Abstract:A SERS active gold nanostar layer on the surface of ITO glass slip has been prepared by a low-cost electrostatically assisted APTES-functionalized surface-assembly method for SERS analysis. The two-dimensional morphology of the SERS substrate was examined by scanning electron microscopy. Comparative analysis revealed that the optical characteristics and SERS efficiency of these substrates varied as a function of nanostar morphology. It was found that the substrate assembled with the longest branches of nanostars generated the best SERS efficiency, whether the excitation source is 785 or 633 nm. The potential use of these substrates in detection applications was also investigated by using Nile blue A and Rhodamine 6G. The detection limits are 5 × 10(-11) M and 1 × 10(-9) M, respectively, when using the 785 nm excitation source. Apart from this high enhancement effect, the substrate here also shows extremely good reproducibility at the same time. All of these indicate that gold nanostars are a very good structure for SERS substrate assembly.
Facile synthesis of gold nanostars over a wide size range and their excellent surface enhanced Raman scattering and fluorescence quenching properties

Abstract:Surface enhanced Raman scattering (SERS) is an excellent technique for detecting the trace concentration of targets. However, SERS based detection of fluorescent targets is tricky as the fluorescence signal can severely interfere with the SERS signal upon resonance excitation. This can limit the trace detection capability of SERS. Herein, the authors demonstrate that Au nanostars (NSs) are excellent for fluorescence quenching and trace detection by SERS. The NS based SERS substrate was used to measure the R6G concentrations as low as 10 pM upon laser excitation that are in resonance with the fluorescence absorption. The authors used a simple seed-mediated and surfactant assisted method to synthesize AuNSs in a wide size range. The size of the NSs can be tuned from 55 nm to about 1 μm simply by varying the seed to HAuCl4 ratio in the growth solution.

Abstract:Gold nanoparticles have the potential to be used in biomedical applications from diagnostics to drug delivery. However, interactions of gold nanoparticles with different biomolecules in the cellular environment result in the formation of a “protein corona”—a layer of protein formed around a nanoparticle, which induces changes in the properties of nanoparticles. In this work we developed methods to reproducibly synthesize spheroidal and star-shaped gold nanoparticles, and carried out a physico-chemical characterization of synthesized anionic gold nanospheroids and gold nanostars through transmission electron microscopy (TEM), dynamic light scattering (DLS), zeta potential (ZP), nanoparticles tracking analysis (NTA), ultraviolet-visible (UV–Vis) spectroscopy and estimates of surface-enhanced Raman spectroscopy (SERS) signal enhancement ability. We analyzed how they interact with proteins after pre-incubation with bovine serum albumin (BSA) via UV–Vis, DLS, ZP, NTA, SERS, cryogenic TEM (cryo-TEM) and circular dichroism (CD) spectroscopy. The tests demonstrated that the protein adsorption on the particles’ surfaces was different for spheroidal and star shaped particles. In our experiments, star shaped particles limited the protein corona formation at SERS “hot spots”. This benefits the small-molecule sensing of nanostars in biological media. This work adds more understanding about protein corona formation on gold nanoparticles of different shapes in biological media, and therefore guides design of particles for studies in vitro and in vivo.

Abstract:This paper reports a simple, one-pot, template-free synthesis of flower-like Au nanoparticles (three-dimensional branched nanoparticles with more than 10 tips) with high yield and good size monodispersity at room temperature. The size of the Au nanoflowers could be tuned by controlling the composition of the starting reaction mixture. The key synthesis strategy was to use a common Good's buffer, HEPES, as a weak reducing and particle stabilizing agent to confine the growth of the Au nanocrystals in the special reaction region of limited ligand protection (LLP). Time-course measurements by UV-vis spectroscopy and TEM were used to follow the reaction progress and the evolution of the flower-like shape. The Au nanoflowers exhibited strong surface-enhanced effects which were utilized in the design of an efficient, stable, and nontoxic Raman-active tag for in vivo applications.

