Abstract:levels of ST6Gal-I mRNA and protein.Lgr5 and Axin2 are downstream of b-catenin and require Wnt signaling for expression.To determine whether ST6Gal-I expression also is dependent on Wnt signaling, organoids were cultured for 3-4 days in reduced concentrations of the Wnt-conditioned media necessary for in vitro organoid growth and analyzed for Lgr5 and Axin2 mRNA, as well as ST6Gal-I mRNA and protein expression.Similar to Lgr5 and Axin2, ST6Gal-I expression decreased with diminishing Wnt signaling (p=0.004),suggesting ST6Gal-I expression is at least in part regulated by the Wnt pathway.Importantly, the differentiation of gastric and colonic organoids into epithelial cell monolayers sharply decreased ST6Gal-I mRNA (p=0.0008) and protein expression as well as Lgr5 (p<0.0001) and Axin2 (p =0.001) mRNA expression, suggesting that ST6Gal-I expression reflects epithelial cell stemness but is significantly reduced in differentiated cells.However, survival of human epithelial cell organoids was not dependent on ST6-Gal-I expression since a lentiviral knockdown of ST6Gal-I exhibited growth characteristics similar to non-transduced organoids.Additionally, knockdown of ST6Gal-I in a tamoxifen-induced Cre mouse model did not affect mucosal epithelial stem cell expression of Lgr5 and Axin2 mRNA in vitro.Conclusions: ST6Gal-I expression correlates strongly with mucosal epithelial cell stemness and may play a role in the differentiation of epithelial cells in mucosal cancers.
