Abstract:We present a focus-adjustable endoscopic probe for optical coherence tomography (OCT), which is able to acquire images with different focal planes and overcome depth-of-focus limitations by image fusing. The use of a two-way shape-memory-alloy spring enables the probe to adjust working distance over 1.5 mm, providing a large scanning range with high resolution and no sensitivity loss. Equipped with a homemade hollow-core ultrasonic motor, the probe is capable of performing an unobstructed 360 deg field-of-view distal scanning. Both the axial resolution and the best lateral resolution are ∼4 μm, with a sensitivity of 100.3 dB. Spectral-domain OCT imaging of phantom and biological tissues with the probe is also demonstrated.

Abstract:Minimized probe is a common requirement in endoscopic optical coherence tomography (OCT). We introduce the development of mainstream designs based on ball lens, fiber lens, graded index fiber, free form lens and free-lens, summarize their advantages and disadvantages, and put forward some suggestions for miniaturization of the probe. The development of probe with extended depth of focus (DOF) poses significance on imaging subcellular structure of human internal organs. We review several important techniques of DOF extension suitable for miniature probes, among which the probe based on mode interference is believed to have great potential because of its easy fabrication, compact structure, high light transmission efficiency, optimized working distance and DOF, and uniformity of axial light intensity.

Abstract:We present a novel miniature endoscopic optical coherence tomography (OCT) probe based on an electrothermal microelectromechanical (MEMS) mirror and a C-lens. The MEMS mirror has a relatively large mirror plate of 0.5 mm × 0.5 mm on a small chip size of 1.5 mm × 1.3 mm, leading to the outer diameter of the endoscopic probe down to only 2.5 mm so that the probe is capable of being inserted through the biopsy channel of a conventional endoscope. A long focal length of 12 mm is achieved by properly designing the C-lens. Compared to commonly used GRIN lenses, C-lenses have advantages of much smaller sensitivities of the working distance and spot size to machining variations. The C-lens based probe can scan a large field of view (FOV) of 6 mm × 6 mm at only 5-V drive voltage. A swept source endoscopic OCT system is constructed using this miniature probe. Experiments show that the corresponding lateral and axial resolutions are about 50 and 14 μm, respectively. Various samples including human fingers, onions, and cancerous colon tissue are imaged and the result suggests this large-working-distance, large-FOV endoscopic probe is promising for applications in large tubular organs and large-area lesions.

Abstract:It is difficult to maintain high transverse resolution over an increased depth range using miniature probes for optical coherence tomography (OCT) due to the rapid divergence of light and the space limitation. To solve this problem, we introduce a fiber-based filter in the proposed probe to manipulate its output beam. Significant mode interference (MI) is exploited to enhance the depth of focus (DOF), and the mode phase difference is tuned to achieve a uniform axial intensity within the DOE The magnified MI field instead of the diffracted one is adopted as the final pupil filter in the probe to increase its working distance (WD). The probe is fabricated with a diameter of 125 mu m and a total length of 2.6 mm for its distal fiber optics. Compared to the conventional probe with similar minimal lateral resolution of better than 4.4 mu m, the proposed probe achieves two times that of the DOF gain and 1.7 times that of the WD. Improvements in performance of the probe are demonstrated by OCT imaging using a fresh lemon and human skin. With merits of enhanced imaging quality and easy fabrication, the proposed probe poses great potential for important applications, especially for endoscopic imaging of human internal organs in vivo. Tapered Structure Based All-Fiber Probe for Endoscopic Optical Coherence Tomography

