Abstract:PURPOSE:Neural tube defects (NTDs) are one of the most prevalent and the most severe congenital malformations worldwide. Studies have confirmed that folic acid supplementation could effectively reduce NTDs risk, but the genetic mechanism remains unclear. In this study, we explored association of single nucleotide polymorphisms (SNP) within folate metabolic pathway genes with NTDs in Han population of Northern China.METHODS:We performed a case-control study to compare genotype and allele distributions of SNPs in 152 patients with NTDs and 169 controls. A total of 16 SNPs within five genes were genotyped by the Sequenom MassARRAY assay.RESULTS:Our results indicated that three SNPs associated significantly with NTDs (P<0.05). For rs2236225 within MTHFD1, children with allele A or genotype AA had a high NTDs risk (OR=1.500, 95%CI=1.061~2.120; OR=2.862, 95%CI=1.022~8.015, respectively). For rs1801133 within MTHFR, NTDs risk markedly increased in patients with allele T or genotype TT (OR=1.552, 95%CI=1.130~2.131; OR=2.344, 95%CI=1.233~4.457, respectively). For rs1801394 within MTRR, children carrying allele G and genotype GG had a higher NTDs risk (OR=1.533, 95%CI=1.102~2.188; OR=2.355, 95%CI=1.044~5.312, respectively).CONCLUSIONS:Our results suggest that rs2236225 of MTHFD1 gene, rs1801133 of MTHFR gene and rs1801394 of MTRR gene were associated with NTDs in Han population of Northern China.

Abstract:Objective To analyze the association between single nucleotide polymorphisms (SNPs) of folate metabolism pathway genes and neural tube defects(NTDs) in children of Han population in northern China.Methods One hundred and fifty-two NTDs patients and 169 controls in a case-control study were examined.Sixteen SNPs in 5 genes were genotyped by Sequenom MassARRAY.Prediction of effects of mutations on protein function was performed with SIFT and PolyPhen programs.Results The distribution of allele and genotype frequencies of methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) rs2236225 loci was significantly different between NTDs group and the healthy control group (x2 =5.292,6.080;P =0.021,0.048),and the children with A allele and AA genotype were associated with increased NTDs risk compared with G allele and GG genotype (OR =1.500,95％ CI 1.061-2.120;OR =2.862,95％ CI 1.022-8.015).The distribution of allele and genotype frequencies of methylenetetrahydrofolate reductase (MTHFR) rs1801133 loci was significantly different between NTDs group and the healthy control group(x2 =7.403,6.973;P =0.007,0.031),and the children with T allele and TF genotype were associated with increased NTDs risk compared with C allele and CC genotype(OR =1.552,95％ CI 1.130-2.131;OR =2.344,95％ CI 1.233-4.457).The distribution of allele and genotype frequencies of methionine synthase reductase(MTRR) rs1801394 loci was significantly different between NTDs group and the healthy control group(x2 =6.365,6.219;P =0.012,0.045),and the children with G allele and GG genotype were associated with increased NTDs risk compared with A allele and AA genotype (OR =1.553,95 ％ CI 1.102 ～ 2.188;OR =2.355,95 ％ CI 1.044-5.312).There was no difference in other loci among different genotypes or alleles.SIFT and Polyphen programs showed that the prediction results were damaging and probably damaging (MTHFR rs1801133 and MTRR rs1801394).Conclusions The rs2236225 of MTHFD1 gene,rs1801133 of MTHFR gene and rs1801394 of MTRR gene are associated with NTDs in children of Han population in Northern China.The further results confirmed that the genetic variation of folate metabolism pathway genes is one of NTDs susceptibility factors.

Correlation analysis of MTHFD1 gene polymorphisms and neural tube defects in Han population of Northern China

