Abstract:Direct application of available well-defined materials for mass spectrometry analysis represents a significant step in the combination of materials science and analytical chemistry. The present study introduces a facile vacuum filtration method for directly coating different commercially available materials onto the surface of filter paper. High sensitivity and quantitation precision have been demonstrated for different analytes including therapeutic drugs, proteins and volatile compounds in various matrices using the obtained papers as substrates for paper spray mass spectrometry.

Abstract:A highly integrated polystyrene (PS) microfluidic chip coupled to electrospray ionization mass spectrometry for on-chip protein digestion and online analysis was developed. The immobilized enzymatic microreactor for on-chip protein digestion was integrated onto microchip via the novel method of region-selective UV-modification combined with glutaraldehyde-based immobilization. The micro film electric contact for applying high voltage was prepared on chips by using UV-directed electroless plating technique. A micro-tip was machined at the end of main channel, serving as the interface between microchip and mass spectrometric detector. On-chip digestion and online detection of protein was carried out by coupling the microchip with mass spectrometry (MS). The influences of methanol flow rate in side channel on the stability of spray and intensity of signals were investigated systematically. Also the influence of sample flow rate on the performance of immobilized enzymatic reactor were investigated. Stable spray was obtained at the spray voltage of 2.8-3.0kV and the methanol flow rate of 500-700nLmin(-1) with the relative standard deviation (RSD) of total ion current (TIC) less than 10%. The influence of sample flow rate on the performance of immobilized enzymatic reactor was also studied. The sequence coverage of protein identification decreased with the increase of flow rate of the sample solution. A sequence coverage of 96% was obtained with immobilized enzymatic reactor at the sample flow rate of 100nLmin(-1) with the reaction time of 8.4min. It could detect cytochrome c as low as 10μgmL(-1) with the developed system. No obvious decrease in protein digestion efficiency was observed after the chip continuously performed for 4h and stored for 15d.

Abstract:Paper spray has been developed as a fast sampling ionization method for direct analysis of raw biological and chemical samples using mass spectrometry (MS). Quantitation of therapeutic drugs in blood samples at high accuracy has also been achieved using paper spray MS without traditional sample preparation or chromatographic separation. The paper spray ionization is a process integrated with a fast extraction of the analyte from the raw sample by a solvent, the transport of the extracted analytes on the paper, and a spray ionization at the tip of the paper substrate with a high voltage applied. In this study, the influence on the analytical performance by the solvent–substrate systems and the selection of the elution methods was investigated. The protein hemoglobin could be observed from fresh blood samples on silanized paper or from dried blood spots on silica-coated paper. The on-paper separation of the chemicals during the paper spray was characterized through the analysis of a mixture of the methyl violet 2B and methylene blue. The mode of applying the spray solvent was found to have a significant impact on the separation. The results in this study led to a better understanding of the analyte elution, on-paper separation, as well as the ionization processes of the paper spray. This study also helps in establishing a guideline for optimizing the analytical performance of paper spray for direct analysis of target analytes using mass spectrometry.

Abstract:To analyze compounds in complicated matrixes using mass spectrometry, we describe a novel ambient ionization approach, termed paper assisted ultrasonic spray ionization (PAUSI). The ionization process is based on the ultrasonic vibration of the piezoelectric ceramic disk, on which the samples are placed. Porous materials are utilized to generate fine initial droplet, which could alleviate matrix effect during ionization process for complicated matrix. PAUSI was evaluated as an attractive tool to screen analytes from complicated matrixes, such as (1) bovine serum with NaCl 150 g/L, (2) viscous samples, and (3) biological fluid, without any sample preparation. Moreover, it provides great advantage in simplifying the mass spectrometry analysis process, and the ionization device is inexpensive and easy to operate.

Abstract:Paper spray ionization has been developed as a direct, fast and low-cost sampling and ionization method for qualitative and quantitative mass spectrometric (MS) analysis of complex mixtures. Analyte ions are generated by applying a high voltage and a small volume (∼10μL) of spray solvent onto a porous substrate. The sample can be preloaded onto the paper or mixed into the spray solution. The geometry of the paper and the method of supplying the necessary internal standard are important factors that affect the ionization efficiency and subsequently the sensitivity and quantitation accuracy of the analytical data. As the cut angle of the paper tip is changed, the spray plume, the total spray current and the electric field intensity at the tip all vary correspondingly, with resulting differences in signal intensity. Sample load is another important factor for obtaining a stable MS signal and accurate quantitative results. The optimal sample load was found to be dependent on the paper size. The dissolution and spray process was also investigated and analyte transfer on paper was shown to be largely associated with bulk solution flow toward the spray tip. The information gathered from these systematic studies provides guidance for the design and optimization of a disposable sample cartridge for paper spray MS, a device which potentially is suitable for fast clinical analysis, especially for point-of-care diagnostics.

