Abstract:The presence of extracellular polymeric substance (EPS) plays a vital role in the accumulation and toxicity of nanoparticles to microorganisms, in which the involved processes and mechanisms are still waiting to be revealed. Herein, we specifically investigated the interfacial interaction between titanium dioxide nanoparticles (nTiO2) and algae (Chlorella pyrenoidosa) with/without EPS and the effect of EPS on algal cell internalization of nTiO2. Results showed that the presence of EPS on cell surface promoted heteroaggregation between nTiO2 and algal cells, and induced more nTiO2 accumulation on algal surface; however, algal cell internalization of nTiO2 was limited by the presence of EPS. Pearson correlation analysis further proved that the presence of EPS had a positive effect on the surface accumulation of nTiO2 and a negative effect on the internalization of nTiO2. More than 60% of cell internalized nTiO2 entered algal cells through the energy dependent endocytosis pathway. It is interesting to find that anatase nTiO2 (nTiO2-A) entered algal cells mainly through the clathrin dependent endocytosis, while rutile nTiO2 (nTiO2-R) mainly through the dynamin dependent endocytosis. This difference could be due to the different affinities of nTiO2-A and nTiO2-R to the mediating receptors referring to different endocytic pathways. The removal of EPS activated the associated mediating pathways, allowing more nTiO2 to be internalized. These findings address the role of EPS on the interaction between nTiO2 and algae and promote a deeper understanding of the ecological effect of nTiO2.
Insights into the Regulation Mechanisms of Algal Extracellular Polymeric Substances Secretion Upon the Exposures to Anatase and Rutile TiO2 Nanoparticles

Abstract:As an important part of extracellular secondary metabolites, extracellular polymeric substances (EPS) can play a significant role in protecting cells from the threat of exogenous substances, including nanoparticles (NPs). However, the regulation mechanisms of EPS secretion under NPs exposure remain largely unknown. This study investigated the signaling pathways and molecular responses related to EPS secretion of algae (Chlorella pyrenoidosa) upon the exposures to anatase and rutile TiO2 NPs (nTiO2-A and nTiO2-R, respectively) at two similar toxic (20% and 50% of algal growth inhibition) concentrations. The results showed that EPS responded to nTiO2 stress via excess secretion and compositional variation, and nTiO2-A induced more EPS secretion than nTiO2-R at similar toxicity concentrations. The up-regulation of the Ca2+ signaling pathway might play a greater role in promoting EPS secretion under nTiO2-R exposure compared with nTiO2-A exposure, while the significantly increased intracellular ROS could mainly account for the increased EPS secretion under nTiO2-A exposure. The up-regulated genes related to biological synthesis and protein metabolism and the enhanced biosynthetic metabolism might be the direct causes of the increased EPS secretion. The increased ROS could have a greater effect on the amino acid metabolism and related genes upon the exposure to nTiO2-A than nTiO2-R to induce more EPS secretion. More serious membrane damage caused by nTiO2-R than nTiO2-A would affect the intracellular inositol phospholipid metabolism more severely, while the inositol phospholipid pathway and Ca2+ signaling pathway might agree and communicate with each other inherently to regulate EPS secretion upon nTiO2-R exposure. The findings address the regulation mechanisms of algal EPS secretion under nTiO2 exposure and provide new insights into algal bio-responses to nTiO2 exposure.
The Role of Exopolymeric Substances in the Bioaccumulation and Toxicity of Ag Nanoparticles to Algae.

