Abstract:Abstract Background : Whole ovarian transplantation has the potential to restore fertility in cancer patients, but ovarian ischemia-reperfusion injury following transplantation causes decreased graft function. Melatonin protects against antioxidant damage and has anti-inflammatory effects, but its effects in whole ovarian transplantation have not been investigated. Objective :This study was aimed to verify the beneficial antioxidant and anti-inflammatory effects of melatonin in whole ovarian transplantation. Methods : The cryopreserved whole ovaries were allotransplanted in LEWIS rats. Forty rats were randomly divided into 8 groups: control group, sham surgery group, saline group; low-dose (25 mg/kg) melatonin group; high-dose (50 mg/kg) melatonin group; melatonin (50 mg/kg) + ML385 group; melatonin (50 mg/kg) + luzindole group, and melatonin+ 4P-PDOT group. The estrous cycle recovery was evaluated by vaginal exfoliative cell monitoring and serum hormone. Follicle morphology was observed by HE. The levels of eoxidative stress factors, antioxidant factors, and inflammatory factors in both serum and ovarian tissues were measured by ELISA, RT-qPCR, western blot and fenton detection. RT-qPCR, western blot and immunofluorescence assays were used to measure the levels of MT1 and Nrf2. Results : The rats in high-dose and low-dose melatonin groups recovered estrous cycle faster and lost fewer follicles, and the serum endocrine hormone levels were close to normal. The serum and ovarian tissue antioxidant capacity were significant higher, while the levels of inflammatory factors were significant lower in the high-dose and low-dose melatonin groups. In addition, the melatonin receptor MT1 was found to be involved in antioxidant and anti-inflammatory processes. Melatonin also triggered the Nrf2/ARE pathway activity via receptor MT1. Blocking Nrf2 or MT1 receptors could eliminate the beneficial effects of melatonin on whole transplanted ovaries. These findings suggest that melatonin can attenuate oxidative stress injury and inflammatory responses in whole transplanted ovaries via the MT1/Nrf2/ARE signaling pathway, thereby effectively protecting whole transplanted ovarian function.

Abstract:The progress achieved in anticancer therapy in recent years has been paralleled by an increase in the survival of women with cancer globally. Nonetheless, the gonadotoxic impact of anticancer drugs has led to ovarian failure in treated women. While there are documented cases of successful ovarian tissue transplants resulting in restored fertility and childbirth, challenges persist, including suboptimal functional recovery and limited graft lifespan. Melatonin, an inert hormone primarily secreted by the mammalian pineal gland, exhibits diverse physiological functions, including antioxidative, anti-inflammatory, anti-apoptotic, and angiogenesis-regulating properties. Consequently, researchers have explored melatonin as a modulator to enhance graft function recovery in ovarian transplantation experiments, yielding promising outcomes. This review examines the relevant literature, consolidating findings that underscore the positive effects of melatonin in safeguarding the morphology and structure of transplanted ovarian tissues, facilitating graft function recovery, and extending lifespan. The amassed evidence supports the consideration of melatonin as a prospective protective agent for human ovarian tissue transplantation in the future.

Transplantation of intact rat gonads using vascular anastomosis: effects of cryopreservation, ischaemia and genotype

