A wireless acoustic wave sensor modified with biocompatible polymer for glucose monitoring

Abstract:The dynamic monitoring of vivo blood glucose acts an important role for diabetic patients to ensure timely healthcare and help to prevent from multiple complications caused by diabetes. This report describes a new non-enzyme glucose sensor based on wireless quartz crystal microbalance (QCM) that can be adapted for use in implanted devices. The chitosan-based enzyme-free boronic-acid glucose sensor improves biocompatibility considering for future vivo implanting. The optimal response of the sensor is in chitosan: 4-Formylphenylboronic acid (4-FPBA) =3:2. In the case of pH 7.4, the sensor has good linear response to glucose and selectivity of sensor to fructose, mannose, urea, and lactic acid performs well under physiological conditions. At physiological temperature, the linear response was also repeated and the average response time was about 4 min. The characteristics of this sensor indicate that it can be applied to dynamic monitoring of implantable blood glucose concentrations.

Abstract:Different hydrogels of poly(acrylamide-co-3-acrylamido phenylboronic acid-co-chitosan grafted maleic acid) (P(AM-co-AAPBA-co-CSMA)s) were synthesized using poly(ethylene glycol) diacrylate (PEGDA) as a crosslinker to serve for glucose sensing and insulin delivery. The structure and morphology of the hydrogels, named as CSPBA were studied by FTIR and SEM, while the mechanical properties were tested using dynamic mechanical analysis (DMA) and universal testing machine. The prepared hydrogels shrinked at low glucose concentration due to the 2:1 boronate-glucose binding, and swelled at high glucose concentration because of 1:1 boronate-glucose complexation. Both binding mechanisms are useful for glucose sensing and insulin delivery. The integration of CSMA into hydrogels network not only enhanced the response to glucose at physiological pH, but also improved the mechanical properties and increased the encapsulation efficiency of the prepared hydrogels. These CSPBA may find potential as implantable hydrogels in applications were continuous glucose monitoring and controlled release is beneficial.
Continuous Glucose Monitoring Enabled by Fluorescent Nanodiamond Boronic Hydrogel

Abstract:Continuous glucose monitoring (CGM) allows diabetic patients to better maintain blood glucose level. In this proof‐of‐concept study, a skin mountable device is developed with fluorescent nanodiamond hydrogel, presenting a new strategy of fluorescence based CGM toward treatment and control of diabetes. Continuous monitoring of glucose allows diabetic patients to better maintain blood glucose level by altering insulin dosage or diet according to prevailing glucose values and thus to prevent potential hyperglycemia and hypoglycemia. However, current continuous glucose monitoring (CGM) relies mostly on enzyme electrodes or micro‐dialysis probes, which suffer from insufficient stability, susceptibility to corrosion of electrodes, weak or inconsistent correlation, and inevitable interference. A fluorescence‐based glucose sensor in the skin will likely be more stable, have improved sensitivity, and can resolve the issues of electrochemical interference from the tissue. This study develops a fluorescent nanodiamond boronic hydrogel system in porous microneedles for CGM. Fluorescent nanodiamond is one of the most photostable fluorophores with superior biocompatibility. When surface functionalized, the fluorescent nanodiamond can integrate with boronic polymer and form a hydrogel, which can produce fluorescent signals in response to environmental glucose concentration. In this proof‐of‐concept study, the strategy for building a miniatured device with fluorescent nanodiamond hydrogel is developed. The device demonstrates remarkable long‐term photo and signal stability in vivo with both small and large animal models. This study presents a new strategy of fluorescence based CGM toward treatment and control of diabetes.