Abstract:Exopolymeric substances (EPS) have an important role in bioaccumulation and toxicity of nanoparticles (NPs) to algae, which warrants specific studies. The interaction of EPS with citrate and polyvinyl pyrrolidone (PVP) coated AgNPs (C-AgNPs and P-AgNPs, respectively) and its roles in bioaccumulation and toxicity of the AgNPs to Chlorella pyrenoidosa were investigated. The amino and aromatic carboxylic groups in the EPS were involved in the EPS-AgNP interactions. Compared with Ag(+), C-AgNPs had comparable total bioaccumulation but greater absorption by intact algae with EPS; P-AgNPs had the smallest total bioaccumulation and were mainly adsorbed on algal surfaces. With EPS removed, the total bioaccumulations and surface adsorptions for the three Ag species decreased but the cell internalizations increased; the 96 h half growth inhibition concentrations decreased, indicating EPS alleviated the algal toxicity of Ag. The cell-internalized but not the adsorbed AgNPs could contribute to the nanotoxicity. The EPS could bind both AgNPs and Ag(+), and thus inhibited the cell internalization and the nanotoxicity. However, the EPS-bound Ag on the cell surfaces would migrate along with the algae and be biologically amplified in the aquatic food chains, presenting ecological risks. These results are helpful for understanding the fate and ecological effects of NPs.
Heteroagglomeration of Oxide Nanoparticles with Algal Cells: Effects of Particle Type, Ionic Strength and Ph

Abstract:Discharged oxide nanoparticles (NPs) have been shown toxic to unicellular algae, yet the research on heteroagglomeration between NPs and cells as an important precondition of the toxicity is scarce. This study for the first time investigated heteroagglomerations between various NPs and algal cells (Chlorella pyrenoidosa) and analyzed influencing factors including pH and ionic strength (IS) through cosettling experiment, transmission electron microscopic (TEM) observation, and Derjaguin–Landau–Verwey–Overbeek (DLVO) calculation. The examined NPs included anatase and rutile TiO2, microporous and spherical SiO2, and α-form and β-form Al2O3. The results of cosettling experiments coincided well with the TEM observations, whereas the DLVO theory could only partly explain the NP–cell heteroagglomerations. The NP–cell heteroagglomeration for rutile TiO2 and β-form Al2O3 was weak and insensitive to pH or IS. Preferential heteroagglomeration occurred at low pH or high IS for microporous SiO2, while marked heteroagglomeration only occurred under the neutral and low IS condition for anatase TiO2. The heteroagglomeration for spherical SiO2 was insensitive to pH but increased with increasing IS, while the heteroagglomeration for α-form Al2O3 occurred at low pH and irrelevant to IS. The work will shed new light on the bionano interfacial interaction and help to understand biological effects of NPs.
Distinct Toxic Interactions of TiO2 Nanoparticles with Four Coexisting Organochlorine Contaminants on Algae

Abstract:Engineered nanoparticles are increasingly discharged into the environment. After discharge, these nanoparticles can interact with co-existing organic contaminants, resulting in a phenomena referred to as 'joint toxicity'. This study evaluated joint toxicities of TiO2 nanoparticles (TiO2NPs) with four different (atrazine, hexachlorobenzene, pentachlorobenzene, and 3,3',4,4'-tetrachlorobiphenyl) organochlorine contaminants (OCs) toward algae (Chlorella pyrenoidosa). The potential mechanisms underlying the joint toxicity were discussed, including TiO2NPs-OC interactions, effects of TiO2NPs and OCs on biophysicochemical properties of algae and effects of TiO2NPs and OCs on each other's bioaccumulation in algae. The results indicate that coexposure led to a synergistic effect on the joint toxicity for TiO2NPs-atrazine, antagonistic effect for TiO2NPs-hexachlorobenzene and TiO2NPs-3,3',4,4'-tetrachlorobiphenyl, and an additive effect for TiO2NPs-pentachlorobenzene. There was nearly no adsorption of OCs by TiO2NPs, and the physicochemical properties of TiO2NPs were largely unaltered by the presence of OCs. However, both OCs and NPs affected the biophysicochemical properties of algal cells and thereby influenced the cell surface binding and/or internalization. TiO2NPs significantly increased the bioaccumulation of each OC. However, with the exception of atrazine, the bioaccumulation of TiO2NPs decreased when used with each OC. The distinct joint toxicity outcomes were a result of the balance between the increased toxicities of OCs (increased bioaccumulations) and the altered toxicity of TiO2NPs (bioaccumulation can either increase or decrease). These results can significantly improve our understanding of the potential environmental risks associated with NPs.
Physicochemical Transformation and Algal Toxicity of Engineered Nanoparticles in Surface Water Samples.

