Abstract:The aim of this study was to investigate the independent and interactive effectiveness of epigeic earthworms and arbuscular mycorrhizal (AM) fungi as soil conditioners in modifying the properties of salt-affected soil and increasing maize productivity. Treatments included maize plants inoculated vs. not inoculated with AM fungi and treated with or without earthworms. The mycorrhizal colonization, soil pH, electric conductivity (EC), nutrient availabilities and uptakes, catalase, soil microbial biomass C, and Shannon-Wiener index (H) for microbial communities from T-RFLP profiles were measured at harvest. The results showed that erthworms and AM fungi interactively decreased soil pH and EC, and increased the N, P and K content of maize shoot and root and their biomass. Earthworms and AM fungi interactively increased soil organic C, catalase activity and microbial biomass C in the saline alkali soil. Earthworms significantly decreased bacterial diversity, while earthworms and AM fungi interaction significantly increased fungal diversity. Pontibacter and Methylobacteriu were dominant bacteria, and Stachybotrys and Trichoderma were dominant fungi for all treatments. Earthworm addition significantly improved the abundance of Pontibacter and decreased the abundance of Methylobacteriu, which might result in higher soil P availablity and N absorption, respectively. Earthworms, AM fungi and their interaction increased the abundance of Trichoderma but decreased the abundance of Stachybotrys. Maize biomass and nutrient uptakes correlated significantly with the abundance of Trichoderma and Stachybotrys. In conclusion, earthworms, mycorrhiza and their interaction may have a potential role in elevating the chemical and biological properties to alleviate salinity and improve crop productivity in salt-affected soils. Abstract:Under salt stress, the root system is the first barrier in plant resistance. Endodermal suberin in plant roots can act as a diffusion barrier to effectively control ion uptake under abiotic stress. Soil biological interaction can affect plant root anatomic characters. In this study, the effects of earthworms ( Eisenia foetida ) and arbuscular mycorrhizal (AM) fungi ( Funneliformis mosseae ) on the maize root diffusion barrier in saline-alkali soil and the distribution characteristics of Na + were investigated. Maize seedlings grown in soil amended with earthworms and AM fungus were measured, and FY088 fluorescence staining was used to locate the accumulation of suberin in the endodermis. Scanning electron microscopy–energy dispersive X-ray spectrometry (SEM‒EDS) scanning was used to analyze the distribution of Na + in the cross-section of roots, and inductively coupled plasma-optical emission spectrometry (ICP-OES) was used to measure the mineral absorption of plants. The results showed that the total biomass of maize seedlings under salt stress increased by 70.4% with the amendment of earthworms and AM fungus, compared with the group without the amendment. Earthworms and AM fungus significantly changed the root anatomy of maize seedlings in saline-alkali soil increasing the accumulation of suberin layers in maize endodermis, effectively changing the distribution characteristics of Na + in root cross-section, reducing the transfer of Na + to stele, and increasing the K/Na ratio in roots and shoots by 59.8% and 73.3%, respectively. Our research showed that in saline-alkali soil, earthworms and AM fungus mediated root development and anatomical structure changes of maize, which was beneficial to maintain ion homeostasis of maize seedlings and improve root salt tolerance. This study provides new evidence for earthworm–mycorrhizal interaction to improve plant salt tolerance in saline-alkali soil.

Abstract:Trichoderma longibrachiatum sp. are free-living filamentous fungi which are common in agro-ecosystems. However, few studies thus far have examined the interaction between Trichoderma longibrachiatum and arbuscular mycorrhizal (AM) fungi in saline soil and their potential for improving plant stress tolerance. Here, single, dual-inoculated (T. longibrachiatum MF, AM fungal community or Glomus sp.), and non-inoculated maize (Zea may L.) were subjected to different salinity levels (0, 75, 150, and 225 mM NaCl) to test the synergistic effects of dual inoculants on maize plants in different salt stress conditions. Plant performance and metabolic profiles were compared to find the molecular mechanisms underlying plant protection against salt stress. The first experiment revealed that dual inoculation of an AM fungal community and T. longibrachiatum MF improved the biomass and K+/Na+ ratio in maize under non-saline conditions, and generally enhanced AM fungal growth in root and soil under all but the 225 mM NaCl conditions. However, MF inoculant did not influence the structure of AM fungal communities in maize roots. In the second experiment, dual inoculation of Glomus sp. and T. longibrachiatum MF increased maize plant biomass, K+/Na+ ratio, and AM fungal growth in root and soil significantly at both 0 and 75 mM NaCl conditions. We identified metabolic compounds differentially accumulated in dual-inoculated maize that may underline their enhanced maize plant tolerance to increasing soil salinity. Our data suggested that the combination of Glomus sp. and T. longibrachiatum leads to interactions, which may play a potential role in alleviating the stress and improve crop productivity in salt-affected soils.

