Abstract:Structurally complex natural products have been a fruitful source for the discovery and development of new drugs. In an effort to construct a compound collection populated by architecturally complex members with unique scaffolds, we have used the natural product limonin as a starting point. Limonin is an abundant triterpenoid natural product and, through alteration of its heptacyclic core ring system using short synthetic sequences, a collection of 98 compounds was created, including multiple members with novel ring systems. The reactions leveraged in the construction of these compounds include novel ring cleavage, rearrangements, and cyclizations, and this work is highlighted by the discovery of a novel B-ring cleavage reaction, a unique B/C-ring rearrangement, an atypical D-ring cyclization, among others. Computational analysis shows that 52 different scaffolds/ring systems were produced during the course of this work, of which 36 are unprecedented. Phenotypic screening and structure-activity relationships identified compounds with activity against a panel of cancer cell lines.

Abstract:The lamellarins, a group of natural pyrrole alkaloids with interesting anticancer activities, were firstly isolated from a prosobranch mollus Lamellaria sp.. Due to their multidrug resistance (MDR) reversal in some cancer cell lines, total synthesis and structure modifications of lamellarins to provide drug candidates for the treatment of cancer have become one of the most promising project in the area of novel anticancer drug discovery. In this article, the latest results and progresses regarding the synthesis and modification of lamellarins are reviewed.

Abstract:The highly enantioselective preparation of pharmacologically interesting hexahydropyridazine derivatives based on a multicomponent cascade reaction is described. This one-pot approach utilizes an organocatalytic Michael reaction followed by intermolecular α-amination and intramolecular hemiaminalization to yield a chiral pyridazine backbone with contiguous stereogenic centers and multiple functional groups in good yield and with high stereoselectivity. Compounds synthesized by this method potently inhibited proliferation of MCF-7 breast cancer cells. Mechanistic studies suggest that compound 5 c exerts these anticancer effects by inducing apoptosis through extracellular signal related kinase (ERK)- and poly(adenosine diphosphate ribose) polymerase (PARP)-regulated pathways, as well as mitochondrial pathways.

Abstract:Four 18 (13 → 14)-abeo limonoids, five cedrelone limonoids, and five walsurin limonoids were isolated from the fruits of Walsura robusta together with 21 known compounds. Their structures were determined by extensive studies of their one- and two-dimensional NMR spectra and mass spectroscopy results. Among all the isolated compounds, the absolute configurations of 11-oxo-dihydrocedrelone, walsuronoid B, dysoxylumosin G, and walsunoid H were confirmed by single crystal X-ray diffraction. Selected compounds were evaluated for their reversal of multidrug resistance (MDR) phenotype in human breast cancer cells in vitro. Walsurin A showed significant effect in sensitization of these resistant cancer cells at non-toxic concentration to doxorubicin with the modulation factor of 62.76.

Abstract:Limonoids , a class of abundant natural tetracyclic triterpenoids, present diverse biological activity and provide a versatile platform amenable by chemical modifications for clinical use. Among all of the limonoids isolated from natural sources, obacunone, nomilin , and limonin are the primary hub of limonoid-based chemical modification research. To date, more than 800 limonoids analogs have been synthesized, some of which possess promising biological activities. This review not only discusses the synthesis of limonoid derivatives as promising therapeutic candidates and details the pharmacological studies of their underlying mechanisms from 2002 to 2022, but also proposes a preliminary limonoid synthetic structure-activity relationship (SAR) and provides future direction of limonoid derivatization research.

Sesquiterpene dimers with a rare skeleton from Chloranthus holostegius exhibiting multidrug resistance reversal activity in vitro and in vivo

Abstract:Two previously unreported lindenane sesquiterpene dimers (1 and 2) with a rare skeleton containing an oxaspiro[4.5]decane moiety were isolated from the roots of Chloranthus holostegius var. trichoneurus. Their structures were elucidated by HRESIMS, NMR, ECD, and NMR quantum chemical calculations, along with DP4+ probability analysis. In bioassay, compound 1 exhibited significant activity to reverse the multidrug resistance (MDR)in MCF-7/ADR cells, with an IC50 value of 4.4 μM. Further mechanistic studies revealed that compound 1 combined with doxorubicin could induce apoptosis of MCF-7/ADR cells and block the cell cycle in the G2/M phase. Mechanistically, compound 1 could inhibit the efflux function of P-glycoprotein (P-gp) using the zebrafish model. Finally, the enhanced chemotherapeutic effects of doxorubicin were further confirmed by in vivo zebrafish xenograft experiments.