Abstract:Mass spectrometry (MS) has been a powerful essential tool for proteomic analysis, due to its structural identification and throughput. However, the MS analysis of complex biological samples is severely hampered by the complexity of the samples, the low abundance and poor ionization efficiency of analytes and the coexistence of abundant interfering species in the sample matrices. An additional step to selectively enrich target proteins or peptides prior to the MS analysis is therefore critically necessary. Molecularly imprinted polymers (MIPs), as chemically synthesized receptors produced through polymerization of functional monomer(s) in the presence of a template compound, exhibit affinity and specificity to the template. Also, they are easy to prepare, stable and cost efficient. In recent a decade, MIPs have been demonstrated as promising sorbents for sample pretreatment in proteomics research. In this review, we survey recent advances in the combination of MIP-based affinity extraction and MS for targeted proteomic analysis. Various imprinting strategies for unmodified and post-translationally modified proteins and peptides are discussed. The binding properties of the prepared MIPs and the performance of the hyphenated approach are highlighted. Finally, we brief the perspectives and future challenges of the molecular imprinting technology for targeted proteomics analysis.
Detecting the Therapeutic Drugs in Blood Samples Through PDMS-printed Paper Spray Mass Spectrometry.

Abstract:In this paper, paper microfluidic channel fabricated by directly screen-printing of polydimethylsiloxane (PDMS) is proposed for paper spray mass spectrometry analysis of therapeutic drugs in the blood samples. Compared with traditional paper spray, PDMS-printed paper spray (PP-PS) allows fluid to flow to the tip of paper with less sample loss which significantly improved the signal intensity of target compounds in blood samples. As paper can reduce the matrix effect, PP-PS also has a greater advantage than electro-spray Ionization (ESI) when directly analyzing complex biological sample in terms of the detection efficiency. Linearity and limits of detection (LOD) were evaluated for five psychotropic drugs: olanzapine, quetiapine, 9-hydroxyrisperidone, clozapine, risperidone. As a result, PP-PS improved the signal intensity of the psychotropic drugs at a concentration of 250 ng/ml in blood samples by a factor of 2-5 times and lowered the relative standard deviation (RSD) by a factor of 2-5.6 times compared with traditional paper spray. And PP-PS also improved signal intensity by a factor of 9-33 times compared with ESI. Quantitative experiments of PP-PS mass spectrometry indicated that the linear range was 5-500 ng/ml and the LOD were improved by a factor of 5-71 times for all these drugs compared with traditional paper spray. In addition, PP-PS was applied to the home-made miniaturized mass spectrometer and the precursor ions of all five psychotropic drugs (250 ng/ml) in the mass spectrometry results were obtained as well. These could prove that PP-PS has the potential to analyze complex biological samples in application on the miniaturized mass spectrometer which can be used outside the laboratory.

Abstract:Paper spray ionization mass spectrometry (PSI-MS) is a new technology of mass spectrometry.This method uses low-cost paper as the carrier,and solid or liquid sample is added to a triangular paper substrate.Through the application of high voltage,the target molecules in the high voltage electric field are driven to reach the paper tip,then a charged spray for PSI-MS is generated.This technology combines the characteristics of ambient mass spectrometry (AMS) and electrospray ionization (ESI) technology,and can analyze sample or substance attached to the sample surface in an open atmospheric environment.It has many advantages such as low cost,high throughput,simple,real-time and fast,and it can also simplify the work processes,improving the usability of mass spectrometry equipment.Herein,a general overview of PSI-MS is given in this paper,including the principle,influencing factors (paper substrate,spray solution,elution mode,and internal standard),the improvement and development of the technology,and corresponding applications in various fields (i.e.biological samples,phytochemistry,variety identification,food safety,and environmental analysis) in recent years.Characteristics and advantages of this technology are introduced in details.A number of studies have proved that PSI-MS is a widely used analytical technique with great prospects.Finally,the shortcomings of PSI-MS in terms of accurate quantification,carrier selection and field analysis are discussed,and some ideas for its improvement are put forward.