Abstract:Most studies on the behavior and toxicity of engineered nanoparticles (NPs) have been conducted in artificial water with well-controlled conditions, which are dramatically different from natural waters with complex compositions. To better understand the fate and toxicity of NPs in the natural water environment, physicochemical transformations of four NPs (TiO2, ZnO, Ag, and carbon nanotubes (CNTs)) and their toxicities towards a unicellular green alga (Chlorella pyrenoidosa) in four fresh water and one seawater sample were investigated. Results indicated that water chemistry had profound effects on aggregation, dissolution, and algal toxicity of the NPs. The strongest homoaggregation of the NPs was associated with the highest ionic strength, but no obvious correlation was observed between the homoaggregation of NPs and pH or dissolved organic matter content of the water samples. The greatest dissolution of ZnO NPs also occurred in seawater with the highest ionic strength, while the dissolution of Ag NPs varied differently from ZnO NPs. The released Zn(2+) and especially Ag(+) mainly accounted for the algal toxicity of ZnO and Ag NPs, respectively. The NP-cell heteroagglomeration occurred generally for CNTs and Ag NPs, which contributed to the observed nanotoxicity. However, there was no significant correlation between the observed nanotoxicity and the type of NP or the water chemistry. It was thus concluded that the physicochemical transformations and algal toxicities of NPs in the natural water samples were caused by the combined effects of complex water quality parameters rather than any single influencing factor alone. These results will increase our knowledge on the fate and effects of NPs in the aquatic environment.

Abstract:In order to assess the overall risk posed by engineered nanoparticles (ENPs), the biological effects of this emergent pollutant to aquatic ecosystems must be evaluated. We present findings from studies conducted with a diversity of ENPs (metallic, quantum dots) on a variety of freshwater and marine algae (phytoplankton) illustrating both their direct and indirect effects. We show that in general, while the surface properties of ENPs govern their aggregation behavior and ionic strength controls their dissolution, exopolymeric substances (EPS) produced by algae determine their potential to be toxic and thereby movement through the water column and food web. The production of EPS reduces the impact of ENPs (bioavailability and toxicity) and/or their ions on cellular activities of algae. It does not however directly reduce the aggregation and/or solubility of ENPs but rather affects their stability. Complicating understanding of these interactions is the great assortment of surface coatings for ENPs. This perspective is intended to highlight our current knowledge and the need for future research particularly focused on determining the fate and transport of ENPs in the aquatic environment.
Interactions of Metal Oxide Nanoparticles with Extracellular Polymeric Substances (EPS) of Algal Aggregates in an Eutrophic Ecosystem

Abstract:Nanoparticle binding with organic ligands in natural waters is important to understand their environmental behavior and fate. In this study, the interaction of two metal oxide nanoparticles (nano-CuO and nano-Fe3O4) with extracellular polymeric substances (EPS) from algal aggregates was studied through using fluorescence emission-excitation matrix (EEM) quenching titration combined with parallel factor (PARAFAC) analysis. Spectroscopy analysis detected two protein-like peaks in the loosely bound EPS (LB-EPS) fraction, while both protein-, fulvic- and humic-like peaks can be found in the tightly bound EPS (TB-EPS) fraction. PARAFAC analysis identified three independent fluorescence components, including one tryptophan-, one tyrosine- and one humic-like component, from 68 original EEM spectra. A quenching experiment showed that humic-like components in the TB-EPS fraction exhibited higher nanoparticle binding capacities (logK(M)>6.23) than that in the LB-EPS fraction (logK(M):<5.72), while tryptophan-like components were characterized with a high nano-CuO affinity in the LB-EPS fraction (logK(M): 5.66). Further analysis revealed that the LB-EPS fraction was responsible for the transportation of nano-CuO (logK(M) > 5.66), while the mobility of nano-Fe3O4 would be influenced by organic ligands in the TB-EPS fraction (logK(M) > 6.28). Based on the results, the toxicity of nanoparticles in the eutrophic waters would be higher than those in the oligotrophic ecosystems. Combined Toxicity of Silver Nanoparticles with Hematite or Plastic Nanoparticles Toward Two Freshwater Algae.

