Abstract:The mussel byssus has long been a source of inspiration for the adhesion community. Recently, adhesive synergy between flanking lysine (Lys, K) and 3,4-Dihydroxyphenylalanine (DOPA, Y) residues in the mussel foot proteins (Mfps) has been highlighted. However, the complex topological relationship of DOPA and Lys as well as the interfacial adhesive roles of other amino acids have been understudied. Herein, we study adhesion of Lys and DOPA-containing peptides to organic and inorganic substrates using single-molecule force spectroscopy (SMFS). We show that a modest increase in peptide length, from KY to (KY)3, increases adhesion strength to TiO2. Surprisingly, further increase in peptide length offers no additional benefit. Additionally, comparison of adhesion of dipeptides containing Lys and either DOPA (KY) or phenylalanine (KF) shows that DOPA is stronger and more versatile. We furthermore demonstrate that incorporating a nonadhesive spacer between (KY) repeats can mimic the hidden length in the Mfp and act as an effective strategy to dissipate energy.
Universal and Stable Slippery Coatings: Chemical Combination Induced Adhesive-Lubricant Cooperation

Abstract:Liquid lubricant of low affinity makes slippery coatings widely used in lubricating, anti-biofouling, anti-icing, fluid guiding, and drag reduction. Two critical challenges, however, remain in the practical application of slippery coatings consisting of liquid lubricants: (1) universality regardless of roughness and chemical composition of substrates, (2) stability of surface lubricity against evaporation. Herein, a chemical method is reported to create a universal and stable slippery lubricant-adhesive cooperated coating (SLACC) through acid catalyzed dehydration reaction between the phenolic hydroxyl of polydopamine (PDA), with universal (for challenge-1) and strong (for challenge-2) adhesion properties, and liquid-like polydimethylsiloxane (PDMS), with lubricant properties. Through overlying PDMS on PDA, a spatial gradient interpenetration of chemical combined PDA and PDMS leaving lubricant PDMS at the outermost of coating is achieved. This structure contributes to the following performances of SLACC: nearly universality suitable for 100 different abiotic or biotic substrates and stability sustainable for long-term usages, UV radiating, refrigerating, hot air drying, freeze drying, knife scratch and abrasion. This proposed strategy is envisioned anti-fouling from plane to tube and exhibits drag reduction in confined space.
Adaptive synergy between catechol and lysine promotes wet adhesion by surface salt displacement

Abstract:Keeping it sticky when wet Some biological molecules are remarkably sticky, even to surfaces submerged in water. Mussel adhesion, for example, is based on the overproduction of dihydroxyphenylalanine (DOPA) and proteins with a high abundance of cationic amine residues such as lysine. Using bacterial iron chelators consisting of paired DOPA and lysine groups as analogs for the mussel proteins, Maier et al. show that these two functional groups synergistically enhance interfacial adhesion (see the Perspective by Wilker). Abstract:Mussels can strongly adhere to hydrophilic minerals in sea habitats by secreting adhesive proteins. The adhesion ability of these proteins is often attributed to the presence of Dopa derived from posttranslational modification of Tyr, whereas the contribution of structural feature is overlooked. It remains largely unknown how adhesive proteins overcome the surface-bound water layer to establish underwater adhesion. Here, we use molecular dynamics simulations to probe the conformations of adhesive protein Pvfp-5. and its salt-tolerant underwater adhesion on superhydrophilic mica. Dopa and positively charged basic residues form pairs, in this intrinsically disordered protein, and these residue pairs can lead to firm surface binding. Our simulations further suggest that the unmodified Tyr shows similar functions on surface adhesion by forming pairing structure with a positively charged residue. We confirm the presence of these residue pairs and verify the strong binding ability of unmodified proteins using nuclear magnetic resonance spectroscopy and lap shear tests.