Abstract:Multi-branched gold nanostars with fractal feature were synthesized using the Triton X-100 participant seed-growth method. By increasing the amount of ascorbic acid, the branch length of gold nanostars could be greatly increased. It has been interesting to find that the secondary growth of new branches takes place from the elementary structure when the aspect ratio of the branches is greater than 8.0 and the corresponding plasmon absorption wavelength is greater than 900 nm. Raman activity of the gold nanostar films has been investigated by using the 4-mercaptobenzoic acid (4-MBA) as Raman active probe. Experimental results show that the surface-enhanced Raman scattering (SERS) ability of the gold nanostars could be efficiently improved when the fractal structure appears. The physical mechanism has been attributed to the intense increased secondary branch number and the increased “hot spots”. These unique multi-branched gold nanostars with fractal feature and great SERS activity should have great potential in sensing applications.

Abstract:We have developed a highly sensitive SERS-based sandwich immunoassay by utilizing two derivatives of gold nanostars simultaneously. One is densely packed self-assembled substrates of gold nanostars and the other is labeled immune nanostar aggregates. Taking advantage of the electromagnetic field coupling between tips of adjacent individual nanostars, the self-assembled substrate of gold nanostars exhibited a better SERS performance than that of gold nanoparticles. On the other hand, the immuno-aggregates made of gold stars also showed an improved SERS activity compared to those made of gold nanoparticles. Thus, by combining the self-assembled substrates of gold nanostars and immuno-aggregates of gold nanostars, highly sensitive sandwich immunoassays were obtained. The experimental results show that there existed a linear correlation between the concentration of antigens and the prominent peak intensity of SERS signals. The limit of detection (LOD) is ∼10 fg mL-1, 5 to 6 orders smaller than that of the conventional ELISA protocols. Such a structure will have potential wide-range applications in biological sensing and quantitative biochemical analysis.

Abstract:One of the main challenges for highly sensitive surface-enhanced Raman scattering (SERS) detection is the noise interference of fluorescence signals arising from the analyte molecules. Here we used three types of gold nanostars (GNSs) SERS probes treated by different surface modification methods to reveal the simultaneously existed Raman scattering enhancement and inhibiting fluorescence behaviors during the SERS detection process. As the distance between the metal nanostructures and the analyte molecules can be well controlled by these three surface modification methods, we demonstrated that the fluorescence signals can be either quenched or enhanced during the detection. We found that fluorescence quenching will occur when analyte molecules are closely contacted to the surface of GNSs, leading to a similar to 100 fold enhancement of the SERS sensitivity. An optimized Raman signal detection limit, as low as the level of 10(-11) M, were achieved when Rhodamine 6 G were used as the analyte. The presented fluorescence-free GNSs SERS substrates with plentiful hot spots and controllable surface plasmon resonance wavelengths, fabricated using a cost-effective self-assembling method, can be very competitive candidates for high-sensitive SERS applications.
Monodispersed mesoscopic star-shaped gold particles via silver-ion-assisted multi-directional growth for highly sensitive SERS-active substrates

Abstract:Surface-enhanced Raman scattering (SERS) exploits localized surface plasmon resonances in metallic nanostructures to significantly amplify Raman signals and perform ultrasensitive analyses. A critical factor for SERS-based analysis systems is the formation of numerous electromagnetic hot spots within the nanostructures, which represent regions with highly concentrated fields emerging from excited localized surface plasmons. These intense hotspot fields can amplify the Raman signal by several orders of magnitude, facilitating analyte detection at extremely low concentrations and highly sensitive molecular identification at the single-nanoparticle level. In this study, mesoscopic star-shaped gold particles (gold mesostars) were synthesized using a three-step seed-mediated growth approach coupled with the addition of silver ions. Our study confirms the successful synthesis of gold mesostars with numerous sharp tips via the multi-directional growth effect induced by the underpotential deposition of silver adatoms (AgUPD) onto the gold surfaces. The AgUPD process affects the nanocrystal growth kinetics of the noble metal and its morphological evolution, thereby leading to intricate nanostructures with high-index facets and protruding tips or branches. Mesoscopic gold particles with a distinctive star-like morphology featuring multiple sharp projections from the central core were synthesized by exploiting this phenomenon. Sharp tips of the gold mesostars facilitate intense localized electromagnetic fields, which result in strong SERS enhancements at the single-particle level. Electromagnetic fields can be further enhanced by interparticle hot spots in addition to the intraparticle local field enhancements when arranged in multilayered arrays on substrates, rendering these arrays as highly efficient SERS-active substrates with improved sensitivity. Evaluation using Raman-tagged analytes revealed a higher SERS signal intensity compared to that of individual mesostars because of interparticle hot spots enhancements. These substrates enabled analyte detection at a concentration of 10- 9 M, demonstrating their remarkable sensitivity for trace analysis applications.