Abstract:In the natural environment, the interactions of different types of nanoparticles (NPs) may alter their toxicity, thus masking their true environmental effects. This study investigated the toxicity of silver NPs (AgNPs) combined with hematite (HemNPs) or polystyrene (PsNPs) NPs toward the freshwater algae Chlamydomonas reinhardtii and Ochromonas danica. The former has a cell wall and cannot internalize these NPs, while the latter without a cell wall can. Therefore, the toxicity of AgNPs toward C. reinhardtii was attributed to the released Ag ions, while AgNPs had direct toxic effects on O. danica. Moreover, nontoxic HemNPs ameliorated AgNP toxicity toward C. reinhardtii, by decreasing the bioavailability of Ag ions through adsorption. Despite their role as Ag-ion carriers, HemNPs still reduced the toxicity of AgNPs toward O. danica by competitively inhibiting AgNP uptake. In both algae, Ag accumulation fully accounted for the combined toxicity of AgNPs and HemNPs. However, the combined toxicity of AgNPs and PsNPs was complicated by their significant individual toxicities and the synergistic interactions of these particles with the algae, regardless of differences in Ag accumulation. Overall, in environmental assessments, considerations of the combined toxicity of dissimilar NPs will allow more accurate assessments of their environmental risks.
Surfactants Present Complex Joint Effects on the Toxicities of Metal Oxide Nanoparticles.

Abstract:The potential toxicities of nanoparticles (NPs) have been intensively discussed over the past decade. In addition to their single toxicities, NPs can interact with other environmental chemicals and thereby exert joint effects on biological systems and the environment. The present study investigated the combined toxicities of NPs and surfactants, which are among the chemicals that most likely coexist with NPs. Photobacterium phosphoreum was employed as the model organism. The results indicate that surfactants with different ion types can alter the properties of NPs (i.e., particle size and surface charge) in different ways and present complex joint effects on NP toxicities. Mixtures of different NPs and surfactants exhibited antagonistic, synergistic, and additive effects. In particular, the toxicity of ZnO was observed to result from its dissolved Zn(2+); thus, the joint effects of the ZnO NPs and surfactants can be explained by the interactions between the Zn ions and the surfactants. Our study suggests that the potential hazards caused by mixtures of NPs and surfactants are different from those caused by single NPs. Because surfactants are extensively used in the field of nanotechnology and are likely to coexist with NPs in natural waters, the ecological risk assessments of NPs should consider the impacts of surfactants.
Unraveling the risks of nAl2O3 on harmful algal blooms: Insights from paralytic shellfish toxins production of Alexandrium tamarense

Abstract:As one of the commonly used and cost-effective nanomaterials, nanosized aluminum oxide (nAl2O3) posses unique properties and chemical stability. However, its extensive use and resultant dissemination into aquatic ecosystems prompt concerns over the proliferation and repercussions of harmful algal blooms, particularly those caused by dinoflagellates producing toxins. This study investigated the sub-chronic effects of nAl2O3 on growth, physiological activities, and paralytic shellfish toxins (PSTs) production in Alexandrium tamarense. Results showed dose-dependent inhibition in growth (EC50 = 20.6 mg L-1), esterase activity, and photosynthetic efficiency (Fv/Fm) during the sub-chronic exposure (13-day). The internalization of nAl2O3 in microalgal cells and the significant decrease in the total cellular PSTs content were observed under high nAl2O3 concentrations (>40 mg L-1). The study also demonstrated a clear decrease in the content of some derivatives of PSTs (GTX5, C1/2, and GTX2/3) with the increase in nAl2O3 concentrations, accompanied by the induction of an unknown derivative. Excessive ROS production, dissolved Al, and physical inhibition were suggested as potential mechanisms for nAl2O3 toxicity and changes in PSTs toxin profile. Overall, this research enhances our understanding of the potentiated risks and threats on the possible concurrent events of toxic dinoflagellate, such as Alexandrium species and nanoparticles in aquatic environments.

