Abstract:Owing to the great role of glucose in biological systems and food industry, it is significant to maintain the dynamic balance of organisms. Various methods widely reported are focused on the glucose detection with high sensitivity and low detection limit. However, the sensor with ultra-low concentration sensitivity is not suitable for the direct detection of high concentration of glucose in human serum and food samples. Additionally, the application of ratiometric fluorescence probe is usually limited by the narrow range of color changes and insensitivity color change of the target. Here, a novel dual-emission ratiometric fluorescence probe with green to red fluorescence intensity of 6:1 was first constructed by fluorescein isothiocyanate (FITC) with green fluorescence as signal unit and silica coated quantum dot (2) with red fluorescence as the stable internal standard. The glucose was oxidized to generate H2O2 by the catalysis of glucose oxidase, which can oxidize Fe2+ to form Fe3+ ion. The formation of Fe3+ ion will cause the quenching of FITC fluorescence, which can realize the continuous fluorescence color change from green to yellow to orange to red for the qualitative and quantitative detection of blood glucose and fruit juice. The proposed method is cheap, simple, intuitive and has a wide detection range for glucose detection, which is of significance in designing the dual-emission ratiometric fluorescence sensor with wide range of color change.

Abstract:It is highly demanded and challenging to develop small-molecular fluorescent sensors for the ratiometric detection of pH fluctuation. A ratiometric fluorescent pH sensor was constructed by integrating a pH-sensitive fluorophore with a pH-insensitive hemicyanine group. Its linear and reversible ratiometric fluorescent response from pH 7.0 to 8.8 and 6.0 to 9.0 respectively makes this sensor suitable for the practical tracking of pH fluctuation in live cells. With this sensor, stimulated pH fluctuation has been successfully tracked in a ratiometric manner via fluorescence imaging with low dark toxicity.

Abstract:In this work, 3-aminophenylboronic acid (APBA) functionalized dual-emission quantum dots (QDs) were prepared and utilized as the ratiometric fluorescence probe for the onsite naked eye determination of glucose. The unique ratiometric fluorescence probe, which combined ratiometric fluorescence technique with boric acid functional materials into one system, has been prepared by hybridizing red-emitting CdTe QDs (r-QDs) and green-emitting CdTe QDs (g-QDs). The fluorescence of the embedded r-QDs stays constant, whereas the g-QDs functionalized with APBA can selectively bind glucose by the chemistry of boronic acid and cis diol compounds, leading to the fluorescence of g-QDs quenched due to the surface quenching states induced mechanism and resulting in continuous fluorescence color changes (from green to red) of the ratiometric fluorescence probe system. Under optimum conditions, the proposed ratiometric probe was used for selective detection of glucose in a concentration range of 0.1-2.0 mmol/L with a detection limit of 4.5 timol/L. Moreover, the developed ratiometric fluorescence probe was successfully applied to the detection of glucose in human serum samples. The present study provides an efficient and facile ratiometric fluorescence probe for rapid, convenient, sensitive and selective visual identification of glucose in blood glucose sensing without the need of complex equipment. Abstract:A novel direct readout colorimetric optical glucose sensor strip was constructed based on a three-layer film, including a green-emitted CdTe/CdS quantum dots (QDs) layer as a stable color background, a red-fluorescent platinum-porphyrin oxygen-sensing layer and a glucose oxidase layer. The sensor achieved high resolution (up to 0.2 mmol L(-1)) glucose determination with a detection range from 0 to 3.0 mmol L(-1). A "glucose ruler" which acts as a glucose standard colorimetric card was obtained. Glucose concentration could easily be directly readout using the "glucose ruler", which made the glucose determination rapid, convenient and easy. The effects of pH, salinity and temperature were systematically investigated. The prepared sensor was finally applied for glucose sample analysis, compared with the "glucose ruler", accurate results could be directly readout.

