy (CDT)/photocatalytic therapy (PCT)/photothermal therapy (PTT) all triggered by a single NIR-II laser. The dpGCS NCs feature excellent NIR-II plasmonic and PT properties, which guarantee their capabilities of NIR-II PA and PT imaging for real-time visual observation of tumor size and location during cancer treatment. Additionally, the TME-activated in situ •OH production via dpGCS NC-catalyzed Fenton-like reaction is further enhanced by the NIR-II irradiation, while photoexcited plasmonic hole-induced formation of extra •OH is also evidenced for PCT. Both in vitro and in vivo experiments confirm remarkable therapeutic efficacy of the present phototheranostic platform under NIR-II laser through the CDT/PCT/PTT trimodal combination therapy, achieving complete inhibition of tumor growth in tumor-bearing mice after administration of dpGCS NCs plus a single NIR-II laser irradiation. This work provides a distinctive paradigm for the development of NIR-II phototheranostic platforms for imaging-guided cancer therapy using a single laser.

Abstract:Molecular imaging-guided precision photothermal therapy has shown tremendous potential for invasively thermal ablation with solid tumors. However, it is still a pressing clinical need to develop precise and deep-tissue multi-scale imaging technologies easily integrated into a single system to boost accurate photothermal treatments. Herein, a SERS/NIR-II optical nanoprobe assembled by gold nanostars, Raman molecular tags, and silver sulfide quantum dots through silica bridges (named AuDAg2S) is reported that allows the combination of the fingerprint-style surface-enhanced Raman scattering (SERS) imaging and second region near-infrared (NIR-II) in-depth biological fluorescence imaging. It can also yield strong localized surface plasmon resonance for a satisfactory photothermal conversion efficiency of 67.1% at 1064 nm to effectively kill CT26 colon cancer cells. Moreover, the AuDAg2S nanoprobes can enhance the detection sensitivity up to the picomolar level and increase the depth of the NIR-II fluorescence signal up to 1 cm under the excitation of the near-infrared laser. As a result, the AuDAg2S nanoprobes can achieve multidimensional tumor imaging in levels of living subjects, histology, and single cell, and further guided deep NIR-II photothermal anti-tumor therapy in vivo. This work may offer a promising approach for SERS/NIR-II whole optical imaging for deep tumor photothermal treatment in future clinical applications.
A self-assembled nanoplatform based on Ag2S quantum dots and tellurium nanorods for combined chemo-photothermal therapy guided by H2O2-activated near-infrared-II fluorescence imaging

Abstract:A nanoplatform based on Ag2S quantum dots (QDs) and tellurium nanorods (TeNRs) was developed for combined chemo-photothermal therapy guided by H2O2-activated near-infrared (NIR)-II fluorescence imaging. Polypeptide PC10AGRD-modified TeNRs and Ag2S QDs were co-encapsulated in 4T1 cell membrane to prepare a nanoplatform (CCM@AT). Ag2S QDs and TeNRs in the CCM@AT were used as a fluorescence probe and photosensitizer, and a chemotherapeutic prodrug and quenching agent to quench the fluorescence of Ag2S QDs, respectively. After the CCM@AT was specifically targeted to the tumor site, the TeNRs were dissolved by the high concentration of H2O2 at the tumor site to light up the fluorescence of Ag2S QDs for NIR-II fluorescence imaging. In addition, the generated toxic TeO66- molecules decreased ATP production by selective cancer chemotherapy, which is beneficial for photothermal therapy. The elevated temperature due to photothermal therapy in turn promoted the chemical reaction in chemotherapy. In vitro and in vivo toxicity results showed that the CCM@AT possesses high biocompatibility. Compared to single photothermal therapy and chemotherapy, the synergistic chemo-photothermal therapy can effectively suppress the growth of 4T1 tumor. This all-in-one nanoplatform provides a boulevard for the combination therapy of tumors guided by NIR-II fluorescence imaging. STATEMENT OF SIGNIFICANCE: NIR-II fluorescence imaging shows the characteristics of low tissue absorption, reflection, and scattering, which can greatly reduce the influence of autofluorescence in vivo. However, the non-negligible effect of autofluorescence is still observed in fluorescence imaging in vivo. Therefore, there is an urgent need to develop a strategy of controlled release of fluorescence for accurate imaging and tumor therapy. Here, Ag2S quantum dots (QDs) with NIR-II fluorescence emission and good photothermal conversion efficiency are used as a fluorescence probe and photosensitizer, and tellurium nanorods (TeNRs) are used as a chemotherapeutic prodrug and quenching agent to quench the fluorescence of Ag2S QDs. This multiple nanoplatform provides an inspiration for the combination therapy of tumor guided by NIR-II fluorescence imaging.