Abstract:In the past decades, DNA-modified gold nanoparticles have attracted widespread attention due to their novel biochemical properties, such as the excellent biocompatibility, the programmable sequences, and the ability to cooperatively hybridize complementary DNA. Silver triangular nanoparticles (referred to as “nanoprisms” or “NPRs”) is a kind of typical anisotropic nanomaterials that can produce massive localized plasmonic field enhancement at their tips, which makes them one of the ideal SERS substrate. Unfortunately, it has previously been demonstrated that the Ag NPRs are extremely sensitive to the changes of ion concentration, and their surfaces, especially the tips, are easily oxidized, which makes it difficult to obtain stable DNA-modified Ag NPRs, hampering its biomedical applications.
High surface-enhanced Raman scattering performance of individual gold nanoflowers and their application in live cell imaging.

Abstract:Molecular imaging techniques based on surface-enhanced Raman scattering (SERS) face a lack of reproducibility and reliability, thus hampering its practical application. Flower-like gold nanoparticles have strong SERS enhancement performance due to having plenty of hot-spots on their surfaces, and this enhancement is not dependent on the aggregation of the particles. These features make this kind of particle an ideal SERS substrate to improve the reproducibility in SERS imaging. Here, the SERS properties of individual flower-like gold nanoparticles are systematically investigated. The measurements reveal that the enhancement of a single gold nanoparticle is independent of the polarization of the excitation laser with an enhancement factor as high as 108. After capping with Raman signal molecules and folic acid, the gold nanoflowers show strong Raman signal in the living cells, excellent targeting properties, and a high signal-to-noise ratio for SERS imaging.

Abstract:Anisotropic metallic nanoparticles (NPs) possess unique optical properties, which lend them to applications such as surface-enhanced Raman scattering (SERS). However, their preparation by an efficient, biocompatible and high yield synthetic method is still challenging. In this work, we demonstrate a simple and reproducible way to produce chitosan (CS) encapsulated gold-silver nanoflowers by sequentially adding chitosan, chloroauric acid, silver nitrate, and ascorbic acid to water at room temperature. This is a one-pot, seed-and surfactant-free synthetic method, which is simple and credible. CS is used to modulate the size of NPs, while AgNO3 is introduced to improve the monodispersity and homogeneity of NPs. Highly sensitive, spectrally and physically stable SERS tags are developed in virtue of the cooperative effect of CS and Ag+. Cresyl violet (CV) is applied as a Raman reporter to test the SERS property of NPs, and the results demonstrated that the nanoflowers exhibited stronger and more stable SERS signals than those of spherical gold nanoparticles. Importantly, after being modified by tumor cell-specific targeting ligands (folic acid), the sensitive and stable labeled nanoflowers are applied for cancer cell targeting and SERS imaging.
Cu2+-Assisted Synthesis of Ultrasharp and Sub-10 nm Gold Nanostars. Applications in Catalysis, Sensing, and Photothermia

Abstract:Gold nanostars have shown enormous potential as the main enablers of advanced applications ranging from biomedicine to sensing or catalysis. Their unique anisotropic structure featuring sharp spikes that grow from a central core offers enhanced optical capabilities and spectral tunability. Although several synthesis methods yield NSs of different morphologies and sizes up to several hundred nanometers, obtaining small NSs, while maintaining their plasmonic properties in the near-infrared, has proven challenging and elusive. Here, we show that Cu2+ addition during NS synthesis in polyvinylpyrrolidone/dimethylformamide generates more crystallographic defects and promotes the directional growth, giving rise to NSs with a larger number of much sharper spikes. They are also formed at smaller volumes, enabling the generation of ultrasmall nanostars, with a volume as small as 421 nm3 (i.e., 9.2 nm of volume-equivalent diameter), while maintaining a plasmon resonance in the near-infrared. To this end, we systematically evaluate the influence of synthesis parameters on the nanostar size and optical characteristics and demonstrate their properties for applications in catalysis, surface-enhanced Raman spectroscopy sensing, and hyperthermia. The ultrasmall nanostars show excellent attributes in all of them, leveraging their small size to enhance properties related to a higher surface-to-volume ratio or colloidal diffusivity.
Study on the roughen process of branches of AuAg nanostars for the improved Surface-enhanced Raman Scattering (SERS) to detect crystal violet in fish