Abstract:Developing a highly sensitive fluorescence probe for biomolecule detection has a significant meaning in clinical diagnosis and research. Tuning the optical property of fluorescent nanomaterials by heteroatom doping is generally feasible to improve the application performance. In this work, a kind of cesium-doped graphene quantum dots (Cs-GQDs) are synthesized, characterized, and explored as a fluorophore in ultrasensitive determination of H2O2 and glucose. The prepared Cs-GQDs with a quantum yield of 39.8% eradiate intense blue fluorescence under UV irradiation. The emission of Cs-GQDs is almost excitation-independent. Doping Cs into the GQDs leads to a clear blue shift of the fluorescence emission peak, probably related to the increased band gap. In addition, Cs-GQDs show a pH-sensitive behavior under acidic conditions. In the presence of horseradish peroxidase, o-phenylenediamine could react with H2O2 to form a yellow fluorescent compound of 2,3-diaminophenazine (DAP). A ratiometric fluorescence probe toward H2O2 and glucose is established using the fluorescence resonance energy transfer mechanism between DAP and Cs-GQDs. This sensing strategy shows high sensitivity for H2O2 and glucose detection with detection limits of 25 and 23 nM, respectively, and excellent selectivity toward glucose out of various interferences. This method is extended to glucose detection in human serum samples. Abstract:A novel paper-based colorimetric glucose sensor was proposed employing Prussian blue nanoparticles (PB NPs) as mimic peroxidase. The sensor was manufactured by spraying solution containing PB NPs, glucose oxidase and chromogenic agents into a paper, then coating the filter layer and spreading layer on the top. The layer-by-layer structure enabled the sensor detect glucose in whole blood, as well as elimination of the coffee-ring effect which ensure the performance. As a powerful alternative to natural peroxidase, PB NPs showed the mimic enzymatic activity well preserved in dry environment. The manufacture process of the sensor is easy to be industrialized. Under optimal conditions, the sensor exhibited a linear range from 2.5 mM to 25 mM for glucose in blood with satisfactory reproducibility (the coefficient of variant 0.99). Coupled with a handhold device, the PB NPs-based test strip realized the goal of personal operation, user-friendly control, automatic readouts, and data storage, and able to link the Cloud, showing unique potential in clinical application, especially in community-level medical scenarios.

Abstract:Noninvasive and visual monitoring of glucose is highly desirable for diabetes diagnostics and long-term home-based health management. Owing to the correlation of the glucose level between blood and sweat, on-body sweat glucose detection provides potential for noninvasive healthcare but is highly challenging. Herein, we for the first time demonstrate a wearable skin pad based on the ratiometric fluorescent nanohybrid, which can realize noninvasive and visual monitoring of sweat glucose. Luminescent porous silicon (PSi) particles, which have a porous structure and oxidation-responsive photoluminescence decay, are chosen to load (adsorb or entrap) carbon quantum dots (CQDs) for the construction of the dual fluorescence nanohybrid. Bimetallic (Au and Ag) nanoparticles (BiM) are also co-decorated on the PSi particle to improve detection sensitivity by enhancing PSi's initial fluorescence and oxidation kinetics. Owing to the efficient fluorescence resonance energy transfer effect, BiM-CQDs@PSi initially exhibits PSi's red fluorescence with complete quenching of CQDs's blue fluorescence. The oxidation of PSi triggered by hydrogen peroxide (H2O2) weakens the FRET effect and decays PSi's fluorescence, causing ratiometric fluorescence to change from red (PSi) to blue (CQDs). A wearable skin pad is easily fabricated by co-immobilization of BiM-CQDs@PSi and glucose oxidase (GOX) in a transparent and biocompatible chitosan film supported by an adhesive polyurethane membrane. When the skin pad is attached on the body, the same ratiometric fluorescence transition (red -> blue) is observed upon the stimulation of H2O2 generated in GOX-catalyzed oxidation of sweat glucose. Based on the strong correlation between the ratio of the fluorescence change and sweat glucose level, clinical tests toward diabetics and healthy volunteers can clearly indicate hyperglycemia.