Abstract:Background Neural tube defects (NTDs) is a common birth defects worldwide. The methylenetetrahydrofolate dehydrogenase1 (MTHFD1) gene has been proved to play an important role in folate metabolism, which was strongly associated with the increased NTDs risk. The study is aimed to explore the correlation of single nucleotide polymorphisms (SNPs) in MTHFD1 gene with NTDs susceptibility. Methods A case-control study was conducted on children who included 152 NTDs patients and 169 healthy controls. Tag-SNPs were identified in HapMap database. Then, we investigated the association between NTDs and four selected tag-SNPs in MTHFD1 gene: rs1950902, rs2236225, rs2236224, rs11849530. We also performed a meta-analysis based on previous published studies to further evaluate the association. Results The results indicated that rs2236225 polymorphism displayed a significant association with NTDs risk (AA vs. GG: OR = 2.862, 95%CI = 1.022ཞ8.015; GA + AA vs. GG: OR = 1.619, 95%CI = 1.040ཞ2.520; A vs. G: OR = 1.500, 95%CI = 1.061ཞ2.120). In addition, rs2236224 polymorphism was correlated with increased NTDs risk (TT vs. CC: OR = 2.559, 95%CI = 1.128ཞ5.804; CT + TT vs. CC; OR = 1.631, 95%CI = 1.041ཞ2.556; T vs C: OR = 1.489, 95%CI = 1.072ཞ2.068). Further analysis showed the harmful effect of rs2236225 polymorphism was further supported by the result of meta-analysis. Meanwhile, haplotype analysis results showed that A-A and T-A haplotypes were correlated with increased NTDs risk, but C-A haplotype might decrease NTDs risk. Conclusions The results indicated that rs2236225 and rs2236224 polymorphisms of MTHFD1 gene were significantly associated with NTDs susceptibility in Han population of Northern China.

Abstract:OBJECTIVES:The methylenetetrahydrofolate dehydrogenase (MTHFD1) gene, as one of the key genes involved in the folate pathway, has been reported to play a critical role in the pathogenesis of neural tube defects (NTDs). However, the results of published studies are contradictory and inconclusive. Thus, this meta-analysis aimed to evaluate the effect of the common polymorphism in the MTHFD1 gene, the G1958A (R653Q, dbSNP ID: rs2236225) variant, on the risk of NTDs in all eligible studies. METHODS:Relevant literature published before January 3, 2014 was retrieved from the MEDLINE, EMBASE, Cochrane Library, and CBM databases. Pooled crude odds ratios (ORs) and their corresponding 95% confidence intervals (CIs) were calculated to evaluate the association between the MTHFD1 G1958A polymorphism and NTDs risk. RESULTS:We performed a meta-analysis of nine studies with a total of 4,302 NTDs patients and 4,238 healthy controls. Our results demonstrated a significant correlation between the MTHFD1 G1958A polymorphism and NTDs in an overall meta-analysis. For family-based studies, the study subjects were classified as NTD cases, mothers with NTDs offspring, and fathers with NTDs offspring. We found no association between any of the fathers' genotypes and NTDs, whereas there was a clear excess of the 1958A allele in the mothers of children with NTDs compared with controls individuals. CONCLUSIONS:In summary, our meta-analysis strongly suggests that the MTHFD1 G1958A polymorphism might be associated with maternal risk for NTDs in Caucasian populations. However, the evidence of this association should be interpreted with caution due to the selective nature of publication of genetic association studies.