Abstract:Background: A limited store of ovarian follicles is present at birth and its progressive decline during ageing is hastened by alkylating agents and ionizing radiation during treatment for cancer or autoimmune disease. Oligo- or azoospermia can arise for similar reasons in men. There is some experimental evidence showing that targeted gene deletion or drugs to produce hypogonadotrophism can protect germ cells from wastage. Another strategy for conserving fertility is to cryopreserve ovarian or testicular tissue for subsequent transplantation. To maximize gonadal function, it is desirable to preserve whole gonads for transplantation using vascular anastomosis. Methods and results: We investigated this strategy in the rat model. All freshly isotransplanted ovaries (n = 8) survived and resumed follicle growth and secretion and, although ischaemia for 24 h at 4 degrees C did not disrupt ovarian function, the organs had fewer follicles. Four out of seven (57%) cryopreserved transplants survived for > or =60 days, were ovulatory and one pregnancy was established, but the ovarian reserve was compromised by fewer follicles. Ovarian allotransplants were vigorously rejected, even with moderate immunosuppression using cyclosporin A. On the other hand, only three out of seven (42%) fresh testicular isotransplants had active spermatogenesis, and none of the cryopreserved testes was functional. Conclusions: The effects of gonadectomy in rats can be reversed by isotransplants, but the results are more successful with ovaries than testes, and allotransplants were never successful. Intact cryopreserved ovaries can be restored to function after transplantation with vascular anastomoses.
Melatonin activates Nrf2/HO-1 signalling pathway to antagonizes oxidative stress-induced injury via melatonin receptor 1 (MT1) in cryopreserved mice ovarian tissue

Abstract:Our previous research has shown that melatonin (MLT) can reduce cryopreserved ovarian damage in mice. Yet, the molecular mechanism of MLT protection is still unclear. Some studies have shown that melatonin receptor 1 (MT1) is very important for animal reproductive system. To evaluate whether MLT exerts its protective effect on cryopreserved mice ovarian tissue via MT1, we added antagonist of MT1/MT2 (Luzindor) or antagonist of MT2 (4P-PDOT) to the freezing solution, followed by cryopreservation and thawing of ovarian tissue. The levels of total superoxide dismutase (T-SOD), catalase (CAT), nitric oxide (NO) and malondialdehyde (MDA) were detected. Besides, by using RT-PCR and Western blotting, the expression of Bcl-2, Bax and Nrf2/HO-1 signalling pathway-related proteins was detected. These findings demonstrated that compared with the melatonin group, the addition of Luzindor increased apoptosis, NO and MDA activities, decreased CAT and T-SOD activities and inhibited Nrf2/HO-1 signalling pathway. In conclusion, melatonin can play a protective role in cryopreserved ovarian tissue of mice through MT1 receptor.

Abstract:Human ovarian tissue xenotransplantation emerges as widely used experimental animal model to investigate the various aspects of follicle development and the effect of other related interventions of fertility preservations such as cryopreservation.There are many factors influence the effect of xenotransplantation,such as animal recipiem,ovarian tissue preparation,vascular supply after transplantation and the use of gonadotropms.Although there is some evidence that this procedure might be effective m preservation of future female fertility and restoring ovarian endocrine and reproductive function,it should only be performed under experimental conditions,until the technique is found to be safe and ethical.This article reviews the factors affecting the xenotransplantation of human ovarian tissue and the recent progress in this paper.
Assessment of apoptosis and oxidative stress in cryopreserved ovary after grafting in fibrin-alginate scaffold with endothelial cells and melatonin in wistar rats

