Abstract:Chronic cerebral hypoperfusion (CCH) is a major risk factor for cognitive decline and degenerative processes. Shunaoxin dropping pill (SNX) has been clinically used to treat cerebrovascular diseases. However, the effect and mechanism of SNX in treating CCH-induced cognitive impairment remain unclear. In this study, CCH was induced in rats using permanent bilateral common carotid artery ligation (2-VO). CCH rats were characterized by impaired spatial learning and memory ability, as well as increased oxidative stress and inflammation in the hippocampus. Additionally, CCH rats had reduced richness and biodiversity of fecal microbiota, which showed a strong correlation with altered serum metabolites. SNX significantly improved the cognitive impairment and restored the dysbiosis of fecal microbiota and serum metabolites in CCH rats. Notably, SNX did not prevent cognitive impairment in antibiotics-treated CCH rats. Our findings suggest that the microbiota-gut-brain axis is a promising therapeutic target for the treatment of CCH-induced cognitive impairment.

Abstract:Background: Shexiang Baoxin Pill (SBP), a formulated traditional Chinese medicine (TCM), has been widely used to treat cardiovascular diseases for years. This herbal mixture has been shown to promote differentiation of cultured neuronal cells. Here, we aimed to investigate the effects of SBP in attenuating cognitive impairment in APP/PS1 transgenic mice.Methods: Ethanol and water extracts of SBP, denoted as SBPEtOH and SBPwater, were standardized and applied onto cultured rat pheochromocytoma PC12 cells. The potential effect of SBPEtOH extract in attenuating the cognitive impairments in APP/PS1 transgenic mice was shown by following lines of evidence: (i) inhibition of Aβ fibril formation, (ii) suppression of secretions of cytokines, and (iii) improvement of behavioral tests by Morris water maze.Results: SBPwater and SBPEtOH inhibited the formation of β-amyloid fibrils and protected the Aβ-induced cytotoxicity in cultured PC12 cells. In APP/PS1 transgenic mice, the treatment of SBPEtOH inhibited expressions of NO, NOS, AChE, as well as aggregation of Aβ. Besides, the levels of pro-inflammatory cytokines were suppressed by SBP treatment in the transgenic mice. Importantly, the behavioral tests by Morris Water maze indicated that SBP attenuated cognitive impairments in APP/PS1 transgenic mice.Conclusion: The current result has supported the notion that SPB might ameliorate the cognitive impairment through multiple targets, suggesting that SBP could be considered as a promising anti-AD agent.

Abstract:Objective:To observe the Huanglong pill on D-galactose induced subacute aging model independent activity,memory and resistance to fatigue and SOD activity.Methods: 40 ICR mice,male and female in half,were randomly divided into 4 groups,namely control group,model group,Huanglong pills,anti-Fu-chun tablet group.Huanglong Pill to 2.5g/kg,anti-Fu Chun-chip group 0.7g/kg,ig are 0.2mL/10g,model group and control group was given the same volume of distilled water,once a day for gavage 49 days.Administration of the same model group and treatment group mice were injected daily neck back 5% D-galactose,the control group injected with normal saline,0.5mL / only,continuous injection of 49 days.Huanglong pill on the D-galactose induced subacute aging mice spontaneous activity,learning and memory and learning platform memory forgotten,swimming time and the activity of serum SOD.Results:Huanglong pill could result D-galactose induced subacute aging mice significantly increased the number of standing,compared with model group,significant difference P0.05;significantly enhance the learning and memory and forgetting to extend the time for learning and memory,could step down test latency extended the number of errors decreased,and the model group P0.05;aging mice was significantly prolonged the swimming time,and the model group P0.05;increased serum SOD activity,compared with the model group P0.05.Conclusion:Huanglong pills can improve the D-galactose induced subacute aging mice of many signs,has some anti-aging effect.