Abstract:Salt stress has become a severe global problem, and salinity is one of the most important abiotic factors limiting plant growth and yield. It is known that arbuscular mycorrhizal (AM) fungi decrease plant yield losses under salinity. With the aim of determining whether AM inoculation would give an advantage to root development under salt stress, a greenhouse experiment was carried out with AM or without AM fungi. Maize plants were grown in a sand and soil mixture with 5 NaCl levels (0, 0.5, 1.0, 1.5, and 2.0 g/kg dry substrate) for 55 days, following 15 days of nonsaline pretreatment. At all salt levels, mycorrhizal plants had higher dry shoot and root mass, higher root activity, and lower root to shoot ratios than non-mycorrhizal plants. In salt-free soil, root length, root surface area, root volume, and number of root tips and forks were significantly larger in mycorrhizal plants than in non-mycorrhizal plants, whereas, under salt stress, average root diameter and root volume of mycorrhizal plants were larger than those of non-mycorrhizal plants. Regardless of the NaCl level, mycorrhizal plants had lower specific root length, lower percentage of root length in the 0-0.2 mm diameter class, and higher percentage of root length in both the 0.2-0.4 mm and 0.4-0.6 mm diameter classes, which suggests that the root system shows a significant shift towards a thicker root system when maize plants were inoculated with Glomus mosseae (Nicolson & Gerdemann). The results presented here indicate that the improvements in root activity and the coarse root system of mycorrhizal maize may help in alleviating salt stress on the plant.

Abstract:Salt stress is one of the most important abiotic stresses that restrict crop growth. Plant root-related microorganisms are important in moderating abiotic stress in plants. In this study, changes in microbial community composition in the different ecological niches of maize roots were studied to evaluate whether earthworms and mycorrhizal fungi could improve the salt tolerance of maize. This experiment was a 2 × 2 factorial design representing all the combinations of arbuscular mycorrhizal (AM) fungi (2.5 g/kg) with earthworms (10/pot) in saline soil. After culture, rhizosphere, rhizoplane and endosphere bacteria of maize were collected, and community assembly was analyzed and co-occurrence network was constructed. The regulating effect of earthworm-arbuscular mycorrhiza fungi (AMF) on bacterial community gradually decreased along the rhizosphere-rhizoplane-endosphere continuum. Earthworms and AMF are able to regulate key species in bacterial co-occurrence networks, thereby influencing the composition and function of the microbiome. Members of Rhizobiaceae and Gemmatimonadaceae, as key species, potentially regulate rhizosphere bacterial communities with earthworms and AMF. On the rhizoplane, AMF may specifically enriched bacterial communities with functions related to plant P absorption and maintenance of plant K-Na ratio, thereby maintaining plant nutrient homeostasis. Soil nitrate nitrogen content, plant K-Na ratio and root abscisic acid content were correlated with a large number of microbial network modules, and had potential causal relationship with rhizosphere, rhizoplane and endosphere bacterial community changes, which played an important role in the biomass accumulation of maize seedlings in saline soil. These results will provide a theoretical basis for the study of soil root-microbe interactions.
Enhancement of Soil Phosphorus Bioavailability by Arbuscular Mycorrhizae and Earthworms Through Regulating Soil Bacterial Community and Plant Nutrient Balance under Salt Stress

