Abstract:It is an undeniable fact that the effect of marine toxins becomes increasingly serious to people. Paralytic shellfish poisons (PSP) are well-known sodium channel-blocking marine toxins, which block the conduction of nerve impulses and lead to a series of neurological disorders symptoms. Similarly, as inhibitors of serine/threonine protein phosphatases type 1 (PP1) and 2A (PP2A), diarrheic shellfish poisons (DSP) can destroy cytoskeleton and affect intracellular signal transduction, which causes some symptoms of gastrointestinal food allergy. Since the conventional methods to detect marine toxins had some disadvantages (e.g., complicated pretreatment, time-consuming detection, huge instruments and off-site test), bionic electronic eye (Bionic e-Eye) was designed and studied for the in-field fast measurement and real-time on-line analysis to marine toxins. Bionic e-Eye installed the homemade software-iPlate and completed the integration of image acquisition and further data processing. With the cooperation of ELISA, Bionic e-Eye used two color models-HSV and RGB to evaluate its bioanalytical performance in saxitoxin (STX) assay and okadaic acid (OA), which were the standard representations of PSP and DSP, respectively. Moreover, the microtiter plate reader (MTPR) was compared with Bionic e-Eye in the detection of marine toxins. The results demonstrated that Bionic e-Eye using saturation had the comparable precision, dynamic range, detection limit and sensitivity to MTPR in the two assays. Therefore, Bionic e-Eye has great potential to be a field-test platform and becomes the supplement of conventional off-site detection methods in measurement domain of marine toxins. Design of In￣Suit High Throughput Pretreatment and Detection System for Diarrhetic Shellfish Poisons

Abstract:In order to achieve the requirement of diarrhetic shellfish poison(DSP)in￣suit detection,we design a high throughput pretreatment system and a mobile terminal based detection system. Our system can make fast and accurate DSP detection by analyzing image of chromogenic reaction come from ELISA method. The rate of recovery by our pretreatment system and by hand are 89% and 93% respectively,it means that our pretreatment system can meet the follow￣up detection requirement. Detect the prevented samples by our detection system, the standard deviation is approximately 0.13 and average rate of recovery is 89.5%,it can satisfy the accuracy and repeatability of detection. In the detection of actual samples,comparing our results with the results come from microplate reader,it shows that our pretreatment and detection system can do a fast and accurate in￣suit work and provide a new method for the detection.
New Handheld Strip Analyzer for On-Site Rapid Detection of Paralytic Shellfish Poisoning Based on a Smartphone

Abstract:Saxitoxin( STX) are common marine toxins which can affect human health through food chain by accumu-lating in shellfishes. It is urgently required to have a convenient method for on-site detection of marine toxins to a-void the poisoning incidents that have occurred frequently. In this work,a handheld analysis meter system coopera-ting with competitive immunoassay strips is designed for rapid detection of STX. The smartphone is used as light de-tector,for image acquisition and data processing by a specific application. The 3D-printed portable accessory of sm-artphone is fixed for test strip. The method is simple and fast,with a detection limit of STX of 5.2 ng/mL,respec-tively. It can provide quantitative analysis of STX in the range of 5.2 ng/mL~100 ng/mL. Based on the smartphone system and biochemical analysis,it will be promising tool for on-site rapid detection for paralytic shellfish poisoning.

Abstract:Saxitoxin (STX) is one of the paralytic shellfish poisons (PSP) that endanger people’s health. It is necessary to develop methods for the on-site rapid detection for STX in order to prevent safety accidents. An enzyme-linked immunosorbent assay (ELISA) is timesaving and effective, but it is not suitable for large-scale in-field tests due to the expensiveness of commercial ELISA kits and the bulkiness of a microtiter plate reader (MTPR). In this study, a portable smartphone-based colorimetric analyzer (SBCA) with a cost-effictive enhanced gold nanoparticle-based ELISA (EGNB-ELISA) was proposed for STX detection. In a bicinchoninic acid (BCA) protein assay (R2 = 0.9939) and a glucose assay (R2 = 0.9937), SBCA was shown to be in good agreement with MTPR. EGNB-ELISA had a 12.5-fold lower detection limit (0.4 ng/mL) and a lower detection range (1 - 50 ng/mL, Y = 0.4037X + 0.3564, R2 = 0.9797) than the classical ELISA. The recovery rate ranged over 89.1 - 112.2%. The whole detection system, combining both homemade SBCA and ENGB-ELISA, is expected to satisfy the needs of on-site STX sample tests to guarantee seafood safety.
The Research of Biosensor and Bionic e-Eye for Rapid Detection of Okadaic Acid on-Site

