Abstract:Biocatalytic reactions in living cells involve complex transformations in the spatially confined microenvironments. Inspired by biological transformation processes, we demonstrate effective biocatalytic cascade driven photodynamic therapy in tumor-bearing mice by the integration of an artificial enzyme (ultrasmall Au nanoparticles) with upconversion nanoparticles (NaYF4@NaYb0.92F4:Er0.08@NaYF4)zirconium/iron porphyrin metal-organic framework core-shell nanoparticles (UMOF NPs) which act as biocatalysts and nanoreactors. The construction of core-shell UMOF NPs are realized by using a unique "solvent-assisted self-assembly" method. The integration of ultrasmall AuNPs on the UMOFs matrix leads to glucose depletion, providing Au-mediated cancer therapy via glucose oxidase like catalytic activity. Meanwhile, the UMOF matrix acts as a near-infrared (NIR) light photon-activated singlet oxygen generator through a continuous supply of oxygen via hydrogen peroxide decomposition upon irradiation. Such kinds of biocatalysts offer exciting opportunities for biomedical, catalytical ,and energy applications.

Abstract:Two new porphyrin-based organic compounds (Por and ZnPor) were synthesized by introducing hydrophilic polyethylene glycol chains and pentafluorobenzene moieties onto the parent porphyrin structure. After self-assembling into nanoparticles, the absorption spectrum of (Zn)Por NPs broadened and red-shifted to some extent, relative to that of organic molecules. Meanwhile, the fluorescence of organic molecule nanoparticles was quenched significantly, which facilitated the nonradiative thermal generation for potential applications in photothermal cancer therapy. Por NPs and ZnPor NPs presented spherical structure with average diameter about 100 run, endowing them with tumor targeting properties based on the enhanced permeability and retention (EPR) effect. Due to the heavy atom effect, ZnPor NPs presented the higher efficiency of ROS generation than that of Por NPs. In contrast, Por NPs exhibited the better photothermal effect relative to that of ZnPor NPs under irradiation of a 635-nm laser. The photothermal conversion efficiency of Por NPs was calculated to be 16.34%. The in vitro experiments suggested that Por NPs and ZnPor NPs could enter tumor cells efficiently with good biocompatibility and exhibited high photocytotoxicity with IC50 of 7.3 mu g/mL and 3.0 mu g/mL, respectively. Thus, the as-prepared porphyrin nanomaterials can be used as potential photosensitizers for cancer photodynamic/photothermal synergistic therapy in vivo, benefiting from their good biocompatibility, strong near-infrared absorption, and high photodynamic and photothermal effects.

Abstract:Photosensitizers are light-sensitive molecules that are highly hydrophobic, which poses a challenge to their use for photodynamic therapy. Hence, considerable efforts have been made to develop carriers for the delivery of PSs. Herein, we synthesized a new theranostic nanoagent (CQDs@PtPor) through the electrostatic interaction between the tetraplatinated porphyrin complex (PtPor) and the negatively charged CQDs. The size and morphology of as-prepared CQDs and CQDs@PtPor were characterized by a series of methods, such as XRD, TEM, XPS, and FTIR spectroscopy. The CQDs@PtPor composite integrates the optical properties of CQDs and the anticancer function of porphyrin into a single unit. The spectral results suggested the effective resonance energy transfer from CQDs to PtPor in the CQDs@PtPor composite. Impressively, the CQDs@PtPor composite showed the stronger PDT effect than that of organic molecular PtPor, suggesting that CQDs@PtPor is advantageous over the conventional formulation, attributable to the enhanced efficiency of 1O2 production of PtPor by CQDs. Thus, this CQDs-based drug nanocarrier exhibited enhanced tumor-inhibition efficacy as well as low side effects in vitro, showing significant application potential in the cancer therapy.