Abstract:Background: To systematically evaluate the association of MTHFR genetic polymorphisms, maternal folic acid intake, and the time when folic acid intake was started with the risk of congenital heart disease (CHD) and investigated the role of their interaction on infant CHD risk in Chinese populations. Methods: A case–control study involving 592 CHD cases, 617 health controls, and their mothers was performed. The exposures of interest were single nucleotide polymorphisms (SNPs) of the MTHFR gene, maternal folic acid use, and the time when folic acid use was started. We applied the logistic regression model to explore the strength of association. Results: Our findings showed that mothers lacking folic acid intake had a significantly higher risk of CHD in offspring (aOR = 2.00; 95%CI: 1.34–2.98). Mothers who started to use folic acid from the first trimester of the fetation (aOR = 1.65; 95% CI: 1.22–2.23) or from the second trimester of the fetation (aOR = 7.77; 95% CI: 2.52–23.96), compared with those starting to use folic acid from 3 months previous to the conception, were at a significantly higher risk of CHD in offspring. Genetic variants at rs2066470 (AA vs. GG: aOR = 5.09, 95%CI: 1.99–13.03), rs1801133 (AA vs. GG: aOR = 2.49, 95%CI: 1.58–3.93), and rs1801131 (TG vs. TT: aOR = 1.84, 95%CI: 1.36–2.50; GG vs. TT: aOR = 3.58, 95%CI: 1.68–7.63) were significantly associated with the risk of CHD based on the multivariate analysis. Additionally, statistically significant interactions between maternal folic acid intake and genetic variants of the MTHFR gene at rs1801133 and rs1801131 were observed. Conclusion: An association of maternal folic acid intake and the time when intake was started with the risk of CHD in offspring was found. What's more, maternal folic acid fortification may help counteract partial of the risks of CHD in offspring attributable to MTHFR genetic mutations. Abstract:Objective To investigate the polymorphism of folate metabolism pathway genes in pregnant women with congenital heart diseases ( CHD), and to explore the relationship between the gene polymorphisms and the susceptibility to CHD. Methods According to the inclusion criteria, Collected 320 cases of fetal congenital heart disease diagnosed by color Doppler echocardiography. From peripheral venous blood, PCR-RELP method was used to detect the polymorphism of MTHFR677 and MTHFR1298 loci. Results (1) Gene loci polymorphism: In the control group and CHD group, C and T allele、A and C alleles all have differences in the 2 groups was statistically significant ( χ2= 16. 589, P ＜ 0. 001; χ2=5. 078, P = 0. 020); (2) Genotype polymorphism: In the control group and CHD group, CC, CT, TT genotype have differences in the both groups was statistically significant ( χ2= 15. 282, P ＜ 0. 001). AA, AC and CC genotype have no statistically significant difference in 2 groups ( χ2= 5. 092, P = 0. 080); (3) In the study of stratified: Tetralogy of fallot group CC, CT and TT genotype and AA, AC and CC genotype, with the control group there were statistical significance (χ2= 7. 794, P = 0. 020; χ2= 8. 998, P = 0. 010); Ventricular septal defect group CC, CT, TT genotype, with the control group there were statistical significance ( χ2= 10. 407, P ＜ 0. 001), And AA, AC and CC genotype, with the control group there were no statistical significance ( χ2= 0. 667, P = 0. 720). Conclusion The MTHFR C677 T polymorphism of mother is associated with congenital heart disease in children. Hierarchical research tips that the MTHFR C677 T polymorphism of mother is associated with occurrence of tetra logy of fallot and ventricular septal defect in children. The other results in the case group and the control group MTHFR A1298 C genotype although no obvious difference, but the A/C allele is having the significant difference, prompt allele A/C to children is A risk factor of CHD.
Polymorphisms in FZD3 and FZD6 genes and risk of neural tube defects in a northern Han Chinese population

Abstract:Neural tube defects (NTDs) are the most common and severe malformations of the central nervous system. The association of single nucleotide polymorphisms (SNPs) of the Frizzled 3 ( FZD3 ) and Frizzled 6 ( FZD6 ) genes and NTDs in the Han population of northern China was principally studied. One synonymous SNP (rs2241802) in FZD3 gene and three nonsynonymous SNPs (rs827528, rs3808553 and rs12549394) in FZD6 gene were analyzed by polymerase chain reaction (PCR) and sequencing methods in 135 NTD patients and 135 normal controls. The allele, genotype and haplotype frequencies were calculated and analyzed to examine the relationship between FZD3/FZD6 SNPs and NTDs. Both T allele and TT genotype frequencies of the FZD6 rs3808553 loci in the NTDs group were significantly higher than those in the controls, and children with T allele and TT genotype were associated with increased NTDs risk (OR = 1.575, 95 % CI 1.112–2.230, P = 0.010 and OR = 2.811, 95 % CI 1.325–5.967, P = 0.023, respectively). There were no differences among different genotypes or alleles in other three SNPs. Haplotypes A-G-C and A-T-C in FZD6 were found associated with NTDs in the case–control study (OR = 0.560, 95 % CI 0.378–0.830, P = 0.004 and OR = 1.670, 95 % CI 1.126–2.475, P = 0.011, respectively). The rs3808553 of FZD6 is obviously associated with NTDs in Han population of northern China. The TT genotype may increase risk for NTDs.
Association of Genetic Polymorphisms of De Novo Nucleotide Biosynthesis with Increased Chd Susceptibility in the Northern Chinese Population