Abstract:Marine mussels are able to firmly affix on various wet surfaces by the overproduction of special mussel foot proteins (mfps). Abundant fundamental studies have been conducted to understand the molecular basis of mussel adhesion, where the catecholic amino acid, l-3,4-dihydroxyphenylalanine (DOPA) has been found to play the major role. These studies continue to inspire the engineering of novel adhesives and coatings with improved underwater performances. Despite the fact that the recent advances of adhesives and coatings inspired by mussel adhesive proteins have been intensively reviewed in literature, the fundamental biochemical and biophysical studies on the origin of the strong and versatile wet adhesion have not been fully covered. In this review, we show how the force measurements at the molecular level by surface force apparatus (SFA) and single molecule atomic force microscopy (AFM) can be used to reveal the direct link between DOPA and the wet adhesion strength of mussel proteins. We highlight a few important technical details that are critical to the successful experimental design. We also summarize many new insights going beyond DOPA adhesion, such as the surface environment and protein sequence dependent synergistic and cooperative binding. We also provide a perspective on a few uncharted but outstanding questions for future studies. A comprehensive understanding on mussel adhesion will be beneficial to the design of novel synthetic wet adhesives for various biomedical applications.

Abstract:Using AFM based single-molecule force spectroscopy, we studied the synergy between Dopa and lysine for wet adhesion on titania (TiO2) and mica surfaces. We found that the binding forces for lysine-Dopa dipeptides are significantly higher when the positive charge of lysine is unprotected on both surfaces. However, such a synergistic effect is absent when the sequence is reversed. We attribute such differential synergistic effects to their distinct structure for load distribution within the molecules, which may represent a general principle for synergistic strong adhesion.
A wet adhesion strategy via synergistic cation–π and hydrogen bonding interactions of antifouling zwitterions and mussel-inspired binding moieties

Abstract:3,4-Dihydroxyphenylalanine (Dopa) is a mussel-inspired, unique and versatile adhesive moiety, while zwitterions are well-known for their antifouling and repellent properties. It is still unclear whether zwitterionic surfaces could effectively prevent the adhesion of Dopa units, and a zwitterion-Dopa interaction mechanism may lead to development of a novel wet adhesion strategy. Here we report single-molecule force spectroscopy (SMFS) between Dopa and zwitterionic molecules of opposite dipole orientations, with a much stronger bond dissociation energy for zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC, similar to 19.4 k(B)T) than for [2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl)ammonium hydroxide (DMAPS, similar to 10.8 k(B)T). The strong MPC-Dopa interaction could be attributed to the synergy of cation-pi interaction and hydrogen bonding, but the DMAPS-Dopa interaction could only be attributed to hydrogen bonding, as further demonstrated by density functional theory (DFT) computation. Based on the orientation-mediated zwitterion-Dopa binding mechanism, we design a polymer containing both MPC and Dopa moieties as a promising underwater adhesive with a measured cohesion of 7.2-14.1 mJ m(-2), even stronger than reported cation-pi-facilitated underwater adhesives. The elucidated molecular interaction principles shed light on designing novel zwitterion-Dopa-mediated materials with wide applications in various fields such as tissue engineering scaffolds, cell-based screening and underwater adhesives.

Abstract:The glue proteins secreted by marine mussels bind strongly to virtually all inorganic and organic surfaces in aqueous environments in which most adhesives function poorly. Studies of these functionally unique proteins have revealed the presence of the unusual amino acid 3,4-dihydroxy- l -phenylalanine (dopa), which is formed by posttranslational modification of tyrosine. However, the detailed binding mechanisms of dopa remain unknown, and the chemical basis for mussels’ ability to adhere to both inorganic and organic surfaces has never been fully explained. Herein, we report a single-molecule study of the substrate and oxidation-dependent adhesive properties of dopa. Atomic force microscopy (AFM) measurements of a single dopa residue contacting a wet metal oxide surface reveal a surprisingly high strength yet fully reversible, noncovalent interaction. The magnitude of the bond dissociation energy as well as the inability to observe this interaction with tyrosine suggests that dopa is critical to adhesion and that the binding mechanism is not hydrogen bond formation. Oxidation of dopa, as occurs during curing of the secreted mussel glue, dramatically reduces the strength of the interaction to metal oxide but results in high strength irreversible covalent bond formation to an organic surface. A new picture of the interfacial adhesive role of dopa emerges from these studies, in which dopa exploits a remarkable combination of high strength and chemical multifunctionality to accomplish adhesion to substrates of widely varying composition from organic to metallic.