Abstract:Based onEnzyme-linked immunosorbent assay(ELISA),test kit,namely,enzyme sensor and the corre-sponding instrument was prepared,Bionic e-Eye for rapid,effective and quantitative detection of okadaic acid(OA) on-site. The detection limit of the kit is 0.19μg/L and the range is among 0.2μg/L and 5μg/L. The recovery rate is among 75%and 125%and the accuracy is within±15%. The performance of Bionic e-Eye is consistent with micro-plate reader and high performance liquid chromatography(HPLC). In addition,without artificial calculation,the instrument can show the results directly on the screen,which is very suitable for rapid detection,and will be a new technology for rapid and quantitative detection of OA on site.

Abstract:Okadaic acid (OA) is one of the marine toxins that are widely distributed and harmful to humans. However, the current detection methods for OA involve complex procedures, need long detection time, and rely on large-scale laboratory equipment. In this work, a multi-channel handheld automatic spectrometer (MHAS) based on a spectral sensor was developed with the advantages of small size, simple operation and low cost. It could achieve rapid detection within 30 s and a wide spectral detection range of 470-780 nm with a broadband LED as the light source and a microplate containing 8 wells as a sample cell. Moreover, through the combination of gold nanoparticles (AuNPs) and aptamer-OA34, a highly sensitive and rapid system for OA detection was established with a LOD of 1.80 μg L-1 and a wide detection range of 20-10 000 μg L-1, which is comparable to a microplate reader. Compared with other studies, the proposed MHAS realized rapid on-site detection of OA with a wider detection range, shorter detection time and higher portability. Therefore, the MHAS promises to be a stable and efficient optical detection instrument for on-site detection in the fields of food safety, disease diagnosis and environmental monitoring.

Abstract:Vitamin C (VC) maintains the activity of sulfhydrylase and the reduced state of glutathione, and acts as a detoxifying agent. It is widely found in plant tissues as well as in fresh fruits and vegetables. Accurate determination of vitamin C content is of great significance for diet and health care. In this work, MnO2 nanoflakes as the biomimetic oxidase was used to react with VC and TMB was used as a chromogenic agent. The color of the final solution reflects the concentration of VC. And the bionic E-eye as a portable in-situ detection platform was designed to analyzed the color change in 10 s by photographing. By optimizing experimental conditions, the linear range of this method for VC detection is from 7.8 μM to 62.5 μM and the detection limit 7.65 μM which is much lower than other methods. Besides, this methods shows good selectivity, short reaction time (10 min) and also was used for VC detection in artificial urine samples, which shows great potential in disease monitoring.

Abstract:A novel system for fast poison detection based on electrical cell-substrate impedance sensing was proposed and fabricated for resolving the problems of marine diarrheic shellfish poison detection. Comparing with tranditional methods,the system has the performance of simple operation,high consistency and high throughput. In this study,the design of system hardware and software were introduced and then its performance was tested. Finally, toxin detection experiments with Neuro-2a,DSP and OA were used to certify the availability of system. The results showed that the system had some attributes such as fast,long-time monitoring,no invasion and high throughput and applied a new detection platform for DSP of marine productions.

Abstract:Molecular diagnosis of genome is one of the major methods for pathogens detection. The commonly used PCR method can realize an exponential amplification of the target gene but is time-consuming. In this work, we proposed a duplex and visual method using rapid PCR combined with molecular beacons to specifically detect two kinds of shrimp pathogens in one reaction tube. We only need to observe the fluorescence change of the reaction tube with naked eye to determine the result. A home-made automatic transfer equipment allows reaction tubes shuttling quickly between two water baths to achieve rapid PCR amplification. A simple device was also designed to present the detection results easily determined with naked eye. This duplex and visual detection method is fast, low-cost and of high specificity. From DNA extraction to results judgment, only 15 min was enough. Infectious Hypodermal and Hematopoietic Necrosis Virus (IHHNV) and Vibrio parahaemolyticus (VP) are two common shrimp pathogens which were chosen as our detection objects. This method may give a possibility to conduct end-point visual duplex detection, which may make a positive influence on the pathogen prevention.