Abstract:Limonoid is derivatives of triterpenoid compound that have a wide variety of structures due to various ring-opening, rearrangements, and high-degree of oxidation. Limonoid is known as compounds that have wide-range of biological activities, including anticancer activity. This research was aimed to determine the chemical structure and cytotoxic activity of limonoid in the n-hexane extract of Chisocheton lasiocarpus fruit. Dried powder of C. lasiocarpus fruit was extracted using methanol followed by fractionated using n-hexane, ethyl acetate, and n-butanol. Five azadirone-type limonoids, 6α-(acetoxy)-14β,15β-epoxyazadirone (1), dysobinin (2), 7α-acetylneotricilenone (3), 6α-O-acetyl-7-deacetylnimocinol (4), and 7α-hyroxyneotricilenone (5), were isolated from the n-hexane extract of Chisocheton lasiocarpus fruit. The chemical structure of all compounds was identified by spectroscopic analysis, including 1D, 2D-NMR, IR, UV and HR-TOF MS as well as by comparison with previous reported spectra data. Compounds 1-5 were investigated from this plant for first time. Abstract:Twelve new limonoids, trichilitins A—L (1—12), together with ten reported congeners (13—22) were isolated from Trichilia connaroides. Compounds 6, 7, 13, 14, and 19 exhibited significant cytotoxic effects, especially 7 exhibited significant cytotoxic effects against HCT‐116 with an IC50 value of 0.035 μmol·L–1 and induced cell cycle arrest by promoting the expression levels of p53 and p21. Comprehensive Summary Twelve new limonoids (1—12), named trichilitins A—L, were isolated from the leaves and twigs of Trichilia connaroides, together with ten known compounds (13—22). The structures were elucidated by extensive spectroscopic investigations, X‐ray diffraction analyses, and ECD calculations. Compound 1, which belongs to a unique class of ring B‐seco limonoid, has been identified as 6/7/6/5 tetracyclic due to a key Baeyer‐Villiger oxidation. Compounds 2—7 were identified as ring intact limonoids, while compounds 8—10 were established as ring D‐seco ones, and 11 and 12 were determined to be rearranged ones. All of the compounds were tested for cytotoxicity against three human tumor cell lines (HCT‐116, NCl‐H1975, and SH‐SY5Y). Compounds 6, 7, 13, 14, and 19 exhibited significant cytotoxic effects, especially 7 exhibited significant cytotoxic effects against HCT‐116 with an IC50 value of 0.035 μmol·L–1 and was more active than the positive control, doxorubicin with an IC50 value of 0.20 μmol·L–1. Compound 7 effectively induced apoptosis of HCT‐116, which was associated with S‐phase cell cycle arrest. Furthermore, the Western blot analysis showed that compound 7 could induce cell cycle arrest by promoting the expression levels of p53 and p21.

Abstract:Phenotypic screening of high quality compound library is an effective strategy to discover novel bioactive molecules. Previously, we developed the divergent organocatalytic cascade approach to efficiently construct a focused library with scaffold diversity and successfully identified a novel spiropyrazolone antitumor scaffold. Herein, a series of spiropyrazolone derivatives were designed, synthesized and assayed. Most of them showed good in vitro antitumor activity with a broad spectrum. Preliminary structure-activity relationship for the substitutions and the stereo configuration were obtained. Compound 5k showed good antitumor activity and could effectively induce cancer cell apoptosis, which represents a good starting point for the development of novel antitumor agents. Limonoids and their anti-proliferative and anti-aromatase properties in human breast cancer cells