Abstract:Diabetes mellitus is a chronic metabolic disorder, requiring vigilant monitoring of blood glucose levels. In this study, an injectable fluorescent enzymatic hydrogel was designed for rapid glucose detection. The leakage-free glucose-responsive hydrogel was constructed by the covalent linkage of a multi-arm poly-(ethylene glycol) (PEG), bovine serum albumin (BSA), glucose oxidase (GOx), and 4-(aminomethyl)-6,7-dimethoxycoumarin (Coumarin-NH2). The GOx serves as glucose-recognition element and the pH-sensitive Coumarin-NH2 as a fluorescence turn-on reporter. The material properties of the fluorescent hydrogel were systematically characterized which show high elasticity with good mechanical strength. Upon the addition of glucose, the as-developed fluorescent hydrogel shows a fast response time, good sensitivity, and good reproducibility at physiological pH and ambient temperature. The glucose-sensing mechanism is based on the oxidation of the glucose by GOx that generates protons to change the local pH. Consequently, protonation of the covalently immobilized and pH-sensitive Coumarin-NH2 turns on the fluorescence of the coumarin. The fluorescence hydrogel developed holds great promise as an injectable, implantable glucose-sensing biomaterials for in vivo continuous glucose monitoring.

Abstract:It is significant for people with diabetes to know their body's real-time glucose level, which can guide the diagnosis and treatment. Therefore, it is necessary to research continuous glucose monitoring (CGM) as it gives us real-time information about our health condition and its dynamic changes. Here, we report a novel hydrogel optical fiber fluorescence sensor segmentally functionalized with fluorescein derivative and CdTe QDs/3-APBA, which can continuously monitor pH and glucose simultaneously. In the glucose detection section, the complexation of PBA and glucose will expand the local hydrogel and decrease the fluorescence of the quantum dots. The fluorescence can be transmitted to the detector by the hydrogel optical fiber in real time. As the complexation reaction and the swelling–deswelling of the hydrogel are all reversible, the dynamic change of glucose concentration can be monitored. For pH detection, the fluorescein attached to another segment of the hydrogel exhibits different protolytic forms when pH changes and the fluorescence changes correspondingly. The significance of pH detection is compensation for pH errors in glucose detection because the reaction between PBA and glucose is sensitive to pH. The emission peaks of the two detection units are 517 nm and 594 nm, respectively, so there is no signal interference between them. The sensor can continuously monitor glucose in 0–20 mM and pH in 5.4–7.8. The advantages of this sensor are multi-parameter simultaneous detection, transmission-detection integration, real-time dynamic detection, and good biocompatibility.

Abstract:Optical sensors have attracted a great deal of interest for glucose detection. However, their practical applications for continuous glucose monitoring are still constrained by operational reliability in subcutaneous tissues. Here, we show an implantable hydrogel platform embedded with luminescent polymer dots (Pdots) for sensitive and long-term glucose monitoring. We use Pdot transducer in a polyacrylamide hydrogel matrix to construct an implantable platform. The hydrogel-Pdot transducer showed bright luminescence with ratiometric response to glucose changes. The <i>in vitro</i> and <i>in vivo</i> sensitivities of the hydrogel implant were enhanced by varying the enzyme concentration and injection volume. After implantation, the hydrogel with Pdot transducer remained at the implanted site without migration for 1 month and can be removed from the subcutaneous tissue for further analysis. Our results indicate that the hydrogel-Pdot platform maintains the intrinsic sensing property with excellent stability during 1 month implantation, while fibrous capsule formation on the implant in some cases needs to be solved for long-term continuous glucose monitoring.

Abstract:Glucose monitoring sensors with high softness and flexibility are critical for the developments of wearable and implantable healthcare devices that enable diagnosis, prognosis, and management of diabetes. The design and implementation of such sensors have been extensively exploited by electrochemical strategies, which, however, suffer from poor reusability and complex modification procedures, and necessitate frequent calibration or sensor replacement due to enzymatic reaction instability. Here, a soft and plasmonic hydrogel optical sensor is created for quantitative and continuous glucose monitoring under physiological conditions. The optical sensor consists of a flexible optical fiber made from composites of gold nanoparticles and glucose-responsive hydrogels. The reversible binding of glucose to the nanocomposite optical fiber results in dynamic volume expansion of the hydrogel matrix, which modulates the localized surface plasmon resonance effect, enabling glucose to be quantified from the light transmission. To achieve robust readout, a dual-wavelength differential approach is employed to endow the sensor with self calibration capability. We show that the sensor is reversible and reusable for detecting physiological glucose levels with high linearity and negligible hysteresis. The soft and flexible glucose sensor holds great promises of serving as a minimally-invasive probe for point-of-care glucose monitoring in clinics.
Designing Tough Hydrogel Shells for Glucose Sensing