Abstract:Dopa (L-3,4-dihydroxyphenylalanine) is a key chemical signature of mussel adhesive proteins, but its susceptibility to oxidation has limited mechanistic investigations as well as practical translation to wet adhesion technology. To investigate peptidyl-Dopa oxidation, the highly diverse chemical environment of Dopa in mussel adhesive proteins was simplified to a peptidyl-Dopa analogue, N-acetyl-Dopa ethyl ester. On the basis of cyclic voltammetry and UV-vis spectroscopy, the Dopa oxidation product at neutral to alkaline pH was shown to be alpha,beta-dehydro-Dopa (Delta D), a vinylcatecholic tautomer of Dopa-quinone. Delta D exhibited an adsorption capacity on TiO2 20-fold higher than that of the Dopa homologue in the quartz crystal microbalance. Cyclic voltammetry confirmed the spontaneity of Delta D formation in mussel foot protein 3F at neutral pH that is coupled to a change in protein secondary structure from random coil to beta-sheet. A more complete characterization of Delta D reactivity adds a significant new perspective to mussel adhesive chemistry and the design of synthetic bioinspired adhesives.
Molecular Interaction Mechanisms Between Lubricant-Infused Slippery Surfaces and Mussel-Inspired Polydopamine Adhesive and DOPA Moiety

Abstract:Lubricant-infused slippery surfaces have recently emerged as promising antifouling coatings, showing potential against proteins, cells, and marine mussels. However, a comprehensive understanding of the molecular binding behaviors and interaction strength of foulants to these surfaces is lacking. In this work, mussel-inspired chemistry based on catechol-containing chemicals including 3,4-dihydroxyphenylalanine (DOPA) and polydopamine (PDA) is employed to investigate the antifouling performance and repellence mechanisms of fluorinated-based slippery surface, and the correlated interaction mechanisms are probed using atomic force microscopy (AFM). Intermolecular force measurements and deposition experiments between PDA and the surface reveal the ability of lubricant film to inhibit the contact of PDA particles with the substrate. Moreover, the binding mechanisms and bond dissociation energy between a single DOPA moiety and the lubricant-infused slippery surface are quantitatively investigated employing single-molecule force spectroscopy based on AFM (SM-AFM), which reveal that the infused lubricant layer can remarkably influence the dissociation forces and weaken the binding strength between DOPA and underneath per-fluorinated monolayer surface. This work provides new nanomechanical insights into the fundamental antifouling mechanisms of the lubricant-infused slippery surfaces against mussel-derived adhesive chemicals, with important implications for the design of lubricant-infused materials and other novel antifouling platforms for various bioengineering and engineering applications.

Abstract:Mussel foot proteins (Mfps) secreted in the byssal plaque of marine mussels are widely researched for their relevance to mussel adhesion in water. As the abundant residue in the amino acid sequences of major adhesive proteins, 3,4-dihydroxyphenylalanine (Dopa) or its catecholic moiety plays a key role in both Mfp binding to surface and cohesive cross-linking of Mfps in byssal plaques. The binding performance of an Mfp significantly depends on the content and redox state of Dopa, whereas the types of interaction vary in line with different surface chemistries and pH conditions. Thorough understanding of mussel adhesion from a molecular perspective is crucial to promote the application of synthetic mussel-bionic adhesives. This article presents a brief review of the research progress on the adhesion mechanisms of Mfps, which further emphasizes the contributions of Dopa-mediated interactions and considers other amino acids and factors. The involved inter- and intramolecular interactions are responsible for not only the diverse adhesion capacities of an adhesive byssal plaque as mussel’s adhesion precursor but also the formation and properties of the plaque structure.
Molecular Dynamics Simulations on the Adhesion of DOPA to Self-assembled Monolayers