Abstract:Dengzhan Shengmai (DZSM), a traditional Chinese medicine formulation, has been administered extensively to elderly individuals with cognitive impairment (CI). However, the underlying mechanisms by which Dengzhan Shengmai improves cognitive impairment remains unknown. This study aimed to elucidate the underlying mechanism of the effect of Dengzhan Shengmai on aging-associated cognitive impairment via a comprehensive combination of transcriptomics and microbiota assessment. Dengzhan Shengmai was orally administered to a D-galactose-induced aging mouse model, and evaluation with an open field task (OFT), Morris water maze (MWM), and histopathological staining was performed. Transcriptomics and 16S rDNA sequencing were applied to elucidate the mechanism of Dengzhan Shengmai in alleviating cognitive deficits, and enzyme-linked immunosorbent assay (ELISA), quantitative real-time polymerase chain reaction (PCR), and immunofluorescence were employed to verify the results. The results first confirmed the therapeutic effects of Dengzhan Shengmai against cognitive defects; specifically, Dengzhan Shengmai improved learning and impairment, suppressed neuro loss, and increased Nissl body morphology repair. Comprehensive integrated transcriptomics and microbiota analysis indicated that chemokine CXC motif receptor 4 (CXCR4) and its ligand CXC chemokine ligand 12 (CXCL12) were targets for improving cognitive impairments with Dengzhan Shengmai and also indirectly suppressed the intestinal flora composition. Furthermore, in vivo results confirmed that Dengzhan Shengmai suppressed the expression of CXC motif receptor 4, CXC chemokine ligand 12, and inflammatory cytokines. This suggested that Dengzhan Shengmai inhibited CXC chemokine ligand 12/CXC motif receptor 4 expression and modulated intestinal microbiome composition by influencing inflammatory factors. Thus, Dengzhan Shengmai improves aging-related cognitive impairment effects via decreased CXC chemokine ligand 12/CXC motif receptor 4 and inflammatory factor modulation to improve gut microbiota composition.

Effect of Shunaoxin Dropping Pills on Neurocognitive Function of Patients with Vascular Cognitive Impairment of None Dementia

Abstract:Objective To evaluate the effect of Shunaoxin dropping pills on neuropsychological features of patients with vascular cognitive impairment of none dementia.Methods One hundred and three outpatients with vascular cognitive impairment of none dementia,collected from November 2014 to January 2015,were enrolled in our study;they were divided into two groups:Shunaoxin treatment group (n=50) received Shunaoxin pills and conventional therapy and control group (n=53) received conventional therapy;conventional therapy included anti-platelet therapy (clopidogrel 75 mg once daily),control of pressure (target value=140/90 mmHg) and control of blood fat (atorvastatin 20 mg once daily).Shunaoxin pills (four pills/time for three times per day) were taken orally by the patients from the Shunaoxin treatment group for consecutive three months.Their neuropsychological features were evaluated by Montreal cognitive assessment (MoCA),digital span test (DST),auditory verbal learning test (AVLT),trail making test (TMT),verbal fluency test (VFT) and Stroop test before treatment and 3 months after treatment.Results There were 47 available cases in both two groups,respectively.As compared with control group,Shunaoxin treatment group had significantly increased MoCA total scores,scores of MoCA-language and MoCA-delayed recall,and VLT scores (learning,recall and recognition,P＜0.05),while there were no significant changes in the rest parts of MoCA,TMT,VFT and Stroop test (P＞0.05).Conclusion Based on the therapy of anti-platelet,control of pressure and blood fat,3 months therapy of Shunaoxin dropping pills can improve general cognition,especially in verbal learning,delayed recall and verbal repeat.
Shenqi Xingnao Granules Ameliorates Cognitive Impairments and Alzheimer’s Disease-Like Pathologies in APP/PS1 Mouse Model

