Abstract:Regioselective acetylation of per-O-TMS carbohydrates was achieved preferentially at the 6 position or 1,6-position under mild conditions involving the eco-friendly solvent acetonitrile, at room temperature, in ambient atmosphere and in a shorter time. Good or moderate yields were obtained via 4-dimethylaminopyridine, without auxiliary equipment. A single α-O-Acetyl acetylation anomer was exclusively defined for the involved substrates. A 6-O-monoacetate derivative was applied and used as a stable glycosyl donor in the disaccharides construction. The methodology was successful for a range of substrates, which include the following: d-lactose, d-trehalose, d-galactose, methyl α-d-galactose, d-glucose, d-mannose, d-xylose, and l-fucose.

Abstract:The synthesis of 2, 4-diisopropyl-5,5-dimethyl-1,3-dioxane through the acetalization of isobutyraldehyde with 2, 2, 4-trimethyl-1,3-pentanediol (TMPD) catalyzed by the novel carbon based acid was first carried out. High conversion (≥98%) and specific selectivity were obtained using the novel carbon based acid, which kept high activity after it was reused 5 times. Moreover, the catalyst could be used to catalyze the acetalization and ketalization of different aldehydes and ketones with superior yield. The yield of several products was over 90%. The novel heterogeneous catalyst has the distinct advantages of high activity, strikingly simple workup procedure, non-pollution, and reusability, which will contribute to the success of the green process greatly.

Abstract:Aryl carboxylic acids are among the most abundant substrates in chemical synthesis and represent a perfect example of a traceless directing group that is central to many processes in the preparation of pharmaceuticals, natural products and polymers. Herein, we describe a highly selective method for the direct step-down reduction of carboxylic acids to arenes, proceeding via well-defined Pd(0)/(ii) catalytic cycle. The method shows a remarkably broad substrate scope, enabling to direct the classical acyl reduction towards selective decarbonylation by a redox-neutral mechanism. The utility of this reaction is highlighted in the direct defunctionalization of pharmaceuticals and natural products, and further emphasized in a range of traceless processes using removable carboxylic acids under mild, redox-neutral conditions orthogonal to protodecarboxylation. Extensive DFT computations were conducted to demonstrate preferred selectivity for the reversible oxidative addition and indicated that a versatile hydrogen atom transfer (HAT) pathway is operable.

Abstract:A novel synthesis of acyclic cis-enediynes 2 has been established by an acid-catalyzed rearrangement of 1,2-diyn-2-propen-1-ols 1 possessing a C-3-aryl group in the presence of water, alcohols, or thiols. Reactivity of allyl alcohols and regio- and cis/trans diastereoselectivity of the allylic migration were examined. In the presence of(+/-)-10-camphorsulfonic acid (CSA), the parent allyl alcohol 5 and the C-3-methyl-substituted 9 failed to give enediynes, whereas the C-3-aryl-substituted 12 and 29 underwent the allylic rearrangement to provide predominantly cis-enediynes 16 and 31 at room temperature or below. Under similar acidic conditions, enediyne alcohol 13 produced 16b and 16d with the same regio and cis/trans diastereoselectivity observed for 12. Allyl alcohol 30, an isomer of 29, also provided enediynes 31c and 32c after a prolonged reaction (90 h) at room temperature in the presence of CSA and EI;OH. These results suggested that the same allylic cations were obtained from allyl alcohols 12 and 13 or 29 and 30 even though the ease of ionization differed for each substrate. Involvement of allylic cations in the product-forming step was confirmed by the finding that chiral allyl alcohols (-)-12 and (-)-18c furnished racemic products. In general, the p-MeOPh-substituted allyl alcohol 29 gave a better regioselectivity than the Ph-substituted 12. In the reactions with alcohols, the regioisomeric ratios were 100:0 (31:33) for 29 and ca. 96:4 (16:17) for 12; the ratios decreased to ca. 90:10 (31:33) for 29 and ca. 10:30 (16:17) for 12 when thiols were used. The cis/trans diastereoselectivity is higher for allyl alcohol 12 (100% for 16 at 20 degrees C) compared to that for 29 (31:32 = 80:20-94:6 at 0 degrees C). Computational calculations at the RHF/3-21G level, carried out on the model compounds and allylic cations, indicated that nucleophilic trapping takes place preferentially at the Cg carbon to form the thermodynamically much more stable enediynes. Under the best reaction conditions (1 equiv of CSA and 2 equiv of EtOH in CH2Cl2, 20 degrees C), a number of acyclic cis-enediynes can be synthesized in three steps from the commercially available alpha-bromocinnamaldehyde (10).

Abstract:A simple and efficient method was developed for the synthesis of 2,20-arylmethylene dicyclohexane-1,3-dione derivatives via the Knoevenagel–Michael cascade reactions of aromatic aldehydes and 1,3-cyclic diketones catalyzed by ‘‘Amano’’ lipase DF, which expands the application field of enzyme catalytic promiscuity. This protocol provides several advantages over the traditional chemical synthesis, such as simple work-up procedure, high yields（up to 94%） and environmental friendliness.