Abstract:Congenital heart disease (CHD) is one of most prevalent birth defects in the world. However, the underlying molecular mechanism(s) have not been fully understood. Here we report that increased CHD susceptibility is associated with genetic polymorphisms for de novo nucleotide biosynthesis in northern Chinese population, which has been reported with lower plasma folate levels. Nine tagSNPs of four genes (GART, ATIC, MTHFD1 and SHMT1) in de novo nucleotide biosynthesis were sequenced in 802 sporadic CHD patients and 1093 controls from two Han Chinese populations, located in north China (Shandong) and South China (Shanghai), respectively. Six SNPs were found to be significantly associated with CHDs or septation defects only in the Shandong population dataset, but none displayed significant association with any CHDs in the Shanghai population dataset as well as in the combined dataset. We also showed that the minor A allele of rs7279549 in GART reduced transcriptional activity and displayed lower affinity for unknown transcription factor(s), demonstrating the allele is a functional risk factor for CHD in Shandong population. Our study indicates that dysregulation of de novo nucleotide biosynthesis pathway may conditionally contribute to CHD pathogenesis in northern Chinese.
The Relationship Between Frizzled 6 Gene Polymorphisms and Neural Tube Defects in Children of Northern Han Chinese Population

Abstract:: To investigate the association of MTHFR C677T, MTHFR A1298C and MTRR A66G polymorphisms with birth defects in southern Chinese population. Genotyping was performed by Fluorescence Quantitative Analyzer using the Sequencing Reaction Universal Kit. Association analysis method was used to explore the relationship between genetic polymorphisms in MTHFR , MTRR gene and birth defects. Our results showed that serum folic acid level of genotype TT in MTHFR C677T was significantly lower than other genotypes, while homocysteine level significantly higher compared with CC and CT ( P < 0.05). In addition, genotype GG in MTRR A66G might also promote homocysteine accumulation ( P < 0.05). Results of logistic regression represented that MTHFR C677T, MTHFR A1298C, and MTRR A66G polymorphisms were not important or independent risk factors for predicting birth defects. Besides, genotype distribution of MTHFR C677T was significantly different in normal and abnormal pregnancy population, and genotype TT might affect folic acid metabolism and promote homocysteine accumulation. However, MTHFR C677T, MTHFR A1298C, and MTRR A66G polymorphisms were not critical or independent risk factors for predicting birth defects in this study. the dosage of folic acid 14) . In this study, we ex-plored the relationship between the key enzyme genes MTHFR and MTRR of folic acid metabolism and birth defects. Our results showed the genotype distribution of MTHFR C677T, MTHFR A1298C and MTRR A66G polymorphisms was significantly different between normal and abnormal pregnancy populations in southern China. Genotype TT in MTHFR C677T might not be conducive to folic acid metabolism and normal pregnancy, while promoting homocysteine accumulation. Abstract:Objective To explore the association between maternal methylene tetrahydrofolate reductase (MTHFR) C677T polymorphism and neural tube defects (NTDs). Methods CBM, VIP, CNKI, Wanfang, PubMed and Web of Science databases from set up to March, 2014 were electronically searched to identify case-control studies on the relationship between maternal MTHFR C677T polymorphism and NTDs. The data were quantitatively analyzed by RevMan 5.0 software. Results A total of 25 studies were selected including 2 282 cases and 3 420 controls. Overall, the pooled OR (with 95%CI) under co-dominant model and allele contrast were 2.28(1.60-3.24), 1.25(1.02-1.53) and 1.42(1.21-1.67). Subgroup analysis showed significant association between maternal MTHFR C677T polymorphism and NTDs susceptibility in Asian populations. Conclusion The present meta-analysis suggests that MTHFR C677T polymorphism is significantly associated with maternal risk for NTDs, especially in Asian populations.
Genetic Variants in the Folate Pathway and the Risk of Neural Tube Defects: A Meta-Analysis of the Published Literature

Abstract:BACKGROUND: Neural Tube Defects (NTDs) are among the most prevalent and most severe congenital malformations worldwide. Polymorphisms in key genes involving the folate pathway have been reported to be associated with the risk of NTDs. However, the results from these published studies are conflicting. We surveyed the literature (1996-2011) and performed a comprehensive meta-analysis to provide empirical evidence on the association. METHODS AND FINDINGS: We investigated the effects of 5 genetic variants from 47 study populations, for a total of 85 case-control comparisons MTHFR C677T (42 studies; 4374 cases, 7232 controls), MTHFR A1298C (22 studies; 2602 cases, 4070 controls), MTR A2756G (9 studies; 843 cases, 1006 controls), MTRR A66G (8 studies; 703 cases, 1572 controls), and RFC-1 A80G (4 studies; 1107 cases, 1585 controls). We found a convincing evidence of dominant effects of MTHFR C677T (OR 1.23; 95%CI 1.07-1.42) and suggestive evidence of RFC-1 A80G (OR 1.55; 95%CI 1.24-1.92). However, we found no significant effects of MTHFR A1298C, MTR A2756G, MTRR A66G in risk of NTDs in dominant, recessive or in allelic models. CONCLUSIONS: Our meta-analysis strongly suggested a significant association of the variant MTHFR C677T and a suggestive association of RFC-1 A80G with increased risk of NTDs. However, other variants involved in folate pathway do not demonstrate any evidence for a significant marginal association on susceptibility to NTDs.
Single Nucleotide Polymorphisms of the Maternal Msx2 Gene and Their Association with Fetal Neural Tube Defects in Han Ethnic Group in Shanxi Province, China.