Abstract:Objective: Alzheimer's disease (AD) is along with cognitive decline due to amyloid-p (Ap) plaques, tau hyperphosphorylation, and neuron loss. Shenqi Xingnao Granules (SQXN), a traditional Chinese medicine, significantly ameliorated the cognitive function and daily living abilities of patients with AD. However, till date, no study has investigated the mechanism of action of SQXN on AD. The present study aimed to verify the effects of SQXN treatment on cognitive impairments and AD-like pathologies in APP/PS1 mice. Methods: Four-month-old APP/PS1 transgenic (Tg) mice were randomly divided into a model group and SQXN-treated (3.5, 7, 14 g/kg per day) groups. Learning-memory abilities were determined by Morris water maze and object recognition test. All mice were sacrificed and the brain samples were collected after 75 d. The soluble Ap contents were detected by Elisa kit; The levels of expression of NeuN, APP, phosphorylated tau and related protein were measured by Western blotting; The inflammation factors were detected by the proinflammatory panel kit. Results: Four-month-old APP/PS1 mice were administered SQXN by oral gavage for 2.5 months. Using the Morris water maze tests and Novel object recognition, we found that SQXN restored behavioral deficits in the experimental group of Tg mice when compared with the controls. SQXN also inhibited neuronal loss (NeuN marker). SQXN treatment decreased soluble Ap42 through inhibiting the expression of sAPPp and BACE-1 without regulating full-length amyloid precursor protein (FL APP). Insulin degrading enzyme (IDE), the Ap degrading enzyme, were increased by SQXN. In addition, SQXN reduced hyperphosphory-lated tau protein levels and prevented excessive activation of p-GSK-3p in the brain of APP/PS1 mice. Compared with APP/PS1 transgenic negative mice, IFN-y, IL-1p, IL-2, IL-4, IL-5, IL-6, IL-12p70, KC/GRO and TNF-x were not obviously changed in the brain of 6.5-month-old APP/PS1 transgenic (Tg) mice. However, SQXN could inhibited the expression of IL-2. Conclusion: These results demonstrate that SQXN ameliorates the cognitive impairments in APP/PS1 mice. The possible mechanisms involve its inhibition of neuronal loss, soluble Ap deposition, tau hyperphosphorylation and inflammation. (C) 2020 Tianjin Press of Chinese Herbal Medicines. Danggui-Shaoyao-San Ameliorates Cognition Deficits and Attenuates Oxidative Stress-Related Neuronal Apoptosis in D-Galactose-Induced Senescent Mice

Abstract:Ethnopharmacological relevance: Danggui-Shaoyao-San (DSS), a famous traditional Chinese medicine formula consisting of six herbal medicines, has been used to treat gynecological disorders and neural dysfunctions.Aim of the study: The present study was carried out to investigate the effects of DSS on cognitive ability and oxidative stress-related neuronal apoptosis in the hippocampus of aging mice induced by D-galactose (D-gal) to elucidate the underlying molecular mechanisms.Materials and methods: Ethanol extract of DSS (DE) were orally administered to D-gal-induced senescent mice for six weeks. The cognitive ability was determined by the methods of step-down type passive avoidance test and Morris water maze test. The activities of superoxide dismutase (SOD) and nitric oxide synthase (NOS), and levels of carbonyl protein (CP), glutathione (GSH), malondialdehyde (MDA) and nitric oxide (NO) were also examined. Furthermore, the expression of apoptotic related proteins in hippocampus of D-gal-treated mice, such as Bcl-2, Bax and caspase-3 proteins, were determined by immunohistochemistry.Results: DE at the doses of 1.8, 3.6 and 7.2 g/kg significantly enhanced the cognitive performances and restored the abnormal activities of SOD and NOS and levels of CP. MDA, GSH and NO induced by D-gal. Moreover, the neural apoptosis in the hippocampus of D-gal-treated mice was improved by DE through regulating the expression of Bcl-2, Bax and caspase-3.Conclusion: These results demonstrate that DE has neuroprotective effects in D-gal-induced senescent mice by ameliorating oxidative stress induced neuronal apoptosis in the brain. Shikonin Ameliorates D-galactose-induced Oxidative Stress and Cognitive Impairment in Mice Via the MAPK and Nuclear Factor-Κb Signaling Pathway

