Abstract:Optical imaging is important for understanding brain function. However, established methods with high spatiotemporal resolution are limited by the potential for laser damage to living tissues. We describe an adaptive femtosecond excitation source that only illuminates the region of interest, which leads to a 30-fold reduction in the power requirement for two- or three-photon imaging of brain activity in awake mice for improved high-speed longitudinal neuroimaging. An adaptive excitation source enables two- and three-photon imaging of the awake mouse brain with high spatial and temporal resolution at 30-fold-reduced laser power relative to conventional approaches.
High-speed, Large Field-of-view and Deep Imaging with an Adaptive Excitation Source

Abstract:Conventional multiphoton microscopy uses periodically pulsed sources as excitation and the sample is illuminated uniformly by the laser. While necessary for structural imaging, monitoring dynamic biological functions such as neuronal activity in the brain typically only requires imaging of the region of interest (ROI), e.g., the neurons. The adaptive excitation source enables imaging of the region of interest only. It reduces the requirement for the output power of the excitation source (by at least an order of magnitude) and simultaneously reduces the excitation power to the sample for obtaining the necessary information (e.g., neuronal activity). We demonstrate three-photon imaging of brain activity in awake transgenic mice (jRGECO1a), with highest speed (30 frames/s), large field-of-view (620x620 μm/512x512 pixels) and deep penetration (750 μm beneath the dura).

Abstract:Two-photon excitation processes were first introduced by Göppert-Mayer in 1931 [1]. About two decades ago, multiphoton microscopy (MPM) was invented [2]. Despite the development of tomography [3] and hybrid imaging technologies [4-6], MPM remains the most powerful and widely used technique for high resolution deep tissue imaging [7]. However, as the imaging depth increases the images are increasingly degraded, due to inhomogeneous optics properties of biological samples. In recent years, several methods are demonstrated to recover nearly-diffraction-limited performance in deep tissue twophoton imaging by using adaptive optics [8, 9]. However, these methods are realized with the cost of additional imaging acquisition time.

Abstract:Two-photon microscopy has been widely used in neuroscience due to its deeper penetration depth compared with confocal microscopy. However, as imaging depth increases, its imaging quality will also degrade dramatically because of the accumulated aberration and scattered light. Here we report a high-resolution two-photon microscopy deep into the scattering medium, by structure illuminating the sample in the focal volume and demodulating the fluorescent signal thereafter. Our results show that compared with conventional two-photon microscopy the maximum imaging depth increases by hundreds micron in the mouse brain, and even in the surficial layer the imaging resolution is improved by around 15%.Two-photon microscopy has been widely used in neuroscience due to its deeper penetration depth compared with confocal microscopy. Adaptive optics is one of the most powerful tools to recover diffraction-limited resolution deep into the brain. However, it takes additional time for wavefront aberration measurement and compensation. Here we report a high-resolution two-photon microscopy deep into the scattering medium, by structure illuminating the sample in the focal volume and demodulating the fluorescent signal thereafter. The special structure-illumination pattern is design to make the focal point scanning sinusoidally only in the small focal volume. Here we first theoretically analyze the image formation of the structure illuminated two-photon microscopy (SITP) with D-shaped pupils and one annular one circular pupils, respectively. Their intensity points spread functions are investigated and the three-dimensional (3D) optical transfer functions (OTF) are provided. Then, the inertial-free scanning capability of SITP is demonstrated. After that, the ability of rejecting out-of-focus background and reducing noise is demonstrated. Our experimental results show that compared with conventional two-photon microscopy the maximum imaging depth increases by hundreds micron in the mouse brain, and even in the surficial layer the imaging resolution is improved by around 15%. Moreover, no additional time will be taken during to optical aberration and scattered light compensation / rejection.