Abstract:Integrating multiple functionalities of near-infrared second window fluorescence imaging (NIR-II FLI), chemotherapy, and photothermal treatment (PTT) into a single molecule is desirable but still a highly challenging task. Herein, inspired by the results that hyperthermia can enhance the cytotoxicity of some alkylating agents, we designed and synthesized the novel compound NM. By introducing nitrogen mustard's active moiety bis(2-chlorethyl)amino into Donor-Acceptor-Donor (D-A-D) electronic structure, the unimolecular system not only behaviored as a chemotherapeutic agent but also exhibited good PTT and NIR-II FLI abilities. The hydrophobic agent NM was encapsulated by DSPE-PEG2000 to generate the nano-platform NM-NPs. The current study on in vitro and in vivo experiments indicated that NM-NPs make vessels visualize clearly in the NIR-II zone and achieve complete tumor elimination through chemo-photothermal synergistic treatment. Overall, this study provides a new innovative strategy for developing superior, versatile phototheranostics for cancer theranostics.

Abstract:We designed novel diketopyrrolopyrrole polymer based nanoparticles (DPP-IID-FA), which exhibited strong light absorption and excellent photothermal conversion in the NIR optical window, and displayed high biocompatibility and photostability. Furthermore, our nanoparticles could be efficiently uptaken by cancer cells and exhibited outstanding anticancer ability both in vitro and in vivo under NIR-II laser irradiation.

Abstract:As a promising anti-tumoral approach, photothermal therapy (PTT) with NIR-II (1000–1700 nm) absorption could penetrate deeper and generate more efficient hyperthermia in contrast to traditional PTT under 808 nm irradiation. As for optical tracking, NIR-II fluorescence imaging (FI) and NIR-II photoacoustic imaging (PAI) are the two emerging research hotspots. Unfortunately, each single modality has its intrinsic limitations and current research to integrate them in one-component system is still in infancy. Our development of all-in-one nanoplatforms aims to ultimately improve the efficacy of targeted hyperthermia, and incorporate the benefits of NIR-II FI and NIR-II PAI for tracking guidance. Herein, we designed a novel semiconducting polymer P2, and further encapsulated it with amphiphilic PEGylated phospholipid modified with a targeting peptide RGD to ultimately generate P2 based nanoparticles (P2NPs). As a single-component phototheranostic nanoplatform, P2NPs possessed intense NIR-II absorption, excellent NIR-II fluorescence, strong NIR-II PA signals, and high local hyperthermia capability simultaneously. Notably, due to the favorable binding capacity to osteosarcoma (OS) cells, in vitro studies revealed outstanding PTT efficacy of P2NPs against 143B cells under 1064 nm laser. Further work validated excellent dual-modal NIR-II FI/NIR-II PAI tracking and strong PTT tumor ablation in both subcutaneous and orthotopic OS mice models with negligible biotoxicity in vivo . Consequently, these findings warranted P2NPs as a promising 1064 nm activatable all-in-one nanoplatform to perform dual-modal NIR-II FI/NIR-II PAI-guided orthotopic tumoral treatment, providing a new approach for theranostics, therapeutic tracking, and the beyond in other diseases.

Abstract:NIR-IIa fluorescence imaging (FI) and NIR-II photothermal therapy (PTT) have gained popularity due to the advantages of high temporal and spatial resolution and deep penetration. However, the hyperthermia (>48 °C) of conventional PTT with nonspecific warming and thermal diffusion may inevitably cause damage to healthy tissues or organs surrounding the tumor. Therefore, it is highly desirable to provide effective cancer treatment by implementing mild photothermal therapy (mPTT) at mild temperatures with lower laser power density. Here, the nanotheranostic platform FN@P-GA NPs with NIR-II absorption and NIR-IIa emission was developed by constructing J-aggregates. FN@P-GA possesses good biocompatibility, favorable NIR-IIa FI performance, decent stability, and high photothermal conversion efficiency (57.6 %), which lays a solid foundation for FI-guided mPTT. Due to its ability to effectively down-regulate the expression of HSP90 and reduce cellular thermoresistance to kill cancer cells, FN@P-GA successfully achieved NIR-IIa FI-guided mPTT and demonstrated its potent anti-tumor effect under 1064 nm laser irradiation at mild temperature and low power density (0.3 W/cm 2 ).