Abstract:Oxidative stress acts as the major causative factor for various age-associated neurodegenerative diseases, triggering cognitive and memory impairments. In the present study, the underlying neuroprotective mechanism governing how shikonin acts against D-galactose (D-gal)-induced memory impairment, neuroinflammation and neuron damage was examined. The results revealed that chronic administration of D-gal [150 mg/kg intraperitoneally (i.p.)] in mice caused cognitive and memory impairments, as determined by Morris water-maze test. Shikonin treatment, however, alleviated D-gal-induced memory impairment and reversed the D-gal-induced neural damage and apoptosis. Furthermore, western blotting and the results of morphological analysis revealed that shikonin treatments markedly reduced D-gal induced neuroinflammation through inhibition of astrocytosis as determined by glial fibrillary acidic protein (GFAP) detection, and downregulating other inflammatory mediators, including tumor necrosis factor-alpha (TNF-alpha), interleukin-1 beta (IL-1 beta), and IL-6. Moreover, shikonin treatment led to inhibition of the activation of nuclear factor-kappa B (NF-kappa B) and the phosphorylation of mitogen-activated protein kinases (MAPKs), preventing neurodegeneration. Hence, taken together, the results of the present study suggested that shikonin attenuated D-gal-induced memory impairment, neuroinflammation and neurodegeneration, possibly via the NF-kappa B/mitogen-activated protein kinase (MAPK) pathway. Our data suggest that shikonin could be a promising, endogenous and compatible antioxidant candidate for age-associated neurodegenerative diseases, including Alzheimer's disease.
Kai-Xin-San, a Chinese Herbal Decoction, Accelerates the Degradation of β-Amyloid by Enhancing the Expression of Neprilysin in Rats

Abstract:This study aimed to investigate the mechanisms of Kai-Xin-San (KXS, a famous Chinese herbal decoction used to treat amnesia) on the degradation of Aβ and further elucidate the mechanism of KXS on Aβ-induced memory dysfunction. After pretreatment with KXS (1.08 g/kg/day) for two weeks, Aβ 42 (2 μL, 200 μM) was injected into rat hippocampus to induce cognitive dysfunction. Morris water maze (MWM) test was developed to evaluate cognitive performance in rats. Hippocampal neurons were observed by histological staining using Hematoxylin-Eosin (HE) methods. Levels of exogenous Aβ 42, which was injected into the hippocampus, were continually measured through a special Enzyme-linked immunoassay (ELISA) kit to observe the catabolic process of Aβ in the brain. Similarly, Aβ degradation in PC12 cells was also investigated using the ELISA kit. The expressions of Aβ degeneration enzymes, including neprilysin (NEP), angiotensin-converting enzyme (ACE), and endothelin-converting enzyme (ECE), were detected by western blotting to elucidate Aβ reduction mechanism. Our results showed that KXS prevented Aβ 42-induced cognitive impairment and attenuated hippocampus neuronal damage caused by Aβ 42. Moreover, KXS could accelerate Aβ 42 degradation in Aβ 42 injected rats. Furthermore, NEP, an Aβ degradation enzyme, was increased in the hippocampus while ECE and ACE, other two Aβ-degrading enzymes, were not changed. KXS accelerated Aβ degradation in PC12 cells. Our findings revealed that KXS facilitated the degradation of Aβ 42 by increasing the expression of NEP in rat hippocampus. By reducing the Aβ burdens, KXS protected hippocampal neurons, leading to the improvement of cognitive function in rats.

Abstract:Alzheimer’s disease (AD) is the most common form of dementia in the elderly. It may be caused by oxidative stress, inflammation, and cerebrovascular dysfunctions in the brain. LongShengZhi Capsule (LSZ), a traditional Chinese medicine, has been approved by the China Food and Drug Administration for treatment of patients with cardiovascular/cerebrovascular disease. LSZ contains several neuroprotective ingredients, including Hirudo, Astmgali Radix, Carthami Flos (Honghua), Persicae Semen (Taoren), Acori Tatarinowii Rhizoma (Shichangpu) , and Acanthopanax Senticosus (Ciwujia) . In this study, we aimed to determine the effect of LSZ on the AD process. Double transgenic mice expressing the amyloid-β precursor protein and mutant human presenilin 1 (APP/PS1) to model AD were treated with LSZ for 7 months starting at 2 months of age. LSZ significantly improved the cognition of the mice without adverse effects, indicating its high degree of safety and efficacy after a long-term treatment. LSZ reduced AD biomarker Aβ plaque accumulation by inhibiting β-secretase and γ-secretase gene expression. LSZ also reduced p-Tau expression, cell death, and inflammation in the brain. Consistently, in vitro , LSZ ethanol extract enhanced neuronal viability by reducing L-glutamic acid-induced oxidative stress and inflammation in HT-22 cells. LSZ exerted antioxidative effects by enhancing superoxide dismutase and glutathione peroxidase expression, reduced Aβ accumulation by inhibiting β-secretase and γ-secretase mRNA expression, and decreased p-Tau level by inhibiting NF-κB-mediated inflammation. It also demonstrated neuroprotective effects by regulating the Fas cell surface death receptor/B-cell lymphoma 2/p53 pathway. Taken together, our study demonstrates the antioxidative stress, anti-inflammatory, and neuroprotective effects of LSZ in the AD-like pathological process and suggests it could be a potential medicine for AD treatment.