Abstract:Two‐photon microscopy (2PM) is one of the most widely used tools for in vivo deep tissue imaging. However, the spatial resolution and penetration depth are still limited due to the strong scattering background. Here we demonstrate a two‐photon focal modulation microscopy. By utilizing the modulation and demodulation techniques, background rejection capability is enhanced, thus spatial resolution and imaging penetration depth are improved. Compared with 2PM, the transverse resolution is increased by 70%, while the axial resolution is increased to 2‐fold. Furthermore, when applied in conventional 2PM mode, it can achieve inertial‐free scanning in either transverse or axial direction with in principle unlimited scanning speed. Finally, we applied 2PFMM in thick scattering samples to further examine the imaging performance. The results show that the signal‐to‐background ratio of 2PFMM can be improved up to five times of 2PM at the depth of 500 μm. Abstract:We present a two-photon fluorescence microscope designed for high-speed imaging of neural activity in cellular resolution. Our microscope uses a new adaptive sampling scheme with line illumination. Instead of building images pixel by pixel via scanning a diffraction-limited spot across the sample, our scheme only illuminates the regions of interest (i.e., neuronal cell bodies), and samples a large area of them in a single measurement. Such a scheme significantly increases the imaging speed and reduces the overall laser power on the brain tissue. Using this approach, we performed high-speed imaging of the neural activity of mouse cortex . Our method provides a new sampling strategy in laser-scanning two-photon microscopy, and will be powerful for high-throughput imaging of neural activity.

High-resolution Focus-Tunable One-Photon Miniaturized Fluorescence Microscope for Imaging in Freely Moving Animals

Abstract:The relationship between neural activity, brain function, and corresponding biological behaviors remains a significant challenge in neuroscience. Exploring this relationship needs various optical imaging techniques to acquire real-time data with high spatial resolution. A promising technology recently is wearable miniaturized microscopes (mini scopes), which enable long-term neural activity recording in freely moving animals. However, most one-photon mini-scopes have limitations for imaging in-depth with high resolution and large field of view (FOV). To address this, we developed a one-photon miniaturized fluorescence microscope (1P-miniFM), intended for imaging of live brain neurons in free-behaving animals at subcellular level (~1.2 μm). We conducted specially designed optical path, achieving an imaging FOV of ~700 × 400 μm. In addition, we incorporated an electrowetting lens (EWL) to achieve a wide range of ~300 μm z-axis scanning with little resolution loss. 1P-miniFM is compact (11 × 17 × 24 mm) and lightweight (~2.9 g), causing little impediment to animals’ spontaneous behaviors. With genetically encoded calcium indicator GCaMP6s, we monitored neuron activities in secondary motor cortex (M2) during consecutive pain-related and sensory stimulations. We found that M2 neurons are key components and exhibit distinct variations in the response patterns. 1P-miniFM has potential as an excellent tool to explore relationships between neuron network and animal behaviors.

Abstract:As an important nonlinear optical imaging method, multiphoton excitation has demonstrated its wide application in biomedical imaging. Although shorter pulse can induce a higher nonlinear effect, most applications are still restricted in 100 fs region due to the existence of high-order dispersion. Through the compensation of high-order dispersion with multiphoton intrapulse interference, we successfully introduce 10 fs pulse into multiphoton biomedical imaging. We find that a deeper penetration and a correct excitation of multiple dyes can be obtained with this method.

Abstract:To improve imaging speed, multifocal excitation is widely adopted as a parallel strategy in laser-scanning microscopy. Specifically, axial multifocal microscopy is popular in neuroscience as it enables functional imaging of neurons in multiple depths simultaneously. However, previous phase searching algorithms for axial multi-foci generation generally generate foci of uniform intensities, which cannot compensate the scattering-induced power loss in deep tissue and causes inhomogeneous excitation. Here, we propose a novel adaptive optimization-based phase-searching method (AdaPS) to generate axial multi-foci with arbitrary intensity modulations for scattering-induced loss compensation. By adopting Adaptive Moment Estimation (Adam) as the searching algorithm, our method could escape from unsatisfactory local minima and stably converge to the optimal phase pattern with errors at least an order of magnitude lower. We validate AdaPS through both numerical simulations and experiments and demonstrate that AdaPS could provide uniform multi-depth imaging in scattering phantom and enable high-fidelity multi-depth recordings of neural network dynamics in mouse brain in vivo.
Efficient, broadly-tunable source of megawatt pulses for multiphoton microscopy based on self-phase modulation in argon-filled hollow-core fiber