Abstract:For the in-suit detection of the paralytic shellfish poison(PSP),a system based on mobile terminal is de-signed to realize high-throughput quick analysis of the toxin. Chromogenic reaction of PSP kit with the toxin can achieve the concentration within appropriate scope. A darkroom with an area light source can provide a stable test-ing environment. A mobile terminal with high megapixel camera for data collection and processing can realize accu-rate analysis of the testing results and efficient interaction. Finally,toxin detection experiments with shellfish sam-ples used to certify the availability of the system. The results showed that the system had several attributes such as in-suit fast detection,simple operation,high sensitivity and applied a new detection platform for PSP of food.

Abstract:Okadaic acid (OA) and saxitoxin (STX) are typical toxins of diarrhetic shellfish poisoning (DSP) and paralytic shellfish poisoning (PSP), respectively, which are highly toxic marine toxins threatening human health and environmental safety. OA is a potent inhibitor of serine/threonine protein phosphatases that can cause cellular death, while STX is an inhibitor of sodium channel that can lead to neurological damage. In this work, a dual functional cardiomyocyte-based biosensor was proposed to detect DSP and PSP toxins by monitoring the viability and electrophysiology of cardiomyocytes. The results showed that the viability of cardiomyocytes was sensitive to the OA and STX, resulting in significant changes of the electrophysiological properties, including amplitude, firing rate and duration of the extracellular field potential (EFP). The detection limits of the hybrid-biosensor are as low as 7.16 ng/mL for OA and 5.19 ng/mL for STX. In summary, all of the results indicate that the dual functional cardiomyocyte-based hybrid-biosensor will be a promising and utility tool for shellfish toxin detection.

Abstract:Diarrhetic shellfish poisoning (DSP) toxins are potent marine biotoxins. It can cause a severe gastrointestinal illness by the consumption of mussels contaminated by DSP toxins. New methods for effectively and rapidly detecting DSP toxins-contaminated mussels are required. In this study, we used near-infrared (NIR) reflection spectroscopy combined with pattern recognition methods to detect DSP toxins. In the range of 950-1700 nm, the spectral data of healthy mussels and DSP toxins-contaminated mussels were acquired. To select optimal waveband subsets, a waveband selection algorithm with a Gaussian membership function based on fuzzy rough set theory was applied. Considering that detecting DSP toxins-contaminated mussels from healthy mussels was an imbalanced classification problem, an improved approach of twin support vector machines (TWSVM) was explored, which is based on a centered kernel alignment. The influences of parameters of the waveband selection algorithm and regularization hyperparameters of the improved TWSVM (ITWSVM) on the performance of models were analyzed. Compared to conventional SVM, TWSVM, and other state-of-the-art algorithms (such as multi-layer perceptron, extreme gradient boosting and adaptive boosting), our proposed model exhibited better performance in detecting DSP toxins and was little affected by the imbalance ratio. For the proposed model, the F-measure reached 0.9886, and detection accuracy reached 98.83%. We explored the physical basis for the detection model by analyzing the relationship between the occurrence of overtone and combination bands and selected wavebands. This study supports NIR spectroscopy as an innovative, rapid, and convenient analytical method to detect DSP toxins in mussels.

A Clickase-Mediated Immunoassay Based on Nanopore and Bionic Signal Labels for Ultrasensitive, Portable, and On-Site Detection of Ricin