Abstract:Objective: To investigate the effects of Zuogui Pill on blood anti-oxidative abilities and DNA damage of lymphocytes in aging rats induced by D-galactose.Methods: 30 male SD(2 months old) rats were equally divided into three groups randomly: young control group,old group,Zuogui Pill group.The rats in old group were administered D-galactose 400mg.kg-1.d-1 by intraperitoneal injection for 60 days consecutively and the rats in young control group were administered the equal volum normal sodium.And the intervention group were given Zuogui Pill with 4.97g.kg-1.d-1by intragastric administration at the same time.The SOD activity and MDA content in serum were measured.Single Cell Gel Electrophoresis(SCGE) was used to detect the DNA damage of lymphocytes.Results: Compared with young control group,the SOD activity were decreased and the content of MDA were increased in serum of the old group(P<0.01).The DNA damage of lymphocytes was increased(P<0.01).The variations above in aging rats were restored statistically in Zuogui Pill group.Conclusion: Zuogui Pill could slow down the aging progress through improving the blood anti-oxidative ability and decreasing the DNA damage of lymphocytes.
Inhibitory Action of Kaixin Powder on Nonenzymatic Glycosylation and Free Radicals in Aging Rat Induced by D-gal

Abstract:Objective To study the effects of Kaixin Powder on nonenzymatic glycosylation and free radicals in aging rat.Methods Mimetic dementia model induced by D-galactose was used,and the ability of learning and memory was measured by water maze task.The contents of AGEs and MDA as well as activities of SOD in brains and livers were determined.Results The results showed that Kaixin Powder could increase the correct percentage in the water maze task.Meanwhile it was found that Kaixin Powder markedly elevated SOD activity,and could reduce AGEs and MDA contents in brain and liver.Conclusion Kaixin Powder may exhibit its anti-aging effects by,at least partially,inhibiting nonenzymatic glycosylation and reducing free radicals.

Abstract:To observe the effect of Shenqiwan on learning and memory functions in senile model mice induced by D-galactose,40 Wistar rats were dividied into control group,model group,treating group 1(low dosage),treating group2(high dosage).The aging models were induced with D-galactose.The memory function was tested with passage way water maze.The activity of superoxide dismutase(SOD),content of malodialdehyde(MDA)and glutathione peroxidase(GSH-PX) were measured.After sixty days treating with Shenqiwan,the swimming time significantly shortened and the correct number increased significantly in Shenqiwan groups compared with the model group.The activities of SOD and GSH-PX increased and the content of MDA decreased in Shenqiwan groups compared with that in the model group.Shenqiwan can improve the learning and memory functions in aging rats induced with D-glalactose and resist lipid peroxidation in brain tissue.

Abstract:Angelica sinensis (AS, Danggui in Chinese) is an important herbal component of various traditional formulae for the management of asthenia and its tonic effects. Although AS has been shown to ameliorate cognitive damage and nerve toxicity in D-galactose (D-gal)-elicited senescent mice brain, its effects on liver and kidney injury have not yet been explored. In this work, mice were subjected to hypodermic injection with D-gal (200 mg/kg) and orally gavaged with AS (20, 40, or 80 mg/kg) once a day for 8 successive weeks. Results revealed that AS significantly improved liver and kidney function as assessed by organ index and functional parameters. In addition, AS pretreatment effectively ameliorated the histological deterioration. AS attenuated the MDA level and markedly enhanced the activities and gene expressions of antioxidative enzymes, namely Cu, Zn-SOD, CAT, and GPx. Furthermore, AS markedly inhibited the D-gal-mediated increment of expressions of inflammatory cytokines iNOS, COX-2, IκBα, p-IκBα, and p65 and promoted the IκBα expression level in both hepatic and renal tissues. In sum, AS pretreatment could effectively guard the liver and kidney of mice from D-gal-induced injury, and the underlying mechanism was deemed to be intimately related to attenuating oxidative response and inflammatory stress.
Neuroprotective effects of the aqueous extract of the Chinese medicine Danggui-Shaoyao-san on aged mice