Abstract:An exciting recent development for deep-tissue imaging with cellular resolution is three-photon fluorescence microscopy (3PM) with excitation at long wavelengths (1300 and 1700 nm). In the last few years, long-wavelength 3PM has driven rapid progress in deep-tissue imaging beyond the depth limit of two-photon microscopy, with impacts in neuroscience, immunology, and cancer biology. However, wide adoption of 3PM faces challenges. Three-photon excitation (3PE) is naturally weaker than two-photon excitation, which places a premium on ultrashort pulses with high peak power. The inefficiency, complexity, and cost of current sources of these pulses present major barriers to the use of 3PM in typical biomedical research labs. Here, we describe a fiber-based source of femtosecond pulses with multi-megawatt peak power, tunable from 850 nm to 1700 nm. Compressed pulses from a fiber amplifier at 1030~nm are launched into an antiresonant hollow-core fiber filled with argon. By varying only the gas pressure, pulses with hundreds of nanojoules of energy and sub-100 fs duration are obtained at wavelengths between 850 and 1700 nm. This approach is a new route to an efficient, robust, and potentially low-cost source for multiphoton deep-tissue imaging. In particular, 960-nJ and 50-fs pulses are generated at 1300 nm with a conversion efficiency of 10\%. The nearly 20-MW peak power is an order of magnitude higher than the previous best from femtosecond fiber source at 1300~nm. As an example of the capabilities of the source, these pulses are used to image structure and neuronal activity in mouse brain as deep as 1.1 mm below the dura.

Abstract:An enabling technology for the monitoring of neural activity, multiphoton microscopy with Bessel focus scanning is a high-speed volumetric imaging method with subcellular lateral resolution. Similar to many other optical microscopy techniques, however, its axially extended Bessel-like focus experiences sample-induced optical aberrations, which lead to reduced resolution and image contrast at depth. In this study, we demonstrated an adaptive optical Bessel focus scanning multiphoton microscope with pupil-segmentation-based wavefront sensing and highly efficient sample-plane aberration correction. Applying it to mouse brain imaging in vivo, we observed up to threefold signal enhancement for functional spine imaging at depth.

Abstract:Point-scanning two-photon microscopy enables high-resolution imaging within scattering specimens such as the mammalian brain, but sequential acquisition of voxels fundamentally limits its speed. We developed a two-photon imaging technique that scans lines of excitation across a focal plane at multiple angles and computationally recovers high-resolution images, attaining voxel rates of over 1 billion Hz in structured samples. Using a static image as a prior for recording neural activity, we imaged visually evoked and spontaneous glutamate release across hundreds of dendritic spines in mice at depths over 250 µm and frame rates over 1 kHz. Dendritic glutamate transients in anesthetized mice are synchronized within spatially contiguous domains spanning tens of micrometers at frequencies ranging from 1–100 Hz. We demonstrate millisecond-resolved recordings of acetylcholine and voltage indicators, three-dimensional single-particle tracking and imaging in densely labeled cortex. Our method surpasses limits on the speed of raster-scanned imaging imposed by fluorescence lifetime.

Abstract:Abstract Scanning two-photon (2P) fiberscopes (also termed endomicroscopes) have the potential to transform our understanding of how discrete neural activity patterns result in distinct behaviors, as they are capable of high resolution, sub cellular imaging yet small and light enough to allow free movement of mice. However, their acquisition speed is currently suboptimal, due to opto-mechanical size and weight constraints. Here we demonstrate significant advances in 2P fiberscopy that allow high resolution imaging at high speeds (26 fps) in freely-behaving mice. A high-speed scanner and a down-sampling scheme are developed to boost imaging speed, and a deep learning (DL) algorithm is introduced to recover image quality. For the DL algorithm, a two-stage learning transfer strategy is established to generate proper training datasets for enhancing the quality of in vivo images. Implementation enables video-rate imaging at ~26 fps, representing 10-fold improvement in imaging speed over the previous 2P fiberscopy technology while maintaining a high signal-to-noise ratio and imaging resolution. This DL-assisted 2P fiberscope is capable of imaging the arousal-induced activity changes in populations of layer2/3 pyramidal neurons in the primary motor cortex of freely-behaving mice, providing opportunities to define the neural basis of behavior.

Abstract:Two-Photon Microscopy has become an invaluable tool for biological and medical research, providing high sensitivity, molecular specificity, inherent three-dimensional sub-cellular resolution and deep tissue penetration. In terms of imaging speeds, however, mechanical scanners still limit the acquisition rates to typically 10-100 frames per second. Here we present a high-speed non-linear microscope achieving kilohertz frame rates by employing pulse-modulated, rapidly wavelength-swept lasers and inertia-free beam steering through angular dispersion. In combination with a high bandwidth, single-photon sensitive detector, we achieve recording of fluorescent lifetimes at unprecedented speeds of 88 million pixels per second. We show diffraction-limited, multi-modal, Two-Photon fluorescence and fluorescence lifetime (FLIM), microscopy and imaging flow cytometry with a digitally reconfigurable laser, imaging system and data acquisition system. These unprecedented speeds should enable high-speed and high-throughput image-assisted cell sorting.