Abstract:The pH and O 2 are the two important parameters to evaluate environmental conditions, healthcare, and industrial processes. Herein, we developed a ratiometric fluorescent capillary-based dual-functional sensor for simultaneous monitoring of pH and O 2 fluctuations. The fluorescent sensor consists of a pH sensing unit and an O 2 sensing unit on both ends of the capillary inner wall. The ratiometric fluorescent pH sensing unit includes pH-sensitive probe HPTS and the reference dye Ce6, while the O 2 sensing unit is composed of O 2 -sensitive probe RuDP and the reference dye C6. Under single-wavelength excitation at 430 nm, the ratiometric fluorescent dual-functional sensor exhibits highly sensitive, selective and reproducible responses to both pH and O 2 . As for practical aqueous samples, the results about pH and O 2 levels obtained by our sensor are considerable consistent with commercial devices. Meanwhile, the dynamic changes of pH and O 2 levels during glucose catalysis by glucose oxidase can be detected in real-time and accurately. The dual-functional ratiometric fluorescent capillary transducer provides a significant potential platform for environmental monitoring and medical diagnosis.

Ratiometric fluorescent probe based on AuNCs induced AIE for quantification and visual sensing of glucose

Abstract:Blood glucose measurement plays a very important role in clinical diagnosis and fluorescence analysis has attracted extensive attention. A novel ratiometric fluorescent system with aggregation induced emission (AIE) property for the detection of glucose was established in this work. In this system, bovine serum albumin-stabilized Au nanoclusters (BSA-AuNCs) served both as the fluorescent detection probe and the AIE inducer. An AIE molecule, named sodium 1,2-bis [4-(3-sulfonatopropoxyl) phenyl]-1,2-diphenylethene (BSPOTPE), served as fluorescent reference probe. In the presence of H2O2, the fluorescence intensity of BSPOTPE/BSA-Au NCs at 680 nm progressively decreased while that at 490 nm remained constant. Glucose can be catalyzed by glucose oxidase (GOx) and produces H2O2. Therefore, glucose detection can be conveniently achieved by the proposed strategy. The fluorescence intensity change ratio increased linearly with the glucose concentration in the range 1-8 mM. Moreover, the proposed method exhibits a consecutive fluorescence color change ("from red to cyan") to glucose concentration in the range of 1-8 mM under a 365 nm UV lamp and exhibits bright "red" or bright "cyan" in lower glucose concentrations (lower than 3 mM) or high glucose concentrations (higher than 7 mM), respectively. The work offers an ideal rapid clinical diagnostic method for both normal and abnormal blood glucose screening.

Abstract:Glucose and pH are two important indicators of diabetes mellitus. However, their dynamic changes at the same time in brain are still not clear, mainly due to a lack of a single biosensor capable of simultaneous quantification of two species in a live rat brain. In this work, a selective and sensitive ratiometric electrochemical biosensor was developed for simultaneously quantifying glucose and pH using both current and potential outputs in a rat brain of diabetic model. Here, glucose oxidase was first employed as a specific recognition element for both glucose and pH because the active center (FAD) could undergo a 2H+/2e+ process. Moreover, an insensitive molecule toward pH and glucose was used as an inner reference element to provide a built-in correction to improve the accuracy. The ratio between the oxidation peak current density of glucose and that of ABTS gradually increased with increasing concentration of glucose, and showed a good linearity in the range of 0.3-8.2 mM. Meanwhile, the midpotential difference between glucose oxidase and 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) positively shifted with pH decreasing, leading to accurate determination of pH in the linear range of 5.67-7.65. Thus, combined with the unique properties of carbon fiber microelectrode, including easy to insert and good biocompatibility, the developed single biosensor was successfully applied to detect pH and glucose at the same time in hippocampus, striatum, and cortex in a live rat brain of diabetic model.

Abstract:A miniature wearable Raman spectroscopy system used to achieve noninvasive detection of human blood glucose level was developed. A thallium-doped grin lens was employed as the collection lens and a specially designed wearable fiber optic probe was employed to help the stable and convenient collection of Raman spectrum. The glucose solution, 11 rats and 10 healthy human were studied as subjects. In addition, a method of quantitative analysis of Raman spectrum was proposed which using the peak area as the main reference factor while the peak intensity as the auxiliary reference factor to calculate the target concentration. A non-linearized multivariate dominant factor-based partial least squares model was built for different samples to predict glucose level. The results show that the accuracy are 98.1%, 89.3% and 84.4% for glucose solution, rats and human subjects. The system has the advantages of more compact structure, lower cost, better testing stability and convenient for human body to wear, and is feasible and repeatable to achieve the noninvasive detection of human blood glucose accurately.

