Abstract:A new synthesis of the tetrasubstituted tetrahydrofuran fragment of the marine secondary metabolites amphidinolides X and Y is described. The oxygenated chiral quaternary carbon was assembled by asymmetric dihydroxylation in high enantioselectivity and the tetrahydrofuran ring was constructed by an acid-catalyzed 5-endo ring-opening cyclization of the epoxide possessing a vinyl moiety.

Abstract:A facile and highly efficient three-component reaction of thiazole or benzothiazole carbenes, disubstituted ketenes, and activated alkynes is disclosed. With this methodology, a polysubstituted ring system containing furo[2,3-c]thiazepine core can be constructed from simple and readily accessible starting materials in good yields. The scope and limitation of this transformation were investigated in detail by using various thiazole carbene, ketene, and alkyne components. Furthermore, the synthetic utilities of these unique polyheterocyclic compounds were demonstrated via their Diels-Alder reactions with benzynes to furnish thiazepine-fused 7-oxanorbornadiene derivatives in excellent yields.

Abstract:Herein, we devised a synthetic approach for skeletally diversified furfuryl sulfides and 4‐alkylthiopyridines involving the heterocyclization of flexible α‐acyl ketene dithioacetals, enabled by cooperative Pd/Cu catalysis. The overall transformation was initiated by acyl‐directed desulfurative Sonogashira coupling, which was proposed as the key step in surmounting the competitive carbothiolation pathway. This step was followed by 1,6‐addition of either a S‐ or N‐nucleophile and intramolecular cyclization. Notably, the two different annulation reactions featured readily accessible starting materials, selective cleavage and reassembly of multiple chemical bonds, broad functional group tolerance, pharmacologically significant heterocyclic products with high regio‐ and chemoselectivities, and high atom economy.

Abstract:A series of 1,3-cyclodiketone- and tetrahydroepoxyisoindole-fused β-enamino dicarbonyl heterocycles were synthesized via a 1,4-diazabicyclo[2.2.2]octane-catalyzed, CH3NO2-mediated three-component reaction of 1,3-cyclodiketone, furfural, and allylamine in toluene. The target compounds were generated via the formation of β-enamino diketone as a key intermediate, followed by intramolecular Diels-Alder cycloaddition. The prepared molecules bearing a quinoline-2,4-dione moiety could be further brominated with N-bromosuccinimide and diastereoselectively reduced by NaBH4 to afford pyrano[3,2-c]quinolin-5-one-derived heterocycles with six vicinal stereogenic centers.

Abstract:A cyclodimerization of readily accessible acetylenic ketones facilitated by phosphane-borane complex under base is achieved. This methodology allows one pot synthesis of phosphorus-involved tetrasubstituted furans via the construction of a C−P bond and a furan ring within a single procedure. A plausible reaction mechanism is proposed.

Abstract:A series of new, substituted 2,3-dihydropyran derivatives were synthesized by one-pot, three-component reactions of aldehydes, 1,3-dicarbonyl derivatives, and vinyl ethers catalyzed by ethylenediamine diacetate (EDDA) under solvent-free conditions. This new, simple, and efficient, solvent-free, one-pot approach to the substituted 2,3-dihydropyran scaffold starting from simple substrates was found to take place directly.

Abstract:A palladium(II)-catalyzed tandem double-cyclization reaction of 1,omega-bisallenols was developed to form 2,5-dihydrofuran-fused bicyclic skeletons. With "unsymmetric" substrates, the reaction may also be realized with one hydroxyl group being protected as the acetate. Optically active bicyclic products were prepared by applying the Novozym-435 catalyzed kinetic resolution and the tandem double cyclization of these optically active allenol-allenyl acetates. The reaction may proceed via an oxypalladation, insertion, and elimination process.

Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.