Abstract:Danggui-Shaoyao-san (DSS), a famous Chinese complex prescription, has been widely used for gynecological and obstetrical purposes for a long time, and recently it has gained much attention due to its therapeutic efficacy for senile dementia. In the present study, the effects of aqueous extract of Danggui-Shaoyao-san on naturally aged mice were examined to investigate the pharmacological basis for its therapeutic efficacy on senile dementia. The results showed that DSS significantly prolonged latency in a step-through test and increased brain index so as to improve impaired cognitive function of aged mice after oral administration at doses of 250 and 500 mg/kg for 3 months. Using high-performance liquid chromatography technique with electrochemical detection (HPLC-ECD), it was found that DSS increased the content of monoamine neurotransmitters such as norepinephrine (NE), dopamine (DA) and 5-hydroxytryptamine (5-HT) in brains of aged mice. Using transmission electron microscope, it was found that DSS dose-dependently protected the ultrastructure of brain cortex in aged mice. These results indicate that DSS ameliorates memory dysfunction, modulates metabolism of monoamine neurotransmitters and protects the ultrastructure of cortex changed by aging. These findings suggest that DSS may be a useful therapeutic agent for senile dementia, especially Alzheimer's disease (AD).

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Codonopsis pilosula (C. pilosula), also called "Dangshen" in Chinese, is derived from the roots of Codonopsis pilosula (Franch.) Nannf. (C. pilosula), Codonopsis pilosula var. Modesta (Nannf.) L.D.Shen (C. pilosula var. modesta) or Codonopsis pilosula subsp. Tangshen (Oliv.) D.Y.Hong (C. pilosula subsp. tangshen), is a well-known traditional Chinese medicine. It has been regularly used for anti-aging, strengthening the spleen and tonifying the lungs, regulating blood sugar, lowering blood pressure, strengthening the body's immune system, etc. However, the mechanism, by which, C. pilosula exerts its therapeutic effects on brain aging remains unclear.AIM OF THE STUDY: This study aimed to investigate the underlying mechanisms of the protective effects of C. pilosula water extract (CPWE) on the hippocampal tissue of D-galactose-induced aging mice.MATERIALS AND METHODS: In this research, plant taxonomy has been confirmed in the "The Plant List" database (www.theplantlist.org). First, an aging mouse model was established through the intraperitoneal injections of D-galactose solution, and low-, medium-, and high-dose CPWE were administered to mice by gavage for 42 days. Then, the learning and memory abilities of the mice were examined using the Morris water maze tests and step-down test. Hematoxylin and eosin staining was performed to visualize histopathological damage in the hippocampus. A transmission electron microscope was used to observe the ultrastructure of hippocampal neurons. Immunohistochemical staining was performed to examine the expression of glial fibrillary acidic protein (GFAP), the marker protein of astrocyte activation, and autophagy-related proteins, including microtubule-associated protein light chain 3 (LC3) and sequestosome 1 (SQSTM1)/p62, in the hippocampal tissues of mice. Moreover, targeted metabolomic analysis was performed to assess the changes in polar metabolites and short-chain fatty acids in the hippocampus.RESULTS: First, CPWE alleviated cognitive impairment and ameliorated hippocampal tissue damage in aging mice. Furthermore, CPWE markedly alleviated mitochondrial damage, restored the number of autophagosomes, and activated autophagy in the hippocampal tissue of aging mice by increasing the expression of LC3 protein and reducing the expression of p62 protein. Meanwhile, the expression levels of the brain injury marker protein GFAP decreased. Moreover, quantitative targeted metabolomic analysis revealed that CPWE intervention reversed the abnormal levels of L-asparagine, L-glutamic acid, L-glutamine, serotonin hydrochloride, succinic acid, and acetic acid in the hippocampal tissue of aging mice. CPWE also significantly regulated pathways associated with D-glutamine and D-glutamate metabolism, nitrogen metabolism, arginine biosynthesis, alanine, aspartate, and glutamate metabolisms, and aminoacyl-tRNA biosynthesis.CONCLUSIONS: CPWE could improve cognitive and pathological conditions induced by D-galactose in aging mice by activating autophagy and regulating metabolism, thereby slowing down brain aging.