Abstract:Objective: Infertility is a significant public health concern affecting 10-15% of couples. Young women undergoing gonadotoxic treatment are at higher risk of ovarian dysfunction and infertility. To mitigate this risk, ovarian tissue freezing and transplantation have been developed as a novel strategy. However, challenges such as follicular loss and dysfunction during the freezing process, and ovarian damage during transplantation, persist. This study aimed to investigate the potential of using appropriate antifreeze, antioxidant, wound healing, and biological hydrogels to reduce these injuries. Specifically, the effect of fibrin scaffold with endothelial cells and melatonin on apoptotic gene expression and antioxidants in cryopreserved ovaries after transplantation was examined. Methods: A total of 36 adult female wistar rats) 6-8-week-old and weighing from 200 to 220 g) were divided equally into six groups (n = 6): 1) control group (C), 2) transplanted ovarian tissue after vitrification and thawing process (Group 1), 3) transplanted vitrified/thawed ovarian tissue while encapsulated in Fib/Alg hydrogel (Group 2), 4) transplanted vitrified/thawed ovarian tissue while encapsulated in Fib/Alg hydrogel in addition with melatonin (Group 3), 5) transplanted vitrified/thawed ovarian tissue while encapsulated in Fib/Alg hydrogel in addition with endothelial cells (Group 4) and 6) transplanted vitrified/thawed ovarian tissue while encapsulated in Fib/Alg hydrogel in addition with melatonin endothelial cells (Group 5). The ovaries were auto-transplanted in the rats' lumbar region. After 14 days, the ovaries were removed. Antioxidant levels (SOD, GPx, MDA, and TAC) were evaluated using ELISA, and apoptotic gene expressions (Bax/Bcl2 and caspase 3) were analyzed by real-time RT-PCR to determine apoptosis. Results: In the transplanted frozen ovary group, Bax/Bcl2 and caspase 3 gene expression increased significantly (P<0.05), while antioxidant levels (SOD, GPx, MDA, and TAC) decreased. The encapsulated frozen ovary group showed decreased gene expression and increased antioxidant levels. The ovary group encapsulated with fibrin scaffold, endothelial cells, and melatonin had the most significant decrease in gene expression and increase in antioxidant levels (P<0.05). Conclusion: Coordinated action of Fibrin-based scaffold with endothelial cells and melatonin could decrease apoptosis gene expression and increase antioxidant levels in cryopreserved ovaries after transplantation, providing valuable insights into preserving fertility in young women undergoing gonadotoxic treatment.
Melatonin Inhibits Oxidative Stress and Apoptosis in Cryopreserved Ovarian Tissues via Nrf2/HO-1 Signaling Pathway

Abstract:In the field of assisted reproductive technology, female fertility preservation, particularly ovarian tissue cryopreservation in adolescent cancer patients, has attracted much attention. Melatonin (MLT) is well known for its antioxidative and anti-apoptotic properties; however, whether it can ameliorate the cryoinjury and inhibit the generation of reactive oxygen species (ROS) in cryopreserved ovarian tissues (OTs) has not yet been reported. Here, we demonstrated that MLT could protect follicular integrity; prevent cell apoptosis; decrease ROS, malondialdehyde (MDA), and nitric oxide (NO) levels; and increase activities of glutathione peroxidases (GSH-Px), glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD) in cryopreserved OTs. Furthermore, these effects may be related with the activation of the nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway, as evidenced by increased mRNA levels of Nrf2 downstream genes, including heme oxygenase-1 (HO-1), glutathione S-transferase M1 (GSTM1), SOD, and CAT. In summary, MLT can not only directly scavenge ROS but also significantly induce the activation of antioxidative enzymes via the Nrf2 signaling pathway, which is a new mechanism underlying the protection effects of MLT on cryopreserved OTs.

Comparison of melatonin, oxytetracycline, and N-acetylcysteine pre-treatments in autologous intraperitoneal ovarian transplantation in rats

Abstract:This study was aimed at investigating the effects of melatonin, oxytetracycline and N-acetylcysteine on the ovarian follicle reserves and surface epithelium in autologous intraperitoneal ovarian transplantation in rats. Thirty adult female Wistar Albino were selected and randomly divided into six groups (n = 5). Group 1, which was the control group, only had their abdomens opened and closed while Group 2 underwent ovarian transplantation. Group 3, 4, 5 and 6 received 20 μg/kg/IM melatonin, 10 mg/kg/IM oxytetracycline, 150 mg/kg/IP N-Asetil sistein (NAC) and 1% ethanol respectively 15 min before the ovarian transplantation. Vaginal cytology was performed to monitor the estrus phase and the follicle reserve and changes in the surface epithelium were histopathologically evaluated during the preparations. Moreover, cellular apoptosis in tissues was evaluated with immunofluorescence staining of Bcl-2 and Bax. The Bax/Bcl-2 ratio was then calculated as the mean fluorescence intensity (MFI) of Bax and Bcl-2 MFI. Dysplastic change was found only significantly higher in the transplantation group (G2) (p < 0.01). Histopathologically, it was found that the follicle reserve was preserved significantly in the oxytetracycline and melatonin treated group (G3, G4) (p < 0.01). It was also observed that the oxytetracycline treated group (G4) were able to show better preventive effects against dysplastic changes of the surface epithelium. Moreover, the melatonin treated group depicted a low Bax/Bcl-2 ratio compared to the group that only underwent transplantation (G2) (p < 0.01). This study indicated that oxytetracycline and melatonin might be more effective than N-acetylcysteine in protecting against oxidative stress during ovarian transplantation.
Melatonin Induction of HSP90 Expression Exerts Cryoprotective Effect on Ovarian Tissue