Abstract:Aims Soil salinization is an important factor limiting plant phosphorus (P) uptake and crop production. This study aimed to investigate the effects of arbuscular mycorrhizal fungi (AMFs) and earthworms in enhancing soil P bioavailability by regulating soil salt ions and altering the soil bacterial community under salt stress. Methods Treatments with or without earthworms and with or without AMFs in a high-salinity soil were applied. Results The results showed that the maize biomass and plant P, Ca and Mg contents were significantly increased by earthworms and AMF inoculation, and the highest plant P, Ca and Mg contents were observed with earthworm application alone. Earthworms and AMFs significantly decreased the soil stable inorganic P (hydroxyapatite) proportion and increased the soil available dicalcium phosphate proportion. AMFs significantly increased soil phosphatase activity and inorganic P fraction contents. Earthworms and AMFs significantly increased soil bacterial Chao1 and phylogenetic diversity. Structural equation model analysis showed that the most important driver of soil P mineralization was soil bacterial diversity, followed by soil Ca 2+ and total salt concentration. Network analysis suggested that the response of bacteria to soil Ca 2+ but not salt concentration positively correlated with soil P availability. Earthworms and AMFs could stimulate certain bacteria harbouring the phoX alkaline phosphatase gene to increase soil phosphatase activity and soil P availability. Conclusions In conclusion, earthworms and AMFs could enhance soil P bioavailability by stimulating soil P-cycling bacteria to activate soil stable inorganic P and by improving the plant cation nutrient balance under salt stress.
Effects of arbuscular mycorrhizal fungi on carbon assimilation and ecological stoichiometry of maize ( Zea mays ) under combined abiotic stresses

Abstract:Abstract Arbuscular mycorrhizal fungi (AMF) enhance plant tolerance to abiotic stresses like salinity and improve crop yield. However, their effects are variable, and the underlying cause of such variation remains largely unknown. This study aimed to assess how drought modified the AMF effect on plant resistance to high calcium-saline stress. A pot experiment was performed to examine how AMF inoculation affects the growth, photosynthetic activity, nutrient uptake, and C:N:P stoichiometric ratio of maize under high calcium stress and contrasting water conditions. The results showed that high calcium stress significantly reduced mycorrhizal colonization, biomass accumulation, C assimilation rate, and C:N stoichiometric ratio in plant tissues. Besides, the adverse effects of calcium stress on photosynthesis were exacerbated under drought. AMF inoculation profoundly alleviated such reductions under drought and saline stress. However, it barely impacted maize performance when subjected to calcium stress with well-watered conditions. Moreover, watering changed AMF impact on nutrient allocation in plant tissues. Under well-watered conditions, AMF stimulated P accumulation in roots and plant growth, but did not induce leaf P accumulation proportional to C and N, resulting in increased leaf C:P and N:P ratios under high calcium stress. In contrast, AMF decreased N content and the N:P ratio in leaves under drought. Overall, AMF inoculation improved maize resistance to calcium salt through enhanced photosynthesis and modulation of nutrient stoichiometry, particularly under water deficit. This study highlighted the regulatory role of AMF in carbon assimilation and nutrient homeostasis under compound stresses, which provides significant guidance on crop yield improvement in saline and arid regions.

Abstract:Abstract Background Inoculation of arbuscular mycorrhizal (AM) fungi has the potential to alleviate salt stress in host plants through the mitigation of ionic imbalance. However, inoculation effects vary, and the underlying mechanisms remain unclear. Two maize genotypes (JD52, salt-tolerant with large root system, and FSY1, salt-sensitive with small root system) inoculated with or without AM fungus Funneliformis mosseae were grown in pots containing soil amended with 0 or 100 mM NaCl (incrementally added 32 days after sowing, DAS) in a greenhouse. Plants were assessed 59 DAS for plant growth, tissue Na+ and K+ contents, the expression of plant transporter genes responsible for Na+ and/or K+ uptake, translocation or compartmentation, and chloroplast ultrastructure alterations. Results Under 100 mM NaCl, AM plants of both genotypes grew better with denser root systems than non-AM plants. Relative to non-AM plants, the accumulation of Na+ and K+ was decreased in AM plant shoots but increased in AM roots with a decrease in the shoot: root Na+ ratio particularly in FSY1, accompanied by differential regulation of ion transporter genes (i.e., ZmSOS1, ZmHKT1, and ZmNHX). This induced a relatively higher Na+ efflux (recirculating) rate than K+ in AM shoots while the converse outcoming (higher Na+ influx rate than K+) in AM roots. The higher K+: Na+ ratio in AM shoots contributed to the maintenance of structural and functional integrity of chloroplasts in mesophyll cells. Conclusion AM symbiosis improved maize salt tolerance by accelerating Na+ shoot-to-root translocation rate and mediating Na+/K+ distribution between shoots and roots.
[Effect of AM Fungi on Water and Nutrition Status of Corn Plants under Salt Stress].