Abstract:Background Neural tube defects are the most common human birth defects. The causes are multifactorial with complex genetic and environmental factors, although the exact genetic causes are unknown. This research was conducted to study the frequency of Msx2 gene polymorphisms in 59 women with a history of pregnancy with a neural tube defect and in 73 healthy controls. We aimed to determine the effect of this genetic polymorphism on the incidence of neural tube defects in the Han Chinese population.Methods We studied 59 mothers with at least one previous child with a neural tube defect (the case group) and 73 case-control subjects during the same period, from Shanxi Province, China. We analyzed the genotypic distributions and allele frequencies of Msx2 C386T polymorphisms in DNA samples from the case and control groups. A three-dimensional protein model was predicted using Swiss-Pdb Viewer software version 4.0. Disease association was analyzed using chi-square tests.Results Significant differences were observed in the genotypes and allele frequencies of the Msx2 C386T allele between the case and control groups (CT: 32% vs. 15%, P=0.0073 and TT 15% vs. 4%, P=0.013, respectively). Logistic regression analysis showed that the C386T mutation is a potential risk factor for neural tube defects (P < 0.05; OR: 3.466; 95 %Cl: 1.831-6.560). Three-dimensional structure prediction revealed that the Msx2 C386T mutation results in a threonine substitution for methionine at position 129 of exon 2, which might lead to structural mutations or dysfunctions in the protein encoded by Msx2.Conclusion Maternal Msx2 C386T gene polymorphisnns were associated with fetal neural tube defects in Han Chinese women in Shanxi Province. Association of Snps in Genes Involved in Folate Metabolism with the Risk of Congenital Heart Disease

Abstract:Objective: To investigate the association of 12 single nucleotide polymorphisms (SNPs) in folate metabolic genes with congenital heart disease (CHD).Methods: A total of 160 children with CHD and 188 control children were enrolled. Twelve SNPs related to folate metabolism, including CBS-C699T, DHFR-c594+59del19, FOLH1-T1561C, CBS-C699T, DHFR-c594+59del19, GSTO1-C428T, MTHFD-G878A and -G1958A, MTHFR-C677T and -A1298C, MTR-A2756G, MTRR-A66G, NFE2L2-ins1+C11108T, RFC1-G80A, TCN2-C776T and TYMS-1494del6, were genotyped by SNaPShot genotyping technology and confirmed by Sanger sequencing.Results: There were two SNPs including NFE2L2-ins1+C11108T and GST01-C428T and two compound mutants for (MTHFD-G1958A, MTHFR-C677T and MTR-A2756G) and (MTHFD-G1958A, RFC1-G80A and MTR-A2756G), which might increase the risk of CHD, and DHFR-c594+59del19 might decrease the risk of CHD. The CT genotype of NFE2L2-ins1+C11108T, OR = 2.15 (95% CI = [1.07, 4.32], p<0.05). The CT+TT genotype of NFE2L2-ins1+C11108T, OR = 1.98 (95% CI = [1.00, 3.93], p<0.05). The TT genotype of GST01-C428T, OR = 3.49, (95CI% = [1.06, 11.5], p<0.05). The GG genotype of DHFR-c594+59del19, OR = 0.46 (CI% = [0.24, 0.87], p<0.05). The AG+GG genotype of DHFR-c594+59del19, OR = 0.53 (CI% = [0.29, 0.96], p<0.05). The ratios of the two compound mutants for (MTHFD-G1958A, MTHFR-C677T and MTR-A2756G) and (MTHFD-G1958A, RFC1-G80A and MTR-A2756G) in CHD are higher than that in control, p50.05 (OR = 2.968, 95% CI = [1.022, 8.613]).Conclusions: The CT genotype of NFE2L2-ins1+C11108T and the TT genotype of GST01-C428T are susceptible factors for CHD. The AG, GG and (AG+GG) genotypes of DHFR-c594+59del19 are protective genotypes for CHD. Compound mutants for (MTHFD-G1958A, MTHFR-C677T and MTR-A2756G) and (MTHFD-G1958A, RFC1-G80A and MTR-A2756G) may increase the risk of CHD.