Abstract:Phototherapy has emerged as a promising way to treat cancer with high selectivity and non-invasiveness. With a vision towards solving the safety problems of current inorganic materials, tremendous efforts have been dedicated to search for much safer and more efficient organic nanoagents for cancer phototherapy. Herein, we design and synthesize a new porphyrin compound (TPAPor) by covalent conjugation of porphyrin and triphenylamine. The typical donor-acceptor-donor (D-A-D) structure of TPAPor facilitates the red-shift of absorption to the nearinfrared (NIR) region. Subsequently, the amphiphilic organic molecule is self-assembled into nanoparticles (TPAPor NPs) with homogeneous size (about 80 nm), enhanced NIR absorption and excellent biocompatibility. TPAPor NPs display low singlet oxygen (O-1(2)) generation and excellent photothermal conversion efficiency (55.7%) under irradiation of a single NIR laser (690 nm). Moreover, TPAPor NPs could ablate the HeLa cells by NIR laser-induced ROS and heat generation, with the half maximal inhibitory concentrations (IC50) about 18.3 mu g/mL. This study indicates TPAPor NPs are promising nanoagents for cancer therapy through photothermal/photodynamic combination manner.

Abstract:Porphyrins and their derivatives are excellent photosensitizers in photodynamic therapy (PDT). The modification of porphyrin molecules into metal-organic cages (MOCs) is a viable strategy to improve their bioavailability. In this work, MOC C66 based on porphyrin was synthesised by a one-pot self-assembly method. The three-dimensional structure of the metal-organic cage ameliorated the aggregation and self-quenching of porphyrins and increased the molar absorption coefficient in the visible light region, which enhanced the reactive oxygen species (ROS) yield of porphyrins and effectively improved the efficiency of photodynamic therapy. ROS generation ability tests in solution confirmed the improved reactive oxygen capacity of the cage, which showed greater phototoxicity to HeLa and MCF-7 cells in vitro, suggesting a new strategy for future modifications of the simple synthesis of porphyrins as photosensitizers.

Abstract:Pyrrole and porphyrin-derived nanoparticles have great potential use in bioimaging and therapy because of their unique magnetic, optical, and other photophysical properties, whereas the poor solubility in aqueous solution is one of the drawbacks of current photosensitizers for their photodynamic therapy (PDT) applications. Here, we developed a kind of water-soluble porphyrin-based nanoparticles that are coassembled mainly by the electrostatic interaction of anionic porphyrins and cationic tetraphenylmethane derivative. No aggregation-caused quenching (ACQ) was detected for these nanoparticles. In addition, the simple porphyrin transformation into nanoparticles improved their ability to generate reactive oxygen species singlet oxygen (1O2), which is an important factor causing apoptosis. The coassembled water-soluble porphyrin-based nanoparticles exhibited enhanced antitumor efficiency via PDT both in vitro and in vivo.

Abstract:Photodynamic therapy (PDT) takes advantage of photosensitizers (PSs) to generate reactive oxygen species (ROS) for cell killing when excited by light. It has been widely used in clinic for therapy of multiple cancers. Currently, all the FDA-approved PSs, including porphyrin, are all small organic molecules, suffering from aggregation-caused quenching (ACQ) issues in biological environment and lacking tumor targeting capability. Nanoparticles (NPs) with size between 20nm and 200nm possess tumor targeting capability due to the enhanced permeability and retention (EPR) effect. It is urgent to develop a new strategy to form clinical-approved-PSs-based NPs with improved ROS generation capability. In this study, we report a strategy to overwhelm the ACQ of porphyrin by doping it with a type of aggregation-induced emission (AIE) luminogen to produce a binary NPs with high biocompatibility, and enhanced fluorescence and ROS generation capability. Such NPs can be readily synthesized by mixing a porphyrin derivative, Ce6 with a typical AIE luminogen, TPE-Br. Here, our experimental results have demonstrated the feasibility and effectiveness of this strategy, endowing it a great potential in clinical applications.
Ultrasensitive Redox-Responsive Porphyrin-Based Polymeric Nanoparticles for Enhanced Photodynamic Therapy

