Abstract:A novel methylene violet compound (3RAX-NBD) with dual functions of H2S recognition and photodynamic activity was prepared. The 3RAX-NBD has high sensitivity and good selectivity for H2S recognition with a low detection limit (0.135 μM) and a faster reaction rate (k2 = 3.24 M-1S-1). The success of intracellular imaging indicated that 3RAX-NBD is preferentially localized to mitochondria and can be used to further study biological regulatory functions and pathological roles of H2S in living systems. The 3RAX-NBD showed photocytotoxicity against 4T1 cells with its IC50 of 6.40 μΜ under 550 nm laser irradiation, but no toxicity in the darkness. For the treatment of tumor tissues, the average tumor volume in mice ranged from 2399 mm3 to 170 mm3 under losing and laser irradiation. 3RAX-NBD exhibited photodynamic therapeutic efficiency with negligible side effects. Therefore, it suggests that this 3RAX-NBD has potential photodynamic therapy with H2S recognition.
DNA Photocleavage and Binding Modes of Methylene Violet 3RAX and its Derivatives: Effect of Functional Groups

Abstract:With 4'-amino-N,N-diethylaniline and aniline as starting materials, methylene violet 3RAX 1 and its derivatives 2-5 were synthesised. The five compounds were characterised by IR, UV-vis, and H-1 NMR spectroscopy and mass spectrometry. The binding mode between the synthesised compounds and DNA were investigated. The results show that both compounds 1 and 5 bind to DNA by an intercalative mode, while compounds 2-4 interact with DNA through a mixed binding mode involving groove binding and electrostatic interactions. The photocleavage ability of the five compounds to DNA were calculated to be 38, 40, 30, 20, and 13%, respectively, when their concentration was adjusted to 400 mu M. The singlet oxygen production of compounds measured by the 1,3-diphenylisobenzofuran method was consistent with the trend of DNA photocleavage ability. The DNA studies suggest that the binding mode between methylene violet 3RAX and DNA, the ability of methylene violet 3RAX to generate singlet oxygen, and the DNA photocleavage activity could be adjusted through modification of the amino group on methylene violet 3RAX.

Abstract:In recent years, great interest has been focused on the use of photosensitizers (PS) for photodynamic therapy (PDT) as safe and effective anti-tumor drugs. As a good lysosomal-targeted drug, folic acid (FA) is highly interesting as well. [Formula: see text]-methylpyridylporphyrin tailed with folate conjugate (Me-Por-FA) was newly designed and synthesized and the structure was confirmed by UV-vis, IR, 1H NMR, MS and elemental analysis. The interaction of this porphyrin with calf thymus DNA was the intercalative binding mode, which was confirmed by ultraviolet and fluorescence spectra, and the binding constants [Formula: see text] was 6.24 × 104 L/mol. The singlet oxygen (1O[Formula: see text] generated by Me-Por-FA was determined by 1, 3-diphenylisobenzofuran (DPBF) method using tetrapyridylporphyrin (H[Formula: see text]TMPyP) as a comparison with the following order: H2TMPyP > Me-Por-FA. Stained with LysoTracker[Formula: see text] Green DND-26, Me-Por-FA was mainly distributed over the lysosomes during 4 h, but H[Formula: see text]TMPyP was not. This suggests that Me-Por-FA could be developed as a targeted photosensitizer for precise photodynamic therapy.
Novel Naphthalimide Bridged Zinc Porphyrin/bodipy Nanomaterials with D-A Structure for Photodynamic Therapy

Abstract:As a non-invasive cancer therapy method, photodynamic therapy (PDT) shows tremendous promise in clinical cancer treatment. Light-activated singlet oxygen production of photosensitizers (PSs) is the prerequisite for cancer PDT, and the use of organic photosensitizers is always limited by visible light-based activation, hydrophilicity, biocompatibility, selectivity and quantum yield of singlet oxygen. Currently, both zinc porphyrin- and BODIPY-based structures have been widely used in the development of PDT PSs. Here, we developed a novel naphthalimide bridged zinc porphyrin/BODIPY molecule (Por-BDP-1) with two poly(ethylene glycol) (PEG) chains, in which D-A structure was constructed between the naphthalimide group and porphyrin group. After self-assembly into nanoparticles, Por-BDP-1 NPs (Diameter: 122.4 nm) could quench fluorescence in 600-700 nm, bind with calf thymus-DNA, and produce singlet oxygen during light-irradiation (laser: 680 nm, 1.0 W/cm2). In addition, Por-BDP-1 NPs effectively killed HeLa cells with a IC50 value = 44.8 mu g/mL and showed a lower dark toxicity under the same conditions. All our results demonstrated that our naphthalimide bridged zinc porphyrin/BODIPY nano-photosensitizer is a promising nanoagent for PDT in the clinic.