Abstract:Neural tube defects (NTDs) are birth defects of the brain, spine, or spinal cord invoked by the insufficient intake of folic acid in the early stages of pregnancy and have a complex etiology involving both genetic and environmental factors. So the study aimed to explore the association between alterations in maternal one-carbon metabolism and NTDs in the offspring. We conducted a case-control study to get a deeper insight into this association, as well as into the role of genetic polymorphisms. Plasma concentrations of folate, homocysteine (Hcy), S-adenosylmethionine (SAM), S-adenosylhomocysteine (SAH) and genotypes and alleles distributions of 52 SNPs in 8 genes were compared for 61 women with NTDs-affected offspring and 61 women with healthy ones. There were significant differences between groups with regard to plasma folate, SAM, SAH and SAM/SAH levels. Logistic regression results revealed a significant association between maternal plasma folate level and risk of NTDs in the offspring. For MTHFD1 rs2236225 polymorphism, mothers having GA genotype and A allele exhibited an increased risk of NTDs in the offspring (OR = 2.600, 95%CI: 1.227–5.529; OR = 1.847, 95%CI: 1.047–3.259). For MTHFR rs1801133 polymorphism, mothers having TT and CT genotypes were more likely to affect NTDs in the offspring (OR = 4.105, 95%CI: 1.271–13.258; OR = 3.333, 95%CI: 1.068–10.400). Moreover, mothers carrying T allele had a higher risk of NTDs in the offspring (OR = 1.798, 95%CI: 1.070–3.021). For MTRR rs1801394 polymorphism, the frequency of G allele was significantly higher in cases than in controls (OR = 1.763, 95%CI: 1.023–3.036). Mothers with NTDs-affected children had higher AG genotype in RFC1 rs1051226 polymorphism than controls, manifesting an increased risk for NTDs (OR = 3.923, 95%CI: 1.361–11.308). Folic acid deficiency, MTHFD1 rs2236225, MTHFR rs1801133, MTRR rs1801349 and RFC1 rs1051226 polymorphisms may be maternal risk factors of NTDs.

Abstract:Objective To evaluate associations between MTHFD1 gene G1958A polymorphism and the risk of neural tube defects (NTDs).Methods We electronically searched databases including PubMed,The Cochrane Library,Web of Science,CNKI,VIP,and WanFang Data from inception to June 2016 to collect case-control studies of the correlation between the G1958A polymorphism in MTHFD1 and the risk of NTDs.Two reviewers independently screened the studies,extracted data and assessed the risk of bias of included studies,and then,meta-analysis was performed using Stata 12.0 software.Results Thirteen case-control studies were included,involving 1 724 NTDs infants,1 485 mothers and 774 fathers with NTDs offspring.The results of meta-analysis showed that there was significant association between MTHFD1 gene G1958A polymorphism and increased risk of NTDs in infants (AA vs.GG:OR=1.437,95％CI 1.100 to 1.878,P=0.008;AA+AG vs.GG:OR=1.187,95％CI 1.031 to 1.367,P=0.017;A vs.G:OR=l.210,95％CI 1.050 to 1.394,P=0.008).However,there was no association between biparental MTHFD1 gene G1958A polymorphism and NTDs in the offspring.Conclusion The current evidence shows that MTHFD1 gene G1958A polymorphism may be a genetic risk factor for NTDs.Due to the limited quantity and quality of the included studies,more high quality studies are needed to verify the above conclusion.

