Peripheral blood Th17 and Th17/Th2 cells and their association with lung function and biomarkers in asthma-COPD overlap syndrome

Abstract:Background: Recent studies have revealed a novel subpopulation of human circulating CD4+T cells that produce both IL-4 and IL-17A (Th17/Th2 cells), which are present at a higher frequency in severe asthma. Converted from Th17 cells under the IL-4-rich microenvironment, Th17/Th2 cells can be more pathogenic than classic Th17 and Th2 cells. Objective: To analyze the cytokine profiles of circulating CD4+T cells in patients with ACOS, COPD and asthma, with an emphasis on the novel dual-positive Th17/Th2 cells. Methods: Blood samples were obtained from 23 patients with ACOS, 80 with COPD alone and 20 with asthma alone. The frequencies of CD4+T cell subpopulations were examined using intracellular flow cytometry. Results: A significantly increased proportion of Th17 cells was demonstrated in ACOS patients, as compared to patients with COPD and asthma alone, and correlated with FEV1%pred (r=-0.431; p Conclusion: Our results suggest that Th17 inflammation is involved in the pathogenesis of ACOS.