Abstract:A tapered structure based all-fiber endoscopic probe for endoscopic optical coherence tomography (OCT) is presented in this paper. The designation and fabrication of a miniaturized high-performance probe are critical in endoscopic OCT. Compared to the conventional fiber-lens structure based endoscopic probe, the all-fiber probe has a prominent edge in size and flexibility. Due to its lower beam-divergence, the large core multi-mode fiber makes a better fit than a general single mode fiber does when utilized to replace the micro lens in a conventional endoscopic probe as the imaging component. Furthermore, a tapered fiber is introduced as a transition section between the single mode fiber and the large core multi-mode fiber in order to enhance the light transmission efficiency and reduce the rigid length of the probe simultaneously. First, in order to obtain an optimal performance, optical simulation software(Rsoft) is adopted to determine the probe's proper lengths of the tapered section and the large core multi-mode fiber. Second, the all-fiber structure based endoscopic probe is fabricated by means of large core multi-mode fiber tapering, cutting and fusing processes. The beam characterization and insertion loss of the fabricated probe are measured experimentally The probe itself is 250 m, and after covering with a stainless steel protective tube, its outer diameter becomes 325 m. The rigid length of the probe is about 1 cm, which is more flexible and easier for inserting into curved blood vessels. The insertion loss of the probe is measured to be about 0.3 dB. To the best of our knowledge, it is the lowest among all of the all-fiber endoscopic probes. Finally, the probe is integrated with a custom-built swept-source optical coherence tomography system. Imaging of human fingertip and ex-vivo chicken trachea is conducted to demonstrate the key performance parameters of our probe. The effective imaging range of the probe is up to 800 microns in air without the help of any extra mechanism to expand its depth of focus. The probe offers a compact, efficient and flexible candidate for endoscopic optical coherence tomography, which is promising in cardiovascular investigations.

Abstract:We designed and implemented a magnetic-driven scanning (MDS) probe for endoscopic optical coherence tomography (OCT). The probe uses an externally-driven tiny magnet in the distal end to achieve unobstructed 360-degree circumferential scanning at the side of the probe. The design simplifies the scanning part inside the probe and thus allows for easy miniaturization and cost reduction. We made a prototype probe with an outer diameter of 1.4 mm and demonstrated its capability by acquiring OCT images of ex vivo trachea and artery samples from a pigeon. We used a spectrometer-based Fourier-domain OCT system and the system sensitivity with our prototype probe was measured to be 91 dB with an illumination power of 850 μW and A-scan exposure time of 1 ms. The axial and lateral resolutions of the system are 6.5 μm and 8.1 μm, respectively.

Abstract:This paper proposes a novel rotary endoscopic probe for spectral-domain optical coherence tomography (SD-OCT). The probe with a large N.A. objective lens is driven by an ultra-small hollow rectangular ultrasonic motor for circular scanning. Compared to the conventional driven techniques, the hollow ultrasonic motor enables the fiber to pass through its inside. Therefore the fiber, the objective lens and the motor are all at the same side. This enables 360 degree unobstructed imaging without any shadow resulted from power wire as in the conventional motor-driven endoscopic OCT. Moreover, it shortens the length of the rigid tip and enhances the flexibility of the probe. Meanwhile, the ultrasonic motor is robust, simple, quiet and of high torque, very suitable for OCT endoscopic probe. The side length of the motor is 0.7 mm with 5mm in length. The outer diameter of the probe is 1.5mm. A significant improvement in the lateral resolution is demonstrated due to the novel design of the objective lens. A right-angle lens is utilized instead of the traditional right-angle prism as the last optics close to the sample, leading to a reduction of the working distance and an enlargement of the N.A. of the objective lens. It is demonstrated that the endoscopic SD-OCT system achieves an axial resolution of ~7μm, a lateral resolution of ~6μm and a SNR of ~96dB.

Abstract:A high-efficiency lensless all-fiber probe for optical coherence tomography (OCT) is presented. The probe is composed of a segment of large-core multimode fiber (MMF), a segment of tapered MMF, and a length of single-mode fiber (SMF). A controllable output beam can be designed by a simple adjustment of its probe structure parameters (PSPs), instead of the selection of fibers with different optical parameters. A side-view probe with a diameter of 340 mu m and a rigid length of 6.37 mm was fabricated, which provides an effective imaging range of similar to 0.6 mm with a full width at half-maximum beam diameter of less than 30 mu m. The insertion loss of the probe was measured to be 0.81 dB, ensuring a high sensitivity of 102.25 dB. Satisfactory images were obtained by the probe-based OCT system, demonstrating the feasibility of the probe for endoscopic OCT applications. (C) 2017 Optical Society of America