Abstract:The integration of sampling and instant metabolite readout can fundamentally elevate patient compliance. To circumvent the need for complex in-lab apparatus, here, an all-in-one sampling and display transdermal colorimetric microneedle patch was developed for sensing hyperglycemia in mice. The coloration of 3,3',5,5'-tetramethylbenzidine (TMB) is triggered by the cascade enzymatic reactions of glucose oxidase (GOx) and horseradish peroxidase (HRP) at abnormally high glucose levels. The HRP in the upper layer is biomineralized with calcium phosphate (CaP) shell to add a pH responsive feature for increased sensitivity as well as protection from nonspecific reactions. This colorimetric sensor achieved minimally invasive extraction of the interstitial fluid from mice and converted glucose level to a visible color change promptly. Quantitative red green and blue (RGB) information could be obtained through a scanned image of the microneedle. This costless, portable colorimetric sensor could potentially detect daily glucose levels without blood drawing procedures.

Abstract:A ratiometric fluorescence sensing platform with smartphone was designed for highly sensitive detection of glutathione (GSH), ascorbic acid (AA), and alkaline phosphatase (ALP) activity by modulation the inner filter effect (IFE) between persistent luminescent nanoparticles (PLNPs) and 2, 3-diaminophenazine (DAP). As an oxidase-mimetic nanomaterial, MnO2 nanosheets (MnO2 NS) can oxidize o-phenylenediamine (OPD) to DAP, which exhibited a yellow fluorescence emission signal at 558 nm, whereas the fluorescence of PLNPs at 475 nm was quenched through IFE. The presence of GSH and AA reduced MnO2 NS to Mn2+, which inhibited the oxidation of OPD, and effectively preserved the blue fluorescence of PLNPs at 475 nm. Further, ALP can catalytically hydrolyze L-ascorbic acid 2-phosphate trisodium salt (AAP) to yield AA. Therefore, a simple, high sensitivity and reproducibility, and good selectivity ratiometric fluorescence sensing platform was fabricated to detect GSH, AA and ALP activity by measuring the ratio of the fluorescence intensity of DAP and PLNPs. The linear range for GSH, AA and ALP activity were 1-100 μM, and 1 to 5 mU/mL, and the limit of detection were 0.2 μM, 0.33 μM and 0.3 mU/mL, respectively. Finally, the proposed sensor was utilized in spiked human serum and urine samples detection with satisfactory recoveries. Moreover, the red-green-blue (RGB) intensities of a color image were acquired via scanning measurement analysis with the help of the color-scanning application on smartphones and portable UV lamps, enabling real-time and in-situ detection of ALP activity, GSH, and AA.

Performance Comparison between Intra-Body and Radio Frequency Wireless Communication for Foot Plantar Pressure Measurement

Abstract:The sharp boost in healthcare demand has seen novel developments in health monitoring technologies, such as Intra-body communication (IBC). IBC technology envisions a network of continuously operating sensors that measure critical physical and physiological parameters, such as mobility, heart rate and glucose levels just to name a few. Wireless connectivity in IBC technology is the key to its success as it grants portability and flexibility to the user. While radio frequency (RF) wireless technology has been successfully deployed in most biomedical implementations, they consume a lot of power, are susceptible to electromagnetic interference and have security issues. IBC treats the human body as a transmission medium for transmitting and receiving electrical signals. Two IBC methods have been proposed in the literature: galvanic coupling and capacitive coupling. This paper compares and evaluates a binary frequency-shift keying (BFSK) modulation signal using RF, IBC galvanic and IBC capacitive coupling for foot plantar pressure sensors. IBC has characteristics that could naturally address the issues with RF for biomedical technology. The research targets IBC operating in the band from 1 MHz to 100 MHz, and RF operating at 2.4 GHz. Based on the empirical evidence; for this particular application, galvanic coupling shows more promising performance in power and signal-to-noise ratio measurements.
Glucose Sensing in Human Whole Blood Based on Near-Infrared Phosphors and Outlier Treatment with the Programming Language “R”