Abstract:Stimulus-sensitive nanoparticles (NPs) have been established to widely adapt to remarkable abnormalities under the tumor microenvironment, which can observably enhance the therapeutic efficiency, improve the specific targeting ability and reduce the side effects. Photodynamic therapy (PDT) as a promising non-invasive and selective treatment for cancers through photodynamic reaction can profit from stimulus-sensitive NPs. Herein, a harmonious amphiphilic polymer (PEG-b-PTPPDS-b-PEG) with an extremely sensitive redox response is constructed via click chemistry between N-3-TPPC6-N-3, PEG-N-3 and alkynyl-containing disulfide ester for PDT. This polymer can be self-assembled into micelles with excellent stability, ultra-fast sensitivity of redox-triggered porphyrin release, and significant photodynamic anticancer performance. The redox-triggered dissociation of micelles and the release of porphyrin are much faster than common porphyrin-containing polymer. The bio-distribution and phototoxicity of micelles against A549 cells are measured and evaluated in vitro by flow cytometry, confocal scanning laser microscopy (CLSM) and MTT assay, respectively. The results reveal that PEG-b-PTPPDS-b-PEG micelles can effectively enhance the cellular uptake and cellular internalization of porphyrin and have an extremely low dark toxicity with efficient PDT towards A549 cells. This intracellular responsive nanoparticle provides a potential strategy for anticancer therapeutic application.

Abstract:Photodynamic therapy (PDT) of cancers is usually inefficient due to the relatively low level of oxygen in cancer cells; therefore, it needs to combine with other treatment strategies such as chemotherapy or photothermal therapy (PTT) to achieve the best anticancer efficacy. Although porphyrin-containing materials have been widely studied for PDT, the photothermal effect is rarely reported. Herein, nanoscale porphyrin-containing covalent organic polymers (PCOPs) were produced via a room temperature solution-based aging method. The resulting nanoparticles possess high photothermal conversion efficiency (21.7%) and excellent photodynamic effect. For the first time, the in vitro and in vivo tests indicated an enhanced antitumor efficacy for PCOP with combined PDT and PTT. This study provides an efficient approach to fabricate nanoCOP and also demonstrates the great potential of porphyrin-containing COP for biomedical applications.

Abstract:The development of photodynamic therapy requires access to smart photosensitizers which combine appropriate photophysical and biological properties. Interestingly, supramolecular and dynamic covalent chemistries have recently shown their ability to produce novel architectures and responsive systems through simple self-assembly approaches. Herein, we report the straightforward formation of porphyrin-peptide conjugates and cage compounds which feature on their surface chemical groups promoting cell uptake and specific organelle targeting. We show that they self-assemble, in aqueous media, into positively-charged nanoparticles which generate singlet oxygen upon green light irradiation, while also undergoing a chemically-controlled disassembly due to the presence of reversible covalent linkages. Finally, the biological evaluation in cells revealed that they act as effective photosensitizers and promote synergistic effects in combination with Doxorubicin.

CuPc-Fe@BSA nanocomposite: Intracellular acid-sensitive aggregation for enhanced sonodynamic and chemo-therapy