Abstract:It is of particular importance to develop an effective method that possesses several merits simultaneously of rapid, ultrasensitive, portable, and on-site detection potential for food safety detection. Herein, we propose a clickase-mediated immunoassay based on nanopore and bionic signal labels for the detection of ricin. The introduction of Cu/Cys clickase and nanopore simultaneously effectively addressed the inherent limitations of natural enzymes and colorimetric signal output, respectively. Using this method, bionic signal labels can be easily formed through DNA and Gram-positive bacterial cell wall terminal peptide fragments (labeled by alkynyl and azide, respectively) and vancomycin. Translocation of the D-P@vancomycin through the nanopore generated highly specific oscillation current traces. This method showed a great on-site detection potential and superior analytical performance owing to the combination of the specificity of antibodies, high CuAAC click reaction catalytic efficiency of clickase, ultrasensitivity of the nanopore, and high signal resolution of D-P@vancomycin. Moreover, the practical applicability of the established method was also verified, achieving a limit of detection (LOD) down to 200.9 ag/mL with a wide linear relationship under the optimized conditions. In conclusion, this method is promising for rapid, portable, ultrasensitive, and on-site food safety detection.

Abstract:Okadaic acid (OA) is a marine biotoxin that accumulates in seafood and can cause diarrheic shellfish poisoning if consumed. Accordingly, many countries have established regulatory limits for the content of OA in shellfish. At present, methods used for the detection of marine toxins are time-consuming and labor-intensive. In order to realize rapid, simple, and accurate detection of OA, we developed a novel microfluidic immunosensor based on magnetic beads modified with a highly specific and sensitive monoclonal antibody (mAb) against OA that is used in conjunction with smartphone imaging to realize the rapid detection of OA in shellfish. The method achieves on-site detection results within 1 h with an IC 50 value of 3.30 ng/mL for OA and a limit of detection (LOD) of 0.49 ng/mL. In addition, the analysis of real samples showed that the recoveries for spiked shellfish samples ranged from 84.91% to 95.18%, and the results were confirmed by indirect competitive enzyme-linked immunosorbent assay (icELISA), indicating that the method has good accuracy and precision. Furthermore, the results are reported in a specially designed smartphone app. The microfluidic immunosensor has the advantages of simple operation, rapid detection, and high sensitivity, providing a reliable technical solution for detecting OA residues in shellfish.

A convenient and efficient method is urgently required to perform on-site detection of marine toxins to avoid frequent poisoning incidents. In this work, a smartphone-based system cooperating with competitive immunoassay strips was designed for rapid on-site detection of OA and STX. The smartphone was employed as a light detector for image acquisition and data processing. The 3D-printed portable accessory of the smartphone was utilized to fix the test strips. The homemade APP -iStrip used the pixel scan to obtain the valley value of the pixel curve as the output of the strip. The strip analytical method was easy to operate with a detection limit of 2.800 ng ml(-1) for OA and 9.808 ng ml(-1) for STX. Moreover, with the optimization of the antigen-antibody ratio on the strip, it provided quantitative analysis of OA and STX in the range of 3-20 ng ml (-1) and 10-100 ng ml(-1), respectively. Taking advantage of the smartphone and biochemical analysis, the strip analytical system will be a promising tool for on-site rapid detection of these two toxins.

Abstract:Salmonella, a common foodborne pathogen, causes many cases of foodborne illness and poses a threat to public health worldwide. Immunological detection systems can be combined with nanoparticles to develop sensitive and portable detection technologies for timely screening of Salmonella infections. Here, we developed an antibody-probe-based immuno-N-hydroxysuccinimide (NHS) bead (AIB) system to detect Salmonella. After adding the antibody probe, Salmonella accumulated in the samples on the surfaces of the immuno-NHS beads (INBs), forming a sandwich structure (INB-Salmonella-probes). We demonstrated the utility of our AIB diagnostic system for detecting Salmonella in water, milk, and eggs, with a sensitivity of 9 CFU mL-1 in less than 50 min. The AIB diagnostic system exhibits highly specific detection and no cross-reaction with other similar microbial strains. With no specialized equipment or technical requirements, the AIB diagnostic method can be used for visual, rapid, and point-of-care detection of Salmonella.
Naked-eye based point-of-care detection of E.coli O157: H7 by a signal-amplified microfluidic aptasensor