Abstract:A near-infrared paper-based analytical device (NIR-PAD) for glucose detection in whole blood was based on iridium(III) metal complexes embedded in a three-dimensional (3D) enzyme gel. These complexes emit NIR luminescence, can avoid interference from the color of blood, and increase the sensitivity of sensing glucose. The glucose reaction behaviors of another two different iridium(III) and platinum(II) complexes were also tested. When the glucose solution was added to the device, the oxidation of glucose by glucose oxidase caused oxygen consumption and increased the intensity of the phosphorescence emission. To the best of our knowledge, this is the first time that data have been treated with the programming language "R", which uses Tukey's test to identify the outliers in the data and calculate a median for establishing a calibration curve, in order to improve the accuracy of NIR-PADs for sensing glucose. Compared with other published devices, NIR-PADs exhibit a wider linear range (1-30 mM, [relative emission intensity] = 0.0250[glucose] + 0.0451, and R 2 = 0.9984), a low detection limit (0.7 mM), a short response time (<2 s), and a small sample volume (2 μL). Finally, blood specimens were obtained from 19 patients enrolled in Taipei Veterans General Hospital under an approved IRB protocol (Taiwan; 2017-12-002CC). The sensors exhibited remarkable characteristics for glucose detection in comparison with other methods, including the clinical method in hospitals as well as those without blood sample pretreatment or a dilution factor. The above results confirm that NIR-PAD sensors can be put to practical use for glucose detection.

Label-Free Colorimetric Detection of Urine Glucose Based on Color Fading Using Smartphone Ambient-Light Sensor

Abstract:In this work, a label-free colorimetric assay was developed for the determination of urine glucose using smartphone ambient-light sensor (ALS). Using horseradish peroxidase—hydrogen peroxide—3,3′,5,5′-tetramethylbenzidine (HRP-H2O2-TMB) colored system, quantitative H2O2 was added to samples to-be-determined for deepest color. The presence of glucose oxidase in urine led to the formation of H2O2 and the reduction of TMBred. As a result of this, the color of the urine faded and the solution changed from deep blue to light blue. We measured the illuminance of the transmitted light by a smartphone ambient light sensor, and thereby color changes were used to calculate the content of urine glucose. After method validation, this colorimetric assay was practically applied for the determination of urine samples from diabetic patients. Good linearity was obtained in the range of 0.039–10.000 mg/mL (R2 = 0.998), and a limit of detection was 0.005 mg/mL. Our method was had high accuracy, sensitivity, simplicity, rapidity, and visualization, providing a new sensor to be potentially applicable for point-of-care detection of urine glucose.

Abstract:Glucose performs many essential functions associated with metabolic processes in the living system, and is closely related to many diseases such as diabetes and hypoglycemia. Most of the existing glucose concentration detection methods require complex instruments, which undoubtedly limit its widespread use. Here, we have designed a glucose colorimetric detection system composed of glucose, glucose oxidase (GOD), manganese dioxide nanosheets (MnO2 NSs) and 3,3',5,5' -tetramethylbenzidine (TMB) to achieve colorimetric detection with the naked eye. Compared with the single-color change of the colorimetric method in previous studies, multiple-color changes have been realized. MnO2 NSs, as a kind of nanomaterial imitating oxidase, can directly oxidize TMB to oxTMB. Because oxTMB showed a dark yellow color when strongly oxidized and light blue when weakly oxidized, this feature can achieve multiple-color changes rather than a single-color change, which is helpful for colorimetric observation with the naked eye. Finally, we successfully used MnO2 NSs for colorimetric detection of glucose and realized multiple-color changes, making it easier to achieve colorimetric observation with the naked eye. The linear detection range is 0-4000 mu M and limit of detection is 5.0 mu M. This is not only useful for glucose, but also has an important significance for other experiments considering colorimetric experiments with the naked eye.