Abstract:Our previous study revealed that melatonin (MLT) protected the quality of cryopreserved ovarian tissues in mice. This work was carried out to examine the role of MLT in inducing HSP90 expression of ovarian tissue for achieving cryoprotection. Pieces of ovarian tissues were obtained from 50 female rats treated with MLT at 0, 0.001, 0.01, 0.1, and 1 mM, respectively. After cryopreservation-thawing, HSP90 mRNA and protein level were evaluated using qRT-PCR and western blot. The qRT-PCR results revealed that HSP90 mRNA expression was significantly (p < 0.01) upregulated in MLT-treated groups in comparison with the controls (0 mM). Western blot revealed higher HSP90 protein expression in MLT-treated groups compared to control group (0 mM), thus further confirming that MLT positively affected HSP90 expression. Moreover, 0.1 mM MLT had better effects than other concentrations of MLT. Conclusively, findings in the present work provide a feasible technology for improving cryopreserved ovarian tissue quality through the addition of MLT to elicit HSP90 expression.
Successful fertility following optimized perfusion and cryopreservation of whole ovary and allotransplantation in a premature ovarian insufficiency rat model

Abstract:Background Fertility preservation by whole ovary cryopreservation and transplantation (WOCP&TP) with vascular anastomosis requires successful cryopreservation. In this study, we investigated the possibility of restoring ovarian function and natural fertility after WOCP&TP in a premature ovarian insufficiency (POI) rat model. The influence of cryopreservation on the offspring of rats following WOCP&TP was also explored. Method Rats aged 8-10 weeks were used as donors and recipients for allotransplantation. Fifteen rat whole ovaries were divided into three groups: the optimized group, the conventional group, and the fresh group. Different perfusion modes were used before cryopreservation and after thawing. Whole ovaries were observed by morphologic analysis, immunohistochemical staining, and transferase-mediated deoxyuridine triphosphate nick end-labeling assay. Ovarian function and fertility after WOCP&TP were then observed in 25 cyclophosphamide-induced POI rats for 8 months. Ovarian function was assessed by vaginal smears and blood hormone levels. Fertility restoration was quantified as the live birth rate after mating. The filial generation of rats was mated at 8-10 weeks of age. Offspring were observed for birth defect. Results Histological evaluation demonstrated intact morphology of follicles in all groups, with 77.6% of the total number of follicles identified as intact in the optimized group. The apoptotic rates of ovarian cells in the optimized group were significantly lower than that in the conventional group. Of the 20 live POI rats, 14 (70%) began to recover ovarian function after 2 weeks of transplantation, with normal hormone levels achieved 4 weeks after transplantation. Four of 14 rats were pregnant and delivered live offspring. One rat had a second pregnancy and delivered a second litter of live offspring. When the offspring matured, they were mated, and second and third generations of rats were born. All offspring had no abnormalities in appearance. Conclusions High rates of restoration of ovarian function and natural fertility with multiple generations of offspring were obtained following WOCP&TP in a cyclophosphamide-induced POI rat model by utilizing optimized perfusion. Cryopreservation did not affect the viability of successive generations.
Effects of Two Cryoprotectants on Mouse Ovarian Tissue after Vitrification