Abstract:Fast and sensitive detection of E.coli O157: H7 is significantly essential for clinical management as well as for transmission prevention during disease outbreaks. Though many types of detection strategies have been implemented for measuring E.coli O157: H7, most of them still rely on complex instruments or tedious/laborious setups, which restrict their applications in resource-limited scenarios. Herein, we introduce an eye-based microfluidic aptasensor (EA-Sensor) for fast detection of E.coli O157: H7 without the assist of any instruments. We demonstrate the perfect coupling of aptamer sensing, hybridization chain reaction (HCR)-amplification and a distance-based visualized readout to quantitatively determine the pathogen concentration. We first used gel-electrophoresis assay to evaluate the system and the results proved that E.coli O157: H7 was well recognized by the aptamer and HCR could increase the signal by about 100 folds. In addition, the Aptamer specificity and signal-amplification ability were verified on the EA-Sensor for sensing E.coli O157: H7 by naked eyes. Furthermore, we demonstrated that E.coli O157: H7 in milk could be accurately and conveniently measured with good performance. With the benefits of operation integration and strategy integration, our EA-Sensor shows advantages of high specificity, easy operation, efficient amplification and visualized readout, which offers a favorable point-of-care tool for E.coli O157: H7 or other pathogen detection in resource-constrained settings.

Abstract:Bionic electronic eye (Bionic e-Eye), a developed smartphone-based colorimetric reader system, consists of smartphone or pad (iPhone 4s or iPad 3) as detection instrument and portable accessory as illumination provider integrating with a wide-angle lens, a piece of lowest-power electro luminescent and a custom-made dark hood. A 96-well microtiter plate (MTP) was positioned on the electro luminescent and Bionic e-Eye captures the detection images by the back camera of smartphone. Being similar to human visual system, the hue, saturation and value (HSV, also called hex cone model) color model was employed in image processing algorithm of Bionic e-Eye. Optimized system dimension was determined by the system steadiness experiment of different photograph distances. Moreover, the commercially available bicinchoninic acid (BCA) protein assay and cell counting kit (CCK8) assay were carried out to evaluate this Bionic e-Eye. Analytical performance of Bionic e-Eye had the better precision, higher sensitivity than microtiter plate reader (MTPR) and previous smartphone-based colorimetric reader for both two assays. Also, Bionic e-Eye using optical image detection had simultaneous and synchronous working mode, while MPTR using machine moving detection had asynchronous working mode in high throughput detection. Therefore, Bionic e-Eye will be an ideal point-of-care (POC) colorimetric detection device in the field of clinical application, industrial quality control, environment monitoring, and food assessment.

Abstract:Addressing the critical health concerns posed by domoic acid (DA), a neurotoxic compound produced by toxic marine algae and bioaccumulated in shellfish, necessitates the development of a rapid, precise, and robust detection system. Traditional DA detection methods have stability and sensitivity issues, which hinder effective toxin detection. To overcome these limitations, we developed a novel direct competitive enzyme-linked immunosorbent assay (dc-ELISA) platform that utilizes peptide-immobilized magnetic beads (MGBs/peptide). The affinity peptides identified through phage display and chemically synthesized with biotin labels present an innovative alternative to conventional antibodies for ELISA applications. Streptavidin-modified MGBs were used as the bioreceptor carriers to facilitate magnetic separation and simplify sample preparation, making the MGB/peptide-based dc-ELISA platform an ideal tool for comprehensive monitoring efforts. The developed platform exhibits a detection range of 0.5-10 ng mL-1 and a low limit of detection of 0.29 ng mL-1, offering enhanced sensitivity and cost-effectiveness. Moreover, our developed dc-ELISA demonstrated a high recovery rate when validated with DA-spiked CRM-mussel samples. This method overcomes the limitations of traditional detection techniques and offers a scalable and efficient approach to marine toxin surveillance with improved marine environmental monitoring and public health management.

Abstract:With harmful algal blooms, marine food poisoning caused by marine biotoxins frequently occurs and is life-threatening if severe. However, the conventional detection methods of marine toxins have a few limitations: low sensitivity and high-cost. Therefore, it is necessary to establish a fast and sensitive on-site detection method for real seafood sample. Biosensors based on aptamers, antibodies, and cells have been applied in marine toxins monitoring. This review presents the classification and toxic effects of marine toxins, and recent biosensor for marine toxin detection. In addition, we have compared the superiority and limitation of these biosensors. Finally, challenges and opportunities of biosensors in food safety detection were discussed. Considering the excellent results achieved by the aptasensor in the field of detection, it seems ready to be put into practical applications.
