Diclofenac Loaded Lipid Based Microparticulate Drug Delivery System for Osteoarthritis

Abstract:Lipid based microparticulate drug delivery systems (lipospheres) of drugs exhibit the improved solubility and dissolution which may result in the improvement of the bioavailability. The lipospheres of diclofenac were prepared by complexing diclofenac with dipalmotyl phosphatidylcholine using solvent removal method. The prepared lipospheres were evaluated for particle size, drug content, in vitro dissolution study, in vivo analgesic activity and in vivo anti inflammatory activity. The particle size ranged from 350 +/- 2.90 mu to 2550 +/- 5.60 mu. The lipospheres showed 98.04 +/- 1.50% drug loading. At the end of 12 h the % drug release was found to be 42.58 +/- 1.04% and 91.11 +/- 2.84% for the diclofenac and the lipospheres, respectively. The lipospheres of diclofenac showed excellent analgesic activity with 67.24% analgesia as compared to the standard (diclofenac tablet) which showed only 60.34% analgesia against the control. The prepared lipospheres of diclofenac showed 70.47 +/- 1.24% inhibition of rat paw edema at the end of 24 h as compared to the standard which showed only 61.90 +/- 2.40% inhibition of rat paw edema after 24 h. It was concluded that the lipospheres of diclofenac may be effective in delivering the drug for prolonged period of time with improved dissolution and therapeutic effect in the patients of osteoarthritis.