Abstract:A method to directly identify proteins contained in mixtures by microcolumn reversed-phase liquid chromatography electrospray ionization tandem mass spectrometry (LC/MS/MS) is studied. In this method, the mixture of proteins is digested with a proteolytic enzyme to produce a large collection of peptides. The complex peptide mixture is then separated on-line with a tandem mass spectrometer, acquiring large numbers of tandem mass spectra. The tandem mass spectra are then used to search a protein database to identify the proteins present. Results from standard protein mixtures show that proteins present in simple mixtures can be readily identified with a 30-fold difference in molar quantity, that the identifications are reproducible, and that proteins within the mixture can be identified at low femtomole levels. Based on these studies, methodology has been developed for direct LC/MS/MS analysis of proteins enriched by immunoaffinity precipitation, specific interaction with a protein-protein fusion product, and specific interaction with a macromolecular complex. The approach described in this article provides a rapid method for the direct identification of proteins in mixtures.

Abstract:Paper spray mass spectrometry (PS-MS) is explored as a fast and convenient way for direct analysis of molecules in tissues with minimum sample pretreatment. This technique allows direct detection of different types of compounds such as hormones, lipids, and therapeutic drugs in short total analysis times (less than 1 min) using a small volume of tissue sample (typically 1 mm(3) or less). The tissue sample could be obtained by needle aspiration biopsy, by punch biopsy, or by rubbing a thin tissue section across the paper. Abstract:In this paper, particle mass spectrometry (PMS), which consists of an aerodynamic desorption/ionization (AD) source, a quadrupole ion trap (QIT) mass analyzer, and a charge detector, was exploited to characterize the protein adsorption on microparticles based on the mass variations of microparticles before and after protein adsorption. This method is simple and has low sample cost. Importantly, its mass resolution is good enough to distinguish the microparticles with and without protein. For the adsorption of bovine serum albumin (BSA) on 3 mu m porous poly styrene-divinylbenzene (poly S-DVB), the minimum mass increase that can be resolved by PMS corresponds to 128 fg (1.8 ng/cm(2)) or 1.17 x 10(6) BSA molecules on each poly S-DVB particle. With PMS, the adsorption process of BSA on poly S-DVB spheres was successfully characterized, and the obtained maximum adsorption capacity q(m) and dissociation constant K-d were consistent with that determined by the conventional depletion method. In addition, the influence of surface modification of silica particles on the enzyme immobilization was evaluated. Compared with C-4 (propyldimethylsilane), C-8 (octyldimethylsilane), and Ph (phenyldimethylchlorosilane), the CN (cyanoethyldimethylchlorosilane) functionalized silica particles were screened to be most beneficial for the immobilization of both lysozyme and trypsin.

Abstract:Paper spray is developed as a direct sampling ionization method for mass spectrometric analysis of complex mixtures. Ions of analyte are generated by applying a high voltage to a paper triangle wetted with a small volume (<10 microL) of solution. Samples can be preloaded onto the paper, added with the wetting solution, or transferred from surfaces using the paper as a wipe. It is demonstrated that paper spray is applicable to the analysis of a wide variety of compounds, including small organic compounds, peptides, and proteins. Procedures are developed for analysis of dried biofluid spots and applied to therapeutic drug monitoring with whole blood samples and to illicit drug detection in raw urine samples. Limits of detection of 50 ng/mL (or 20 pg absolute) are achieved for atenolol in bovine blood. The combination of sample collection from surfaces and paper spray ionization also enables fast chemical screening at high sensitivity, for example 100 pg of heroin distributed on a surface and agrochemicals on fruit peels are detectable. Online derivatization with a preloaded reagent is demonstrated for analysis of cholesterol in human serum. The combination of paper spray with miniature mass spectrometers offers a powerful impetus to wide application of mass spectrometry in nonlaboratory environments.

Abstract:Current detection methods for paper-based analytical devices (PADs) rely on spectroscopic and electrochemical properties, which place special requirements on the analyte or need analyte labeling. Here, ion-transmission mass spectrometry (MS) was proposed for coupling with PADs to enable rapid in situ MS analysis of the sample on paper. The sample was analyzed directly on paper via analyte ionization by ions transmitted through the paper, generated by a low-temperature plasma probe. Prior to MS analysis, the sample can be separated by paper electrophoresis or by paper chromatography, among a variety of other features offered by PADs. The versatility of this technique was demonstrated by MS analysis of a paper microarray, a mixture of amino acids, and whole blood doped with drugs on PADs.