Abstract:Due to their rigid π-conjugated macrocyclic structure, organic sonosensitizers face significant aggregation in physiological conditions, hindering the production of reactive oxygen species (ROS). An acid-sensitive nanoassembly was developed to address this issue and enhance sonodynamic therapy (SDT) and emission. Initially, copper phthalocyanine (CuPc) was activated using a H2SO4-assisted hydrothermal method to introduce multiple functional groups (-COOH, -OH, and -SO3H), disrupting strong π-π stacking and promoting ROS generation and emission. Subsequently, negatively charged CuPc-SO4 was incorporated into bovine serum albumin (BSA) to form CuPc-Fe@BSA nanoparticles (10 nm) with Fe3+ ions serving as linkers. In acidic conditions, protonation of CuPc-SO4 and BSA weakened the interactions, leading to Fe3+ release and nanostructure dissociation. Protonated CuPc-SO4 tended to self-aggregate into nanorods. This acidity-sensitive aggregation is vital for achieving specific accumulation within the tumor microenvironment (TME), thereby enhancing retention and SDT efficacy. Prior to this, the nanocomposites demonstrated cycling stability under neutral conditions. Additionally, the released Fe ions exhibited mimicry of glutathione peroxidase and peroxidase activity for chemotherapy (CDT). The synergistic effect of SDT and CDT increased intracellular oxidative stress, causing mitochondrial injury and ferroptosis. Furthermore, the combined therapy induced immunogenic cell death (ICD), effectively activating anticancer immune responses and suppressing metastasis and recurrence.

Abstract:The development of intelligent nano-platforms with high tumor-targeting capability, enhanced solubility, and efficacious photochemotherapy is urgently demanded for clinical anticancer treatments. Herein, we report an amphiphilic porphyrin ( P ) to complex with camptothecin ( CPT ) to form supra-amphiphile-assembled nanoparticles (). The host–guest complexation between P and CPT promotes the singlet oxygen production of P and enhances the solubility of CPT , which should be beneficial to improve PDT efficiency and chemotherapy effect. Cancer cell membrane ( M ) is further used to camouflage , which endows the resulting "M with excellent stability, homologous targeting ability, and specific co-delivering of the chemotherapeutic drug and photosensitizer to tumor cells. The presented work takes advantage of the unique structure and intrinsic performance of P to construct an integrative co-delivery system with the assistance of membrane-coating technology, which not only extends the application scopes of porphyrin photosensitizers but also provides a promising nanoplatform for cancer therapy.

Abstract:In this study, we designed an effective nanoplatform to improve the photodynamic therapy (PDT) of porphyrins. Combining a porphyrin-based metallacage (PM), hyaluronidase (HAase) and DSPE-mPEG2000 together, the nanoparticle (PM@HAase-mPEG) showed enhanced PDT efficacy. The PM improved the stability of the porphyrin, avoided its aggregation and provided cavities to concentrate oxygen molecules, which was beneficial for enhancing PDT. HAase degraded HA to increase the intracellular accumulation of nanoparticles, normalized blood vessels and relieved hypoxia in tumors. PM@HAase-mPEG inhibited the growth of tumors in a 4T1 mouse model by the generated singlet oxygen with excellent PDT efficacy. This study resolved the problems of the instability of PSs, less cellular accumulation of drugs, and tumor hypoxia that limited the anti-tumor application of PDT.

Abstract:Self-assembled small molecules, as a novel form of drug presentation, have splendid capabilities for water stability and cell endocytosis. Photodynamic therapy (PDT) is regarded as a promising cancer treatment because it is less invasive and has fewer side effects. In this work, an amphiphilic glycyrrhetinic acid-porphyrin conjugate (TPP-GA) was designed and synthesized, and this could self-assemble into nanoparticles (TPP-GA NPs) in water. The morphology of the TPP-GA NPs was observed using transmission electron microscopy (TEM) and their stability in water was revealed by dynamic light scattering (DLS). The PDT effect of the TPP-GA NPs was confirmed by means of in vitro experiment, after photostability verification. It could be conceived that the application of TPP-GA NPs for PDT is a feasible choice and this work shows progress in the self-assembly of amphiphilic small organic molecules.

Abstract:Porphyrin-based covalent organic polymer nanoparticles were synthesized via a Schiff base reaction at room temperature. The resulting product showed a good photodynamic effect and a high photothermal conversion efficiency (34.88%). Both in vitro and in vivo experiments demonstrated the enhanced antitumor efficacy via synergistic photodynamic and photothermal therapy.

