Abstract:A combinatorial nickel-catalyzed monofluoroalkylation of aryl bromides with the industrial raw regent ethyl chlorofluoroacetate has been developed. The two key factors to successful conversion are the combination of nickel with readily available nitrogen and phosphine ligands and the using of a mixture of different solvents. Mechanistic investigations indicated a new zinc regent might generated in situ and be involved in the reaction process.

Abstract:A nickel-catalyzed direct cross-coupling of unactivated aryl fluorides with aryl bromides is realized. The one-pot reaction, which avoids the use of preformed and sensitive organometallic reagents, proceeds effectively via C-F bond cleavage at room temperature in THF in the presence of the phosphine ligand and magnesium powder (with or without TMSCl) to produce the desired biaryls in modest to good yields.

Abstract:A combinatorial nickel‐catalyzed direct trifluoroethylation of aryl iodides with an industrial raw material CF3CH2I has been developed, demonstrating high efficiency, excellent functional‐group compatibility, especially with large sterically hindered groups. Abstract:As a unique organofluorine fragment, gem-difluoromethylated motifs have received widespread attention. Here, a convenient and efficient synthesis of aryldifluoromethyl aryl ethers (ArCF 2 OAr') was established via Nickel-catalyzed aryloxydifluoromethylation with arylboronic acids. This approach features easily accessible starting materials, good tolerance of functionalities, and mild reaction conditions. Diverse late-stage difluoromethylation of many pharmaceuticals and natural products were readily realized. Notably, a new difluoromethylated PD-1/PD-L1 immune checkpoint inhibitor was conveniently synthesized and showed both improved metabolic stability and enhanced antitumor efficacy. Preliminary mechanistic studies suggested the involvement of a Ni(I/III) catalytic cycle.

The utility of this method is demonstrated by the monofluoromethylation of a borylated and acyl-protected derivative of the statin drug ezetimibe. Mechanistic investigations indicate that a fluoromethyl radical is involved in the Ni(I)/Ni(III) catalytic cycle.

Abstract:The use of abundant and inexpensive fluorine feedstocks to synthesize fluorinated compounds is a promising strategy that has not been extensively investigated. Dibromodifluoromethane (CF2Br2) is an inexpensive fluorine source that has rarely been used for C-H fluoroalkylation. This study reveals an iridium-catalyzed, tunable strategy for synthesizing acyl fluorides and difluorobromomethylated products using CF2Br2. To achieve the desired products, this process only requires the change of solvent (from DMSO to 1,4-dioxane) under blue LED illumination. A variety of arenes and heteroarenes with electron-donating substituents were successfully used, yielding the corresponding products in moderate to good yields. Mechanistic experiments revealed that DMSO served a dual role, functioning as both solvent and nucleophilic reagent in C-H fluorocarbonylation.

Abstract:A combinatorial nickel-catalyzed cross-coupling between arylboronic acids and unactived 1-fluoro-1-iodoalkanes has been developed, which demonstrated high efficiency, mild conditions, and excellent functional-group compatibility. Readily available nitrogen and phosphine ligands were combined with a nitrogen source, which in situ generated a variety of easily tunable catalysts to promote the fluoroalkylation for broad scopes of both coupling partners. This new strategy on combinatorial catalysis offers new solutions for nickel-catalyzed cross-coupling reactions.

Abstract:A new radical difluoromethylation was developed by using inexpensive and readily available difluoroacetic anhydride and N-phenyl-4-methylbenzenesulfonamide for the first time. The reaction of arylboronic acids with the new difluoromethylation reagent, N-phenyl-N-tosyldifluoroacetamide, proceeded smoothly in the presence of palladium catalyst to provide difluoromethylarenes in satisfactory to excellent yields. The electronic property (electron-donating or electron-withdrawing) of the substituent linked to the aromatic ring did not considerably influence the reactivity of arylboronic acid. Various groups, including the synthetically useful functional groups Cl, CN, and NO2, were tolerated well under the current reaction conditions.

Abstract:The nickel-catalyzed multicomponent dicarbofunctionalization of alkenes represents one of the most straightforward approaches to complex organofluorine compounds. However, the catalytic carbofluoroalkylation of electron-deficient alkenes remains underdeveloped. Here, we report a nickel-catalyzed three-component aryldifluoroalkylation of industry-relevant acrylonitrile and acrylate with arylboronic acids and difluoroalkyl halides. The reaction can extend to a four-component nickel-catalyzed carbonylation reaction under 1 atm CO gas. This modular catalytic synthesis features synthetic convenience, high functional group tolerance, and simple components to construct complex difluoroalkylated compounds expediently. The synthetic utility has also been demonstrated by the diversified transformations of the resulting products into medicinal interest fluorinated compounds.

Abstract:Comprehensive Summary Herein we report nickel‐catalyzed cross‐coupling of gem‐difluorinated cyclopropanes with boronic acids, providing the corresponding arylated 2‐fluoroallylic scaffolds. This approach used commercially available phosphine ligand Xantphos to obtain monofluorinated alkenes with high regioselectivity and Z‐stereoselectivity. Mechanistic studies proposed a Ni(II)‐fluoroallyl pathway and excluded the radical pathway. A variety of C(sp3)-CF2H containing compounds were prepared through a hydrodifluoromethylation process with good efficiency under mild conditions. In addition, various gem-difluoroalkenes bearing CF2H or 1o, 2o, 3o alkyl group were synthesized from trifluoromethyl alkenes via a defluorination process using this new established approach.

Abstract:Herein, we describe a nickel-catalyzed reductive decarboxylative difluoromethylation reaction of alkenes using inexpensive and easy-to-handle difluoroacetic anhydride (DFAA)/pyridine N-oxide reagent system. A variety of C(sp3)-CF2H containing compounds were prepared through a hydrodifluoromethylation process. Besides, various gem-difluoroalkenes bearing CF2H group were synthesized via defluorinative reductive cross-coupling process from trifluoromethyl-substituted alkenes using this new reaction system. Difluoroacetic anhydride has been then extended to other common alkyl anhydrides, and the corresponding hydroalkylation and defluoroalkylation processes have been successfully achieved. This method features broad substrate scope, good functional group tolerance as well as high efficiency.

Abstract:Herein, we describe a nickel-catalyzed divergent formylation and carboxylation reaction of aryl halides with isocyanides. A rich array of aromatic aldehydes and carboxylic acids can be, respectively, accessed in moderate to good yields. Some sensitive functional groups such as hydroxyl, iodine, cyano, and indolyl are fairly tolerant of nickel catalysis. In the carboxylation reactions, the combination of isocyanide and H2O is first employed as a promising carbonyl surrogate instead of gaseous CO and CO2.

Abstract:The defluoroborylation of fluoroarenes by chromium-catalyzed cleavage of unactivated C–F bonds is described. The reaction uses HBpin as the boron source, low-cost and commercially available chromium salt as the precatalyst, and terpyridine as crucial ligand, providing a protocol with atom-efficient benefits and a wide range of applicable substrates for the functionalization of aryl C–F bonds. Preliminary mechanistic studies indicate an unprecedant Cr-catalyzed magnesation of unactivated C–F bond occurred. The generated arylmagnesium intermediates then participated in the subsequent borylation reaction. The application of the stratey in the preparation of valuable derivatives is demonstrated by the late-stage functionalization of boronate ester groups.