Abstract:Under NaCl stress, the dry matter production of corn plants inoculated with or without arbuscular mycorrhizal (AM) fungus (Glomus mosseae) was decreased, but the decrement for non-mycorrhizal plants was 10% higher than that for mycorrhizal ones. Under salt stress condition, the dry weights of root system and aboveground part of mycorrhizal corn and its leaf water potential were higher than those of non-mycorrhizal corn, while the proline content of mycorrhizal corn was less than that of non-mycorrhizal corn. The contribution of hypha to P uptake of plants decreased from 45.3% to 42.6%, while the effect of AM fungi on plant growth increased from 30.9% to 63.5% under salt stress condition. The above-mentioned results indicated that the mechanism that AM fungi enhance the salt-resistance of corn is related with the improvements of water and P nutrition conditions. Meanwhile, it was found whether under salt stress or not, the ratio of P accumulation of root system to aboveground part of mycorrhizal corn was higher than that of non-mycorrhizal corn, indicating that the infection of AM fungi changed the P distribution pattern in plant bodies, which is beneficial to increase the salt-resistance of plants.
Improved Tolerance of Maize Plants to Salt Stress by Arbuscular Mycorrhiza is Related to Higher Accumulation of Soluble Sugars in Roots

Abstract:The effect of colonization with the arbuscular mycorrhizal (AM) fungus Glomus mosseae (Nicol. & Gerd.) Gerdemann & Trappe on the growth and physiology of NaCl-stressed maize plants (Zea mays L. cv. Yedan 13) was examined in the greenhouse. Maize plants were grown in sand with 0 or 100 mM NaCl and at two phosphorus (P) (0.05 and 0.1 mM) levels for 34 days, following 34 days of non-saline pre-treatment. Mycorrhizal plants maintained higher root and shoot dry weights. Concentrations of chlorophyll, P and soluble sugars were higher than in non-mycorrhizal plants under given NaCl and P levels. Sodium concentration in roots or shoots was similar in mycorrhizal and non-mycorrhizal plants. Mycorrhizal plants had higher electrolyte concentrations in roots and lower electrolyte leakage from roots than non-mycorrhizal plants under given NaCl and P levels. Although plants in the low P plus AM fungus treatment and those with high P minus AM fungus had similar P concentrations, the mycorrhizal plants still had higher dry weights, soluble sugars and electrolyte concentrations in roots. Similar relationships were observed regardless of the presence or absence of salt stress. Higher soluble sugars and electrolyte concentrations in mycorrhizal plants suggested a higher osmoregulating capacity of these plants. Alleviation of salt stress of a host plant by AM colonization appears not to be a specific effect. Furthermore, higher requirement for carbohydrates by AM fungi induces higher soluble sugar accumulation in host root tissues, which is independent of improvement in plant P status and enhances resistance to salt-induced osmotic stress in the mycorrhizal plant.
Earthworm and Arbuscular Mycorrhizal Fungi Improve Plant Hormones and Antioxidant Enzymes to Alleviate Simulated Acid Rain Stress