Abstract:Hydrogel shells are created with double‐network hydrogel using triple‐emulsion template. The shells provide high mechanical stability, optical transparency, and well‐defined permeation threshold, serving as a promising platform for enzyme‐based microsensors. A fluorometric glucose sensor is demonstrated by encapsulating glucose oxidase in the core and pH‐responsive dye in the shell, which exhibits excellent stability against mechanical stress, drying, and proteolysis. Conventional hydrogel microcapsules often suffer from inadequate mechanical stability, hindering their use. Here, water‐cored double‐network (DN) hydrogel shells are designed, formed by polyacrylamide and calcium alginate networks using triple‐emulsion templates. These DN hydrogel shells offer robust mechanical stability, optical transparency, and a precisely‐defined cut‐off threshold. The feasibility of this platform is demonstrated through the development of a fluorometric glucose sensor. Glucose oxidase is enclosed within the water core, while a pH‐responsive fluorescent dye is incorporated into the DN shells. Glucose diffuses into the core through the DN shells, where the glucose oxidase converts glucose into gluconic acid, leading to pH reduction and a subsequent decrease in fluorescence intensity of DN shells. Additionally, the pH‐sensitive colorant dissolved in the medium enables visual pH assessment. Thus, glucose levels can be determined using both fluorometric and colorimetric methods. Notably, the DN shells exhibit exceptional stability, enduring intense mechanical stress and cycles of drying and rehydration without leakage. Moreover, the DN shells act as effective barriers, safeguarding glucose oxidase against proteolysis by large disruptive proteins, like pancreatin. This versatile DN shell platform extends beyond glucose oxidase encapsulation, serving as a foundation for various capsule sensors utilizing enzymes and heterogeneous catalysts.

Abstract:The healing of complex diabetic wounds with hyperglycemic microenvironment and bacterial infection was considered an important clinical issue. In this study, glucose oxidase (GOx) and gold nanoclusters (AuNCs) were encapsulated in quaternary carboxymethyl chitosan (QCMCS)/sodium alginate oxide (OSA) hydrogels, and were immersed in tannic acid (TA) solution to achieve antioxidant, antibacterial, pro-angiogenesis, pro-collagen deposition and real-time monitoring functions. In vitro studies showed that TA-QCMCS/OSA@GOx@AuNCs hydrogels had inhibition rates of 98.99% and 99.99% against S. aureus and E. coli, respectively, and the survival rate of mouse fibroblasts (L929) was over 95%. In vivo studies showed that TA-QCMCS/OSA@GOx@AuNCs hydrogels was 97.28% effective in healing diabetic wounds. In addition, image signals from TA-QCMCS/OSA@GOx@AuNCs hydrogels can be collected in real-time to accurately obtain glucose concentration values of diabetic wounds and reflect the healing status of diabetic wounds in a timely manner. The results showed that TA-QCMCS/OSA@GOx@AuNCs hydrogels provides a novel idea for real-time monitoring of diabetic wounds treatment.