Abstract:A series of (4-cyanophenyl)porphyrins have been prepared and structurally characterized by H-1 NMR, IR and UV/Vis spectroscopy, mass spectrometry, and elemental analysis. Abstract:Photodynamic therapy (PDT) is a promising cancer treatment approach with the advantages of low toxicity and noninvasive characteristics. In this study, a series of metalloporphyrin–indomethacin conjugates tethered with poly(ethylene glycol) (PEG) chains were prepared and characterized. The singlet oxygen production of the conjugates was evaluated through 2′, 7′-dichlorofluorescin (DCFH) method. Because of the heavy atom effect, the metal porphyrin complexes exhibited the higher singlet oxygen ( 1 O 2 ) quantum yield than that of free base porphyrin. The order of 1 O 2 yield of the synthesized porphyrins was PtPor > PdPor > ZnPor > Por. The MTT assay using HeLa cells verified the low cytotoxicity of porphyrin–indomethacin conjugates in the dark. Upon irradiation, the platinated porphyrin (PtPor) showed the highest therapeutic activity among these conjugates, probably due to its high efficiency of 1 O 2 generation. The cellular uptake and subcellular localization of the conjugates were further evaluated through a confocal laser scanning microscope. The results showed that the conjugates were primarily localized in the lysosomes of HeLa cells.

Abstract:Lysosome, an organelle which contains a number of hydrolases and hydrogen ions, plays a crucial role in cellular survival and apoptosis. If selectively destroy lysosomes membrane, inner hydrolases and hydrogen ions will leak and induce cell death. In this work, three lysosome-targeting fluorescent probes (HCL 1-3, heptamethine cyanine lysosomal-targeting probe) were designed, synthesized and developed for photodynamic therapy. Piperazine and N, N-dimethyl structures made HCL 1-3 have good lysosome targeting ability while Pearson's correlation co-efficients reached 0.85, 0.87 and 0.78. It can be concluded from MTT test, HCL 1-3 have high photo cytotoxicity and low dark cytotoxicity from MTT test. Calcein/PI staining assays also supported cytotoxicity of HCL 1-3 under light conditions. In vivo experiments, HCL 2 accumulated in tumor and a strong fluorescence signal was observed at 12 h post injection. All results showed that our experiments provide help and new ideas for cyanine dyes in cancer treatment.

Abstract:Through a new synthetic route, three ruthenium-phenanthroline porphyrins (RPP1, RPP2 and RPP3) were prepared. Their photophysical and photochemical properties, such as DNA photocleavage activity, singlet-oxygen photogeneration and two-photon absorption (2PA) were evaluated. These porphyrins 1-3 had substantial photocleavage activities, with 71%, 74% and 38% observed at 20 mu M. The porphyrins with different substituents on phenathroline group had similar singlet oxygen quantum yields, with Phi(Delta) values at 0.52, 0.47 and 0.41, respectively. The 2PA cross-section (sigma((2))) values of RPP 1-3, measured by the Z-scan method, were calculated to be 152, 172 and 135 GM, respectively, which were around 5-fold higher than that of H2TPP. Thus, these porphyrins, with their good photocleavage activities, O-1(2) quantum yields and high 2PA cross section, suggest great potential as photodynamic therapeutic agents.
Synthesis and evaluation of 5,15-diaryltetrabenzoporphyrins as photosensitizers for photo-diagnosis and photodynamic activity of tumors

Abstract:Photodynamic therapy (PDT) is a well-established treatment modality, typically conducted with single-wavelength irradiation, which may not always be optimal for varying tumor locations and sizes. To address this, photosensitizers with absorption wavelengths ranging from 550 to 760 nm are being explored. Herein, a series of 5,15-diaryltetrabenzoporphyrins (Ar2TBPs) were synthesized. All compounds displayed obvious absorption at 550-700 nm (especially at ∼668 nm), intense fluorescence, efficient generation of singlet oxygen and good photodynamic antitumor effects. Notably, compound I3 (5,15-bis[(4-carboxymethoxy)phenyl]tetrabenzoporphyrin) showed excellent cytotoxicity against Eca-109 cell line upon red light irradiation, with an IC50 value of 0.45 μM, and phototherapeutic index of 25.8. Flow cytometry revealed that I3 could induce distinct cell apoptosis. In vivo studies revealed that compound I3 selectively accumulated at tumor site and exhibited outstanding PDT effect with antitumor activity under single-time administration and light irradiation, and revealed more efficiency than the clinical photosensitizer Verteporfin. These findings underscore the considerable promise of I3 as a robust theranostic agent, offering capabilities in real-time fluorescence imaging and serving as a potent photosensitizer for personalized and precise photodynamic therapy of tumors.