Abstract:Urinary glucose determination using a glucose test strip is simple and convenient in daily self-monitoring of diabetes. However, diabetic patients exhibit acquired impaired color vision (ICV), which results in the inability to discriminate between hues. Even with the assistance of a color chart, it is still not easy for these patients to read the urinary glucose results with the naked eye. In this study, a smartphone camera using an image-based colorimetric detection method was successfully developed for quantitative analysis of urine glucose. A horseradish peroxidase-hydrogen peroxide-3,3′5,5′-tetramethylbenzidine (HRP–H2O2–TMB) system was optimized for a reliable and gradual color fading process via a glucose oxidase (GOD) catalyzed oxidation reaction. The color changes of the peroxidase-H2O2 enzymatic reactions in the 96-well microplate were captured by a smartphone RGB camera with subsequent detection of red, green, and blue (RGB) intensities decreasing at each image pixel. The highly quantitative relationships between the glucose concentrations and the color characteristic values of the blue channel of the captured images were successfully established. The high accuracy of this method was demonstrated in urine glucose measurements with a linear response over the 0.039 mg mL−1 to 10.000 mg mL−1 glucose concentration range and a 0.009 mg mL−1 detection limit. The method has great potential as a point-of-need platform for diabetic patients with defective color vision and features high accuracy and low cost.

Abstract:The colorimetric detection of glucose typically involves a peroxidase reaction producing a color, which is then recorded and analyzed. However, enzyme detection has difficulties with purification and storage. In addition, replacing enzyme detection with chemical methods involves time-consuming steps such as centrifugation and purification and the optical instruments used for colorimetric detection are often bulky and not portable. In this study, ammonium metavanadate and sulfuric acid were used to prepare the detection solution instead of peroxidase to produce color. We also analyzed the effect of different concentrations of detection solution on absorbance sensitivity. Finally, a flip chip blue Mini-LEDs miniaturized optical instrument (FC blue Mini-LEDs MOI) was designed for glucose detection using optics fiber, collimating lenses, a miniaturized spectrometer, and an FC Blue Mini-LEDs with a center wavelength of 459 nm. While detecting glucose solutions in the concentration range of 0.1–10 mM by the developed MOI, the regression equation of y = 0.0941x + 0.1341, R 2 of 0.9744, the limit of detection was 2.15 mM, and the limit of quantification was 7.163 mM. Furthermore, the preparation of the detection solution only takes 10 min, and the absorbance sensitivity of the optimized detection solution could be increased by 2.3 times. The detection solution remained stable with only a 0.6% decrease in absorbance compared to the original after storing it in a refrigerated environment at 3 °C for 14 days. The method proposed in this study for detecting glucose using FC blue light Mini-LEDs MOI reduces the use of peroxidase. In addition, it has a wide detection range that includes blood as well as non-invasive saliva and tear fluids, providing patients with a miniaturized, highly sensitive, and quantifiable glucose detection system.

Abstract:It is significant for people with diabetes to know their body's real-time glucose level, which can guide the diagnosis and treatment. Therefore, it is necessary to research continuous glucose monitoring (CGM) as it gives us real-time information about our health condition and its dynamic changes. Here, we report a novel hydrogel optical fiber fluorescence sensor segmentally functionalized with fluorescein derivative and CdTe QDs/3-APBA, which can continuously monitor pH and glucose simultaneously. In the glucose detection section, the complexation of PBA and glucose will expand the local hydrogel and decrease the fluorescence of the quantum dots. The fluorescence can be transmitted to the detector by the hydrogel optical fiber in real time. As the complexation reaction and the swelling–deswelling of the hydrogel are all reversible, the dynamic change of glucose concentration can be monitored. For pH detection, the fluorescein attached to another segment of the hydrogel exhibits different protolytic forms when pH changes and the fluorescence changes correspondingly. The significance of pH detection is compensation for pH errors in glucose detection because the reaction between PBA and glucose is sensitive to pH. The emission peaks of the two detection units are 517 nm and 594 nm, respectively, so there is no signal interference between them. The sensor can continuously monitor glucose in 0–20 mM and pH in 5.4–7.8. The advantages of this sensor are multi-parameter simultaneous detection, transmission-detection integration, real-time dynamic detection, and good biocompatibility.