Abstract:The techniques used to precisely tune the morphologies of noble metal nanoparticles are of vital importance as complex three-dimensional nanostructures can generate many "hot spots" for Surface-enhanced Raman Scattering (SERS). Especially, the SERS properties of star-shaped nanoparticles are closely related to the position, number and morphology of branches, making the control over the final morphology of them more important. By adding different reducing agents during the growth process of AuAg nanostars, we accurately controlled the morphology of branches, and synthesized three kinds of hyperbranched star-shaped nanostructures: Lamellar nanostars, Sea urchin-shaped nanostars and Serrated nanostars. The SERS performance of the hyperbranched nanostars were all stronger than that of common AuAg nanostars. Among them, Lamellar nanostars gained the best SERS enhanced factor about 1×10 7 due to the built-in nanogaps and sharp edges. The SERS detection probes prepared with purified Lamellar nanostars had high uniformity and stability, and showed excellent sensitivity in detecting crystal violet in fish, with detection range of 0.01-10 nM and detection limit of 0.015 nM. The results of this work not only provide guidance for the construction of complex Au and Ag bimetallic nanostructures, but also supply a superior material to develop highly sensitive optical chemical sensors and biosensors.

Electrochemical seeded growth of multiple shapes of gold nanostructures and their application as SERS substrates

Abstract:Surface-enhanced Raman scattering (SERS) is an optical detection method which has the advantages of high specificity and sensitivity. Gold nanostructures are always used to trace detection of chemical molecules because they can produce SERS effect and they are Biocompatible. Our researches used electrochemical and seeded methods to control the growth of nanostructures in two steps. The first step was the nucleation and seed formation of gold on the indium tin oxide (ITO) surface. The second step is the situ growth of gold nanostructures based on seeds. Regulating related parameters of two steps respectively, gold nanostructures with multiple shapes could be synthesized controllably. We used Nile blue A (NBA) as signal molecules to detect the SERS-activity of these gold nanostructures with multiple shapes respectively. The results showed that the gold nanostructures were stable during six months, well reproducible and the SERS-activity of nanostructures depended on the intensity and density of "hot spots". The gold nanoparticles array containing high intensity and density of hot spots has the strongest SERS-activity. This method is simple, easily controlled, inexpensive, environment friendly and does not need any templates, surfactants or coating agent. It has great potential to be used for large scale synthesis and application of electrochemical and SERS detection in biomedical field. © 2013 Springer-Verlag.
Synthesis of Lipophilic Gold Nanorod Superparticles and Their Size-Dependent SERS Performances

Abstract:Gold nanorods are widely used in Surface-Enhanced Raman Scattering (SERS) applications due to their strong localized surface plasmon resonance (LSPR). The self-assembly of gold nanorods further expands their applications and introduces new ensemble properties. In this paper, two types of gold nanorods with similar lengths but significantly different diameters were used for making superparticles. The gold nanorods were first surface-modified with thiol-terminated polystyrene (PS-SH), and then assembled into superparticles through emulsion volatilization. To test the SERS performances of the gold nanorod superparticles, nile red was used as a dye molecule to test the localization of the SERS performance of the single superparticles. It was found that as the size of the superparticles increased, the SERS performances also improved, with the final Raman signal intensity reaching up to 2*106. When the size of the superparticles was the same, the SERS performance was stronger for superparticles composed of gold nanorods with larger diameters. Additionally, a structure similar to the superparticles was designed for FDTD simulations, and the simulated results were highly consistent with the experimental results, further supporting our conclusions.