Abstract:Objective To study the association of single nucleotide polymorphisms (SNPs) of the VANGL1 gene and neural tube defects (NTDs) in the Han population of northern China.Methods The SNPs were genotyped and sequenced by polymerase chain reaction (PCR) in 135 NTD patients and paired normal controls.The genotypes and allele frequencies were calculated and analyzed to examine the correlation between VANGL1 SNPs and NTDs.Prediction of 3-D protein structure was conducted by CPHmodels platform.Results We found two nonsynonymous SNPs in the VANGL1 gene which lead to amino acid changes (rs4839469 c.346G＞A p.Ala116Thr; rs34059106 c.1040A＞C p.Glu347Ala).Both G＞A (Ala116Thr) and G＞C (Ala116Pro) polymorphisms at codon 116 of VANGL1 gene were identified.Significant associations were found between the VANGL1 codon 116 polymorphism and NTDs (P =0.036 and 0.010 for allele and genotype frequencies,respectively).The odd ratio of the GC genotype was 9.750.The 3-D protein structure prediction showed that the VANGL1 gene codon 116 Pro mutant had shortened a-helix than that of wild type.The Glu347Ala variant was not detected in both groups.Conclusions The rs4839469,not the rs34059106 of VANGL1 is associated with NTDs in Han population of northern China.The GC genotype may increase risk for NTDs.

Abstract:OBJECTIVE: To determine whether 5,10-methylenetetrahydrofolate reductase A1298C polymorphism is associated with neural tube defect susceptibility.STUDY DESIGN: Computerized literature searches of the PubMed, Embase, and Medline database were conducted to identify all eligible articles. Study subjects were classified as neural tube defect patients, mothers with neural tube defect offspring and fathers with neural tube defect offspring. The pooled odds ratios with 95% confidence interval (CI) were calculated by fixed-effects model or random-effects model.RESULTS: Twenty-three case-control studies were finally included. The pooled results showed no significant association between 5,10-methylenetetrahydrofolate reductase A1298C polymorphism and neural tube defect susceptibility in any genetic contrast among the 3 groups, except 1 (AC vs AA in neural tube defect patients: odds ratio, 1.16; 95% CI, 1.10-1.32; P = .03), which, however, turned out to be of no statistical significance in the subgroup of the white population (odds ratio, 1.14; 95% CI, 0.98-1.31; P = .08).CONCLUSION: This metaanalysis suggests that 5,10-methylenetetrahydrofolate reductase A1298C polymorphism is not associated with neural tube defect susceptibility in the white population.
Genome-Wide Association Study Identifies Genetic Polymorphisms for Folate-Related Biomarkers in Chinese Preconception Women

Abstract:Background: Single-nucleotide polymorphism (SNP) allele frequencies, dietary habits, and folate status and their associations vary across ethnic populations. Little is known about the SNPs accounting for variations of folate-related biomarkers for Chinese preparing-for-pregnant females. Objectives: We aimed to identify SNPs contributing to RBC and serum folate, vitamin B-12, and homocysteine concentrations in Chinese female preconception population. Methods: A genome-wide association study was conducted on 1000 randomly selected preconception Chinese women from the Shanghai Preconception Cohort. SNPs were genotyped using Illumina chips, and associations with biomarkers were assessed using simple linear regression models under the assumption of an additive genetic model. Genome-wide significance was considered at P < 10-7. Results: The MTHFR rs1801133 was the major genetic coding variant contributing to RBC folate, serum folate, and homocysteine concentrations (P = 2.28 × 10-16; P = 8.85 × 10-8, and P = 2.46 × 10-13, repsectively). It is associated with increased RBC folate (β: 0.154 per additional risk allele after log transform), decreased serum folate (β: -0.951 per additional risk allele), and increased serum homocysteine concentrations (β: 1.153 per additional risk allele). The predominant SNP associated with serum folate was rs147162222 in NTRK2 (P = 2.55 × 10-8), although that associated with homocysteine was rs77025184 located between PDE7B and LINC00271 (P = 4.91 × 10-17). For vitamin B-12, FUT2 rs1047781 was the dominant genetic variant (P = 1.59 × 10-10). The numbers of signals with a P value of <10-7 for RBC folate, serum folate, vitamin B-12, and homocysteine were 12, 18, 8, and 614, respectively. Conclusions: This study represents the first genome-wide association study focusing on folate-related biomarkers in a Chinese preparing-for-pregnant female population. The contributions of dominent SNPs to each biomarker are partly different from other populations. The rs1801133 (C677T) in MTHFR is the predominant genetic variant contributing to RBC folate and rs1047781 (A385T) in FUT2 as the primary one explaining vitamin B-12. Notably, the intronic rs147162222 and noncoding rs77025184 are the predominant SNPs for serum folate and homocysteine, respectively.