Abstract:Abstract Aims Acid rain is considered one of the three most serious environmental disasters worldwide, disrupting the normal physiological metabolism of plants and inhibiting their growth. As important parts of soil biota community, both earthworms and arbuscular mycorrhizal fungi can promote the growth of plants under adverse conditions. However, whether them can improve the stress tolerance of plants under simulated acid rain stress remains to be explored.Methods Illumina high-throughput sequencing was used to conduct a relevant redundancy analysis (RDA) of soil microbial community structure and plant growth factors, and a structural equation model (SEM) was constructed for maize biomass and a biological index to study the mechanisms by which earthworms and mycorrhizal fungi affect maize stress resistance under simulated acid rain stress. Results Earthworms promoted the absorption of soil organic matter by maize; promoted the growth of the root system; and increased the hormone levels of GA3, ABA and IAA; ultimately improving the stress resistance of maize. Mycorrhizal fungi increased the relative abundance of plant growth-promoting rhizosphere bacteria, increased the levels of plant hormones and antioxidant enzymes, and improved the stress resistance of maize. Earthworms promoted infection by mycorrhizal fungi, and the interaction between earthworms and mycorrhizal fungi increased the root IAA content and the Shannon index of rhizosphere bacteria.Conclusions Both earthworms and mycorrhizal fungi can improve the stress resistance of maize through underground regulation. They interacted in increasing the root IAA content and the Shannon index of rhizosphere bacteria and alleviated the simulated acid rain stress of the aboveground part of the maize.
Influence of Arbuscular Mycorrhizae on Photosynthesis and Water Status of Maize Plants under Salt Stress

Abstract:The influence of arbuscular mycorrhizal (AM) fungus Glomus mosseae on characteristics of the growth, water status, chlorophyll concentration, gas exchange, and chlorophyll fluorescence of maize plants under salt stress was studied in the greenhouse. Maize plants were grown in sand and soil mixture with five NaCl levels (0, 0.5, 1.0, 1.5, and 2.0 g/kg dry substrate) for 55 days, following 15 days of non-saline pretreatment. Under salt stress, mycorrhizal maize plants had higher dry weight of shoot and root, higher relative chlorophyll content, better water status (decreased water saturation deficit, increased water use efficiency, and relative water content), higher gas exchange capacity (increased photosynthetic rate, stomatal conductance and transpiration rate, and decreased intercellular CO2 concentration), higher non-photochemistry efficiency [increased non-photochemical quenching values (NPQ)], and higher photochemistry efficiency [increased the maximum quantum yield in the dark-adapted state (Fv/Fm), the maximum quantum yield in the light-adapted sate (Fv′/Fm′), the actual quantum yield in the light-adapted steady state (ϕPSII) and the photochemical quenching values (qP)], compared with non-mycorrhizal maize plants. In addition, AM symbiosis could trigger the regulation of the energy biturcation between photochemical and non-photochemical events reflected in the deexcitation rate constants (kN, kN′, kP, and kP′). All the results show that G. mosseae alleviates the deleterious effect of salt stress on plant growth, through improving plant water status, chlorophyll concentration, and photosynthetic capacity, while the influence of AM symbiosis on photosynthetic capacity of maize plants can be indirectly affected by soil salinity and mycorrhizae-mediated enhancement of water status, but not by the mycorrhizae-mediated enhancement of chlorophyll concentration and plant biomass.

Earthworms promoted infection by mycorrhizal fungi, and the interaction between earthworms and mycorrhizal fungi increased the root IAA content and the Shannon index of rhizosphere bacteria and alleviated the simulated acid rain stress of the aboveground part of the maize. This funding is crucial to understanding above- and belowground feedback in plant-soil systems under abiotic stress.
Influence of Arbuscular Mycorrhiza on Organic Solutes in Maize Leaves under Salt Stress