Abstract:With the rapid development and versatile applications of nanotechnology, a large quantity of nanoparticles (NPs) has been released into the environment. It has been one of the most popular studies that concerning the toxic effect of NPs to the algae in aquatic ecosystem. The membrane integrity of algal cells was assessed using different methods when algae were treated by CuO NPs in the absence and presence of Suwannee river fulvic acid (SRFA), one of DOM. We found SRFA could increase the toxicity of CuO NPs to cell membrane due to the more stable NPs suspension in the presence of SRFA. Results additionally indicate that maybe oxidative damage caused by CuO NPs was the primary reason that led to membrane structure and function failed.
The Influence of Dissolved and Surface-Bound Humic Acid on the Toxicity of TiO2 Nanoparticles to Chlorella Sp.

Abstract:NOM is likely to coat TiO2 nanoparticles (nano-TiO2) discharged into the aquatic environment and influence the nanotoxicity to aquatic organisms, which however has not been well investigated. This study explored the influence of nanoparticle surface-bound humic acid (HA, as a model NOM) as well as dissolved HA on the toxicity of nano-TiO2 to Chlorella sp., with a specific focus on adhesion of the nanoparticles to the algae. Results showed that nano-TiO2 and the dissolved HA could inhibit the algal growth with an IC50 of 4.9 and 8.4 mg L-1, respectively, while both dissolved and nanoparticle surface-bound HA could significantly alleviate the algal toxicity of nano-TiO2. IC50 of nano-TiO2 increased to 18 mg L-1 in the presence of 5 mg L-1 of the dissolved HA and to 48 mg L-1 as the result of surface-saturation by HA. Co-precipitation experiment and transmission electron microscopy observation revealed that both dissolved and nanoparticle surface-bound HA prevented the adhesion of nano-TiO2 to the algal cells due to the increased electrosteric repulsion. The generation of intracellular reactive oxygen species (ROS) was significantly limited by the dissolved and nanoparticle surface-bound HA. The prevention of adhesion and inhibition of ROS generation could account for the HA-mitigated nanotoxicity. (C) 2012 Elsevier Ltd. All rights reserved.
Interactions of CuO nanoparticles with the algae Chlorella pyrenoidosa: adhesion, uptake, and toxicity

Abstract:The potential adverse effects of CuO nanoparticles (NPs) have increasingly attracted attention. Combining electron microscopic and toxicological investigations, we determined the adhesion, uptake, and toxicity of CuO NPs to eukaryotic alga Chlorella pyrenoidosa. CuO NPs were toxic to C. pyrenoidosa, with a 72 h EC50 of 45.7 mg/L. Scanning electron microscopy showed that CuO NPs were attached onto the surface of the algal cells and interacted with extracellular polymeric substances (EPS) excreted by the organisms. Transmission electron microscopy (TEM) showed that EPS layer of algae was thickened by nearly 4-fold after CuO NPs exposure, suggesting a possible protective mechanism. In spite of the thickening of EPS layer, CuO NPs were still internalized by endocytosis and were stored in algal vacuoles. TEM and electron diffraction analysis confirmed that the internalized CuO NPs were transformed to Cu2O NPs (d-spacing, ∼0.213 nm) with an average size approximately 5 nm. The toxicity investigation demonstrated that severe membrane damage was observed after attachment of CuO NPs with algae. Reactive oxygen species generation and mitochondrial depolarization were also noted upon exposure to CuO NPs. This work provides useful information on understanding the role of NPs-algae physical interactions in nanotoxicity.
Toxic Effects of Different Coating-Related Functionalized Nanoparticles on Aquatic Organisms