Abstract:Lemons are a widely used citrus crop and have shown several potential health benefits. In the present study, the mechanism and effectiveness of the anti-cancer and anti-aromatase properties of limonoids were investigated for the first time. Defatted lemon (Citrus lemon L. Burm) seed powder was extracted with ethyl acetate (EtOAc) and methanol (MeOH) for 16 h each, successively. These extracts were fractionated using 1D (silica) and 2D (ion exchange and SP-70 columns) column chromatography to obtain nine limonoids. The compounds were identified by TLC, HPLC, and LC-MS techniques. A panel of 9 purified limonoids, including limonin, nomilin, obacunone, limonexic acid (LNA), isolimonexic acid (ILNA), nomilinic acid glucoside (NAG), deacetyl nomilinic acid glucoside (DNAG), limonin glucoside (LG) and obacunone glucoside (OG) as well as 4 modified compounds such as limonin methoxime (LM), limonin oxime (LO), defuran limonin (DL), and defuran nomilin (DN), were screened for their cytotoxicity on estrogen receptor (ER)-positive (MCF-7) or ER-negative (MDA-MB-231) human breast cancer cells. We further tested the mechanism of the anti-proliferative activity of limonoids using an in vitro aromatase enzyme assay and western blot with anti-caspase-7. Among the tested limonoids, 11 limonoids exhibited cytotoxicity on MCF-7 whereas 8 limonoids showed cytotoxicity against the MDA-MB-231 cell lines. Although most of the limonoids showed anti-aromatase activity, the inhibition of proliferation was not related to the anti-aromatase activity. On the other hand, the anti-proliferative activity was significantly correlated with caspase-7 activation by limonoids. Our findings indicated that the citrus limonoids may have potential for the prevention of estrogen-responsive breast cancer (MCF-7) via caspase-7 dependent pathways.

Abstract:Inspired by potent antiproliferative xanthone natural products and so far limited examples of derived bioactive agents, a structure activity study of architecturally novel types of xanthones is reported. Their preparation was enabled in a short and divergent manner by a modular chlorination in combination with optimized protocols for a polar condensation and a hetero‐cyclization. Application of these procedures allowed for the synthesis of various polyhalogenated representatives (including mixed bromo/chloro xanthones) that were obtained in up to fourfold improved yields as compared to previous procedures. Subsequent Suzuki coupling of either halide enabled access to phenyl‐ and chloro‐bearing xanthones, which may be functionalized at four out of five non‐hydroxylated positions. Antiproliferative assays against breast cancer cell lines revealed potent activities of some of these simplified analogs that are in the range of pharmaceutically used anticancer drug doxorubicin.

Abstract:Nine dimeric bile acid-amino acid conjugates as the mimic of cycle peptide and they have been synthesized in good yield and their structures were well confirmed by H-1 NMR, C-13 NMR and mass spectra. The ESI-MSn technique was used to probe the conformation of these molecules. The preliminary bioactivity tests showed that some of the title compounds could inhibit the growth of human breast cancer cell MCF-7. The antitumor activity probably derived from the unique sandwich-type amphiphilic conformation.

Abstract:In the present study, a series of natural bioactive (D)-Limonene-based 1,2,3-triazole 14a - e hybrids were synthesized by copper (II)-catalyzed [3 + 2] cycloaddition reaction. Synthetic products are characterized by spectroscopic techniques ( 1 H- and 13 C NMR). Our objective is to evaluate the anticancer activity against human cancer cell lines HT-1080, A-549, MCF-7 and MDA-MB-231. Indeed, we found that the majority of 1,2,3-triazole hybrids 14a - e showed average cytotoxic activity towards all conquering cells tested with IC 50 values between 13.03 ± 0.67 and 41.66 ± 1.98 μM, except in the case of 14d , which represents IC 50 values 13.03 ± 0.67 μM and 15.98 ± 2.03 μM respectively against HT-1080 and A-549 cells.