Abstract:In this study, we use cationic imidazole as the targeting group and phenoxazine as the electron donor, attaching phenoxazine to porphyrin via two different linking methods to synthesize two different phenoxazine-porphyrin photosensitizers (PSs), P1 and P2 , intended for photodynamic therapy (PDT) applications. Additionally, we synthesized a molecule, P3 , that does not contain the cationic imidazole group, to serve as a control PS. We comprehensively evaluate the performance of P1 and P2 through steady-state and transient spectroscopy analysis, theoretical calculations, photooxidation experiments, and in vitro cell experiments. The results showed that P1 and P2 exhibit excellent water solubility, longer triplet state lifetimes ( P1 at 200 μs, P2 at 170 μs), outstanding photostability, higher photooxidation rates and quantum yields of singlet oxygen ( 1 O 2 ). Notably, in the photooxidation experiment, P1 's photooxidation rate is 1.1 times that of P2 , and 1.5 times that of tetraphenyl zinc porphyrin (ZnTPP) without phenothiazine. This may be due to the larger Gibbs free energy difference between the singlet and triplet excited states in P1 (Δ G S-T of P1 is 0.87 eV, compared to 0.83 eV for P2 ), which results in a greater driving force for intersystem crossing (ISC) in P1 than in P2 . The introduction of the cationic imidazole group in P1 and P2 significantly enhances water solubility and tumor cell uptake compared to P3 , particularly showing higher intracellular accumulation and 1 O 2 generation capabilities in HepG2 tumor cells. Flow cytometry test results further confirmed the significant photocytotoxicity of P1 and P2 , with a marked decrease in survival rates after light exposure, where the survival rate of P1 nanoparticles (NPs) dropped to 41.7%, P2 NPs to 35.1%, and P3 NPs to 75.0%. The findings of this study provide crucial theoretical support for the design of new PSs.

Abstract:Porphyrin derivatives serve as photocatalysts in reversible-deactivation radical polymerization and as photosensitizers in photodynamic therapy (PDT). Herein, a triple function porphyrin, ZnTPPC6Br, was synthesized as photocatalyst and initiator for photoATRP. Oxygen-tolerant photoATRP produced fructose-based star-shaped glycopolymers as targeted photosensitizers for PDT. ZnTPPC6Br/CuII/PMDETA could synthesize polymer photosensitizers with predictable Mn and low Ð. Mechanistic studies unveiled the transition of ZnTPPC6Br from a singlet excited state (1PC*) to a triplet excited state (3PC*), enabling the activator CuI/L generation and initiates photoATRP. The excess ligands facilitate the active species return to the ground state, while the presence of DMSO assisting in oxygen depletion. Three fructose-based monomers with different polymerizable groups (acrylated, methacrylated, and p-vinylbenzoated) were employed to scale up polymerization, yielding glycopolymeric photosensitizers post-deprotection. In vitro cellular studies showed enhanced PDT efficacy of glycopolymeric photosensitizers against MCF-7 cells, attributed to specific GLUT5 binding for targeted endocytosis, highlighting their potential for precise cancer treatment compared to L929 cells. The multifunctional capabilities of ZnTPPC6Br are anticipated to serve as a strategic avenue for the advancement of polymer photosensitizers with potential PDT applications.

Abstract:In the pursuit of new potent photosensitizers (PSs) for photodynamic therapy (PDT) with better efficacy, a series of 5,15-diaryltetranaphtho (Otvagin et al., 2022; Pham et al., 2021) [2,3]porphyrins (Ar 2 TNPs) with two or four carboxyalkoxy groups were designed, synthesized, and evaluated. These new compounds exhibited strong, broad and red-shifted UV–vis absorptions at 729 nm and other strong absorptions at 446, 475, 650, 659, 714 nm for tumors and other diseases of varying sizes and depths. They possess high molar extinction coefficients (0.95 × 10 5 –1.48 × 10 5 M −1 cm −1 ), good singlet oxygen quantum yields and photodynamic antitumor effects towards Eca-109 cells in vitro . It is suggested that the extension of porphyrin with naphthalene into Ar 2 TNP results into remarkable improvement of photophysical characteristics, while the introduction of carboxyalkoxy groups on meso -phenyl can significantly improve the solubility and photodynamic effects in vitro and in vivo . Notably, compound II 3 can localize primarily in lysosomes of Eca-109 cells and induce substantial cell apoptosis after PDT. It can also selectively accumulate in tumor tissues and be traced real-timely through in vivo fluorescence imaging with distinctive inhibition of tumor growth. Therefore, compound II 3 deserves to be considered as a promising PDT drug candidate for individualized tumor real-time tracing and treatment.