Abstract:Introduction: Optical coherence tomography (OCT) is a pivotal imaging modality in ophthalmology for real-time, in vivo visualization of retinal structures. To enhance the capability and safety of OCT, this study focuses on the development of a micro intraocular OCT probe. The demand for minimal invasiveness and precise imaging drives the need for advanced probe designs that can access tight and sensitive areas, such as the ocular sclera. Methods: A novel OCT probe was engineered using a piezoelectric tube with quartered electrodes to drive Lissajous scanning movements at the end of a single-mode fiber. This design allows the probe to enter the eyeball through a scleral opening. Structural innovation enables the outer diameter of the endoscopic OCT probe to be adjusted from 13G (2.41 mm) to 25G (0.51 mm), accommodating various imaging field sizes and ensuring compatibility with different scleral incisions. Results: The fabricated micro intraocular OCT probe successfully performed preliminary imaging experiments on in vivo fingers. The Lissajous scanning facilitated comprehensive coverage of the target area, enhancing the imaging capabilities. Discussion: The integration of a piezoelectric tube with quartered outside electrodes into the OCT probe design proved effective for achieving precise control over scanning movements and adaptability to different surgical needs. The design characteristics and practical applications demonstrated the probe's potential in clinical settings.

Abstract:In this paper, we present a distal-scanning common path probe for optical coherence tomography (OCT) equipped with a hollow ultrasonic motor and a simple and specially designed beam-splitter. This novel probe proves to be able to effectively circumvent polarization and dispersion mismatch caused by fiber motion and is more robust to a variety of interfering factors during the imaging process, experimentally compared to a conventional noncommon path probe. Furthermore, our design counteracts the attenuation of backscattering with depth and the fall-off of the signal, resulting in a more balanced signal range and greater imaging depth. Spectral-domain OCT imaging of phantom and biological tissue is also demonstrated with a sensitivity of similar to 100dB and a lateral resolution of similar to 3 mu m. This low-cost probe offers simplified system configuration and excellent robustness, and is therefore particularly suitable for clinical diagnosis as one-off medical apparatus.

Abstract:Although methods have been proposed to maintain high transverse resolution over an increased depth range, it is not straightforward to scale down the bulk-optic solutions to minimized probes of optical coherence tomography (OCT). In this paper, we propose a high-efficient fiber-based filter in an all-fiber OCT probe to realize an extended depth of focus (DOF) while maintaining a high transverse resolution. Mode interference in the probe is exploited to modulate the complex field with controllable radial distribution. The principle of DOF extension by the fiber-based filter is theoretically analyzed. Numerical simulations are conducted to evaluate the performances of the designed probes. A DOF extension ratio of 2.6 over conventional Gaussian beam is obtainable in one proposed probe under a focused beam diameter of 4.6μm. Coupling efficiencies of internal interfaces of the proposed probe are below −40 dB except the last probe–air interface, which can also be depressed to be −44 dB after minor modification in lengths for the filter. Length tolerance of the proposed probe is determined to be −28∕+20μm, which is readily satisfied in fabrication. With the merits of extended-DOF, high-resolution, high-efficiency and easy-fabrication, the proposed probe is promising in endoscopic applications.

