Late Breaking Abstract - Membrane Bile Acid Receptor Contributes to the Proliferation of Lung Cancer Cells Induced by Lung Fibroblast-Tumor Cells Interaction Though TLR4/NF-kB Signaling Pathway

Abstract:Cancer associated fibroblasts (CAFs) are a critical component of the tumor microenvironment (TME). However, the underlying mechanism of CAFs in the progression of lung cancer has not been entirely elucidated. We hypothesized that membrane bile acid receptor (M-BAR) promotes the complex interaction between CAFs and lung cancer cells. In our study, human lung cancer cells (A549 cells) were co-cultured with lung fibroblasts (HPF cells) for 24, 48, 72 and 96 hours in a special 96-well plate in vitro. In addition, A549 cells were retro-virally transfected to establish stable cell lines overexpressing M-BAR. The results showed that the expression of M-BAR in the overexpressed group was significantly higher, compared with that in the control in A549 cells (P<0.05). The cell proliferation ability in the A549 cells was improved when co-cultured with HPF cells (P<0.05). The overexpression of M-BAR promoted the proliferation abilities and the expressions of TLR4 and phosphorylated NF-kB increased in the A549 cells with M-BAR overexpressing in the co-culture system as well as the increase of the TLR4 levels (P<0.05). However, silencing of the TLR4 attenuated the M-BAR‑induced proliferation and the expressions of TLR4 and phosphorylated NF-kB decreased. Therefore, this study suggests that M-BAR accelerates to the proliferation of NSCLC induced by lung fibroblast-tumor cells interaction though TLR4/NF-kB signaling pathway, which may become a novel target in the diagnosis, treatment and prognosis of patients with lung cancer.