Abstract:A pot experiment was conducted to examine the effect of the arbuscular mycorrhizal (AM) fungus, Glomus mosseae, on plant biomass and organic solute accumulation in maize leaves. Maize plants were grown in sand and soil mixture with three NaCl levels (0, 0.5, and 1.0 g kg−1 dry substrate) for 55 days, after 15 days of establishment under non-saline conditions. At all salinity levels, mycorrhizal plants had higher biomass and higher accumulation of organic solutes in leaves, which were dominated by soluble sugars, reducing sugars, soluble protein, and organic acids in both mycorrhizal and non-mycorrhizal plants. The relative abundance of free amino acids and proline in total organic solutes was lower in mycorrhizal than in non-mycorrhizal plants, while that of reducing sugars was higher. In addition, the AM symbiosis raised the concentrations of soluble sugars, reducing sugars, soluble protein, total organic acids, oxalic acid, fumaric acid, acetic acid, malic acid, and citric acid and decreased the concentrations of total free amino acids, proline, formic acid, and succinic acid in maize leaves. In mycorrhizal plants, the dominant organic acid was oxalic acid, while in non-mycorrhizal plants, the dominant organic acid was succinic acid. All the results presented here indicate that the accumulation of organic solutes in leaves is a specific physiological response of maize plants to the AM symbiosis, which could mitigate the negative impact of soil salinity on plant productivity.

Abstract:Both the effects of salinity on ecology research of arbuscular mycorrhizal fungi(AMF) and growth effect of AMF on plants were depicted.The progresses in physiology and molecular mechanism of salt tolerance induced by AMF were reviewed in this paper,which were including nutrition absorption,osmosis adjustment,anti-oxidative enzymes,hormones and gene expression.Some unsolved problems in AMF research areas were also mentioned.

Abstract:The interactive impacts of arbuscular mycorrhizal fungi (AMF, Glomus intraradices ) and earthworms ( Aporrectodea trapezoides ) on maize ( Zea mays L.) growth and nutrient uptake were studied under near natural conditions with pots buried in the soil of a maize field. Treatments included maize plants inoculated vs. not inoculated with AMF, treated or not treated with earthworms, at low (25 mg kg −1 ) or high (175 mg kg −1 ) P fertilization rate. Wheat straw was added as feed for earthworms. Root colonization, mycorrhiza structure, plant biomass and N and P contents of shoots and roots, soil available P and NO 3 − –N concentrations, and soil microbial biomass C and N were measured at harvest. Results indicated that mycorrhizal colonization increased markedly in maize inoculated with AMF especially at low P rate, which was further enhanced by the addition of earthworms. AMF and earthworms interactively increased maize shoot and root biomass as well as N and P uptake but decreased soil NO 3 − –N and available P concentrations at harvest. Earthworm and AMF interaction also increased soil microbial biomass C, which probably improved root N and P contents and indirectly increased the shoot N and P uptake. At low P rate, soil N mobilization by earthworms might have reduced potential N competition by arbuscular mycorrhizal hyphae, resulting in greater plant shoot and root biomass. Earthworms and AMF interactively enhanced soil N and P availability, leading to greater nutrient uptake and plant growth.

Abstract:The effects of arbuscular mycorrhizal fungi(AMF)on plant growth and osmotic adjustment matter content of pot cultured tomato under NaCl stress(0.5%and 1%)were studied.The results showed that AMF-inoculation significantly increased tomato plant growth,promoted the accumulation of soluble sugar in leaves and roots,increased soluble protein in leaves and proline content in roots under salt stress,so AMF enhanced salt tolerance of tomato plants.The accumulation of soluble sugar,soluble protein and root proline induced by AMF played an important role in osmotic adjustment mechanism of enhanced salt tolerance in mycorrhizal tomato.

Abstract:Arbuscular mycorrhizal fungi (AMF) was considered as a biotechnological tool for plant stress tolerance improvement and degraded ecosystem restoration. However, the variations in the effects of AMF species and abundance on plant performance under stress condition have to be still investigated. The study was conducted to evaluate the mechanisms of five AMF species, single or mixture, on Leymus chinensis stress tolerance along a saline-alkaline gradient. The results showed that AMF enhanced plant stress tolerance by promoting plant growth, enhancing nutrient absorption, maintaining ion balance, and improving photosynthetic efficiency. Funneliformis mosseae, Rhizophagus intraradices and the mixture treatments had more beneficial effects than Diversispora versiformis and Acaulospora scrobiculata inoculations. Plant grown under high stress level exhibited more benefits from AMF symbiosis. Our study suggests that not only presence of AMF, but also the species and abundance should be considered to reveal the exact effects of AMF on plant saline-alkaline tolerance and degraded grassland restoration.