Abstract:In this work, gold nanostars (AuNSs) with size around 90 nm were prepared through an easy one-step method. They show excellent catalytic activity and large surface-enhanced Raman scattering (SERS) activity at the same time. Surprisingly, they exhibited different catalytic performance on the reduction of aromatic nitro compounds with different substituents on the para position. To understand such a difference, the SERS spectra were recorded, showing that the molecular orientation of reactants on the gold surface were different. We anticipate that this research will help to understand the relationship of the molecular orientation with the catalytic activity of gold nanoparticles. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:In this work, the novel tetrapod gold nanostars with four specific numbers of sharp tips were synthesized with good uniformity, high yield and chemical stability. The size of tetrapod gold nanostars has been increased effectively by increasing the amount of HAuCl4 or DEA added to the reaction solution, and the corresponding in-plane localized surface plasmon resonance (LSPR) band can shift from 700 to 1100 nm. In order to combine the advantages of different metals, the Ag-coated tetrapod gold nanostars were prepared with different Ag coating thicknesses by increasing the volume of AgNO3. With the increase of Ag coating thickness, the morphology of the nanoparticles has changed from gold nanostars with four sharp tips to bimetallic cuboids with four sharp tips and finally to crore-shell cuboids completely. The surface-enhanced Raman scattering (SERS) activity of Ag-coated tetrapod gold nanostars has also been improved with the different Ag coating thicknesses. Combined the tip effect and the enhancement effect of Ag, the SERS activity has the strongest enhancement when the four sharp tips of tetrapod gold nanostars are not completely coated by the Ag layer but still exposed to the outsides. For tetrapod gold nanostars with larger size to grow Ag-coated tetrapod gold nanostars, the bimetallic cuboids with four sharp tips nanostructure have the biggest SERS enhancement factor (EF) of 1.73 × 106 for Rhodamine 6G (R6G), making it an ideal candidate in SERS-based sensing application.
Preparation And Raman Enhancement Properties Of Gold Nanostars

Abstract:Gold nanostars (GNSs) have a series of sharp tips structures, which will produce strong hot spots and have great application potential in Raman enhancement. In this paper, muti-tip GNSs have been prepared experimentally, and the control techniques of their tip and size have been mastered. For the first time, a fast and efficient self-assembly technique without additives has been developed, and a series of Surface Enhanced Raman Scattering (SERS) substrates have been successfully prepared by using this technique. The effect of different GNSs density of substrates on SERS signal is further studied experimentally. The results show that the SERS signal is closely related to the density of particles in the substrate. The higher density of GNSs in the substrate, the more hot spots covered by the incident light plate, and the greater contribution to the SERS signal.

Abstract:This work presents the synthesis and characterization of Au nanostars (AuNSs) and demonstrates their application as surface enhanced Raman scattering (SERS)-activity tags for cellular imaging and sensing. Nile blue A (NBA) and bovine serum albumin (BSA) were used as Raman reporter molecules and capping materials, respectively. The SERS-activity tags were tested on human lung adenocarcinoma cell (A549) and alveolar type II cell (AT II) and found to present a low level of cytotoxicity and high chemical stability. These SERS-activity tags not only can be applied in multiplexed cellular imaging, including dark field imaging, transmission electron microscopy (TEM) and SERS imaging, but also can be used for cellular sensing. The SERS spectra clearly identified cellular important components such as proteins, nucleic acids, lipids, and carbohydrates. This study also shows that endocytosis is the main channel of tags internalized in cells. The AuNSs exhibiting strong surface enhanced Raman effects are utilized in the design of an efficient, stable SERS-activity tag for intracellular applications.

Abstract:Gold nanostars, possessing multiple sharp spikes, have emerged as promising plasmonic particles in the field of ultrasensitive sensing. We have developed a water-based method for high-yield synthesis of size-tunable anisotropic gold nanoparticles with a varying number of spiky surface protrusions, and performed systematic experimental and theoretical analyses of the optical properties of the single gold nanostars by characterizing them simultaneously with scanning electron microscopy and dark-field scattering spectroscopy. The morphologies and corresponding scattering spectra of the individual gold nanostars have been compared with electromagnetic simulations of the plasmonic resonances utilizing the finite-difference time-domain (FDTD) method. The study provides a correlation between the experimental and calculated scattering spectra and charge distributions of the different plasmon modes in the individual gold nanostars with varying numbers and relative orientations of surface protrusions. Our results provide guidelines for choosing gold nanostars with a proper number of spikes and appropriate dimensions of the core and arms for particular plasmonic applications as well as for further developing preparation methods of multispiked metal nanoparticles.
