Cytoprotective Effects of CHBP, a Novel Erythropoietin Derivative Peptide, against H2O2-Induced Oxidative Stress in Human Renal Tubular Epithelial Cell

Abstract:LHBP is a novel erythropiotin derivative peptide that have been proved to attenuate renal ischemia reperfusion injury of mice. However, whether LHBP have protective effect against H2O2-induced oxidative stress on human renal tubular epithelial cell in vitro is still unknown. Therefore, the protective effects of LHBP on human renal tubular epithelial cell under oxidative stress and its molecular mechanisms were investigated. Human renal tubular epithelial cells were serum-free starved for 24 h and then exposed to 500uM hydrogen peroxide (H2O2) in serum-free DMEM/F12 for 1 h, LHBP was treated at the same time. After 24 h serum-free DMEM/F12 treatment, we observed a decrease of cell viability assessed by the MTT assay and an increased apoptotic by flow cytometric analysis using the Annexin V and the PI double-staining system. The expression levels of pro-apoptotic factor caspase-3 and anti-apoptotic signal PI3K/AKT were detected by western blot. The results showed that LHBP had a moderate anti-oxidative effect on human renal tubular epithelial cells. Co-incubation with LHBP increased the cell viability, markedly reduced the apoptosis induced by H2O2. And its protective effect is mediated by its inhibition of inflammation and tubular epithelium apoptosis. Furthermore, LHBP could up-regulate the expression of PI3K and AKT. These results suggest that LHBP prevents H2O2-induced cell injury through anti-apoptosis via the PI3K/AKT pathways.