Abstract:Abstract Carbon based polymer dots have piqued attention of researchers because of excellent biocompatibility, and good solubility. Most of the p-dots are able to generate ROS which is effective for photodynamic therapy for the treatment of cancer while some photosensitizers such as porphyrins possess some drawbacks such as hydrophobicity, and dark toxicity. Therefore in this study we conjugated red emission carbon based polymer with pyropheophorbide-α through amide condensation and π–π stacking. One pot synthesis of the conjugate was successfully achieved. Their photophysiological properties were studied and structures were characterized by FT-IR, TEM and 1 HNMR. pH- sensitivity of the conjugates was confirmed using fluorescence and UV–vis spectroscopy. Photo toxicity and dark toxicity of the prepared conjugates were evaluated in human esophageal cancer cell line (Eca-109). Hemocompatibility of the synthesized conjugates was evaluated and proved that the conjugates are safe to use for the treatment of tumor. Our results showed the PS doped p-dots had less dark toxicity and increased light toxicity as well as ROS generation was high as compared to precursor drug. Therefore, incorporation of p-dots to porphyrin improved biocompatibility and enhanced the photodynamic effect.

Abstract:A photosensitizer with high phototoxicity, suitable amphipathy and low dark toxicity could play a pivotal role in photodynamic therapy (PDT). In this study, a facile and versatile approach was adopted to synthesize a series of novel fluorinated hematoporphyrin ether derivatives (I1-I5 and II1-II4), and the photodynamic activities of these compounds were studied. Compared to hematoporphyrin monomethyl ether (HMME), all PSs showed preferable photodynamic activity against A549 lung tumor cells. The longest visible absorption wavelength of these compounds was approximately 622 nm. Among them, II3 revealed the highest singlet oxygen yield (0.0957 min-1), the strongest phototoxicity (IC50=1.24 μM), the lowest dark toxicity in vitro, and exhibited excellent anti-tumor effects in vivo. So compound II3 could act as new drug candidate for photodynamic therapy.

Abstract:Herein, two mitochondria-targeting photosensitizers (PSs, CCVJ-Mito-1 and CCVJ-Mito-2) that exhibit turn-on fluorescence response towards increasing viscosity were reported. Notably, CCVJ-Mito-2 exhibits absorption in the near-infrared (NIR) region, which can be employed as a NIR PS targeting mitochondria and a fluorescent probe for tracking of mitochondrial viscosity changes during photodynamic therapy (PDT). This dual functional PS can help to shed light on the dynamic changes of cellular microenvironment during PDT and further guide the PDT process.

Herein, a novel meso phosphate-substituted pyronin PY-P and its nanoparticles (PY-P NPs) were prepared as an efficient type I organic photosensitizer. The in vivo data prove that PY-P NPs have outstanding low dark toxicity but high photocytotoxicity under hypoxia (<1% O2).

Abstract:Porphyrin and its derivatives have become the second-generation photosensitizers for photodynamic therapy (PDT). Three water-soluble porphyrins containing halogen atoms (F, Cl, Br) were synthesized by N-pyridyl substituted alkylation reaction. We found that P3 has the highest singlet oxygen generation capacity and the results of density functional theory (DFT) further support this result. MTT assay was used to study cytotoxicity, which proved that P1-P3 have good phototoxicity and high selectivity. Fluorescence imaging and intracellular reactive oxygen species illustrate cell apoptosis and reactive oxygen species (ROS) production. High ROS production, excellent phototoxicity ensure that photosensitizers become potential candidates for PDT drugs.
Tailored Engineering of Novel Xanthonium Polymethine Dyes for Synergetic PDT and PTT Triggered by 1064 nm Laser toward Deep‐Seated Tumors

Abstract:Small molecular dye that simultaneously exerts dual PDT/PTT effects as well as florescence imaging triggered by a single NIR‐II light has never been reported to date. Apart from the huge challenge in pushing absorption profile into NIR‐II region, fine‐tuning dyes' excited state via rational structure design to meet all three functions, especially oxygen photosensitization, remains the most prominent throttle. Herein, five novel NIR‐II dyes (BHs) are productively developed by strategically conjugating dyad innovative xanthonium with sequentially extended polymethine bridges, enabling intense absorption from 890 to 1206 nm, significantly 400 nm longer than conventional cyanine dyes with same polymethines. More importantly, owning to high resonance and favorable excited state energy population induced by greater rigidity via ring‐fused amino, BH 1024 exhibits best singlet oxygen generation capability, moderate photothermal heating, and considerable fluorescence under 1064 nm laser irradiation. Furthermore, BH 1024 is encapsulated into folate‐functionalized polymer, which demonstrated a synergetic PDT/PTT effect in vitro and in vivo, eventually achieving solid tumors elimination under NIR‐II fluorescence guide. As far as it is known, this is the first time small molecular dyes for NIR‐II PDT or NIR‐II PDT/PTT are explored and designed.