Abstract:Mussels use a variety of 3,4-dihydroxy-phenylalanine(DOPA) rich proteins specifically tailored to adhering onto wet surfaces. To facilitate the development of next generation of aqueous adhesives, in this work, umbrella sampling and weighted histogram analysis method was used to calculate the binding free energy of DOPA on different self-assembled monolayers; steered molecular dynamics simulations were performed to study the desorption force of DOPA on them. Simulation results show that, the adhesion free energy of DOPA on the negatively charged COO--SAM surface is larger than that on the positively charged NH3+-SAM surface. DOPA is more easily adhered to the negative charged surfaces; on the charged surface, the higher adhesion free energy, indicated higher stability. Further analysis of the orientation distribution of DOPA on different surfaces reveals different interaction mechanisms between DOPA and different surfaces. It binds on the hydrophobic surface through benzene ring, while on the hydrophilic surface through hydroxyl. DOPA interacts with the negatively charged surface and the positively charged surface by amino and carboxyl groups, respectively. Comparison of desorption forces for DOPA on different self-assembled monolayers indicates that desorption forces on charged surfacesare greater than those on neutral surfaces, which is consistent with the trend of adhesion. Among the neutral SAMs, we also find that on the hydrophobic CH3-SAM, the desorption force is the maximum, indicating stronger adhesion stability. With the increase of hydrophobicity, both desorption force and adhesion stability increases. This work helps to understand adhesion mechanism of mussel on different surfaces, and provide theoretical insights for developing new underwater adhesives.
Ternary Synergy of Lys, Dopa, and Phe Results in Strong Cohesion of Peptide Films.

Abstract:Synergistic interactions between 3,4-dihydroxyphe-nylalanine (Dopa, Y*), cationic residues, and the aromatic rings have been recently highlighted as influential factors that enhance the underwater adhesion strength of mussel foot proteins and their derivatives. In this study, we report the first ever evidence of a cation-catechol-benzene ternary synergy between Y*, lysine (Lys, K), and phenylalanine (Phe, F) in adhesive peptides. We synthesized three hexapeptides containing a different combination of Y*, K, and F, i.e., (KY*)3, (KF)3, and (KY*F)2, respectively, exploring the relationship between the cohesive performance and molecular architecture of peptides. The peptide with the (KY*F)2 sequence displays the strongest underwater cohesion energy of 10.3 +/- 0.3 mJ m-2 from direct nanoscale surface force measurements. Combined with molecular dynamics simulation, we demonstrated that there are more bonding interactions (including cation-pi, pi-pi, and hydrogen bond interactions) in (KY*F)2 compared to the other two peptides. In addition, peptide (KY*F)2 still shows the strongest cohesive energies of 7.6 +/- 0.7 and 3.7 +/- 0.5 mJ m-2 in acidic and high-ionic strength environments, respectively, although the cohesive energy decreases compared to the value in pure water. Our results further explain the underwater cohesion mechanisms combining multiple interactions and offer insights on designing Dopa containing underwater adhesives.

Abstract:When it comes to bonding in an aqueous environment, intertidal marine organisms such as mussels, sandcastle worms, barnacles and polychaetes are providing inspiration for the assembly of synthetic molecules into the polymeric adhesives. Although intense demands in various life or industrial applications and many mussel-inspired systems have been shown to develop coatings and hydrogels, exploring the mimetic mussel adhesive system through the control of composition and polymer molecular weight is demonstrated less often. In this report, we showed a simple approach to synthesize mussel-inspired adhesive, P(N-butyl acrylate-co-l-Dopa-methacrylate anhydride), which possesses characteristics of simple synthesis, easy-using, no extra-crosslinker need, and excellent adhesion behaviour on diverse substrates. Adhesion behaviour was examined using low- and high-surface-energy substrates (e.g. PTFE and steel, respectively). Furthermore, the fabricated adhesives exhibited excellent underwater adhesive behaviour for steel in various aqueous solutions, including ion ((IO4)− and Mn3+), acid and alkali solutions (pH = 4, 6.7 and 8, respectively). The outstanding fabricated adhesives take advantage of the catechol functionality and wettability of the resulting polymer. These insights should help us to design next-generation underwater adhesive systems as well as open up a number of future research designs for mussel-inspired adhesives with performance not accessed previously.
Single Molecule Evidence for the Adaptive Binding of DOPA to Different Wet Surfaces.