Abstract:Objective: To observe the effects of two cryoprotectants on morphology and function of mouse ovarian tissues after vitrification. Methods: A total of 23 four-week old ICR female mice were randomly assigned to four groups: non-frozen (n=6), ovariectomized (n=5), EG40 (n=6) and ED20 (n=6). Ovarian tissues were vitrified with ethylene glycol (EG) or EG combined with dimethyl sulphoxide (DMSO) as cryoprotectants. Some of the vitrified tissues were transplanted under kidney capsules, the others were examined histologically after frozen and thawed. Estrus cycle was observed 5 days after ovarian autografting. The recovered autografts were collected and examined histologically one month after transplantation. Results: In the EG40, ED20 and non-frozen groups, the initiation rate of estrus were all 100%, but the initiation days of estrus were 10.2±1.2 d, 8.0±0.9 d and 6.8±1.0 d respectively. The initiation days of estrus in the EG40 group were significantly more than that in the non-frozen group (P<0.01). There was no significant difference between the ED20 and the non-frozen groups (P>0.05). The follicles at different development stages in autografts were normal morphology, but the number of follicles in vitrified ovarian tissues was less than that in non-frozen tissues. Conclusion: The vitrification with EG and DMSO as cryoprotectants could effectively preserve mouse ovarian tissues.
Effect of Exogenous Melatonin on the Development of Mice Ovarian Follicles and Follicular Angiogenesis

Abstract:In mammalian, the periodic growth and development of ovarian follicles constitutes the physiological basis of female estrus and ovulation. Concomitantly, follicular angiogenesis exerts a pivotal role in the growth of ovarian follicles. Melatonin (N-acetyl-5-methoxytryptamine, Mel), exists in follicle fluid, was suggested to affect the development of follicles and angiogenesis. This research was conducted to investigate the effects and mechanisms of Mel on the development of ovarian follicles and its angiogenesis. In total, 40 ICR mice at age of 3 weeks were allocated into four groups at liberty: control, Mel, FSH and FSH + Mel for a 12-day trial. Ovaries were collected at 8:00 a.m. on Day 13 for detecting the development of ovarian follicles and angiogenesis. Results indicated that Mel promoted the development of ovarian follicles of 50–250 μm (secondary follicles) and periphery angiogenesis, while FSH remarkably increased the number of antral follicles and periphery angiogenesis. Mechanically, Mel and FSH may regulate the expression of VEGF and antioxidant enzymes in different follicular stages. In conclusion, Mel primarily acted on the secondary follicles, while FSH mainly promoted the development of antral follicles. They both conduced to related periphery angiogenesis by increasing the expression of VEGF. These findings may provide new targets for the regulating of follicular development.
Protective Effects Of Melatonin Agonist On Radiotherapy-Induced Ovarian Damage In Rats

Abstract:Objective: To investigate whether melatonin agonist has protective effects on radiotherapy-induced ovarian damage in rats. Methods: Twenty-four SD rats were divided into 4 groups, receiving normal saline (n=6), 200 cGy radiation + normal saline (n=6), 200 cGy radiation + MT (50 mg/kg) (n=6) and 200 cGy radiation + MT (100 mg/kg) (n=6), respectively. All rats were decapitated at two weeks after radiotherapy. Hydroxyl free radical (center dot OH) levels in blood were detected as the indicator of free radical damage. Levels of serum follicle-stimulating hormone (FSH), estradiol (E2) and the number of follicles were measured as the indicators of ovarian reserve, mitochondrial membrane potential (Delta Psi), cytochrome C (cyt c) levels and caspase3 activity were assayed as indicators of mitochondrial pathway. Results: Radiotherapy caused a significant increase in the levels of center dot OH, FSH, cyt c and the activity of caspase3, meanwhile the level of E2, the number of follicles and.. decrease in the Rad group (P<0.05); however in Rad + MT group, these changes were abrogated (P<0.05). Conclusion: melatonin can diminish radiotherapy-induced ovarian damage in female rats, it may be related to the mechanism that MT inhibit mitochondrial pathway induced by radiation.