Abstract:Smart flexible sensing materials and devices for wearable health monitoring, are considered as prospective and promising areas in healthcare that are currently in high demand in the market. Conductive hydrogel-based sensors, which closely resemble human tissue, encounter notable challenges in attaining high electronic conduction, robust mechanical properties, biocompatibility, and intimate tissue adhesion simultaneously. In this work, a poly(2-acrylamide-2-methylpropane sulfonic acid- co -acrylamide)/bacterial cellulose (P(AMPS- co -AM)/BC) hydrogel with a particular interpenetrating and interlocking networked structure is reported. This hydrogel possesses all the mentioned intriguing properties plus easy degradability. It is realized by utilizing the interaction between amino, amide, and sulfonic groups on polymer molecular chains and polyhydroxy groups on bacterial cellulose. This hydrogel exhibits high strength (1.28 MPa) and tensile strain (525%), remarkable tissue adhesion (697 kPa), exceptional antibacterial property and low cytotoxicity. Furthermore, it can degrade in the natural environment when exposed to UV light irradiation. The hydrogel-based strain sensors have demonstrated efficient human motion detection with a broad strain detection range (0-360%) and a high sensitivity ( GF =11.36). Moreover, this hydrogel can be assembled into Triboelectric Nanogenerator for monitoring biological signals. This work presents a new avenue for human-machine interaction and electronic robotic skins.

Abstract:A novel glucose-responsive hydrogel system based on dynamic covalent chemistry and inclusion complexation was described. Hydrogels are formed by simply mixing the solutions of three components: poly(ethylene oxide)-b-poly vinyl alcohol (PEO-b-PVA) diblock polymer, alpha-cyclodextrin (alpha-CD) and phenylboronic acid (PBA)-terminated PEO crosslinker. Dynamic covalent bonds between PVA and PBA provide sugar-responsive crosslinking, and the inclusion complexation between PEO and alpha-CD can promote hydrogel formation and enhance hydrogel stability. The ratios of the three components have a remarkable effect on the gelation time and the mechanical properties of the final gels. In rheological measurements, the hydrogels are demonstrated to possess solid-like behaviour and good structural recovery ability after yielding. The sugar-responsiveness of the hydrogels was examined by protein loading and release experiments, and the results indicate that this property is also dependent on the compositions of the gels; at a proper component ratio, a new glucose-responsive hydrogel system operating at physiological pH can be obtained. The combination of good biocompatibility of the three components and the easy preparation of hydrogels with tunable glucose-responsiveness may enable an alternative design of hydrogel systems that finds potential applications in biomedical and pharmaceutical fields, such as treatment of diabetes.
An integrated portable bio-monitoring system based on tough hydrogels for comprehensive detection of physiological activities

Abstract:Advanced soft ion-conducting hydrogels have been developed rapidly in the integrated portable health monitoring equipment due to their higher sensitivity, sensory traits, tunable conductivity, and stretchability for physiological activities and personal healthcare detection. However, traditional hydrogel conductors are normally susceptible to large deformation and strong mechanical stress, which leads to inferior electro-mechanical stability for real application scenarios. Herein, a strong ionically conductive hydrogel (poly(vinyl alcohol)-boric acid-glycerol/sodium alginate-calcium chloride/electrolyte ions (PBG/SC/EI)) was designed by engineering the covalently and ionically crosslinked networks followed by the salting-out effect to further enhance the mechanical strength and ionic conductivity of the hydrogel. Owing to the collective effects of the energy-dissipation mechanism and salting-out effect, the designed PBG/SC/EI with excellent structural integrity and robustness exhibits exceptional mechanical properties (elongation at break for 559.1% and tensile strength of 869.4 kPa) and high ionic conductivity (1.618 S·m −1 ). As such, the PBG/SC/EI strain sensor features high sensitivity (gauge factor = 2.29), which can effectively monitor various kinds of human motions (joint motions, facial micro-expression, faint respiration, and voice recognition). Meanwhile, the hydrogel-based Zn∥MnO 2 battery delivers a high capacity of 267.2 mAh·g −1 and a maximal energy density of 356.8 Wh·kg −1 associated with good cycle performance of 71.8% capacity retention after 8000 cycles. Additionally, an integrated bio-monitoring system with the sensor and Zn∥MnO 2 battery can accurately identify diverse physiological activities in a real-time and noninvasive way. This work presents a feasible strategy for designing high-performance conductive hydrogels for highly-reliable integrated bio-monitoring systems with excellent practicability.