Abstract:Paper spray for direct mass spectrometry analysis is introduced in this chapter. The fundamental study of the mechanism and the characterization of the analytical performance are reviewed for paper spray mass spectrometry (PS-MS). An important aspect in development of paper spray is its capability for quantitation at high precision for analytes in complex samples, which is important for a wide range of applications in many different fields with mandatory criteria in quantitation. Its potential for point-of-care (POC) analysis is discussed with recent implementation with miniature MS analytical systems. Three other ambient ionization methods associated with paper spray, including leaf spray, tissue spray, and extraction spray ionization are also introduced in this chapter.
Direct Analysis of Proteins in Physiological Samples with Three-Dimensional Paper-Based Isoelectric Focusing

Abstract:Point-of-care testing (POCT) of proteins is crucial for on-site and rapid diagnosis of diseases in community and family medicine. Microfluidic paper-based analytical devices (µPADs) have attracted growing attention in POCT of proteins, while the practical applications of µPADs in the analysis of proteins in physiological samples is still limited as physiological samples are usually high-salt and complex. Herein, we developed a three-dimensional (3D) paper-based isoelectric focusing (IEF) platform, which is composed of power supply, reservoirs and separation channel make by origami and stacking of cellulose acetate membrane, to simultaneously separate and enrich proteins in both low-salt samples and high-salt samples, especially for directly pretreating and analyzing proteins in physiological samples. Under the optimized experimental conditions, standard proteins (Phy and BHb) were separated in 18 min under a 36.0 V power supply and obtained 10-fold enrichment using the 3D paper-based IEF platform. Then, the capability of the 3D paper-based IEF platform with using a 12.0 V battery as power supply for direct pre-treatment of high-salt samples was proven through separating three standard proteins in saline (0.9 % NaCl) with separation resolution (SR) >1.29. Through further coupling with colorimetric and lateral flow strip measurements, the 3D paper-based IEF platform was applied to directly pre-treat and quantitatively analyze the two disease biomarkers, i.e., microalbuminuria and C-reactive proteins in clinical human serum. The analytical results had the relative deviations of <8.4% and <13.1% to clinical results, respectively. As the first report of the paper-based IEF platform for directly pretreating high-salt samples, this work proposes a new strategy to minimize the difficulty of directly processing high-salt samples with traditional IEF system and provides a versatile, miniaturized and low voltage demand analytical platform for POCT of proteins in physiological samples. Abstract:Rationale Accurate measurement of trace compounds in blood samples is important in clinical diagnosis and life science. Ambient ionization mass spectrometry, however, suffers from the matrix effect when dealing with complex samples such as blood. Therefore, it is important to reduce the matrix effects in blood samples. Methods A low-cost and disposable Teflon tube was used as a platform to precipitate the protein in blood. The analytes are extracted into organic solvent, and the precipitated protein can be adsorbed by the chromatography paper inserted. Therefore, the Teflon tube after precipitation can be directly subjected to paper spray ionization mass spectrometry, achieving one-step analysis of blood. Results High sensitivity and satisfactory stability were achieved for pharmaceuticals, acids, and endogenic metabolites in blood. The absolute signal intensities of characteristic product ions of the tested analytes were 8-20 times higher after protein precipitation than those obtained using paper spray. Detection limits and quantitative performance were evaluated for three drugs: carbamazepine, metformin, and tioconazole. In addition, the limits of detection and quantitation were improved 9-14- and 8-12-fold, respectively. Conclusions Protein precipitation coupled to paper spray with a tube and then to mass spectrometry was successfully achieved and applied in the one-step analysis of trace compounds in blood samples. The experimental results showed that this method was sensitive, stable, convenient, and economic for the direct analysis of blood.

Abstract:Giventhat protein peptide powders (PPPs) from different biologicalsources were inherited with diverse healthcare functions, which arousedadulteration of PPPs. A high-throughput and rapid methodology, unitedmulti-molecular infrared (MM-IR) spectroscopy with data fusion, coulddetermine the types and component content of PPPs from seven sourcesas examples. The chemical fingerprints of PPPs were thoroughly interpretedby tri-step infrared (IR) spectroscopy, and the defined spectral fingerprintregion of protein peptide, total sugar, and fat was 3600-950cm(-1), which constituted MIR finger-print region.Moreover, the mid-level data fusion model was of great applicabilityin qualitative analysis, in which the F1-score reached 1 and the totalaccuracy was 100%, and a robust quantitative model was establishedwith excellent predictive capacity (R (p): 0.9935, RMSEP: 1.288, and RPD: 7.97). MM-IR coordinated data fusionstrategies to achieve high-throughput, multi-dimensional analysisof PPPs with better accuracy and robustness which meant a significantpotential for the comprehensive analysis of other powders in foodas well.