Abstract:We present a novel circumferential-scan endoscopic optical coherence tomography (OCT) probe by using a circular array of six electrothermal microelectromechanical (MEMS) mirrors and six C-lenses. The MEMS mirrors have a 0.5 mm × 0.5 mm mirror plate and a chip size of 1.5 mm × 1.3 mm. Each MEMS mirror can scan up to 45° at a voltage of less than 12 V. Six of those mirrors have been successfully packaged to a probe head; full circumferential scans have been demonstrated. Furthermore, each scan unit is composed of a MEMS mirror and a C-lens and the six scan units can be designed with different focal lengths to adapt for lesions with uneven surfaces. Configured with a swept source OCT system, this MEMS array-based circumferential scanning probe has been applied to image a swine's small intestine wrapped on a 20 mm-diameter glass tube. The OCT imaging result shows that this new MEMS endoscopic OCT has promising applications in large tubular organs.
3D-printed axicon enables extended depth-of-focus intravascular optical coherence tomography

Abstract:A fundamental challenge in endoscopy is how to fabricate a small fiber-optic probe that can achieve comparable function to probes with large, complicated optics (e.g., high resolution and extended depth of focus). To achieve high resolution over an extended depth of focus (DOF), the application of needle-like beams has been proposed. However, existing methods using miniaturized needle beam designs fail to adequately correct astigmatism and other monochromatic aberrations, limiting the resolution of at least one axis. Here, we describe a novel approach to realize freeform beam-shaping endoscopic probes via two-photon direct laser writing, also known as micro 3D-printing. We present a design achieving approximately 8-micron resolution with a DOF of >0.8 mm at a central wavelength of 1310 nm. The probe has a diameter of 0.25 mm (without the catheter sheaths) and is fabricated using a single printing step directly on the optical fiber. We demonstrate our device in intravascular imaging of living atherosclerotic pigs at multiple time points, as well as human arteries with plaques ex vivo. This is the first step to enable beam-tailoring endoscopic probes which achieve diffraction-limited resolution over a large DOF.

Abstract:A two-axis scanning MEMS based forward-viewing endoscopic imaging probe has been developed and successfully applied in a swept source OCT system for three-dimensional imaging. The 2-axis MEMS mirror has an aperture size of 0.85 mm by 0.85 mm and a chip size of 1.5 mm by 1.7 mm and the outer diameter of the probe is 5.0 mm. The MEMS mirror achieves large optical scan angle up to 30° at 3 V. The MEMS based forward-imaging probe demonstrates real time 3D OCT images of human nail and rat skin with 10.6 μm axial resolution and 17.5 μm lateral resolution at a frame rate of 50 fps.

Abstract:This paper proposes an endoscopic probe for optical coherence tomography (OCT) applying on side-imaging of internal organs. The probe consists of single-mode fiber, a gradient-index (GRIN) lens, and a mirror of cylindrical wedge shape attaching to a magnetized metal piece by epoxy with a short steel wire. For OCT scanning, we use magnetic field generated by a larger magnet externally to drive the rotation of the magnetized metal. Compare with other probes, our probe design has two distinct advantages: 1) The exit beam will be unobstructed during 360 degree circumference scanning because there are no connecting wires in the scanning part. 2) In principle, the probe can be made very tiny because of the simple structure consisting only the single-mode fiber, GRIN lens, reflection mirror and the magnetized metal piece. We used a spectrometer-based Fourier-domain OCT system and the system sensitivity with our prototype probe was measured to be 91 dB. The axial and lateral resolutions of the system are 6.5 μm and 8.1 μm, respectively.
Modeling, optimization, and validation of an extended-depth-of-field optical coherence tomography probe based on a mirror tunnel

Abstract:The diagnostic capability of high-resolution optical coherence tomography (OCT) may be enhanced by using extended depth-of-field (EDOF) imaging that retains high transverse resolution over long depths. A recently developed mirror-tunnel optical probe design (single-mode fiber to multimode fiber to lens structure) that generates coaxially focused modes has been previously shown to enable EDOF for endoscopic OCT applications. Here, we present ray-tracing optical modeling of this optical configuration, which has the potential to guide performance improvement through optimization. The Huygens wave propagation of the field was traced through probe components with initial lengths. The irradiance along the x-z plane was analyzed, yielding an average full width at half-maximum (FWHM) of 9 µm over a 640 µm DOF (defined as the axial range over which the beam's transverse FWHM is maintained). A custom merit function was then defined, based on the focal region illumination intensity profile that yielded the maximum possible depth having a constrained FWHM size. An orthogonal gradient descent optimization algorithm was then applied using this merit function, using the multimode fiber, spacer, and lens lengths as variables. Optimization resulted in a modeled mean 6 µm FWHM spot diameter over an EDOF of 1 mm. Following optimization, a probe was fabricated, and was validated using a custom-built near-field scanning pinhole beam profiler. The experimental results (6 µm mean FWHM over 800 µm EDOF) showed reasonable correspondence to the simulated predictions, demonstrating the potential utility of optical modeling and optimization for improving EDOF performance in mirror-tunnel endoscopic OCT probes.