Abstract:Inspired by the natural product evodiamine, a novel antitumor indolopyrazinoquinazolinone scaffold was designed by scaffold hopping. Structure-activity relationship studies led to the discovery of compound 15j, which shows low nanomolar inhibitory activity against the HCT116 cell line. Further antitumor mechanism studies indicated that compound 15j acted by the dual inhibition of topoisomerase 1 (Top1) and tubulin and induced apoptosis with G2 cell-cycle arrest. The quaternary ammonium salt of compound 15j (compound 15js) exhibited excellent in vivo antitumor activity (TGI = 66.6%) in the HCT116 xenograft model with low toxicity. Indolopyrazinoquinazolinone derivatives represent promising multitargeting antitumor leads for the development of novel antitumor agents.

Abstract:Synthesis, characterization, and theoretical studies of a novel limonene-bis(isoxazole)-thiazolidinone hybrid, prepared from natural D-limonene in five steps, were conducted. The compounds obtained were successfully identified using HRMS, 1 H- and 13 C-NMR spectral data. Subsequently, in-silico docking and molecular dynamic simulation studies were performed to predict potential interaction modes between the compounds and the active sites of the target hTopo IIα. Both studies indicated that all molecules exhibited good binding affinity towards the hTopo IIα enzyme, similar to the reference drug etoposide, when placed in the enzyme's active site. In addition, in silico ADME/Tox studies were carried out to assess the drug-likeness and pharmacokinetic properties of the molecules. The limonene-bis(isoxazole)-thiazolidinone hybrid derivatives may serve as promising lead compounds for the development of new topoisomerase-targeted anticancer agents.

Abstract:A series of novel 1,5-benzodiazepine-2,4-dione derivatives with C-6 amide substituents were designed and synthesized using three-component reactions. The preliminary assays showed that most of them displayed moderate to good antitumor activities against human lung carcinoma (A549), human breast epithelial carcinoma (MCF-7), human colon carcinoma (HCT116), human cervical carcinoma (Hela) and Lewis lung carcinoma (2LL). Exhilaratingly, the activity level of 6m rivaled that of 5-Fluorouracil (5-Fu) against MCF-7 cell lines, which might be used as novel lead scaffold for potential anticancer development.

Abstract:Artemisinin is a potential anticancer agent with an interesting trioxane sesquiterpene structure. In order to improve the biological activity and metabolic stability of artemisinin, a series of novel ring-contracted artemisinin dimers were synthesized. These dimers were evaluated by MTT assay against six cancer cell lines. Most of the dimmers exhibited improved antiproliferative activities over artemisinin. Especially, compound 8b showed the most pronounced anti-cancer activity for PC12 cancer cells with an IC50 value of 1.56 μM. Thus, PC12 cancer cells were used to further investigate the mechanism of antiproliferation for this series of compounds. Compound 8b arrested cell cycle at G1 phase and induced cell apoptosis via up-regulation of Bad, Bax, caspase-3 and caspase-9 protein expressions while inhibiting the expression of Bcl-xL. The present studies are the first to synthesize the ring-contracted artemisinin as dimers and show that these dimers have potent anti-tumor activities against several cancer cell lines.

Abstract:The tri-ring diterpenes with trans-configuration showed the anti-tumor potency from many previous inverstigations, but the research for the tri-ring diterpene with cis-configuration has not been regarded so far. During our chemical studies on the bioactive compounds from the plants, a novel tri-ring diterpene (+)-7-deoxynimbidiol which attracts our intensive interest for its good anti-tumor activity was isolated from Celastrus hypoleucus. cis-(+/-)-7-Deoxynimbidiol(compound 4), the diastereoisomer of (+)-7-deoxynimbidiol, was total synthesized via eight-step reactions with geranic acid as the starting material, and modified by etherification of the phenolic hydroxyl with the substituted benzyl bromide. Eight novel tri-ring diterpenes (compounds 5a-5h) were obtained. The anti-tumor activities of all the new compounds were evaluated. Compound 5d was ound to have potency against the cultured human-tumor cell line MCF7 (IC50 = 7.49 mu g/mL). the fluorine atom on the benzyl group was speculated to enhance the anti-tumor activities.