Abstract:Background: The clinical efficiency of photodynamic therapy (PDT) in combination with chemotherapy has proven to be a promising strategy for tumor treatment, yet is restricted by the high glutathione (GSH) concentration at the tumor site and nonspecific drug targeting. Objective: The goal of the current research was to create a biocompatible GSH-depleting and tumor- targeting nanoparticle (denoted as DOX/CA@PCN-224@HA) for the combined photodynamic and chemo photo-chemo) therapy. Methods: The nanoparticles were characterized by transmission electron microscopy (TEM). A UV-vis spectrophotometer was used to measure the drug loading efficiency (DE) and encapsulation efficiency (EE). The GSH-depleting ability was measured using Ellman's test. Confocal laser scan microscopy (CLSM) was used to assess the cellular uptake. MTT was adopted to evaluate the cytotoxicity of DOX/CA@PCN-224@HA against 4T1 cells. Results: The altered PCN-224 showed excellent monodispersing with a dimension of approximately 193 nm ± 2 nm in length and 79 nm ± 3 nm in width. The larger and spindle grid-like structure of PCN-224 obtains better dual-drug loading ability (DOX: 20.58% ± 2.60%, CA: 21.81% ± 1.98%) compared with other spherical PCN-224 nanoparticles. The ultimate cumulative drug release rates with hyaluronidase (HAase) were 74% ± 1% (DOX) and 45% ± 2% (CA) after 72 h. DOX/CA@PCN-224@HA showed GSH-consuming capability, which could improve the PDT effect. The drug-loaded nanoparticles could accurately target 4T1 cells through biological evaluations. Moreover, the released DOX and CA display cooperative effects on 4T1 cells in vitro. DOX/CA@PCN-224@HA nanoparticles showed inhibition against 4T1 cells with an IC50 value of 2.71 μg mL-1. Conclusion: This nanosystem displays great potential for tumor-targeted enhanced (photo-chemo) therapy.

Self-assembled IR dye/Mitoxantrone loaded Porphysomes Nanosystem for Enhanced Combinatorial Chemo-Photothermal Cancer Therapy

Abstract:Chemo-photothermal therapy (PTT) is an emerging non-invasive cancer treatment modality. Light-responsive porphysomes (DPP IR Mtx @Lipo NPs) nanosystems ablate breast cancer cells upon oxidative stress and hyperthermia. The unique self-assembled porphysomes were formed spherical shape in the size range of 150 ± 30 nm formed by the co-assembly of porphyrins along with IR 775 and chemotherapeutic drug, Mitoxantrone (Mtx), forming a camouflaged nanosystem (DPP IR Mtx @Lipo NPs, porphysomes). The advent of the prepared porphysomes aids in proper tuning of NIR absorbance improving singlet oxygen species generation among other anticancer drugs. The eminent release of DPP and adjuvant chemo-drug, Mitoxantrone from the self-assembled porphysomes is triggered by IR 775, a NIR photosensitizer upon laser irradiation. These multifunctional DPP IR Mtx @Lipo NPs have an efficient photothermal conversion efficiency of 65.8% as well as bioimaging properties. In-vitro studies in 2D and 3D models showed a significant cell death of 4T1 cells via the apoptotic pathway when irradiated with NIR laser, causing minimal damage to nearby healthy cells. DPP IR Mtx @Lipo NPs exhibited commingled PDT/PTT interdependent via NIR laser exposure, leading to mitochondrial disruption. Interestingly, the transient transfection using p53-GFP in cancer cells followed by DPP IR Mtx @Lipo NPs treatment causes rapid cell death. The activation of p53-dependent apoptosis pathways was vividly expressed, evidenced by the upregulation of Bax and increased pattern of Caspase-3 cleavage. This effect was pronounced upon transfection and induction with DPP IR Mtx @Lipo NPs, particularly in comparison to non-transfected malignant breast cancer 4T1 cells.