Abstract:We developed a compact, optical fiber scanning piezoelectric transducer (PZT) probe for endoscopic and minimally invasive optical coherence tomography (OCT). Compared with previous forward-mount fiber designs, we present a reverse-mount design that achieves a shorter rigid length. The fiber was mounted at the proximal end of a quadruple PZT tube and scanned inside the hollow PZT tube to reduce the probe length. The fiber resonant frequency was 338 Hz using a 17-mm-long fiber. A 0.9 mm fiber deflection was achieved with a driving amplitude of 35 V. Using a GRIN lens-based optical design with a 1.3× magnification, a ∼6 μm spot was scanned over a 1.2 mm diameter field. The probe was encased in a metal hypodermic tube with a ∼25 mm rigid length and covered with a 3.2 mm outer diameter (OD) plastic sheath. Imaging was performed with a swept source OCT system based on a Fourier domain modelocked laser (FDML) light source at a 240 kHz axial scan rate and 8 μm axial resolution (in air). En face OCT imaging of skin in vivo and human colon ex vivo was demonstrated.

Abstract:We present a proof-of-concept for side-viewing endoscopic optical coherence tomography probes designed for real-time imaging. The design employs a commercial rotating micro-engine (Kinetron), with a 1 mm outer diameter and maximum speed of 10,000 rpm, to steer the beam from a GRIN lens-terminated optical fiber (Agiltron), 360 degrees around the probe body. The engine is encapsulated inside a PET tube with an outer diameter of 1.6 mm, and coupled to a swept-source based optical coherence tomography (SS-OCT) system operating at 1300 nm with an A-scan rate of 100 kHz. Large material dispersion mismatch between the reference and sample arms, which would otherwise degrade the axial resolution, is compensated for by using the Master-Slave OCT technique to process the interferograms. This allows for more convenient interchange of probes without exactly matching the fibre length. The probe can be configured using GRIN lenses at the end of the fibers with different focal lengths, and the fiber lengths can differ by several cm. When the probe is changed, the reference path is adjusted, but no dispersion compensation is needed, due to use of the Master Slave method developed by our group. We will demonstrate the performance of such systems with full 360 degree tomographic images of scattering phantoms taken at different probe driving speeds, with different configurations of the probe heads.

Abstract:Optical coherence elastography (OCE) is a functional extension of optical coherence tomography (OCT). It offers high-resolution elasticity assessment with nanoscale tissue displacement sensitivity and high quantification accuracy, promising to enhance diagnostic precision. However, in vivo endoscopic OCE imaging has not been demonstrated yet, which needs to overcome key challenges related to probe miniaturization, high excitation efficiency and speed. This study presents a novel endoscopic OCE system, achieving the first endoscopic OCE imaging in vivo. The system features the smallest integrated OCE probe with an outer diameter of only 0.9 mm (with a 1.2-mm protective tube during imaging). Utilizing a single 38-MHz high-frequency ultrasound transducer, the system induced rapid deformation in tissues with enhanced excitation efficiency. In phantom studies, the OCE quantification results match well with compression testing results, showing the system's high accuracy. The in vivo imaging of the rat vagina demonstrated the system's capability to detect changes in tissue elasticity continually and distinguish between normal tissue, hematomas, and tissue with increased collagen fibers precisely. This research narrows the gap for the clinical implementation of the endoscopic OCE system, offering the potential for the early diagnosis of intraluminal diseases.