Abstract:Multiphoton microscopes are hampered by limited dynamic range, preventing weak sample features from being detected in the presence of strong features, or preventing the capture of unpredictable bursts in sample strength. We present a digital electronic add-on technique that vastly improves the dynamic range of a multiphoton microscope while limiting potential photodamage. The add-on provides real-time negative feedback to regulate the laser power delivered to the sample, and a log representation of the sample strength to accommodate ultrahigh dynamic range without loss of information. No microscope hardware modifications are required, making the technique readily compatible with commercial instruments. Benefits are shown in both structural and in-vivo functional mouse brain imaging applications.

Abstract:We describe a new light-sheet microscopy method for fast, large-scale volumetric imaging. Combining synchronized scanning illumination and oblique imaging over cleared, thick tissue sections in smooth motion, our approach achieves high-speed 3D image acquisition of an entire mouse brain within 2 hours, at a resolution capable of resolving synaptic spines. It is compatible with immunofluorescence labeling, enabling flexible cell-type specific brain mapping, and is readily scalable for large biological samples such as primate brain.

Abstract:Ultrafast imaging is a powerful tool for studying space-time dynamics in photonic material, plasma physics, living cells, and neural activity. Pushing the imaging speed to the quantum limit could reveal extraordinary scenes about the questionable quantization of life and intelligence, or the wave-particle duality of light. However, previous designs of ultrafast photography are intrinsically limited by framing speed. Here, we introduce a new technique based on a multiple non-collinear optical parametric amplifier principle (MOPA), which readily push the frame rate into the area of ten trillion frames per second with higher spatial resolution than 30 line pairs per millimeter. The MOPA imaging is applied to record the femtosecond early evolution of laser-induced plasma grating in air for the first time. Our approach avoids the intrinsic limitations of previous methods, thus can be potentially optimized for higher speed and resolution, opening the way of approaching quantum limits to test the fundamentals.

Abstract:An adaptive pulse shaper controlled by multiphoton intrapulse interference phase scanning (MIIPS) was used, together with a prism-pair, to measure and cancel high-order phase distortions introduced by a high-numerical-aperture objective and other dispersive elements of a two-photon laser-scanning microscope. The delivery of broad-bandwidth (approximately 100 nm), sub-12-fs pulses was confirmed by interferometric autocorrelation measurements at the focal plane. A comparison of two-photon imaging with transform-limited and second-order-dispersion compensated laser pulses of the same energy showed a 6-to-11-fold improvement in the two-photon excitation fluorescence signal when applied to cells and tissue, and up to a 19-fold improvement in the second harmonic generation signal from a rat tendon specimen.

Abstract:Deep tissue multiphoton imaging requires high peak power to enhance signal and low average power to prevent thermal damage. Both goals can be advantageously achieved through laser repetition rate tuning instead of simply adjusting the average power. We show that the ideal repetition rate for deep two-photon imaging in the mouse brain is between 1 and 10 MHz, and we present a fiber-based source with an arbitrarily tunable repetition rate within this range. The performance of the new source is compared to a mode-locked Ti:Sapphire (Ti:S) laser for in vivo imaging of mouse brain vasculature. At 2.5 MHz, the fiber source requires 5.1 times less average power to obtain the same signal as a standard Ti:S laser operating at 80 MHz.

Abstract:Recently, we presented a new approach to create high-speed amplitude modulation of femtosecond laser pulses and tag multiple excitation beams with specific modulation frequencies. In this work, we discuss the utility of this method to record calcium signals in brain tissue with two-photon frequency-division multiplexing (2P-FDM) microscopy. While frequency-multiplexed imaging appears slightly inferior in terms of image quality as compared to conventional two-photon laser scanning microscopy due to shot noise-induced cross-talk between frequency channels, applying this technique to record average signals from regions of interest (ROI) such as neuronal cell bodies was found to be promising. We use phase information associated with each pixel or waveform within a selected ROI to phase-align and recombine the signals into one extended amplitude-modulated waveform. This procedure narrows the frequency detection window, effectively decreasing noise contributions from other frequency channels. Using theoretical analysis, numerical simulations, and in vitro imaging, we demonstrate a reduction of cross-talk by more than an order of magnitude and predict the usefulness of 2P-FDM for functional studies of brain activity.