Abstract:Sperm cells can be damaged during the semen cryopreservation process, decreasing their fertilizing ability. Physical damage and oxidative stress may occur during the freeze-thawing process. Antioxidants such as the native antioxidant melatonin can potentially improve cryopreservation outcomes. In this study, we added melatonin to cryoprotectant to examine its effect on frozen-thawed human sperm. We found that adding 0.1mM melatonin to cryoprotectant significantly increased sperm viability (24.80 ± 0.46% vs. 20.97 ± 1.27%, P < 0.05) and membrane integrity (P < 0.05), and decreased intracellular reactive oxygen species and lipid peroxidation damage. Furthermore, mRNA levels of the transcription factor NF-E2-related factor-2 and its downstream genes were significantly increased. Resistance to oxidative stress was enhanced and expression of the antiapoptotic gene Bcl-2 was increased by inclusion of 0.1mM melatonin in the cryoprotectant. Moreover, 0.1mM melatonin upregulated the expression of heat shock protein 90 (HSP90), which confers resistance to stressors in frozen-thawed sperm. Results obtained upon addition of inhibitors of melatonin receptors (luzindole and 4-P-PDOT) and an HSP90 inhibitor (geldanamycin) in the cryoprotectant demonstrated that melatonin promoted HSP90 translation via the melatonin receptor MT1 and increased adenosine triphosphate levels, thus increasing the viability of thawed sperm.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Research question</h3><p>Whether melatonin can provide non-invasive ovarian protection against damage caused by cis-diamminedichloroplatinum (cisplatin) and preserve fertility for female cancer patients? And what is the possible mechanism if it is the case?</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Design</h3><p>Athymic BALB/c-nu nude tumor-bearing female mice were employed to exclude the antagonistic effect towards cisplatin-based anticancer therapy and promoting cancer effect of melatonin. Sexually mature and newborn C57BL/6 female mice were used to evaluate the potential effects of melatonin towards ovarian follicle pool, pregnant rate and litter number for cisplatin-treated mice and their ovaries were used to perform Immunohistochemistry, TUNEL and gene array to explore the underlying mechanism. We also isolated granulosa cells to confirm the potential protective mechanism of melatonin</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>Not only could melatonin enhance the anti-cancer effect of cisplatin in tumor-bearing nude mice, but also it could reduce ovarian toxicity and preserved long-term fertility in cisplatin treated C57BL/6 female mice. Mechanistically, when co-administrated, melatonin could reduce DNA damage and relieve toxicity of lipid peroxidation in ovary caused by cisplatin. Specially, melatonin could largely removelipid peroxidation in granulosa cells and thus prevent ovarian follicles from being depleted.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3><p>Melatonin has potential to be used as a chemotherapeutic adjuvant to improve anti-cancer outcome and to preserve ovarian function for cisplatin chemotherapy simultaneously. Notably, its DNA protection and antioxidant effects on follicles may benefit female cancer survivors and prevent premature ovarian failure (POF) as well as fertility loss caused by chemotherapy.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">KEY MESSAGE</h3><p>Ovarian reserve preservation and its mechanism of melatonin against cisplatin-induced ovarian damage have been explored. The better control of tumor and better ovarian reserve and fertility of female mice when melatonin was co-administrated with cisplatin indicates melatonin may be a promising fertoprotective agent for cisplatin-treated female cancer patients.</p>