Abstract:Phenylboronic acids (PBAs) have gained considerable interest in recent years due to their recognition for diol-containing molecules such as glucose. Their response to the elevated glucose concentrations can be reported by measuring the change in the size or optical properties of polymers-bearing PBAs. In this context, fast response and good mechanical properties are crucial factors for constructing glucose-responsive sensors. Toward this goal, we have synthesized glucose-responsive nanostructured gels (NSGs) using 3-acrylamidophenylboronic acid and N -isopropylacrylamide. Herein, activated nanogels with controllable size were prepared and used as nano-cross-linkers. The prepared NSGs showed glucose-responsiveness in a remarkable concentration-dependent manner, exhibited high elasticity upon compression and slicing and resisted high level of deformation such as bending, twisting and stretching.

Abstract:Diabetic wound is one of the chronic wounds that is difficult to heal, and effective treatment of it still confronts a great challenge. Monitoring the variation of diabetic wound microenvironment (such as hydrogen peroxide (H2O2)) can understand the wound state and guide the wound management. Herein, we developed a multifunctional hydrogel with the abilities of monitoring the H2O2 concentration, alleviating oxidative stress and promoting wound healing, which was prepared by encapsulating manganese‐containing bioactive glass (MnBG) and CePO4:Tb in biocompatible Gelatin methacryloyl (GelMA) hydrogel (CPT‐MnBG‐Gel). On the one hand, the H2O2‐dependent fluorescence quenching effect of the CePO4:Tb contributed to visible monitoring of the H2O2 concentration of wounds via smartphone imaging, and the CPT‐MnBG‐Gel hydrogel could effectively monitor the H2O2 level of 10.35‐200 μmol L−1. On the other hand, MnBG could alleviate oxidative stress and promote the proliferation, migration and differentiation of fibroblasts and endothelial cells in vitro owing to the bioactive Mn and Si ions, and in vivo evaluation also demonstrated that the CPT‐MnBG‐Gel hydrogels could effectively accelerate wound healing. Abstract:Owing to its rapid response and broad detection range, a phenylboronic acid (PBA)-functionalized hydrogel film-coated quartz crystal microbalance (QCM) sensor is used to non-invasively monitor salivary glucose in diabetic patients. However, nonspecific protein adsorption on the PBA-functionalized hydrogel film can cause dramatic loss of sensitivity and accuracy of the sensor. A traditional zwitterionic polymer surface with ultra-low protein fouling can hinder the interaction of PBA in the hydrogel matrix with glucose molecules owing to its steric hindrance, resulting in poor glucose sensitivity of the sensor. Herein, we developed a novel hydrogel film that enhanced the antifouling properties and sensitivity of the QCM sensor by infiltrating a glucose-sensitive monomer (i.e., PBA) into a zwitterionic polymer brush matrix to form an interpenetrating polymer network (IPN). The IPN hydrogel film could minimize the glucose sensitivity loss since the antifouling polymer distributed in its matrix. Moreover, a stable hydration layer was formed in this film that could prevent water from transporting out of the matrix, thus further improving its antifouling properties and glucose sensitivity. The experimental results confirmed that the IPN hydrogel film possessed excellent resistance to protein fouling by mucin from whole saliva with reductions in adsorption of nearly 88% and could also enhance the glucose sensitivity by nearly 2 fold, compared to the PBA-functionalized hydrogel film. Therefore, the IPN hydrogel film provides improved antifouling properties and sensitivity of the QCM sensor, which paves the way for non-invasive monitoring of low concentrations of glucose in saliva.

Abstract:In this paper, a cellulose-based hydrogel with the ability to lower glucose levels, scavenge reactive oxygen species (ROS), and colorimetrically monitor early lesions was synthesized to address the issue of glucose and H 2 O 2 overexpression on the wound surface. Briefly, boronic acid pinacol ester containing phenolphthalein (BAPE-PPH) was employed as a crosslinker to react with hydroxypropyl methylcellulose (HPMC) to afford hydrogels (HPMC@PPH-Hs) with good biocompatibility. Because glucose has higher affinity for the phenylboronic ester group than cellulose derivatives, the HPMC@PPH-Hs showed excellent glucose-lowering performance. The presence of B-C bonds in the network endows the as-obtained hydrogel with H 2 O 2 sensitivity, and cleavage of the B-C bonds forces the transformation of BAPE-PPH moieties into PPH, giving rise to colorimetric behaviour. Thus, the obtained HPMC@PPH-Hs provide information to enrich the family of smart hydrogels, and its potential application in diabetic wound healing may be desirable.