Qing'E Formula Alleviates the Aging Process in D-galactose-induced Aging Mice.

Abstract:Qing'E formula (QEF) is a clinically used prescription with four ingredients, Eucommiae Cortex, Psoraleae Fructus, Juglandis Semen and Garlic Rhizoma, from the Song dynasty (10th century CE). The present study aimed to investigate the anti-aging effect and mechanisms of QEF on D-galactose-induced aging mice. A mouse subacute aging model was established by subcutaneous injection of D-galactose at the neck consecutively for 8 weeks. Motor activity and memory impairment of the mice were evaluated by the rotarod test and passive avoidance test, respectively. Serum and liver parameters were analyzed with biochemical kits. Hippocampal mRNA and protein expression levels were examined by reverse transcription-quantitative polymerase chain reaction and western blotting, respectively. QEF administration significantly ameliorated the impaired motor and memory of aging mice. In the serum, QEF reduced blood urea nitrogen, creatinine, nitric oxide (NO) and malondialdehyde (MDA) levels, and inhibited alanine aminotransferase and aspartate aminotransferase activities. In the liver, QEF increased the glutathione level, enhanced total antioxidant capacity and catalase activity, deceased NO and MDA production, and reduced NO synthase activity. In the hippocampus, QEF elevated gene expression levels of Klotho, sirtuin 1 (SIRT1), forkhead box transcription factor O3, peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α), insulin-like growth factor-1 and peroxiredoxin-3. QEF increased protein expression levels of Klotho and SIRT1, and decreased that of PGC-1α in the hippocampus. In conclusion, QEF attenuated the aging process in D-galactose-treated mice, which may be mediated through enhancing the antioxidants in the body, protecting renal and hepatic health, and balancing hippocampal expression levels of the longevity-related genes.
Zuogui Pills Improve the Learning and Memory Ability of Brain-Aging Mice through the Estrogen Receptor ERα Methylation Pathway

Abstract:Introduction: Sex hormones are important factors in maintaining brain function and acting as brain protectors. Recent research suggests that neuronal damage in brain aging may be linked to the methylation of the estrogen receptor α (ERα). However, the mechanism of Zuogui Pills (ZGW) in brain-aging ERα DNA methylation and neuronal repair remains unknown. Materials and methods: D-galactose-induced ovary removal mice were used as a model of aging. Changes in estrous cycle were detected in mice by vaginal cell smear. Animal behavior tests, including the Morris water maze (MWM) and new object recognition (NOR) test, were conducted. Hematoxylin-eosin (HE) and Nissl-staining were carried out to assess hippocampal neurogenesis. Enzyme-linked immunosorbent assay (ELISA) was performed for 5- methylcytosine methylation levels, and immunohistochemistry (IHC) and western blotting (WB) experiments were performed to assess ERα/DNA methyltransferase 1 (DNMT1) expression after ZGW treatment. Finally, bisulfite sequencing PCR (BSP) analysis was performed to identify methylated differentially expressed estrogen receptor 1 (ESR1) gene in D-gal-induced senescent neurons before and after ZGW treatment. Results: We found that ERα methylation was involved in the delayed brain ageing process of ZGW. Mechanistically, ZGW can improve the learning and memory ability of brain-aging mice, reduce the expression of 5-methylcytosine (5-mc) in serum, increase the amount of ERα, inhibit the expression of DNMT1, and significantly reduce methylated expression of the ESRI gene. Conclusion: Our data suggested that ZGW slowed down D-gal-induced brain aging in mice, and these results showed that ZGW is beneficial for aging. It may be used for neuronal protection in aging.