Abstract:Melatonin plays pivotal roles in controlling photoperiod-related circadian rhythm and regulating the functions of diverse target tissues as a hormone or an antioxidant agent. However, there is little evidence to demonstrate that melatonin is able to regulate the early ovarian development in the chicken. Here we investigated effects of melatonin on the developmental process from ovarian germ cells to primordial follicles in the chicken. Melatonin was administered from the Day 12 of the embryos till Day 6 of the chicks at 1-100 pg/day. Treatment with melatonin (100 pg/day) induced a decrease of germ cell cysts and follicles number, and the ovarian cortex thickness. In addition, melatonin suppressed the ovarian cells proliferation that was demonstrated by decreased proliferation cell nuclear antigen expression and germ cell marker Dazl protein. Furthermore, melatonin increased the expression of hypothalamic gonadotropin-inhibitory hormone (GnIH) mRNA, but decreased the expression of all mRNAs of hypothalamic gonadotropin-releasing hormone (GnRH) II, pituitary follicle stimulating hormone (FSH)-beta and ovarian FSH receptor, luteinizing hormone (LH) receptor, estrogen receptors alpha and beta, progesterone receptor, steroidogenesis enzymes Cyp19a1 and 3 beta HSDII. These data indicated that melatonin might inhibit the ovarian germ cells number via suppression of the hypothalamus-pituitary-ovarian axis in the chicken, subsequently resulting in the inhibition of primordial follicles formation. Effects of Cryopreservation on Mouse Ovarian Tissue after Vitrification

Abstract:Objective To observe the effects of cryopreservation on morphology of various stage follicles and estradiol secretion of mouse ovarian tissues after vitrification.Methods 23 four-week old ICR female mice were randomly assigned to three groups: non-frozen(n=8),ovariectomized(n=5) and vitrified(n=10). Ovarian tissues were vitrified with ethylene glycol(EG) combined with dimethyl sulphoxide(DMSO) as cryoprotectants.Some of the non-frozen and frozen tissues were examined histologically,the others were transplanted under mice kidney capsules.Estrus cycles were began to observe 5 days after ovarian transplantation,and mouse serum estradiol were detected 30 days after transplantation.Results The number of primordial follicles and that of primary follicles in vitrified tissues were not significant different from that in nonfrozen tissues respectively.However,the percent viability of secondary follicles were(71.76±7.90)% in frozen tissues,compared with(91.67±6.36)% in non-frozen tissues(P0.01).In the vitrified and non-frozen groups,the initiation rate of estrus were all 100%,and the initiation days of estrus were(7.6±1.1)days and(6.6±0.9)days respectively(P0.05).There was no significant difference in estradiol production of ovarian tissues 30 days after transplantation between the frozen and non-frozen groups.Conclusion Vitrified mouse ovarian tissue could maintain similar viability of primordial follicles and primary follicles and estradiol secretion with non-frozen ovarian tissue.

Abstract:Ovarian aging is characterized by gradual declines in oocyte quantity and quality. Melatonin is considered an anti-aging agent due to its cytoprotective actions as an antioxidant. This study examined whether long-term melatonin treatment would delay ovarian aging in mice. Female ICR mice (10 weeks old) were given melatonin-containing water (100 μg/mL; melatonin) or water only until 43 weeks of age. Their oocytes were recovered from the oviduct, and in vitro fertilization was performed. The ovaries were used for a histological analysis of the number of follicles. The mRNA expression of the aging-related sirtuin genes (SIRT1, SIRT3) and the autophagy-related gene (LC3) and the telomere length of the ovarian chromosomes were analyzed. Transcriptome changes in the ovaries were also characterized using microarray. The number of ovulated oocytes decreased with age; however, it was greater in melatonin-treated mice than that from control animals. The decreased fertilization rate and blastocyst rate during aging also were higher in the melatonin-treated mice than in the controls, as were the numbers of primordial, primary, and antral follicles. The mRNA expression of SIRT1 and LC3 and telomere length were enhanced due to melatonin treatment. Seventy-eight genes that were downregulated during aging and upregulated by melatonin were identified by a microarray analysis. Forty of these 78 genes were ribosome-related genes, and a free radical scavenging network was identified. The present results indicate that melatonin delays ovarian aging by multiple mechanisms including antioxidant action, maintaining telomeres, stimulating SIRT expression and ribosome function, and by reducing autophagy.