Abstract:alpha-Hydroxy acids have attracted broad attention because of their prevalence in bioactive molecules and biodegradable polymers, but their conventional syntheses are usually restricted to aromatic substrates, especially, in a stepwise manner. Herein, we disclose the transformation of alkyl and aryl vicinal glycols to alpha-hydroxy acetates in water under the air atmosphere with our solid self-supported NHC-Ir single-site catalyst. Both aliphatic and aromatic glycols are compatible with a much higher catalytic efficiency in the presence of this solid single-site catalyst than other viable molecular catalysts (93% vs <35%) because of the "isolation effect". Remarkably, our catalyst could be reused for 20 runs without an obvious loss in catalytic activity and selectivity. Control experiments and density functional theory calculations reveal that the reaction firstly undergoes a dehydrogenation facilitated by the catalyst, and then it proceeds through an unexpected oxidization relay step by oxygen in the air, leading to the alpha-hydroxy acetate formation. Our protocol can potentially contribute to the valorization of readily available and inexpensive diols.

Abstract:A novel method for the organocatalytic syn diacetoxylation of alkenes has been developed using aryl iodides as efficient catalysts. A broad range of substrates, including electron-rich as well as electron-deficient alkenes, are smoothly transformed by the new procedure, furnishing the desired products in good to excellent yields with high diastereoselectivity (up to >19:1 dr).

Abstract:Odorless 2-(1,3-dithian-2-ylidene)-3-oxobutanoic acid (1c) was prepared and investigated in the thioacetalization of carbonyl compounds as a 1,3-propanedithiol equivalent. The results showed that the thioacetalization of various carbonyl compounds 2 with 1c proceeded smoothly and afforded the corresponding dithioacetals 3 in high yields (up to 99%) in the presence of acetyl chloride at room or reflux temperatures. Moreover, the thioacetalization exhibited high chemoselectivity between aldehydes and ketones. Abstract:Selective syn and anti diacetoxylations of alkenes have been achieved using a PhI(OAc)(2)/BF3 center dot OEt2 system in the presence and absence of water, respectively. A broad range of substrates including electron-deficient alkenes (such as alpha,beta-unsaturated esters) could be elaborated efficiently at room temperature with this methodology, furnishing the desired products in good to excellent yields and diastereoselectivity. In particular, a multigram-scale diastereoselective diacetoxylation of methyl cinnamate (5.00 g) was also accomplished in a few hours, maintaining the same efficiency as small-scale reaction. This novel methodology provides an alternative approach for the preparation of various 1,2-diols.

Abstract:Catalyst-controlled site-selective activation of β- and γ-methylene carbon-hydrogen (C-H) bonds of free carboxylic acids is a long-standing challenge. Here we show that, with a pair of palladium catalysts assembled with quinoline-pyridone ligands of different chelate ring sizes, it is possible to perform highly site-selective monolactonization reactions with a wide range of dicarboxylic acids, generating structurally diverse and synthetically useful γ- and δ-lactones via site-selective β- or γ-methylene C-H activation. The remaining carboxyl group serves as a versatile linchpin for further synthetic applications, as demonstrated by the total synthesis of two natural products, myrotheciumone A and pedicellosine, from abundant dicarboxylic acids.

Abstract:The site-selective functionalization of value-added compounds while implementing atom-economical C–H coupling partners represents an unmet challenge of utmost importance within organic synthesis. Herein, we report a Pd-catalyzed directed C–H acylation of a collection of relevant phenol-containing compounds with ethanol and other alcohols and aldehydes. This tagging technique is distinguished by its water compatibility and predictable regioselectivity and features the use of ethanol as renewable feedstock for the modification of intricate phenols, including estrogens and other top-selling pharmaceuticals. Mechanistic studies support the intermediacy of a challenging 6-membered dimeric palladacycle that undergoes the addition of nucleophilic acyl radical species.

Abstract:Radical-initiated allylation reactions represent a powerful tool for installation on a synthetically useful allyl group for further transformations. In this field, direct allylation of C-H bonds has received considerable attentions due to its more atom-efficient and environmental benign potential. Despite these advances, these strategies still have some limitations, such as utilization of stoichiometric HAT reagents and excessive amount of C-H substrates, incompatibility with aldehydic C(sp2)-H bonds, relaying on the dual catalysis or multiple steps. Herein, a one-step protocol for desulfonylative allylation of C-H bonds of aldehydes and alkanes with allyl sulfones using TBADT as photo-hydrogen atom transfer catalyst is reported. This methodology requires only a single equivalent of C-H substrates and has been applied for the allylation of aldehydic C(sp2)-H, activated or unactivated aliphatic C(sp3)-H with good functional group tolerance. Moreover, this methodology exhibites further potential application for diacrylation of allylic sulfones by slightly adjusting the ratio of reactants, presenting a direct and facile access to the highly substituted 1,5-diketones in good to high yields.

Abstract:Formic acid is efficiently used as a C1 source to directly carboxylate allylic alcohols in the presence of a low loading of palladium catalyst and acetic anhydride as additive to afford β,γ‐unsaturated carboxylic acids with excellent chemo‐, regio‐, and stereoselectivity. The reaction proceeds through a carbonylation process with in situ‐generated carbon monoxide under mild conditions, avoiding the use of high‐pressure gaseous CO. A bisphosphine ligand with a large bite angle (4,5‐bis{diphenylphosphino}‐9,9‐dimethylxanthene, Xantphos) was found to be uniquely effective for this transformation. The regio‐ and stereoconvergence of this reaction is ascribed to the thermodynamically favored isomerization of the allylic electrophile in the presence of the palladium catalyst.

Abstract:The acetalisation of aldehydes with diols to corresponding 1,3-dioxolanes has been accomplished using ionic liquids as catalyst and reaction media. In the reaction process the removal of water produced and other catalysts were not necessary. High conversion and good selectivity were obtained when using HMImBF 4 , which could be easily recycled and reused.