Abstract:A multisubstituted tetrahydrofuran building block bearing three vicinal chiral carbon centers widely exists in a broad spectrum of bioactive natural products, and the development of efficient and convenient methods to establish this skeleton remains a challenging task. Herein, we have developed an efficient method for the construction of significant tetrahydrofuran scaffolds bearing three vicinal and α-quaternary chiral carbon stereocenters through Pd-catalyzed asymmetric [3 + 2] annulation of vinylethylene carbonates with alkenes installed on cyclic N-sulfonyl imines. A series of multisubstituted tetrahydrofuran derivatives are obtained in high efficiencies with excellent enantioselectivities and diastereoselectivities. Density functional theory (DFT) studies are accomplished to rationalize the stereocontrol of the annulation process and disclose that methanol could be applied to stabilize the reactive zwitterionic π-allylpalladium via the H-bond interaction.

Abstract:An effective and convenient strategy for the synthesis of a wide range of biologically imperative novel tetramethyl‐10‐(phenylamino)‐3,4,6,7,9,10‐hexahydroacridine‐1,8(2H,5H)‐dione compound and single crystal XRD studies. The notable advantages like shorter reaction times with excellent yields, environment‐friendly and simple purification process. An efficient and green synthesis of tetramethyl‐10‐(phenylamino)‐3,4,6,7,9,10‐hexahydroacridine‐1,8(2H,5H)‐dione derivatives has been achieved from various substituted aryl aldehydes, 5,5‐dimethyl‐3‐(2‐phenylhydrazinyl)cyclohex‐2‐enone and dimedone in aqueous ethanol medium. Environment‐friendly reaction, excellent yields, operational simplicity and shorter reaction times are the significant features of the present protocol. All the synthesized compounds were characterized by various spectroscopic techniques and X‐ray crystal diffraction studies.

Abstract:We report here a directly catalytic asymmetric method in the construction of CF2H‐containing pyrrolidines. A variety of enantioenriched difluoromethylated pyrrolidine (34 examples, up to 97% ee) were obtained on a 1, 3‐dipole cycloaddition model. The subsequent hydrolysis reaction provides a new difluoromethyl substituted fused ring compound which has certain anticancer activity.

Abstract:A novel asymmetric Pd-catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates and alpha,beta-unsaturated pyrazolones has been developed to afford highly functionalized tetrahydrofuran-fused spiro-heterocyclic skeletons with excellent stereoselectivity. A diverse range of enantioenriched spiro-tetrahydrofuran-pyrazolones, possessing three contiguous stereogenic centers including one all-carbon quaternary, were obtained in high yields and excellent enantioselectivities (up to 90% yield and 98% ee). In addition, the practical application of this protocol was also demonstrated by gram-scale synthesis and further transformations into various compounds. Preliminary mechanistic experiments indicated that the allylic 1, 3-strain was crucial to the stereoselective control. We developed an asymmetric Pd-catalyzed [3 + 2] cycloaddition of alpha,beta-unsaturated pyrazolones with vinylethylene carbonates. An array of spiro-tetrahydrofuran-pyrazolones were obtained in high yields with excellent enantioselectivities.

Abstract:A mixture of an isocyanide and dibenzoylacetylene in dry CH2Cl2 undergoes a smooth addition reaction at ambient temperature to furnish 3-[5-(alkylimino)-3,4-dibenzoyl-2-phenylfuran-2(2H)-yl]- 1-phenylprop-2-yn-1-ones (1 : 2 adduct) and {2,5-bis(alkylimino)-4,7,8a-triphenyl-5H-difuro[2,3-b:3 ',4 '-e]pyran-3(8aH)-yl}(phenyl)methanones (2 : 2 adduct). Single-crystal X-ray analyses conclusively confirmed the structures of the adducts.

Abstract:An N-heterocyclic carbene (NHC)-catalyzed asymmetric [4+2] annulation of (E)-2-benzoyl-3-phenylacrylonitriles with α-chloroaldehydes has been developed. Abstract:An efficient enantioselective protocol has been reported to build highly oxygenated and densely substituted bicyclo[m.n.1] skeletons through intramolecular asymmetric α-arylative desymmetrization of 1,3-diketones. Employing Pd catalyst and FOXAP ligand, various bicyclo[m.n.1] skeleton with different size can be accessed with high enantio- and diastereoselectivities. Utilizing the present method as a key step, formal asymmetric total synthesis of the (-)-parvifoline has been demonstrated.