Abstract:3,4-Dihydroxyphenylalanine (DOPA) is the noncanonical amino acid widely found in mussel holdfast proteins, which is proposed to be responsible for their strong wet adhesion. This feature has also inspired the successful development of a range of DOPA-containing synthetic polymers for wet adhesions and surface coating. Despite the increasing applications of DOPA in material science, the underlying mechanism of DOPA-wet surface interactions remains unclear. In this work, we studied DOPA-surface interactions one bond at a time using atomic force microscope (AFM) based single molecule force spectroscopy. With our recently developed "multiple fishhook" protocol, we were able to perform high-throughput quantification of the binding strength of DOPA to various types of surfaces for the first time. We found that the dissociation forces between DOPA and nine different types of organic and inorganic surfaces are all in the range of 60-90 pN at a pulling speed of 1000 nm s(-1), suggesting the strong and versatile binding capability of DOPA to different types of surfaces. Moreover, by constructing the free energy landscape for the rupture events, we revealed several distinct binding modes between DOPA and different surfaces, which are directly related to the chemistry nature of the surfaces. These results explain the molecular origin of the versatile binding ability of DOPA. Moreover, we could quantitatively predict the relationship between DOPA contents and the binding strength based on the measured rupture kinetics. These serve as the bases for the quantitative prediction of the relationship between DOPA contents and adhesion strength to different wet surfaces, which is important for the design of novel DOPA based materials.
Facile Synthesis of Catechol-Containing Polyacrylamide Copolymers: Synergistic Effects of Amine, Amide and Catechol Residues in Mussel-Inspired Adhesives

Abstract:The straightforward synthesis of polyamide-derived statistical copolymers with catechol, amine, amide and hydroxy residues via free radical polymerization is presented. In particular, catechol, amine and amide residues are present in natural mussel foot proteins, enabling strong underwater adhesion due to synergistic effects where cationic residues displace hydration and ion layers, followed by strong short-rang hydrogen bonding between the catechol or primary amides and SiO2 surfaces. The present study is aimed at investigating whether such synergistic effects also exist for statistical copolymer systems that lack the sequence-defined positioning of functional groups in mussel foot proteins. A series of copolymers is established and the adsorption in saline solutions on SiO2 is determined by quartz crystal microbalance measurements and ellipsometry. These studies confirm a synergy between cationic amine groups with catechol units and primary amide groups via an increased adsorptivity and increased polymer layer thicknesses. Therefore, the free radical polymerization of catechol, amine and amide monomers as shown here may lead to simplified mussel-inspired adhesives that can be prepared with the readily scalable methods required for large-scale applications.

Abstract:Significance Two popular perceptions about the much-mimicked adhesion of mussels are (i) the adhesion depends entirely on 3, 4-dihydroxyphenylalanine (Dopa) groups (benzene derivatives with two H-bonding prongs) and (ii) Dopa can stick to all surfaces. This study shows that both perceptions are incorrect: using three Dopa-containing mussel foot proteins (Mfps) on two chemically different self-assembled monolayers (SAM), we found the highest adhesion on the nonpolar (i.e., hydrophobic) SAMs was exhibited by the Mfp with the most hydrophobic side chains, not the most Dopa. Furthermore, increasing the spacing between the H-bond acceptors in the SAMs prevented the double-pronged H-bonding of Dopa side chains to polar SAMs. These findings clarify the roles of hydrophobic and hydrophilic interactions in both biological and nonbiological adhesion. The adhesion of mussel foot proteins (Mfps) to a variety of specially engineered mineral and metal oxide surfaces has previously been investigated extensively, but the relevance of these studies to adhesion in biological environments remains unknown. Most solid surfaces exposed to seawater or physiological fluids become fouled by organic conditioning films and biofilms within minutes. Understanding the binding mechanisms of Mfps to organic films with known chemical and physical properties therefore is of considerable theoretical and practical interest. Using self-assembled monolayers (SAMs) on atomically smooth gold substrates and the surface forces apparatus, we explored the force–distance profiles and adhesion energies of three different Mfps, Mfp-1, Mfp-3, and Mfp-5, on (i) hydrophobic methyl (CH3)- and (ii) hydrophilic alcohol (OH)-terminated SAM surfaces between pH 3 and pH 7.5. At acidic pH, all three Mfps adhered strongly to the CH3-terminated SAM surfaces via hydrophobic interactions (range of adhesive interaction energy = −4 to −9 mJ/m2) but only weakly to the OH-terminated SAM surfaces through H- bonding (adhesive interaction energy ≤ −0.5 mJ/m2). 3, 4-Dihydroxyphenylalanine (Dopa) residues in Mfps mediate binding to both SAM surface types but do so through different interactions: typical bidentate H-bonding by Dopa is frustrated by the longer spacing of OH-SAMs; in contrast, on CH3-SAMs, Dopa in synergy with other nonpolar residues partitions to the hydrophobic surface. Asymmetry in the distribution of hydrophobic residues in intrinsically unstructured proteins, the distortion of bond geometry between H-bonding surfaces, and the manipulation of physisorbed binding lifetimes represent important concepts for the design of adhesive and nonfouling surfaces.
Deposition and Adhesion of Polydopamine on the Surfaces of Varying Wettability.

