Fgf15/19-Fgfr4 Signalling Pathway Promote Fatty Liver Regeneration in Mice

Abstract:Background: Impaired liver regeneration (LR) in fatty liver was a critical problem to be solved. Our previous study showed that activation of an enterohepatic signal, which was ileal fibroblast growth factor (FGF) 15/19 to hepatic FGFR4 (FGF15/19-FGFR4) signalling promoted liver regeneration and reduced lipid accumulation after hepatectomy in wild type mice. However, the effect of FGF15/19-FGFR4 pathway on fatty liver regeneration is unclear. Methods: Ob/ob mice was used as fatty liver model and two-thirds partial hepatectomy (PH) was performed in ob/ob mice and wild type mice. Recombinant human FGF19 (rhFGF19) was administrated to investigate the effects of FGF15/19-FGFR4 signaling on fatty LR. Serum aminotransferase after PH was tested and western blot was used to detect the expression of FGFR4. Results: After rhFGF19 treatment, higher liver weight / body weight ratio at 36h after PH was observed in ob/ob mice. Serum aminotransferase analysis showed decreased ALT and AST in rhFGF19 treated group. Besides, lower expression of FGFR4 was seen in ob/ob mice compared to wild type mice, especially after suffering PH.Figure: No Caption available.Conclusion: In summary, these results demonstrate that FGF15/19-FGFR4 signalling pathway might promote fatty liver regeneration in vivo. And the related mechanism needs to be further investigated.