Abstract:The peculiar physico-chemical characteristics of nanomaterials (NMs) and the use of different coatings to improve their expected properties result in a huge amount of nanoforms, which vary in chemical composition, size, shape and surface characteristics. This makes it almost impossible to test all the nanoforms available, and efforts have been made to establish grouping or read-across strategies. The aim of this work was to find a behavior pattern of effect among nanoforms of different metallic core nanoparticles (NPs) (TiO2, CeO2 and Ag NP) with the same coatings (sodium citrate, poly (ethylene glycol), dodecylphosphonic acid or oleylamine). Daphnia magna, rainbow trout and two fish cell lines (PLHC-1 and RTH-149) were exposed to a range of concentrations (up to 100 mg/L) of the uncoated or coated NPs. Ag NPs were the most toxic, followed by CeO2 NPs and finally by TiO2 NPs. The results show that a clear pattern of toxicity in the studied species could not be established related to the coatings. However, it was possible to confirm different inter-species sensitivities. RTH-149 was the most sensitive cell line, and Daphnia magna was more sensitive than fish. Moreover, some differences in coating-core interactions were found between the metal oxide and the metal NPs in Daphnia magna.

Exposure Concentration Ratios and Biological Responses Play a Critical Role in Determining the Joint Toxicity of TiO2 Nanoparticles and As(V) to the Organism: the Case Study in Marine Algae Phaeodactylum Tricornutum.

Abstract:Environmental risks of manufactured nanomaterials (MNMs) have been widely investigated while the understanding for joint toxicity mechanism of MNMs with other contaminants is still limited. This limitation may be attributed to variations in the concentration ratios of MNMs and co-existing contaminants in the real environment. To better assess the joint toxicity and clarify its underlying mechanisms, this study exposed Phaeodactylum tricornutum to different concentration combinations of nano-sized titanium dioxide (nTiO2) and As(V) at toxic unit (TU) ratios of 1:4,1:1, and 4:1. The results demonstrated that the joint toxicity modes of nTiO2 and As(V) varied with the TU ratios exhibiting synergism for 1:4, partially addition for 1:1, and antagonism for 4:1. Specifically, at low TU ratio of 1:4, the adsorption of As(V) by nTiO2 together with the subsequent internalization of nTiO2 promoted a significant enrichment of As in algae. Simultaneously, the up-regulation of pst (phosphate transporter) genes in charge of the As(V) transport molecular further exacerbated the enrichment of inorganic As in algae, while the down-regulation of ArsM (arsenite S-adenosylmethionine methyltransferases) genes in charge of the As metabolism inhibited As biotransformation from toxic inorganic to nontoxic organic, causing the aggravated accumulation of toxic inorganic As in algae. At higher TU ratios of 1:1 and 4:1, the accumulation of As decreased in algae due to the higher sedimentation of nTiO2 and thus the lower internalization of As-adsorbed nTiO2, as well as the down-regulation of pst genes restricting the transportation of As(V) into algal cells, which jointly accelerated the As biotransformation from toxic inorganic to nontoxic organic. Our results suggest that more attention should be paid to exposure concentration ratios of MNMs and co-existing contaminants and biological responses including bioavailability, bioaccumulation, biotransformation, which would play a critical role in determining the joint toxicity to the organism.

Abstract:Due to the small size, high mobility and large surface area, nanoplastics (NPs) showed high potential risks to aquatic organisms. This paper reviews the toxicity of NPs to aquatic organism at various trophic levels including bacteria, plankton (algae), zooplankton, benthos, and nekton (fish). The effects at individual level caused by NPs were explained and proved by cytotoxicity and genotoxicity, and the toxicity of NPs beyond individual level was also illustrated. The toxicity of NPs is determined by the size, dosage, and surface property of NPs, as well as environmental factors, the presence of co-contaminants and the sensitivity of tested organisms. Furthermore, the joint effects of NPs with other commonly detected pollutants such as organic pollutants, metals, and nanoparticles etc. were summarized. In order to reflect the toxicity of NPs in the real natural environment, studies on toxicity assessment of NPs with the coexistence of various environmental factors and contaminants, particularly under the concentrations in natural environment are suggested.