Abstract:Mussel-inspired chemistry, particularly the versatile coating capability of polydopamine (PDA), has received much research interest as a promising strategy for fabricating functional coatings in numerous fields. However, the understanding of deposition mechanisms and adhesion behaviors of PDA on different substrates still remains incomplete, significantly limiting the related fundamental research and its practical applications. In this work, a colloidal probe atomic force spectroscopy technique was employed to quantify the interaction forces and adhesion between the PDA coatings and the substrate surfaces with different wettabilities. The surface force measurements and thermodynamic analysis of interaction energy indicate that the surface wettability has a significant influence on the adhesion, deposition behaviors, and morphologies of PDA coatings. Compared with the hydrophilic surfaces, the hydrophobic surfaces exhibit stronger adhesion with the PDA coatings. Furthermore, for the first time, this work demonstrates that ethanol has the capability of effectively displacing the trapped air/vapor layer or the so-called "hydrophobic depletion layer" on the hydrophobic substrate to allow the intimate contact between PDA and the substrate, thus enhancing the adhesion and facilitating the PDA deposition. This work provides new insights into the fundamental PDA deposition mechanism as well as the design and development of versatile mussel-inspired coatings on the substrates of varying hydrophobicity.

Abstract:Although the role of 3,4-dihydroxyphenyl-L-alanine(DOPA)in mussel foot proteins (mfps) in the realization of underwater bonding has been widely recognized, the role of the polarity of the polymer was largely overlooked. Here, by systematically comparing the underwater bonding properties of four mussel-inspired adhesives with different amide/lactam contents but similar catechol contents and molecular weights, we came to the conclusion that the polarity of the polymers also contributes to the strong underwater bonding. With the increase in the amide/lactam contents, the polarity of the polymeric adhesive increases, which correlates to the improved underwater bonding strength. A dielectric constant is introduced to evaluate the polarity of the polymer, which may be used as a guidance for the design of mussel-inspired adhesives with even better underwater bonding properties.

Abstract:The mussel-inspired catechol-based strategy has been well recognized as a promising alternative to design and exploit new generation adhesive materials applicable in many fields, ranging from biomedical adhesives to coatings of biomedical devices and engineering applications. However, in situ achievement of tough adhesion capability to substrate surfaces (e.g., minerals) is severely limited under the physiological environment or seawater condition (namely, relatively high salinity and mild alkalinity). In this work, a facile and versatile approach is proposed to in situ achieve robust wet adhesion in aqueous solutions of high salinity and mild alkalinity, via integrating primary amines into mussel-inspired polydopamine (PDA). By using a surface forces apparatus (SFA), the corresponding interaction behaviors have been systematically investigated. The strong wet adhesion was demonstrated and achieved via a synergetic effect of amine and PDA to the wet surfaces, including the surface salt displacement assisted by primary amine, strong adhesion to substrates facilitated by the catechol groups on PDA moieties, and enhanced cohesion through their cation-π interactions. Our results provide useful insights into the design and development of high-performance underwater adhesives and water-resistance materials.