Abstract:Continuous glucose monitors are crucial for diabetes management, but invasive sampling, signal drift and frequent calibrations restrict their widespread usage. Microneedle sensors are emerging as a minimally-invasive platform for real-time monitoring of clinical parameters in interstitial fluid. Herein, a painless and flexible microneedle sensing patch is constructed by a mechanically-strong microneedle base and a thin layer of fluorescent hydrogel sensor for on-site, accurate, and continuous glucose monitoring. The Förster resonance energy transfer (FRET)-based hydrogel sensors are fabricated by facile photopolymerizations of acryloylated FRET pairs and glucose-specific phenylboronic acid. The optimized hydrogel sensor enables quantification of glucose with reversibility, high selectivity, and signal stability against photobleaching. Poly (ethylene glycol diacrylate)-co-polyacrylamide hydrogel is utilized as the microneedle base, facilitating effective skin piercing and biofluid extraction. The integrated microneedle sensor patch displays a sensitivity of 0.029 mM-1 in the (patho)physiological range, a low detection limit of 0.193 mM, and a response time of 7.7 min in human serum. Hypoglycemia, euglycemia and hyperglycemia are continuously monitored over 6 h simulated meal and rest activities in a porcine skin model. This microneedle sensor with high transdermal analytical performance offers a powerful tool for continuous diabetes monitoring at point-of-care settings.

Abstract:Smart contact lenses for continuous glucose monitoring (CGM) have great potential for huge clinical impact. To date, their development has been limited by challenges in accurate detection of glucose without hysteresis for tear glucose monitoring to track the blood glucose levels. Here, long-term robust CGM in diabetic rabbits is demonstrated by using bimetallic nanocatalysts immobilized in nanoporous hydrogels in smart contact lenses. After redox reaction of glucose oxidase, the nanocatalysts facilitate rapid decomposition of hydrogen peroxide and nanoparticle-mediated charge transfer with drastically improved diffusion via rapid swelling of nanoporous hydrogels. The ocular glucose sensors result in high sensitivity, fast response time, low detection limit, low hysteresis, and rapid sensor warming-up time. In diabetic rabbits, smart contact lens can detect tear glucose levels consistent with blood glucose levels measured by a glucometer and a CGM device, reflecting rapid concentration changes without hysteresis. The CGM in a human demonstrates the feasibility of smart contact lenses for further clinical applications.

Abstract:Conductive hydrogels exhibit tremendous potential for wearable bioelectronics, biosensing, and health monitoring applications, yet concurrently enhancing their biocompatibility and antimicrobial properties remains a long-standing challenge. Herein, we report an all-natural conductive supramolecular hydrogel (GT5-DACD2-B) prepared via the Schiff base reaction between the biofriendly dialdehyde cyclodextrin and gelatin. The potent antibacterial agent fusidic acid (FA) is incorporated through host-guest inclusion, enabling 100% inhibition of Staphylococcus aureus proliferation. The biocompatibility of our hydrogel is bolstered with tannic acid (TA) facilitating antibacterial effects through interactions with gelatin, while borax augments conductivity. This supramolecular hydrogel not only exhibits stable conductivity and rapid response characteristics but also functions as a flexible sensor for monitoring human movement, facial expressions, and speech recognition. Innovatively integrating biocompatibility, antimicrobial activity, and conductivity into a single system, our work pioneers a paradigm for developing multifunctional biosensors with integrated antibacterial functionalities, paving the way for advanced wearable bioelectronics with enhanced safety and multifunctionality.