Abstract:Ubiquitous dissolved organic matter (DOM) from various sources can interact with discharged multiwalled carbon nanotubes (MWCNTs) and pose distinct effects on the toxicity of MWCNTs to aquatic organisms. This study for the first time compared effects of a natural originated humic acid (HA) and two synthetic surfactants (sodium dodecylbenzenesulfonate – SDBS, octyl phenoxy polyethoxyethanol – TX100) on the toxicity of MWCNTs to a unicellular green alga (Chlorella pyrenoidosa). Algal growth inhibition, cell surface hydrophobicity, oxidative stress, MWCNT-cell agglomeration, and cell morphology change and internalization of MWCNTs were assayed to investigate individual and combined toxicities of MWCNTs and DOMs, and mechanisms underlying different effects of DOMs on the toxicity of MWCNTs were specifically addressed. It is shown that SDBS and TX100 were capable of promoting cell internalization of MWCNTs and triggering higher oxidative stress, and thereby increasing the toxicity of MWCNTs; while HA could alleviate the toxicity of MWCNTs through limiting cell internalization of MWCNTs and reducing oxidative stress. The outcomes of this work shed new light on the assessment of ecological toxicity of discharged nanomaterials.
Unraveling the toxicity mechanisms of nanoplastics with various surface modifications on Skeletonema costatum: Cellular and molecular perspectives

Abstract:Nanoplastics are ubiquitous in marine environments, exhibiting high bioavailability and potential toxicity to marine organisms. However, the impacts of nanoplastics with various surface modifications on marine microalgae remain largely unexplored. This study explored the toxicity mechanisms of two nanoplastic types-polystyrene (PS) and polymethyl methacrylate (PMMA)-with distinct surface modifications on Skeletonema costatum at cellular and molecular levels. Results showed that nanoplastics significantly impaired the growth of microalgae, particularly PS-NH2, which caused the most pronounced growth inhibition, reaching 56.99 % after a 96-h exposure at 50 mg/L. Transcriptomic profiling revealed that nanoplastics disrupted the expression of genes predominantly involved in ribosome biogenesis, aminoacyl-tRNA biosynthesis, amino acid metabolism, and carbohydrate metabolism pathways. The integrated biochemical and transcriptomic evidence highlighted that PS-NH2 nanoplastics had the most adverse impact on microalgae, affecting fundamental pathways such as ribosome biogenesis, energy metabolism, photosynthesis, and oxidative stress. Our findings underscore the influence of surface-modified nanoplastics on algal growth and contribute new understanding to the toxicity mechanisms of these nanoplastics in marine microalgae, offering critical information for assessing the risks of emerging pollutants.

Abstract:The present study investigated the impacts of dissolved organic matters (DOM) on joint toxicity involved in zinc oxide nanoparticles (ZnO NPs) and tetrabromobisphenol A (TBBPA) at relevant low-exposure concentrations (< 1 mg/L). It was found that ZnO NPs in single and combined systems exhibited severe inhibition effects on a freshwater microalgae Scenedesmus obliques. However, the presence of DOM slightly alleviated the growth inhibition toxicity induced by the binary mixtures of ZnO NPs and TBBPA. Ultrastructure analysis revealed that ZnO NPs caused structural damage to cells, including plasmolysis, membrane destruction, and the disruption of thylakoid in the chloroplast, regardless of the presence of coexisting substances. Oxidative stress biomarker quantitative analysis and in situ observations indicated that the massive accumulation of reactive oxygen species in the binary mixtures of ZnO NPs and TBBPA caused severe oxidative damage, but the presence of DOM significantly mitigated the damage.
